Abstract

背景技术
癌症给人类造成的痛苦和经济负担是不言而喻的,因此我们一直在不遗余力地与癌症作斗争。但到目前为止,在癌症诊断和治疗的两个关键方面仍然面临巨大的挑战。在诊断方面,视觉成像,例如传统的电脑断层扫描(CT)及磁共振成像(MRI),是最直接的方法,并已普遍应用于临床。荧光成像技术作为一种新兴的成像方式,由于操作简单、灵敏度高、无放射性、无创伤等特点,在临床诊断中显示出巨大的应用前景。与此同时,AIE的光热治疗治疗引起人们的关注,AIE的光热转换效应,不产生副作用。因此通过合理的设计,进一步探索近红外二区AIE在生物医学中应用成为研究热点。
光疗法(PTT或者PDT)是目前治疗肿瘤的一种有效方法,通过非辐射途径的光疗功能,解决癌症治疗。与手术,化疗和放射治疗等常规治疗相比,光疗具有无创性和副作用小的优点,并且可以在FLI的指导下提供精确的肿瘤定位治疗。由于新的作用机制,光疗与其他治疗方法不具有交叉耐药性,因此可以与其他治疗方法结合以最大限度地发挥每种治疗方式的优势。
FLI技术虽然发展迅速,但是目前FLI技术的发展仍然面临两个瓶颈,一个是突破长波长发射实现高穿透深度,另一个是突破高发光强度实现高空间分辨率和低背景干扰。近年来,近红外发光材料的出现打破了传统发光材料在波长上的瓶颈,由于长波长超过1000nm,已被证明具有深度组织穿透能力。虽然目前已经开发出的理想的荧光材料仍然很少,但这类系统通常需要引入大型复杂的π-共轭结构来扩展吸收和发射波长,从而不可避免地造成巨大的合成困难和众所周知的聚集引起的猝灭(ACQ)效应。在这方面,构建具有聚集诱导发光(AIE)特性的主-客体(D-A)型扭曲结构,进一步探索AIE在生物医学中应用。
常见的金纳米材料通常位于近红外一区,这极大限制了其在光治疗肿瘤及成像中的临床应用,并且常见的金纳米材料方法和质量不可控,进一步限制应用。
然而研究出具有靶向作用的近红外二区含有金的AIE存在极大的挑战,这种具有红外二区靶向含金的AIE材料还未见报道。

将适量化合物1(4,8-双(5-溴-3-(己基)-2-噻吩基)-2λ4δ2-苯并[1,2-c:4,5-c']双[1,2,5]噻唑)和适量化合物2(4-甲氧基-N-(4-甲氧基苯基)-N-(4-(4,4,5,5-四甲基-1,3,2-硼酸-2-苯基)苯胺)在THF/水(v/v)混合溶液加入Pd(PPh3)4和K2CO3。在氮气保护下,将混合物在一定温度下搅拌,然后加入水使反应猝灭,并用 CH2Cl2萃取。混合有机提取物在无水Na2SO4上干燥并过滤。通过旋转蒸发去除溶剂。通过柱层析法(硅胶,CH2Cl2/己烷=1:1,v/v)纯化,得到固体浅绿色化合物3。
(2)合成有机化合物TBTP:
加入Pd(PPh3)4(116mg,0.10mmol)和K2CO3(414mg,3.00mmol)的化合物 3(963mg,1.00mmol)和吡啶-4-基硼酸(160mg,1.30mmol)的二氧六烷/四氢呋喃/水(v/v/v:3:2:1)混合物(42mL)。在N2下70℃搅拌12小时,然后加入水(20mL)使反应淬灭,用CH2Cl2(3×20mL)提取。混合有机提取物在无水的 Na2SO4上干燥并过滤。采用旋转蒸发法去除溶剂。产物经硅胶柱层析 (CH2Cl2/MeOH=20:1,v/v)纯化得到化合物TBTP。
(3)合成TBTP-Au:
TBTP-Au的合成:将适量C6F5Au(tht)和TBTP的混合物在CH2Cl2中在保护下,室温搅拌。反应完成后,溶剂被蒸发。粗产物由CH2Cl2/冰乙醚重结晶得到深绿色固体。
(4)合成TBTP-Au-cRGD NPs
合成TBTP-Au-cRGD NPs,将适量DSPE-mPEG溶于适量THF中,并将适量TBTP-Au分子溶于适量THF中。将两组溶液充分混合并使用探针超声波器在60W输出下超声处理60s以形成TBTP-Au NP。将THF在通风柜中用N2蒸发8-16h,加入适量的c-RGD肽,搅拌12h,并通过透析去除多余的 c-RGD,并用超纯水透析6-12次。收集TBTP-Au-cRGD NP供进一步使用。并用同样的方法合成了IR780和TBTP NPs。
步骤(2)所述所用的反应溶剂THF/H2O(v/v=1:1-5:1),其用量为10~40 mL;
步骤(2)所述反应温度在70℃,需要反应时间一般为4-12h。
TBTP-Au-cRGD NPs在波长为808nm的激光照射下,光热转换效率为20.51%,TBTP-Au-cRGD NPs的光热转换效率计算公式如下:

其中,Tmax和Tsurr分别表示最大温度和环境温度,单位为℃;Qdis是由于溶剂和容器对光的吸收所造成的热损失,其值约为0mW;I为激光功率密度,单位为W/cm2;A808是样品在808nm处的吸光度值,无单位;h为热传递系数,单位为W/(cm2·℃);S为容器表面积,单位为m2;τs表示时间常数;mD表示溶剂质量,其值为1g;cD表示水的比热容,其值为4.2J/(g·℃);采用波长为808nm且安全功率为180mW/cm2的激光照射10min,温度上升约 70.8℃。
进一步,合成的肿瘤靶向TBTP-Au-cRGD NPs应用于了肿瘤治疗的研究。
本发明的有益效果如下:
本发明通过在TBTP中引入了经典和简单的五氟苯基(I)金单元,实现近红外二区AIE与金配位结合,该反应条件简便易制备,通过核磁H谱和碳谱,以及质谱确认结构,近红外二区发射在1000-1350nm左右,通过与靶向肽结合形成TBTP-Au-cRGD NP,可以在体内实现特异性的NIR II肿瘤成像,并协同化疗和光热疗法提高癌症治疗。为临床治疗提供了广阔的应用前景。

实施例1
(1)合成TBTP:
将化合物4,8-双(5-溴-3-己基-2-噻吩基)-2λ4δ2-苯并[1,2-c:4,5-c']双[1,2,5] 噻唑,(963mg,1.30mmol)和化合物4-甲氧基-N-(4-甲氧基苯基)-N-(4-(4,4,5,5- 四甲基l-1,3,2-双硼酸酯-2-yl)苯基)苯胺(431mg,1.00mmol)在THF/水(v/v:5: 1)混合物(42ml)中的溶液中加入Pd(PPh3)4(58mg,0.05mmol)和K2CO3(414 mg,3.00mmol)。混合物在70℃下在N2保护下搅拌4小时,然后加入水(20ml) 使反应猝灭,并用CH2Cl2(3×20ml)提取。收集有机提取物在无水Na2SO4上干燥并过滤。通过旋转蒸发去除溶剂。粗品以柱色谱法(硅胶,CH2Cl2/己烷= 1:1,v/v)纯化,得到化合物3为浅绿色固体。
(2)合成TBTP-Au
将C6F5Au(tht)(362mg,0.40mmol)和TBTP(193mg,0.20mmol)的混合物在室温下在,CH2Cl2(30ml)中在N2保护下搅拌48小时。反应完成后,溶剂被蒸发。粗产物由,CH2Cl2/冰乙醚重结晶得到93%收率(247mg)的深绿色固体。
(3)合成TBTP-Au-cRGD NPs
1mg DSPE-mPEG-Mal和1mg DSPE-mPEG溶于1ml THF中,1mg TBTP-Au分子也溶于1ml THF中。将两组溶液充分混合并凝固1分钟。然后,将混合物移到18ml毫升水中,然后使用探针超声波器在60W输出下超声处理60s以形成TBTP-Au NP。将THF在通风柜中用N2蒸发8h,加入5×10-2 M的cRGD肽,搅拌12h,用毫克水透析去除多余的cRGD,用毫克水换6次。收集TBTP-Au-cRGD NPs供进一步使用。用同样的方法合成了ir780和tbtp nps。
实施例2
(1)合成TBTP:

实施例1-2
同实施例1,其不同之处仅在于,2例中没有与五氟苯金反应,没有与cRGD 混合。
体外升降温实验:将浓度为1mg/mL的TBTP-Au-cRGD NPs,用功率强度为180mW/cm2的808nm激光照射。然后采用红外摄像机(FLIR TM A325SC 摄像机)每隔60s测量并记录温度。并在第10min时停止照射,记录温度下降的变化。
为了使纳米材料具有更好的靶向性,1mg DSPE-mPEG-Mal和1mg DSPE-mPEG溶于1ml THF中,1mg TBTP-Au-cRGD分子也溶于1ml THF中。将两组溶液充分混合并凝固1分钟。然后,将混合物移到18ml毫升水中,然后使用探针超声波器在60W输出下超声处理60s以形成TBTP-Au NPs。将 THF在通风柜中用N2蒸发8h,加入5×10-2M的cRGD肽,搅拌12h,用毫克水透析去除多余的cRGD,用毫克水换6次。收集TBTP-Au-cRGD NPs 供进一步使用。
细胞毒性:人乳腺癌细胞(MDA-MB-231)在DMEM(Gibico,USA)培养基中培养,培养基中添加10%胎牛血清,1%青霉素-链霉素液,37℃,5% CO2。此外,小鼠乳腺癌细胞(4T1)在添加10%胎牛血清、1%青霉素-链霉素液的RPMI 1640培养基(procell)中培养,37℃,5% CO2。MCF-10A细胞和4T1细胞分别以每孔0.5×104个细胞的密度接种于96孔板中。孵育24h后,加入含有不同浓度UGs@PEG的新鲜培养基孵育24h。此外,TBTP-Au-cRGD NPs+NIR组的4T1细胞孵育8h后,用808nm,180mW/cm2,10min的激光照射。最后,采用标准MTT法检测细胞活性。
数据分析与讨论:如图1(A)所示,通过TBTP与五氟苯金反应得到 TBTP-Au化合物,通过H谱,C谱以及TOF质谱确定化合物结构。图1(B) 所示,化合物吸收光谱以及荧光光谱,以及良好的二区发射光谱,最大发射波长1035nm。

图3(A)TBTP-Au-cRGD NPs的靶向性研究。将αVβ3整联蛋白结合环状 RGD缀合到TBTP-Au NPs的表面以提供具有高肿瘤特异性的TBTP-Au-cRGD NPs。与MDA-MB-231三阴性乳腺癌细胞孵育12小时后,细胞摄取大量纳米颗粒,主要分布在MDA-MB-231细胞的细胞质中(图3(A))。相比之下, MDA-MB-231细胞在没有cRGD缀合的TBTP-Au NPs处理时显示有限的摄取。为了进一步证实靶向效应是由cRGD-αVβ3整联蛋白识别,我们使用游离cRGD 肽预处理MDA-MB-231细胞,然后在相同条件下孵育TBTP-Au-cRGD。游离 cRGD处理后,MDA-MB-231细胞摄取的纳米颗粒数量明显减少,说明NPs 的靶向能力是通过cRGD整合素介导识别的。荧光强度定量的结果进一步证实了TBTP-Au-cRGD NPs的肿瘤靶向能力(图3(B))。
(I)金复合物已被充分证实通过抑制硫氧还蛋白还原酶(TrxR)的活性来扰乱细胞氧化还原稳态,从而上调活性氧类(ROS)水平,从而诱导细胞自噬和凋亡。为了评估TBTP-Au是否具有通过TrxR抑制提高细胞内ROS水平的能力,我们评估了与TBTP-Au-cRGD NPs孵育4小时后MDA-MB-231细胞的细胞内ROS水平。用商业化的ROS指示剂DCFH-DA对细胞内的ROS进行信号传导,该指示剂经细胞内ROS氧化后能发出明亮的绿光。如图3(C)所示,TBTP-Au-cRGD NPs可被MDA-MB-231细胞有效摄取并分布在细胞质内。通过DCFH-DA的亮绿色荧光和鉴定结果显示,经TBTP-Au-cRGD NPs处理的MDA-MB-231细胞的细胞内ROS水平显着增强,明显高于对照组(图3(D))。与对照组相比,TBTP-cRGD NPs治疗组没有升高(图3(D))。因此,这些结果证实了TBTP-Au-cRGD NPs通过抑制TrxR提高细胞内ROS水平。随后,通过进一步评估体外各种处理后的细胞活力,研究了TBTP-Au-cRGD NPs对 MDA-MB-231细胞的抗癌潜能。在处理后24h,用活/死染色法和MTT法检测 MDA-MB-231细胞的存活率。如图3(E)所示,TBTP-Au-cRGD NPs在黑暗条件下无毒,在暗条件下可以诱导约50%的细胞死亡,表明由TBTP-Au-cRGD NPs诱导的升高的细胞内ROS水平,发挥抗癌作用。用808nm激光(180 mW/cm2)照射10分钟后,TBTP-Au-cRGD NPs表现出更明显的毒性,几乎所有的癌细胞都被杀死。与已知的商用发热剂IR-780相比,TBTP-Au-cRGD NPs 的杀伤效果要高得多,通过以(I)金化合物为基础的化疗和TBTP诱导的PTT 的协同作用,进一步揭示了TBTP-Au-cRGD NPs的优异的抗肿瘤效果。相反,在不存在TBTP-Au-cRGD NPs的情况下,单次激光照射对细胞活力的影响可以忽略不计。此外,TBTP-Au-cRGD NPs的高效抗癌作用通过后续的MTT实验得到了进一步证实。如图3(F)所示,在黑暗条件下,TBTP-cRGD NPs 对细胞无毒性,而TBTP-Au-cRGD NPs对MDA-MB-231细胞有明显的毒性。特别是在暗条件下,TBTP-Au-cRGD NPs处理的细胞在24h、48h和72h的存活率分别为73.49%、67.21%和58.64%,进一步证实了(I)金通过提高细胞内ROS水平诱导产生细胞毒性。激光照射5min(160mW/cm2)后, TBTP-Au-cRGD NPs处理的细胞存活率明显下降,24h、48h和72h的相应存活率分别为22.54%、16.17%和13.89%。而TBTP-Au-cRGD NPs联合5min 激光照射的细胞存活率分别为74.91%、68.80%和63.10%,明显高于 TBTP-Au-cRGD NPs联合激光照射的细胞存活率。基于这些结果,可以得出结论:(I)金的升高ROS水平能力与有效光热转化TBTP-Au-cRGD NPs协同增强化疗和PTT能力。
TBTP-Au-cRGD NPs的NIR-II荧光如图4(A)不同浓度(0.01,0.1,0.25,0.5,1 mg/ml)的TBTP-cRGD NPs和TBTP-Au-cRGD NPs(1000nm长通滤波器, 808nm激发,270mW cm-2,60ms)的NIR-II荧光图像。TBTP–cRGD NPs和 TBTP-Au-cRGD NPs的荧光均随着浓度的增加而逐渐增强,但在相同浓度下 TBTP-Au-cRGD NPs对应的荧光强度远高于配体基团。与TBTP-cRGD NPs 相比,(I)金引入使TBTP-Au-cRGD NPs更卓越的成像性能。将TBTP-Au-cRGDNPs应用于体内NIR-II成像。血流中注射TBTP-Au-cRGD NPs后,小鼠背体和后肢在1000nmLP下成像,如图4(B,C)所示在5分钟时使用 TBTP-Au-cRGD NPs(2mg/ml,100μl)对小鼠背侧(B)和后肢血管(C)进行体内 NIR-II荧光成像,动脉、静脉、毛细血管均清晰可见,因此可以准确绘制血液循环系统。基于TBTP-Au-cRGD NPs优异的体内NIR-II成像能力,随后对肿瘤进行了体内NIR-II成像,用于视觉诊断。为此,我们选择小鼠4T1乳腺肿瘤模型来评估TBTP-Au-cRGD NPs是否可以通过纳米颗粒堆积来分化肿瘤。如图4(D)在不同时间静脉注射TBTP-Au-cRGD NPs(2mg/ml,100μl)后, TBTP-Au-cRGD NPs在荷瘤小鼠中的生物分布如图4(D)所示,在荷瘤裸鼠静脉注射TBTP-Au-cRGD NPs后6h,肿瘤部位明显可见明亮的荧光信号。积累18h后,肿瘤荧光强度达到最大值,信本比高,边界清晰。图4(E)显示了注射后6h、18h、24h、48h和72h肿瘤的NIR-II荧光强度。这些结果表明 TBTP-Au-cRGD NPs具有良好的肿瘤靶向性和蓄积能力。为了进一步验证它们的肿瘤特异性积累,在静脉注射后72h处死小鼠,收集肿瘤和主要器官(心、肝、脾、肺和肾)并进行NIR-II荧光成像。肿瘤具有最强的NIR-II荧光信号。 NIR-II成像强度的定量分析(图4(G))进一步提供了肿瘤对心脏9.05倍、肿瘤对肺4.20倍、肿瘤对肝脏1.21倍、肿瘤对脾脏1.21倍、肿瘤对肠道1.19倍、肿瘤对肾脏1.84倍的荧光比,进一步验证了TBTP-Au-cRGD NPs的高肿瘤特异性。因此,这些结果充分证明TBTP-Au-cRGD NPs优异的NIR-II成像性能和显著的肿瘤特异性。
TBTP-Au-cRGD NPs的体内肿瘤治疗。体外实验表明,TBTP-Au-cRGD NPs可通过化疗和PTT途径发挥高效的协同抗癌治疗作用。为了进一步证明其增强的体内协同抗癌效果,我们采用4T1荷瘤小鼠进行了抗肿瘤生长实验。首先,我们在体内评估了TBTP-Au-cRGD NPs的光热转换效应。如图5(A)所示,静脉注射TBTP-Au-cRGD NPs后,TBTP-Au-cRGD NPs组随着照射时间的延长,肿瘤部位温度有明显升高,从37℃升高到56℃,而“盐水+激光”组的温度变化可以忽略不计。这一结果表明,TBTP-Au-cRGD NPs可以有效地在肿瘤部位积累,并产生足够的肿瘤热疗所需的热量。随后,通过监测肿瘤体积 20天,观察TBTP-Au-cRGD NPs的体内抗肿瘤作用。如图5(B)所示,“IR780+ 激光”组、“TBTP-cRGD NPs+激光”组和“TBTP-Au-cRGD NPs”组与对照组、“生理盐水+激光”组相比,肿瘤生长速度受到部分抑制,且抑制程度几乎相同,表明单一治疗方式的抗癌效率相当。与之形成鲜明对比的是,“TBTP-Au-cRGDNPs+激光”组的肿瘤在5天内被完全消除,这表明TBTP-Au-cRGD NPs在化疗和PTT的协同作用下具有最强大的抗癌能力。图5(C)显示治疗20天后不同组小鼠的肿瘤重量。H&E染色及TUNEL结果显示,“TBTP-Au-cRGD NPs+ 激光”组肿瘤部位坏死区域较其他6组更多(图5(D)),显示出更有效的抑癌能力。说明TBTP-Au-cRGD NPs具有良好的生物相容性和生物安全性。结合 TBTP-Au-cRGD优异的NIR-II显像、肿瘤靶向和肿瘤抑制能力以及良好的生物安全性,可以认为TBTP-Au作为强大的显像剂和化疗PTT剂在临床治疗中具有相当大的应用潜力。**p<0.01,***p<0.001.
TBTP-Au-cRGD NPs的免疫效应。评估通过激活免疫系统诱导全身抗肿瘤免疫,控制转移性肿瘤,防止肿瘤复发为了研究TBTP-Au-cRGD NPs诱导的潜在免疫应答,进行不同组肿瘤组织切片中CD8+T细胞的百分比(图6(A))。 CLSM图像显示,“TBTP-Au-cRGD+激光”组CD8+T细胞百分率最高,其次是“TBTP-cRGD NPs+激光”、“IR780+激光”和“TBTP-Au-cRGD”组。在相同的放大倍数下,用红色荧光计数不同组CD8+T细胞的密度。“TBTP-Au-cRGD NPs +激光”组肿瘤组织中CD8+-T细胞密度较对照组高15倍。相比之下,“TBTP-Au-cRGD NPs”组、“TBTP-cRGD NPs+激光”组和“IR780+激光”组计算的CD8+T细胞密度分别比对照组高3.48倍、4.52倍和4.38倍(图6(B))。因此,“TBTP-Au-cRGD NPs+激光”组CD8+T细胞密度较单一处理组“TBTP-cRGD NPs+激光”组和“TBTP-Au-cRGD NPs”组分别提高3.82倍和4.46倍。这些结果表明,TBTP-Au-cRGD诱导的化疗和PTT联合治疗能触发小鼠的免疫应答,巩固了TBTP-Au-cRGD的高效抗癌作用。此外,钙网蛋白(CRT)从内质网(ER) 转位到细胞膜表面是专业抗原提呈细胞识别癌细胞的重要信号,进而促进树突状细胞成熟,并协助肿瘤相关抗原提呈到淋巴结中的T细胞,最终启动抗肿瘤免疫反应。通过免疫荧光染色检测了TBTP-Au-cRGD联合激光照射后4T1细胞中CRT的表达。钙网蛋白(CRT)染色如图6(C)所示,CLSM荧光图像显示,“TBTP-Au-cRGD+激光”组的CRT标记的绿色荧光最强,定量检测CRT强度显示(图6(D)),CRT表达比对照组高15倍。相比之下,“TBTP-cRGD NPs+laser”和“TBTP-Au-cRGD”组与对照组相比,CRT表达仅中度增加4.52倍和3.48倍。结果表明,TBTP-Au-cRGD NPs协同化疗和PTT可显著触发4T1细胞表面CRT 表达,进而诱导全身免疫反应,使TBTP-Au-cRGD NPs具有持久的抗肿瘤免疫作用。**P<0.01,***P<0.001
结论:通过对NIR-II区分子中引入一个简单的五氟苯基金(I)单元,可以有效地提高吸光度,加强NIR-II荧光成像,并协同AIEgen的多模态抗癌治疗。体外实验结果表明,(I)金的引入增强了吸收能力,使AIE配体TBTP具有明显更高的光热转换效率,更优越的NIR-II成像能力和提高细胞内ROS水平。因此,所制备的TBTP-Au-cRGD NPs能够在体内特异靶向肿瘤NIR-II成像,并协同化疗和PTT进行高效的肿瘤消融,从而触发CRT表达,诱导全身免疫反应,达到持久的抗肿瘤免疫效果。

Claims (10)
1.一种近红外二区AIE化合物的制备方法,其特征在于,包括以下步骤:
合成化合物TBTP;
将所述TBTP与五氟苯基(I)金单元在二氯甲烷中反应获得TBTP-Au。
2.根据权利要求1所述的制备方法,其特征在于,合成化合物TBTP的所述步骤包括:
将4,8-双(5-溴-3-(己基)-2-噻吩基)-2λ4δ2-苯并[1,2-c:4,5-c']双[1,2,5]噻唑)和4-甲氧基-N-(4-甲氧基苯基)-N-(4-(4,4,5,5-四甲基-1,3,2-硼酸-2-苯基)苯胺在THF和水的混合溶液加入Pd(PPh3)4和K2CO3,在氮气保护下,将混合物在预设温度下搅拌,然后加入水使反应猝灭,并用CH2Cl2萃取,在无水Na2SO4上干燥并过滤,通过旋转蒸发去除溶剂;通过柱层析法纯化,得到TBTP。
3.根据权利要求2所述的制备方法,其特征在于,THF和水的用量比(v/v)为3:1~8:1;搅拌时间为4h~12h;CH2Cl2的用量为30mL~60mL;柱层析使用的CH2Cl2和乙烷的比例为2:1~1:2;预设温度为60℃~100℃,搅拌时间为4h~24h;K2CO3溶液的浓度为1.0mg/mL~10.0mg/mL,用量为2mL~40mL。
4.一种近红外二区AIE化合物,其特征在于,所述近红外二区AIE化合物由根据权利要求1-3中任一项所述的近红外二区AIE化合物的制备方法制备。
5.一种近红外二区AIE化合物的制备方法,其特征在于,包括以下步骤:
将根据权利要求1-3中任一项所述的制备方法制备的TBTP-Au溶于适量THF中,将DSPE-mPEG溶于THF中,将两组溶液充分混合并使用探针超声波器处理以形成TBTP-Au NP;
在氮气保护下,将TBTP-Au NP在通风柜中蒸发后加入c-RGD肽,搅拌透析后得到TBTP-Au-cRGD NP。
6.一种近红外二区AIE化合物,其特征在于,所述近红外二区AIE化合物由根据权利要求5所述的近红外二区AIE化合物的制备方法制备。
7.根据权利要求6所述的AIE化合物,其特征在于,所述AIE化合物的近红外吸收峰位于600nm~1000nm,发射峰为于1000nm~1350nm。
8.根据权利要求6所述的AIE化合物,其特征在于,所述AIE化合物的的平均粒径为为96nm。
9.根据权利要求6所述的近红外二区AIE化合物在特异性NIR II肿瘤成像中的非诊断或治疗的应用。
10.根据权利要求6所述的近红外二区AIE化合物在协同化疗和光热疗法进行肿瘤消融中的非诊断或治疗的应用。
