Abstract
本发明提供了PDT化合物及其制备方法和用途。通过轴向配体、离子配体和结构调整的卟啉类化合物形成稳定的配合物,使化合物的吸收光谱实现红移,得到了光毒性强、组织穿透深度大、生物相容性良好的PDT化合物,同时还兼具光热效应和荧光性能,有助于提高治疗效果和降低光动力治疗对机体的损伤。
Description
技术领域
本发明涉及光动力治疗(PDT)领域,特别涉及光敏化合物及其制备方法和用途。
背景技术
光敏化合物在病变组织富集后,用光照激活光敏化合物,激发后的光敏化合物对病灶产生杀伤,达到治疗疾病的目的,这种方法即光动力治疗(PDT)。光敏化合物是光动力治疗中起着决定性作用的化合物。
PDT具有选择性高、无创或创伤微小、可重复治疗、毒性低等优点,在肿瘤的综合治疗中日益显示出重要作用。但是,PDT也存在一些缺点:在PDT中,光敏化合物一般需要和具有发光特性的纳米粒子共同使用,以确定光敏化合物在机体内的聚集情况,使得临床光敏剂成分复杂;另外,临床中依据光敏化合物(或光敏剂)在机体内的半衰期粗略地判断光敏化合物在正常组织中的清除率,以确定光照治疗时机,这也导致治疗选择性低,机体全身光敏副作用强。
另外,在光动力治疗中,光敏化合物的红外波长会显著影响其光毒性和组织穿透深度,红外波长越长,光敏化合物的组织穿透深度越深,越有利于光毒性的发挥。
可用于光动力治疗的化合物为PDT化合物。目前,卟啉类化合物是PDT化合物研究热点,但多数化合物仅适用于诊断或仅适用于光动力治疗,无法同时实现诊断和治疗,组织穿透深度也非常有限。
因此,研究一种兼具诊断和治疗作用的PDT化合物,对简化光敏剂、提高光动力治疗组织穿透深度、减少机体组织伤害具有现实意义。
发明内容
为了解决上述问题,本发明人进行了锐意研究,结果发现:通过对卟啉类化合物的结构进行调整,使化合物的吸收光谱实现红移,得到了光毒性强、组织穿透深度大、生物相容性良好的PDT化合物。不仅可以用于光动力治疗,还兼具光热治疗的联合疗效和荧光成像的优势,可以同时实现诊断和治疗,有利于提高光动力治疗效果和简化光敏剂的组成,从而完成了本发明。
本发明的目的在于提供以下方面:
第一方面,本发明提供一种PDT化合物,或其药学上可接受的盐、溶剂化物、非共价键复合物或前体药物,包括如下结构:
M为离子配体,选自La、Sm、Eu、Gd、Sc、Y、Ce、Pr、Nd、Pm、Tb、Dy、Ho、Er、Tm、Yb、Lu、Zn、Pd、Pt、Rh、Si和Ni中的一种;
X1和X2为五元环残基,其两端的碳原子与卟啉环上的N原子连接成环;
X5、X6、X7、X8各自独立地选自氟、氯、溴、羧基、腈基、硝基、醛基、酰基和酰胺基中的一种;
第二方面,本发明提供了上述PDT化合物的制备方法,包括:
步骤一:无氧条件下,卟啉和离子配体M的乙酰丙酮盐在200~240℃下反应1~3h,得到中间体Ⅰ;中间体Ⅰ与轴向配体L的钠盐在氯仿/甲醇=1/1的混合溶剂中反应,得到中间产物I;
步骤二:
惰性气体气氛下,中间产物I溶于有机溶剂I中,-100~-60℃下滴加还原剂,恢复室温后避光反应至结束,得到PDT化合物。
第三方面,本发明提供了药物组合物,其以上述PDT化合物为活性成分,还包括药学可接受的辅料;
所述药物组合物通过注射给药;
在药物组合物的单位剂型中,活性成分的用量为0.01mg-10g。
第四方面,本发明提供了上述PDT化合物、药学可接受的盐、溶剂化物、非共价键复合物或前体药物,以及以PDT化合物为活性成分的药物组合物在光动力治疗和光热治疗中的用途;
优选地,在制备光动力治疗和/或光热治疗肿瘤药物中的用途。
第五方面,本发明提供了上述PDT化合物、药学可接受的盐、溶剂化物、非共价键复合物或前体物质,在近红外区域700-900nm中,在荧光标记或荧光成像、光声成像中的用途。
根据本发明提供的PDT化合物及其制备方法和用途,具有以下有益效果:
(1)本发明提供的PDT化合物具有类菌绿素的光物理性质,不仅具有第二类光动力治疗能力(即产生单线态氧),还可以产生超氧阴离子自由基、羟基自由基等活性氧物种,进行第一类光动力治疗,不仅有效提高治疗效果,同时减小了对机体的损伤;
(2)本发明提供的PDT化合物三线态能级降低,因而分子不发射磷光,并使非辐射跃迁增强,因而PDT化合物在作为三线态敏化剂产生活性氧物种的同时,也具有更强的光热效应,具有光动力治疗、光热治疗的联合功效;
(3)本发明提供的PDT化合物的吸收光谱在近红外区,因而激发光具有更大的组织穿透深度,有助于在光动力治疗时减小光源对组织的损伤;
(4)本发明提供的PDT化合物具有较强的荧光,同时实现荧光成像、光动力治疗、光热治疗的作用。

下面通过对本发明进行详细说明,本发明的特点和优点将随着这些示例性说明而变得更为清楚、明确。在这里专用的词“示例性”意为“用作例子、实施例或说明性”。这里作为“示例性”所说明的任何实施例不必解释为优于或好于其它实施例。
以下详述本发明。
PDT化合物,或其药学上可接受的盐、溶剂化物、非共价键复合物、配合物或前体药物,包括如下结构:
M为离子配体,选自镧(La)、钐(Sm)、铕(Eu)、钆(Gd)、钪(Sc)、钇(Y)、铈(Ce)、镨(Pr)、钕(Nd)、钷(Pm)、铽(Tb)、镝(Dy)、钬(Ho)、铒(Er)、铥(Tm)、镱(Yb)、镥(Lu)、锌(Zn)、钯(Pd)、铂(Pt)、铑(Rh)、硅(Si)和镍(Ni)离子中的一种;
X1和X2为五元环残基,其两端的碳原子与卟啉环上的N原子连接成环;
X5、X6、X7、X8各自独立地选自氟、氯、溴、羧基、腈基、硝基、醛基、酰基和酰胺基中的一种;
Ar1、Ar2、Ar3、Ar4各自独立地为y1、y2、y3、y4,y5各自独立地选自氢、氟、氯、溴、羟基、氨基、C1-3烷基取代氨基、磺酸基、巯基和C1-3烷基取代硅基中的一种。单键上的(波浪键)表示该单键与其他基团连接。
为了提高PDT化合物的稳定性,优选所述轴向配体L为三脚凳配体,更优选为三(亚磷酸二甲酯)·环戊二烯基合钴(Ⅲ)酸基配体、氘代或卤代三(亚磷酸二甲酯)·环戊二烯基合钴(Ⅲ)酸基配体、三(亚磷酸二乙酯)·环戊二烯基合钴(Ⅲ)酸基配体和三吡唑基氢硼酸配体中的一种。
在一些优选的实施方式中,所述轴向配体L为三(亚磷酸二甲酯)·环戊二烯基合钴(Ⅲ)酸基配体、三(亚磷酸二乙酯)·环戊二烯基合钴(Ⅲ)酸基配体或三吡唑基氢硼酸配体。
离子配体M可以增强化合物分子的稳定性,进一步地,所述离子配体M为金属离子配体,优选为镥(Lu3+)、钆(Gd3+)、铕(Eu3+)、镱(Yb3+)、铒(Er3+)、锌(Zn2+)、钯(Pd2+)、铂(Pt2 +)、铑(Rh3+)和镍(Ni2+)离子中的一种。
在一些优选的实施方式中,所述离子配体M为稀土离子配体,选自镥(Lu3+)、钆(Gd3 +)、铕(Eu3+)、镱(Yb3+)、铒(Er3+)中的一种。这是由于,稀土离子配位后,由于重原子效应,使得分子系间窜越增强,产生活性氧物种能力增强,具有更强的光动力治疗能力;同时分子激发单线态寿命缩短,有利于光声信号的产生。
更优选地,所述离子配体M为镥(Lu3+)或钆(Gd3+),这是由于,与Yb3+等稀土离子相比,Lu3+具有全满的4f轨道,不发生f-f跃迁;Gd3+具有较高的4f-4f跃迁能级,不能被卟啉类化合物敏化,使得这两种金属的配合物不发生配体到金属的传能过程,能作为PDT、PTT(光热治疗)试剂使用。
另外,所述稀土离子镥(Lu3+)或钆(Gd3+)具有适中的旋轨耦合常数,对激发态分布的分配更合理。光照后,稀土配合物能在产生较多ROS(活性氧簇)的同时保留较强的荧光,同时具有荧光成像、光动力治疗、光热治疗效果。
发明人发现,此时,PDT化合物的吸收光谱红移至近红外区(700-900nm),且消光系数增大,使化合物分子具有光声成像的可能;而红移的激发光具有更大的穿透深度,光动力治疗时光源对组织的损伤也有减小;并且,所述PDT化合物还保留了较强的荧光,使其可以作为荧光成像探针使用。
X5、X6、X7、X8各自独立地选自氟、氯、溴、羧基、硝基和醛基中的一种;优选X5、X6、X7、X8各自独立地选自氟、氯或溴。
在一种优选的实施方式中,X5、X6、X7、X8为氟基,氟元素可以提高卟啉大环的反应性,有利于PDT化合物的制备;另外,C-F键的振动能量小于C-H键,可以减少振动弛豫引起的热能损耗,从而提高荧光强度。
Ar1、Ar2、Ar3、Ar4中,优选y1、y2、y3、y4、y5各自独立地选自氢、氟、氯、溴、羟基、氨基和磺酸基中的一种。

本发明还提供了上述PDT化合物的制备方法,包括:
步骤一:无氧条件下,卟啉和离子配体M的乙酰丙酮盐在200~240℃下反应1~3h,得到中间体Ⅰ;中间体Ⅰ与轴向配体L的钠盐在氯仿/甲醇=1/1的混合溶剂中反应,得到中间产物I;
步骤二:
惰性气体气氛下,中间产物I溶于有机溶剂I中,-100~-60℃下滴加还原剂,恢复室温后避光反应至结束,得到PDT化合物。
其中,在步骤一中,优选所述离子配体M的乙酰丙酮盐为钠盐或钾盐,其摩尔量为卟啉摩尔量的1~2倍;卟啉和离子配体M的乙酰丙酮盐在三氯苯中反应至结束,硅胶柱层析分离得到中间体Ⅰ。
在分离中间体Ⅰ时,优选使用石油醚脱除反应溶剂、二氯甲烷脱除未反应完全的反应物,使用二氯甲烷/甲醇=(4~6)/1的混合溶液洗脱中间体Ⅰ;更优选二氯甲烷/甲醇=5/1。
进一步地,中间体Ⅰ与轴向配体L的钠盐的摩尔量之比为1:1,在50~80℃,优选为50~70℃下反应1~3h。
反应完成后,除去反应溶剂,固体用硅胶柱层析分离,石油醚:乙酸乙酯=(10~15):1洗脱,得到中间产物Ⅰ。
为了提高中间产物Ⅰ的纯度,可以将中间产物Ⅰ重结晶处理。在一种优选的实施方式中,可以将中间产物Ⅰ在二氯甲烷/正己烷=(1~20):10的混合溶剂中重结晶,更优选为(1~20):10的混合溶剂。
在步骤二中,所述惰性气体气氛是指氮气气氛、二氧化碳气氛、氦气气氛、氩气气氛或氢气气氛。
所述有机溶剂Ⅰ选自四氢呋喃、2-甲基四氢呋喃、1,4-二氧六环和二氯甲烷中的一种或多种,优选使用四氢呋喃作为反应溶剂。
所述还原剂选自二异丁基氢化铝、三乙氧基氢化铝锂或硼氢化钠,优选二异丁基氢化铝作为还原剂还原中间产物I卟啉大环上的内酯基团,其使用量相对中间产物I而言过量,以使中间产物I充分反应。
在一种优选的实施方式中,二异丁基氢化铝与中间产物I的用量比为0.2~0.35ml/40mg,例如0.25ml/40mg。
二异丁基氢化铝滴加完毕后,反应体系恢复至室温(10~30℃),避光反应1~3h后,淬灭反应。可以使用现有技术中常用的方法淬灭,优选向反应体系中加入水或甲醇使反应淬灭。
优选地,反应结束后去除反应溶剂,固体物质使用硅胶柱层析分离得到PDT化合物。洗脱时,选用石油醚、乙酸乙酯、甲醇、乙醇、丙酮、二氯甲烷和三氯甲烷中的一种或多种作为洗脱液。优选使用石油醚与乙酸乙酯的混合溶液,更优选石油醚:乙酸乙酯=(5~8):1,例如石油醚:乙酸乙酯为7:1。
本发明提供的PDT化合物具有类菌绿素的光物理性质,不仅具有第二类光动力治疗能力(即产生单线态氧),还可以产生超氧阴离子自由基、羟基自由基等活性氧物种,进行第一类光动力治疗。根据文献报道,第一类PDT活性氧物种通常具有更强的杀伤力;因而,本发明提供的PDT化合物不仅有效提高治疗效果,同时减小了对机体的损伤。
另外,本发明提供的PDT化合物三线态能级降低,因而分子不发射磷光,并使非辐射跃迁增强,因而PDT化合物在作为三线态敏化剂产生活性氧物种的同时,也具有更强的光热效应,具有光动力治疗、光热治疗的联合功效。
进一步地,本发明提供的PDT化合物的吸收光谱在近红外区(700-900nm),因而激发光具有更大的组织穿透深度,有助于在光动力治疗时减小光源对组织的损伤。
因此,本发明提供了上述PDT化合物及其药学可接受的盐、溶剂化物、非共价键复合物、配合物或前体物质,在光动力治疗或光热治疗中作为活性成分或药物的用途。
进一步地,所述PDT化合物还具有较强的荧光,可以用于荧光标记,例如,作为荧光成像探针或荧光成像试剂。
在一种优选的实施方式中,所述PDT化合物用于近红外活细胞荧光成像探针,更优选其为定位于溶酶体的近红外活细胞荧光成像探针。
PDT化合物用于光动力治疗时,通过荧光成像可以实时观察PDT化合物在机体内的分布状况,并用于指导诊断和治疗;而光动力和光热的联合功效有助于提高治疗效果,因而,PDT化合物可以同时实现荧光成像、光动力治疗、光热治疗的作用,这是现有技术中其他光动力治疗化合物难以同时做到,有助于简化光动力治疗的光敏剂、提高治疗选择性和减少机体组织伤害。
特别地,所述PDT化合物对肿瘤细胞具有良好的亲和性,可以聚集在活体肿瘤部位,所述PDT化合物在细胞和活体层次都显示出较高的光毒性,具有治疗肿瘤的作用。
因此,本发明提供了所述PDT化合物在制备治疗肿瘤药物中的用途,特别是在光动力治疗或光热治疗肿瘤药物中的用途。
所述肿瘤包括乳腺癌、宫颈癌、肝癌、肺癌、恶性黑色素瘤或鳞状细胞癌。
本发明还提供了一种药物组合物,包括作为活性成分的上述PDT化合物或前述制备方法获得的PDT化合物,以及药学可接受的辅料。所述PDT化合物药学可接受的盐、溶剂化物、非共价键复合物或前体药物,也可以用作药物组合物的活性成分。
优选所述药物组合物注射给药,并制备为预先确定活性成分剂量的各种形式,例如注射溶液、注射乳剂、注射缓释溶液剂、注射混悬剂等常见剂型。
所述注射给药包括静脉注射、动脉注射、肌肉注射和脊椎腔注射,药物组合物中的活性成分通过循环或通过靶向给药聚集到肿瘤部位。
药物组合物中的辅料应是符合用药途径或给药方式的对人体无毒害作用的非活性成分。将活性成分与辅料,例如溶剂、等渗调节剂、表面活性剂、抗氧化剂等,混合后制得注射形式的无菌溶液或分散体系或使用前配制无菌注射用水的无菌粉末。
在一种优选的实施方式中,所述PDT化合物可以通过亲水物质包覆为纳米颗粒,以提高化合物水溶性,利于体内吸收和增强化合物的治疗效果。
优选地,在药物组合物单位剂型中活性成分的用量为0.01mg-10g。
根据病人的年龄、体重、健康状况、饮食、给药途径、联合用药、治疗时间等,具体的用药剂量可能会在病人间不同。一般情况下,在治疗上述疾病时,药物PDT化合物的剂量水平为0.01~500mg/kg体重/每天,或者每个病人每天0.1~20g。
进一步地,本发明提供的PDT化合物的吸收光谱位于近红外区,且消光系数大,还可以用于光声成像。
因此,本发明还提供了上述PDT化合物及其药学可接受的盐、溶剂化物、非共价键复合物或前体物质,在荧光标记或荧光成像、光声成像中的用途,是潜在的荧光标记物、光声成像和荧光成像药剂。
实施例
实施例1化合物Lu-1的合成
步骤1:在Schlenk管中加入Lu(acac)·6H2O(六水合乙酰丙酮镥)0.10mmol,卟啉a(0.05mmol)和8mL溶剂三氯苯,脱气,充入氮气,240℃封管反应1~3h。监测反应体系至无卟啉荧光后,冷却至室温。硅胶柱层析分离,分别用石油醚,二氯甲烷,二氯甲烷/甲醇=5/1洗脱,收集稀土卟啉。将稀土卟啉和配体三(亚磷酸二甲酯)·环戊二烯基合钴(Ⅲ)酸钠(NaLOMe)(0.05mmol)溶解到10mL CHCl3/MeOH=1/1中,60℃反应2h,旋蒸除去溶剂,固体用硅胶柱层析分离,二氯甲烷/正己烷(1~10:10)重结晶,得Lu-a。
步骤2:氮气氛下,Lu-a溶于无水无氧四氢呋喃,-80℃下滴加二异丁基氢化铝(DIBAL),用量以0.25ml/40mg Lu-a计。恢复室温,避光反应1h,加水淬灭反应。旋干溶剂,硅胶柱层析分离,石油醚:乙酸乙酯=7:1洗脱,收集,去除溶剂,得产物Lu-1,红色固体。

FT-IR图谱如图3所示:与未还原化合物(a或Lu-a)相比,分子1600-1800cm-1区域的羰基(C=O)伸缩振动特征峰消失,表明内酯基团的成功还原。同时在3400cm-1附近出现较强的O-H键振动宽峰,表明-OH结构的存在;
UV/Vis(CH2Cl2,25℃):λmax(nm)(logε):341(5.01),396(5.14),503(3.77),543(4.42),697(4.15),753(5.16).
实施例2化合物Lu-2的合成
反应过程与实施例1基本相似,区别在于:
得到Lu-1后,进一步用BF3·Et2O催化,在甲醇溶剂中醚化得到Lu-2。

实施例12纳米颗粒包覆
将1μmol化合物Gd-1(实施例6制备)或Lu-1(实施例1制备)溶解于5mL四氢呋喃,加入20mg介孔氧化硅纳米颗粒(MSN),室温敞口搅拌至溶剂完全挥发。向所得固体中加入5mL水,超声30分钟。离心,水洗3次后,干燥,得到负载了化合物的硅纳米颗粒。
取20mg上述颗粒,加入80mg聚醚F127,加入2mL甲苯和四氢呋喃1:1(体积比)混合溶液,35℃蒸发至溶剂完全挥发,加入2mL水,室温搅拌4小时,静置24小时,取上层清液,即为Gd/Lu-1-MSN的水分散液。
实验例
实验例1光物理性质检测
对实施例1获得的Lu-1和实施例6获得的Gd-1进行紫外可见吸收光谱和发射光谱扫描,结果如图4所示。
由吸收光谱图可知,化合物Lu-1,Gd-1的吸收光谱可以覆盖可见光区和近红外区,并且在700-800nm的近红外区吸收谱带较强。由发射光谱图可知,对化合物进行光激发,化合物的荧光光谱也处于近红外区域。
由此可见,化合物Lu-1和Gd-1在红外成像或活体荧光成像中具有较深的组织穿透深度。
实验例2活性氧物种检测
1)在空气氛围下,实施例1获得的Lu-1和实施例6获得的Gd-1的氯仿溶液在760nm近红外光激发下,在1270nm附近检测到单线态氧的磷光发射,结果如图5中(1)所示,表明分子在光激发下具有产生单线态氧的能力。
2)在空气氛围下,向实施例12制得的Gd/Lu-1-MSN的水分散液中加入二氢乙锭作为超氧阴离子自由基探针,光照(760nm发光二极管灯,7.5mW/cm2)条件下,检测到二氢乙锭被超氧自由基氧化生成的氧化乙啶的荧光,且荧光强度随光照时间延长而增强。结果如图5中(2)所示,表明化合物在光照下能具有产生超氧阴离子自由基的能力。
3)在空气氛围下,向实施例12制得的Gd/Lu-1-MSN的水分散液中加入5-甲基-5-叔丁基甲酰基-吡咯啉氮氧化合物(BPMO)作为羟基自由基捕获剂。在光照(760nm发光二极管灯,7.5mW/cm2)条件下,用电子顺磁共振检测到BMPO羟基加合物信号。结果如图5中(3)所示,表明化合物在光照下能具有产生羟基自由基的能力。
实验例3光热检测
对Gd/Lu-1-MSN的水分散液进行光热转换实验。
80μg/mL(按化合物分子计)Gd/Lu-1-MSN的水分散液光照(760nm发光二极管灯,100mW/cm2)条件下,通过热电偶与红外热成像仪测量分散液温度变化情况,结果如图6中(1)和(2)所示。
由图可知,光照下,Gd/Lu-1-MSN能够使溶液升温,而空白纳米颗粒几乎不能使温度上升,说明化合物Gd-1/Lu-1能使水温明显上升。表明化合物具有较强的光热效应,具有光声成像与光热治疗的潜力。
实验例4细胞光毒性
采用实施例12的方法,用介孔二氧化硅纳米颗粒对化合物进行吸附负载,使之能较好地分散到水溶液中。
实验所用细胞包括HeLa人类宫颈癌细胞、4T1小鼠乳腺癌细胞。细胞培养在添加10%灭活胎牛血清与1%青霉素-链霉素的DMEM完全培养基中进行,培养温度为37℃,培养气氛为5%二氧化碳。
将传代培养的HeLa细胞经胰酶消化后,以适当浓度分散在培养基中。将分散后的HeLa细胞接种至多聚-D-赖氨酸修饰的平底96孔板,使每孔培养基为200μL,细胞数约为104个,保留一组无细胞的培养基进行空白对照。在暗环境中培养细胞24小时后移除培养基,加入100μL新鲜培养基与100μL预先配好的Lu-1,Gd-1制得的培养基溶液,稀释使样品为0.1-5μM的梯度浓度。在暗环境中继续培养24小时后,移除培养基,使用pH=7.4的PBS润洗各孔3次。向各孔加入100μL PBS缓冲液,在相同光强(约7.5mW/cm2)的760nm发光二极管灯下照射30分钟。移除各孔的PBS并使用200μL新鲜培养基替换,继续培养24小时。完成后,移除培养基并使用PBS润洗各孔3次。然后用培养基配置10%的CCK-8试剂(Cell Counting Kit-8),每孔加入100μL,并培养2小时。在此过程中,CCK-8试剂中的2-(2-甲氧基-4-硝基苯基)-3-(4-硝基苯基)-5-(2,4-二磺酸苯)-2H-四唑单钠盐(WST-8)在电子耦合试剂的辅助下可被活细胞还原为黄色的甲瓒产物,使溶液在450nm的吸光度发生变化,且变化值与活细胞数目成正比。使用酶标仪测定各孔在450nm吸收变化,根据以下计算各孵育浓度下细胞存活率:
CV=(As–Ab)/(Ac–Ab)×100%
CV指细胞存活率;As,Ac和Ab分别指的是孵育化合物的细胞的吸光度、空白细胞的吸光度和空白对照的吸光度。
根据各孵育浓度下的细胞存活率计算化合物在不同细胞系中的半数致死浓度IC50,如下:单位为μM
实验例5化合物光毒性机理研究
测试实施例12制得的Gd-1-MSN在缺氧、冷水浴、缺氧与冷水浴同时存在条件下对4T1细胞的光毒性。
在光照时对细胞进行冷水浴处理,使细胞温度维持室温,消除光热效应带来的毒性影响,检测到细胞存活率上升,半数致死浓度上升到大于16μM,结果如图7所示,证明部分细胞毒性来自于光热效应。
在光照前6小时用厌氧产气袋对细胞进行缺氧处理,再进行光照(光照后在正常氧含量情况下孵育24h),观察到细胞存活率上升,半数致死浓度上升到(14.5±0.4)μM,,结果如图7所示,证明部分细胞毒性来自于需氧的光动力效应。
在细胞毒性实验中同时进行缺氧与冷水浴处理,观察到细胞存活率上升,与不进行光照相似,结果如图7所示,表明细胞毒性来自于光热效应与需氧光动力效应的共同作用。
实验例6化合物光毒性诱导凋亡的机理研究
分别使用浓度0,4μM的Gd-1处理HeLa细胞,PCR检测结果如图8所示,在光照条件下,Gd-1可以激活Cleaved PARP蛋白以及Cleaved Caspase3蛋白的表达。如上两种蛋白的激活,证明Gd-1可以激发细胞凋亡信号通路,诱导细胞发生凋亡。而在无光照的条件下,Gd-1不能激活凋亡信号通路,对肿瘤细胞无杀伤作用。
通过流式细胞仪技术,检测上述细胞发生凋亡的现象,结果如图9所示,同样在光照的条件下(+),Gd-1可以诱导发生凋亡的细胞比率上升;而无光的条件下,Gd-1不能诱导细胞发生凋亡。
实验例7细胞荧光成像
采用实施例12的方法,用介孔二氧化硅纳米颗粒对化合物进行吸附负载,使之能较好地分散到水溶液中。
实验所用细胞为HeLa人类宫颈癌细胞。细胞培养在添加10%灭活胎牛血清与1%青霉素-链霉素的DMEM完全培养基中进行,培养温度为37℃,培养气氛为5%二氧化碳。
将传代培养的HeLa细胞经胰酶消化后,以适当浓度分散在培养基中。将分散后的HeLa细胞接种至多聚-D-赖氨酸修饰的载玻片,培养细胞24小时后,加入100μL预先配好的分子Gd-1的培养基溶液,稀释使样品为4μM的浓度。孵育12小时后,加入溶酶体绿色荧光探针,共同孵育15分钟后移除培养基,使用pH=7.4的PBS润洗3次。用ISS集成激光扫描共焦荧光寿命成像系统荧光成像,用超连续激光在740nm处激发,760nm高通滤光片,收集Gd-1分子荧光信号;488nm激光器激发,525/50nm带通滤光片,收集溶酶体探针荧光信号。
结果如图10所示,Gd-1在近红外区域具有较强的荧光信号,与溶酶体探针的信号对比发现能较好地重叠,表明分子具有作为近红外活细胞荧光成像探针的潜力,且定位于溶酶体。
实验例8动物体内荧光成像
活体实验中所有的动物实验都严格遵守中国动物实验的规定进行,使用五周大BALB/C白鼠进行实验,该模型鼠右后腿皮下种植4T1肿瘤。
活体荧光成像所使用的成像仪器是IVIS Spectrum荧光成像系统,可实现高灵敏度生物发光及荧光成像,配置28个高效滤光片,覆盖430-850nm全波段。
实验过程:小鼠尾静脉注射250uL 80μg/mL(以化合物计)Gd-1-MSN的PBS分散液,将小鼠放置在成像仪器中,处于2L/min的氧气和2%的异氟烷的混合气体气氛中,使其麻醉。使用激发波长为740nm进行激发。图像采集波长为820nm,曝光时间为自动。
尾静脉注射化合物24小时后处死小鼠并解剖,取出心、肝、脾、肺、肾、胰、胃、膀胱肌、后肢骨、脑、肿瘤,在成像仪下进行成像。其他条件与活体实验一致。
实验结果由图11所示,化合物进入小鼠以后可实现肿瘤部位的成像,在24小时信号达到最高,并且背景干扰低,周围组织信号弱。解剖实验的结果与活体成像的结果一致,化合物都是定位在肿瘤组织,部分定位于肝脏。这说明化合物的近红外发光性质有效地降低了在活体荧光成像中的背景干扰,即使在活体非解剖的状态下,也可以实现肿瘤的高分辨率荧光成像。
实验例9磁共振成像性质
对Gd-1-MSN进行T1磁共振成像实验,结果如图12所示,由图可知,化合物具有一定的T1磁共振造影能力,纵向弛豫率约1.1s-1·M-1。
实验例10光声成像
活体光声成像所使用的成像仪器是iThera超高分辨率小动物光声成像系统MOSTinvision 128。
实验过程:小鼠尾静脉注射250uL 80μg/mL(以化合物计)Gd-1/Lu-1-MSN的PBS分散液(约1mg/kg),将小鼠放置在成像仪器中,处于2L/min的氧气和2%的异氟烷的混合气体气氛中,使其麻醉。使用激发波长为780nm进行激发。曝光时间为自动。
实验结果由图13所示,化合物进入小鼠以后可实现肿瘤部位的光声成像,在24小时信号达到最高。
实验例11活体光热效应
使用BALB/C小鼠,鼠龄5周,雄性,体重16-25克。每只小鼠在右后肢皮下接种4T1鼠乳腺癌细胞100ul(5×106),一周后进行实验。
小鼠按编号进行随机分组,实验分为空白对照组(PBS-)、给药暗对照组(Gd-1-MSN-)、光照对照组(PBS+)和实验组(Gd-1-MSN+、Lu-1-MSN+),每组7只。给药暗对照组和空白对照组通过尾静脉分别注射250μL PBS和Gd-1-MSN水分散液后不进行干预,光照对照组只进行光照射(760nm发光二极管灯,100mW/cm2);实验组根据小鼠体重以1mg/kg给药,尾静脉注射约250μL 80μg/mL化合物的纳米颗粒分散液。给药后动物避光饲养24h后光照肿瘤部位,每只光照5min。
对3个光照组(PBS+、Gd-1-MSN+和Lu-1-MSN+)在光照过程中,用FLIR E40红外热像仪进行热成像,结果如图14所示,化合物使光照过程中肿瘤部位温度升高,且温度随光照时间延长而上升。表明化合物在活体中同样具有光热效应。
实验例12动物水平药物光治疗实例
实验例11中小鼠治疗完毕不给予避光处理,放入饲养笼中继续饲养,每隔两天用游标卡尺测量肿瘤体积、称量体重。治疗4周。
实验结果如图15所示,通过尾静脉注射给药的方式,化合物Gd-1和Lu-1可以有效抑制皮下肿瘤的生长,小鼠的生存率上升,并且小鼠体重没有明显的减轻;而未给药的对照组小鼠则肿瘤生长迅速。
另外,给药暗对照组的肿瘤生长趋势与未给药组相似,证明了化合物的光疗机理,即通过光照激发才能实现治疗效果。
以上结合具体实施方式和范例性实例对本发明进行了详细说明,不过这些说明并不能理解为对本发明的限制。本领域技术人员理解,在不偏离本发明精神和范围的情况下,可以对本发明技术方案及其实施方式进行多种等价替换、修饰或改进,这些均落入本发明的范围内。本发明的保护范围以所附权利要求为准。
Claims (13)
1.PDT化合物,或其药学上可接受的盐,所述化合物为如下结构:
L为轴向配体,选自三(亚磷酸二甲酯)·环戊二烯基合钴(Ⅲ)酸基配体、氘代或卤代三(亚磷酸二甲酯)·环戊二烯基合钴(Ⅲ)酸基配体、三(亚磷酸二乙酯)·环戊二烯基合钴(Ⅲ)酸基配体中的一种;
M为离子,选自La、Sm、Eu、Gd、Ce、Pr、Nd、Pm、Tb、Dy、Ho、Er、Tm、Yb、Lu中的一种;
X5、X6、X7、X8各自独立地选自氟、氯、溴、羧基、腈基、硝基、醛基、酰基和酰胺基中的一种;
2. 根据权利要求1所述的PDT化合物,其特征在于, 所述离子M为Lu3+、Gd3+、Eu3+、Yb3+、Er3+中的一种。
3.根据权利要求1所述的PDT化合物,其特征在于,X5、X6、X7、X8各自独立地选自氟、氯、溴、羧基、硝基和醛基中的一种;和/或,
Ar1、Ar2、Ar3、Ar4中, y1、y2、y3、y4、y5各自独立地选自氢、氟、氯、溴、羟基、氨基和磺酸基中的一种。
5.根据权利要求1-4之一所述PDT化合物的制备方法,其特征在于,包括:
步骤一:无氧条件下,卟啉衍生物和离子M的乙酰丙酮盐在200~240℃下反应1~3h,得到中间体Ⅰ;中间体Ⅰ与轴向配体L的钠盐在氯仿/甲醇=1/1的混合溶剂中反应,得到中间产物I,所述卟啉衍生物为所述PDT化合物中除去轴向配体L和离子M之外的结构,并将其中X1、X2的C-OH均变为C=O;
步骤二:惰性气体气氛下,中间产物I溶于有机溶剂I中,-100~-60℃下滴加还原剂,恢复室温后避光反应至结束,得到PDT化合物。
6.根据权利要求5所述的制备方法,其特征在于,步骤一中,
卟啉衍生物和离子M的乙酰丙酮盐在三氯苯中反应至结束,硅胶柱层析分离得到中间体Ⅰ;
中间体Ⅰ与轴向配体L的钠盐在50~80℃,反应1~3h,硅胶柱层析分离,石油醚:乙酸乙酯=(10~15):1洗脱,得到中间产物Ⅰ。
7.根据权利要求6所述的制备方法,其特征在于,中间体Ⅰ与轴向配体L的钠盐50~70℃下反应。
8.根据权利要求5所述的制备方法,其特征在于,步骤二中,所述有机溶剂Ⅰ选自四氢呋喃、2-甲基四氢呋喃、1,4-二氧六环和二氯甲烷中的一种或多种,所述还原剂选自二异丁基氢化铝、三乙氧基氢化铝锂或硼氢化钠;
反应结束后去除反应溶剂,固体物质使用柱层析分离得到PDT化合物。
9.根据权利要求8所述的制备方法,其特征在于,所述有机溶剂Ⅰ为四氢呋喃,所述还原剂为二异丁基氢化铝。
10.药物组合物,其以权利要求1-4之一所述PDT化合物或权利要求5-9之一所述方法制得的PDT化合物为活性成分,还包括药学可接受的辅料;
所述药物组合物通过注射给药;
在药物组合物的单位剂型中,活性成分的用量为0.01mg-10g。
11.根据权利要求1-4之一所述PDT化合物、其药学可接受的盐,以及以权利要求1-4之一所述的PDT化合物为活性成分的药物组合物在制备光动力治疗和/或光热治疗的药物中的用途。
12.根据权利要求11所述的用途,其特征在于,所述用途为在制备光动力治疗和光热治疗肿瘤药物中的用途。
13.权利要求1-4之一所述PDT化合物、其药学可接受的盐在近红外区域700-900nm中,在制备荧光标记或荧光成像、光声成像试剂中的用途。
