Abstract
本发明提供一种多肽单体分子及其自组装纳米材料和应用,所述多肽单体分子由疏水分子与多肽通过酰胺键连接,所述多肽包括功能性肽,所述功能性肽包括SEQ ID NO.1~4任意一种所示的氨基酸序列。本发明的多肽单体分子由疏水分子与多肽组成,通过自组装形成α‑螺旋的三维纳米材料。本发明的多肽自组装纳米材料,通过自组装纳米材料的多价键效应的作用,能够快速大量提高病灶部位抗菌材料的局部浓度,具有高效、快速杀菌的功能。

技术领域
本发明属于组装纳米材料技术领域,涉及一种多肽单体分子及其自组装纳米材料和应用。
背景技术
抗生素的滥用导致细菌耐药性进程的加剧,出现了许多广谱耐药菌和超级细菌,已经成为全球性问题。针对耐药性细菌,新型抗生素开发非常困难,因此寻找安全有效的抗生素佐剂,减少抗生素的使用和提高现有抗生素的效果迫在眉睫。因此提出多肽组装纳米材料用作抗生素佐剂辅佐抗生素在生物体内进行高效快速杀菌。
近年来,对于抗生素佐剂领域的应用,抗菌肽等单体分子在抗菌方面有了广泛的发展,但在抗菌效率方面有待发展。
CN112321698A公开了一种抗菌肽及其药物组合物以及应用,所述抗菌肽包括:抗菌肽1、抗菌肽2、抗菌肽3中的任意一种,抗菌肽1的氨基酸序列如SEQ ID No:1所示,抗菌肽2的氨基酸序列如SEQ ID No:2所示,抗菌肽3的氨基酸序列如SEQ ID No:3所示。三种抗菌肽均是以天然抗菌肽PGLa-AM1为基础进行改造而成,具有抗菌效果更强、性能更优的特点。这三种抗菌肽对幽门螺杆菌的抑制效果均强于PGLa-AM1,其中抗菌肽3的广谱抗菌效果最强,并且具有很好的pH响应性,从而更好的与幽门螺杆菌接触,抑制幽门螺杆菌生长。根据该发明提供的三种抗菌肽均具有强抑菌活性、生物相容性、及低成本的优点,有望被广泛应用。但该发明的抗菌肽并不能实现自组装,在应用时往往需要较高的浓度才能达到抗菌的效果。
因此,在本领域中,期待开发一种具有抗菌功能的多肽单体分子及其自组装纳米材料。
发明内容
针对现有技术的不足,本发明的目的在于提供一种多肽单体分子及其自组装纳米材料和应用。本发明提出的具有生物功能的多肽自组装纳米材料的策略成为解决上述瓶颈问题的有效手段。本发明的多肽自组装纳米材料比多肽单体分子能够快速大量提高病灶部位抗菌材料的局部浓度,同时通过自组装纳米材料的多价键效应的作用,体现高效杀菌的优势,成为时下最具潜力的研究方向。
为达到此发明目的,本发明采用以下技术方案:
第一方面,本发明提供一种多肽单体分子,所述多肽单体分子由疏水分子与多肽通过酰胺键连接,所述多肽包括功能性肽,所述功能性肽包括SEQ ID NO.1~4任意一种所示的氨基酸序列。

在本发明中,多肽单体分子可以通过静电吸附作用聚集到细菌周围,然后功能性肽与细菌膜成分通过化学作用相结合,靶向细菌,插入细菌膜中,扰动细菌膜,破坏细菌膜结构,起到杀菌的作用;疏水分子起到平衡亲疏水的作用,使多肽单体分子自组装成三维纳米结构。因此,本发明的多肽单体分子不仅具有抗菌性能,还具有自组装功能,可以通过自组装形成α-螺旋的三维纳米材料。
优选地,所述功能性肽为SEQ ID NO.2所示的氨基酸序列。相比于SEQ ID NO.1、SEQ ID NO.3、SEQ ID NO.4所示的氨基酸序列,当功能性肽为SEQ ID NO.2所示的氨基酸序列时,多肽单体分子具有更好的杀菌效果。
优选地,所述多肽还包括结构性肽,所述结构性肽含有2-8个氨基酸序列,优选SEQID NO.5~7任意一种所示的氨基酸序列,进一步优选SEQ ID NO.6所示的氨基酸序列。

作为本发明的优选技术方案,结构性肽含有2-8(例如2、3、4、5、6、7或8)个氨基酸序列,当多肽单体分子自组装时,多肽单体分子通过范德华力(如氢键作用)形成β-sheet结构,最终组装成纤维状的三维纳米材料。如果多肽单体分子中不含结构性肽,即,仅由疏水分子和功能性肽构成时,多肽单体分子通过亲疏水平衡作用,自组装形成球形的三维纳米材料,其中外部是功能性肽,内部是疏水分子。
优选地,所述结构性肽位于中间,所述疏水分子和功能性肽分别位于结构性肽的两侧。
优选地,所述疏水分子包括软脂酸、硬脂酸或胆固醇中的任意一种,优选软脂酸。
作为本发明的优选技术方案,当软脂酸作为疏水分子时,可以大大提高多肽单体分子的疏水性,达到多肽单体分子亲疏水平衡,有利于自组装纳米材料的构成。
优选地,所述多肽单体分子具有如式Ⅰ~式Ⅵ所示结构:
第二方面,本发明提供一种第一方面所述的多肽单体分子的制备方法,所述制备方法包括以下步骤:
采用多肽固相合成法合成多肽,而后加入疏水分子,将疏水分子连接至多肽上,裂解,后处理,得到所述多肽单体分子。
本发明的多肽单体分子可用多肽合成仪合成,也可人工合成,示例性地,可采用如下制备方法:
(1)使用FMOC策略的多肽固相合成法,以氨基端受FMOC保护的氨基酸为原料,从右到左依次连接多肽序列中所需氨基酸,得到多肽,而后加入疏水分子,将疏水分子连接至多肽上;
合成试剂选用:
(a)载体树脂:wang树脂;
(b)脱保护试剂:括5%无水哌嗪+2%1,8-二氮杂环十一烯-7(DBU)+93%DMF;
(c)缩合反应时使用偶联试剂:5%N-甲基吗啉+95%DMF;
(2)配制裂解液:0.125mL去离子水+0.125mL三异丙基硅烷+4.75mL三氟乙酸,将配制好的裂解液加入装有多肽单体分子的裂解瓶中,裂解瓶中加入磁转子搅拌,将裂解瓶置于0℃冰水浴中,转速300-400r/min,搅拌3h,将多肽单体分子从载体树脂上裂解下来,而后洗涤,干燥,得到所述多肽单体分子。
优选地,所述后处理包括洗涤、干燥。洗涤主要是为了将裂解液去除。
第三方面,本发明提供一种多肽自组装纳米材料,所述多肽自组装纳米材料由第一方面所述的多肽单体分子自组装而成。
优选地,所述多肽自组装纳米材料具有三维纳米结构。
优选地,所述三维纳米结构包括球形结构或纤维状结构。
本发明的多肽自组装纳米材料,一方面,其含有功能性肽,三维纳米材料先通过静电吸附作用聚集到细菌周围,然后功能性肽与细菌膜成分通过化学作用相结合,靶向细菌,插入细菌膜中,通过多价键效应诱导膜曲率改变,来破坏细菌膜,起到杀菌的作用,从而使得多肽自组装纳米材料具有高效杀菌功能;另一方面,相比于单体分子,其能够快速大量提高病灶部位抗菌材料的局部浓度,同时通过自组装纳米材料的多价键效应的作用,体现了高效杀菌的优势。
多价键效应指的是,由于多肽自组装纳米材料由多个多肽单体分子自组装而成,多肽自组装纳米材料中含有多个功能性肽,多个功能性肽与细菌膜相互结合形成多价键效应。
第四方面,本发明提供一种第三方面所述的多肽自组装纳米材料的自组装方法,所述自组装方法包括以下步骤:
将多肽单体分子溶于溶剂中,调节多肽单体分子的浓度达到临界胶束浓度(CMC)值以上,水浴超声,静置,得到所述多肽自组装纳米材料。
优选地,所述将多肽单体分子溶于溶剂中,具体包括以下步骤:先将多肽单体分子溶于助溶剂中,而后再溶于溶剂中。
优选地,所述助溶剂包括二甲基亚砜(DMSO)。本发明对助溶剂的加入量不作限制,只要可以将多肽单体分子充分溶解即可。
优选地,所述溶剂包括去离子水、氯化钠(NaCl)溶液或磷酸盐缓冲盐(PBS)溶液中的任意一种。溶剂的加入量为使得多肽单体分子的浓度达到临界胶束浓度(CMC)值以上。
优选地,所述水浴超声的时间为30-40min,例如30min、35min或40min等。
优选地,所述静置的时间为30-40min,例如30min、35min或40min等。
第五方面,本发明提供第三方面所述的多肽自组装纳米材料在抗生素佐剂中的应用。
本发明的多肽自组装纳米材料可以用作抗生素佐剂,多肽自组装纳米材料中的功能性多肽与细菌膜相互结合,并由于组装后多价键结合效应,引起较大的膜扰动现象,导致细菌膜破裂,此时与佐剂连用的抗生素大量地进入细菌内部,最终导致细菌死亡。该策略可以实现逆转抗生素耐药细菌的耐药性,实现高效杀菌的目的,具有良好的应用前景。
相对于现有技术,本发明具有以下有益效果:
(1)本发明的多肽单体分子由疏水分子与多肽连接而成,既具有杀菌的功能,又可以实现自组装形成三维纳米材料;
(2)相比于单体分子,本发明的多肽自组装纳米材料,能够快速大量提高病灶部位抗菌材料的局部浓度,同时通过自组装纳米材料的多价键效应的作用,体现了高效杀菌的优势。

下面通过具体实施方式来进一步说明本发明的技术方案。本领域技术人员应该明了,所述实施例仅仅是帮助理解本发明,不应视为对本发明的具体限制。
实施例1
在本实施例中提供一种多肽单体分子,所述多肽单体分子由疏水分子与多肽通过酰胺键连接,所述多肽为功能性肽。
其中,疏水分子为软脂酸,功能性肽为SEQ ID NO.2所示的氨基酸序列:KKRAKKFFKKPRVIGVSIPF。
本实施例的多肽单体分子(C16-KKRAKKFFKKPRVIGVSIPF)的结构式如式Ⅰ所示:
制备方法包括如下步骤:
(1)使用FMOC策略的多肽固相合成法,以氨基端受FMOC保护的氨基酸为原料,从右到左依次连接多肽序列中所需氨基酸,得到多肽,而后加入疏水分子,将疏水分子连接至多肽上;
合成试剂选用:
(a)载体树脂:wang树脂;

本实施例提供的多肽单体分子的结构示意图如图1所示。
对本实施例提供的多肽单体分子进行高效液相色谱(HPLC)测试,测试方法如下:将多肽单体分子溶于去离子水,而后通过高效液相色谱仪LC-20A(色谱柱为C18反相色谱柱)进行测试,流动相为:流动相A:0.1%TFA+去离子水,流动相B:0.1%TFA+色谱级乙腈;进样体积为25微升。测试时采用梯度洗脱,0-25min流动相B从20%-90%;25-35min流动相B从90%-100%。本实例提供的多肽单体分子的HPLC谱图如图2所示,从图中可以看出,没有其他杂质峰出现,本实施例制备的多肽单体分子的纯度较高。
对本实施例提供的多肽单体分子进行飞行时间质谱(MALDI-TOF)测试,MALDI-TOF谱图如图3所示,从图中可以看出,已经成功合成目标多肽单体分子。
本实施例还提供一种多肽自组装纳米材料,所述多肽自组装纳米材料由本实施例的多肽单体分子自组装而成,本实施例的多肽自组装纳米材料为球形结构,内部为疏水分子,外部为功能性肽。
自组装方法包括以下步骤:
先将多肽单体分子溶于DMSO中,而后再溶于去离子水中,调节多肽单体分子的浓度达到CMC值以上,水浴超声30min,静置30min,得到所述多肽自组装纳米材料(简称:NAs)。
本实施例提供的多肽自组装纳米材料的透射电子显微镜(TEM)图如图4所示,从图中可以看出,多肽自组装纳米材料的粒径为34.0±4.1nm。
对本实施例提供的多肽自组装纳米材料进行HPLC测试,测试方法如下:将多肽自组装纳米材料溶于去离子水,而后通过高效液相色谱仪LC-20A(色谱柱为C18反相色谱柱)进行测试,流动相为:流动相A:0.1%TFA+去离子水,流动相B:0.1%TFA+色谱级乙腈;进样体积为25微升。测试时采用梯度洗脱,0-25min流动相B从20%-90%;25-35min流动相B从90%-100%。本实施例提供的多肽自组装纳米材料的HPLC谱图如图5所示,从图中可以看出,没有其他杂质峰出现,本实施例制备的多肽自组装纳米材料的纯度较高。

在本实施例中提供一种多肽单体分子,所述多肽单体分子由疏水分子与多肽通过酰胺键连接,所述多肽为功能性肽。

对比例1
本对比例与实施例1不同之处仅在于,将实施例1中多肽单体分子中不包括疏水分子软脂酸,仅由SEQ ID NO.2所示的氨基酸序列:KKRAKKFFKKPRVIGVSIPF构成,制备方法同实施例1。
由于对比例1中的多肽单体分子不含疏水分子,故其无法实现自组装。
将对比例1提供的多肽单体分子KKRAKKFFKKPRVIGVSIPF(简称:Pep20)以及实施例1提供的多肽自组装纳米材料NAs分别与PBS环境下的细菌在37℃下孵育30min后制样,空白组为PBS环境下的细菌,记对比例1提供的Pep20为对照组,实施例1提供的多肽自组装纳米材料NAs为实验组。对比例1提供的Pep20及实施例1提供的NAs与细菌作用的扫描电子显微镜(SEM)图如图7所示,从图中可以看出,空白组中的细菌没有与材料作用,表面光滑,对照组及实验组与细菌孵育30min后,均使细菌表面发生皱缩,但在相同的作用时间后,实验组,即实施例1的多肽自组装纳米材料NAs对细菌膜的破坏更加明显。
对比例1提供的Pep20及实施例1提供的多肽自组装纳米材料NAs靶向细菌膜的激光共聚焦图如图8所示,从图中可以看出,实施例1的多肽自组装纳米材料NAs与细菌膜相互作用,随着时间的增加,可看到NAs明显扰动细菌膜结构,使膜曲率发生变化,破坏细菌膜结构;而随着时间的增加,对比例1的多肽单分子材料Pep20几乎没有使细菌膜曲率发生改变,细菌膜没有明显的形态变化。
将实施例1提供的多肽自组装纳米材料进行最小抑菌浓度(MIC)测试,方法如下:采用OD值(光密度)为0.1的多重耐药的大肠杆菌,在96孔板的孔中分别加入100μL的菌液和100μL不同浓度的多肽自组装纳米材料,在37℃下孵育18h后,用酶标仪测试96孔板中孔的吸光度值,然后将吸光度通过公式计算换算成细菌存活率,即可得到多肽自组装纳米材料的MIC值,实施例1提供的多肽自组装纳米材料的MIC图如图9所示,从图中可以看出,实施例1的多肽自组装纳米材料(NAs)的最小抑菌浓度(MIC)为5.26μM。
对实施例2-6进行同实施例1的测试,结果发现实施例2-6提供的多肽自组装纳米材料同样具有抗菌效果。

Claims (17)
1.一种多肽单体分子,其特征在于,所述多肽单体分子由疏水分子与多肽通过酰胺键连接,所述多肽为功能性肽;
所述功能性肽为SEQ ID NO.2所示的氨基酸序列;
所述疏水分子为软脂酸、硬脂酸或胆固醇中的任意一种。
2.一种多肽单体分子,其特征在于,所述多肽单体分子由疏水分子与多肽通过酰胺键连接,所述多肽由功能性肽和结构性肽组成,所述结构性肽含有2-8个氨基酸序列;
所述结构性肽为SEQ ID NO.6所示的氨基酸序列;
所述功能性肽为SEQ ID NO.2所示的氨基酸序列;
所述疏水分子为软脂酸、硬脂酸或胆固醇中的任意一种。
3.根据权利要求2所述的多肽单体分子,其特征在于,所述结构性肽位于中间,所述疏水分子和功能性肽分别位于结构性肽的两侧。
4.根据权利要求1或2所述的多肽单体分子,其特征在于,所述疏水分子为软脂酸。
5.根据权利要求1或2所述的多肽单体分子,其特征在于,所述多肽单体分子的结构如式Ⅰ、式III~式V所示:
6.根据权利要求1-5中任一项所述的多肽单体分子的制备方法,其特征在于,所述制备方法包括以下步骤:
采用多肽固相合成法合成多肽,而后加入疏水分子,将疏水分子连接至多肽上,裂解,后处理,得到所述多肽单体分子。
7.根据权利要求6所述的制备方法,其特征在于,所述后处理包括洗涤、干燥。
8.一种多肽自组装纳米材料,其特征在于,所述多肽自组装纳米材料由权利要求1-5任一项所述的多肽单体分子自组装而成。
9.根据权利要求8所述的多肽自组装纳米材料,其特征在于,所述多肽自组装纳米材料具有三维纳米结构。
10.根据权利要求9所述的多肽自组装纳米材料,其特征在于,所述三维纳米结构包括球形结构或纤维状结构。
11.根据权利要求8-10任一项所述的多肽自组装纳米材料的自组装方法,其特征在于,所述自组装方法包括以下步骤:
将多肽单体分子溶于溶剂中,调节多肽单体分子的浓度达到临界胶束浓度值以上,水浴超声,静置,得到所述多肽自组装纳米材料。
12.根据权利要求11所述的自组装方法,其特征在于,所述将多肽单体分子溶于溶剂中,具体包括以下步骤:先将多肽单体分子溶于助溶剂中,而后再溶于溶剂中。
13.根据权利要求12所述的自组装方法,其特征在于,所述助溶剂包括二甲基亚砜。
14.根据权利要求12所述的自组装方法,其特征在于,所述溶剂包括去离子水、氯化钠溶液或磷酸盐缓冲盐溶液中的任意一种。
15.根据权利要求11所述的自组装方法,其特征在于,所述水浴超声的时间为30-40min。
16.根据权利要求11所述的自组装方法,其特征在于,所述静置的时间为30-40min。
17.根据权利要求8-10任一项所述的多肽自组装纳米材料在制备抗生素佐剂中的应用。
