Abstract
本发明提供一种消食片及其制备方法,上述消食片包括以下原料及重量份:沙芥菜3‑6份,木香1‑3份,山楂1‑3份,麦芽1‑3份;上述消食片的片剂的规格为0.2‑0.5g/片。上述制备方法包括:称取各原料;再将各原料分别研磨成细粉,过筛后,统一放进混合机里混合均匀,得到原料粉末;再向原料粉末中加入植物淀粉,混合后进行压片即得。本发明消食片具有消食化滞、健脾和胃的功效,兼以补脾健胃、润肠通便、止痛解毒,能调节胃动力,促进胃部排空,避免对脾胃的负面损伤;其制备方法有效提升原料利用率和成品疗效,提高人体生物利用度,减少生产损耗,节约生产成本。

技术领域
本发明涉及保健食品技术领域,尤其涉及一种消食片及其制备方法。
背景技术
随着社会经济的不断提高,人们的生活水平也在不断提高,人们工作越来越忙,饮食品种的种类也变的越来越丰富,而胃肠功能障碍性疾病的发生呈上升并有年轻化的趋势。胃肠功能障碍性疾病的主要临床表现为消化不良,肠胃功能变弱,其症状表现为餐后饱胀感、腹部胀气、嗳气、早饱厌食、反酸恶心、胃腹胀痛、腹泻、便秘、烧灼感和泛酸等。
健胃消食片是中国药典近几版新收载的品种,具有健胃消食的功效。临床上用于脾胃虚弱所致的食积,症见不思饮食、食欲不振、嗳腐酸臭、脘腹胀满;消化不良见上述证候者。口服,可以咀嚼。而市面上有很多用于治疗脾胃虚弱所致的食积,例如健胃消食片、大山楂丸等。目前现有的健胃消食品,经过多年的临床应用和观察后发现:虽然能解决消化不良、脘腹胀闷、食欲不振、食积不化等问题,但是配方较为复杂,入口药味重且酸涩,令人服食口感不佳,并且不具备有解决肉食积滞、食物中毒、止痛、解毒的功效。另外还存在功效单一,有使用禁忌的问题,如:脾虚之人不宜使用大山楂丸,易加重脾胃损伤,且大山楂丸适用于食积在肠胃停滞时间不太长,还没出现嗳气、吞酸、口臭等症状者,若食积日久,则不宜使用。可见,目前的消食片等消食产品并不能满足市场上不同人群的需求。
经过多年的临床应用和观察,需要研发一种具有促进消化功能的消食片,将其作为日常消食服用的保健产品。此外还对消食片的制备方法进行改进,以获得更好的口感并减少不良反应,提高患者的顺应性。
发明内容
第一方面,本发明提供一种消食片,用以解决现有消食片不能消除肉食积滞、功效单一、易加重脾胃损伤的问题。该消食片具有消食化滞、健脾和胃的功效,兼以补脾健胃、润肠通便、止痛解毒,能调节胃动力,促进胃部排空,疗效和顺应性好,人体生物利用度高,能避免对脾胃的负面损伤,安全可靠,无不良反应。
具体地,本发明提供一种消食片,包括以下原料及重量份:沙芥菜3-6份,木香1-3份,山楂1-3份,麦芽1-3份;上述消食片的片剂的规格为0.2-0.5g/片。
进一步设置为,上述消食片的原料及重量份如下:沙芥菜4-6份,木香1-2份,山楂2-3份,麦芽1-2份。
进一步优选为,上述消食片的原料及重量份如下:沙芥菜5份,山楂2份,木香1.5份,麦芽1.5份。
进一步设置为,上述消食片中还包括植物淀粉,上述植物淀粉在片剂中含量为0.05-0.08g/g。
需要说明的是,上述消食片的用量如下:成人每日10-15片,儿童每日4-8片。
需要说明的是,上述消食片能用于制备消食药物和/或消食保健品。
第二方面,本发明提供一种消食片的制备方法,用以解决传统提取、浓缩等工艺的高温高热条件对有效成分的破坏和损伤的问题,有效提升了原料利用率和成品疗效,提高了人体生物利用度,减少了生产损耗,节约了生产成本。
具体地,本发明提供一种消食片的制备方法,包括:
S1.按照以下重量份称取各原料:沙芥菜3-6份,木香1-3份,山楂1-3份,麦芽1-3份;
S2.将上述各原料分别研磨成细粉,过70-100目的筛后,统一放进混合机里混合均匀,得到原料粉末;
S3.向上述原料粉末中加入植物淀粉,混合后进行压片,即得消食片;
上述消食片中,原料粉末含量为0.92-0.95g/g,植物淀粉含量为0.05-0.08g/g。
进一步设置为,压片的成型工艺为湿法制粒压片、干法制粒压片或全粉末压片。
进一步设置为,湿法制粒压片步骤如下:将原料粉末和植物淀粉混合,加入水湿混后,制粒,压片,再干燥至含水量为4-6%,即得消食片。
进一步设置为,干法制粒压片步骤如下:将原料粉末和植物淀粉混合,于超微粉碎机中混合15-30min,制粒,压片,即得消食片。
进一步设置为,全粉末压片步骤如下:将原料粉末和植物淀粉混合,于超微粉碎机中混合15-30min,直接全粉末压片,即得消食片。
需要说明的是,步骤S2.中所得原料粉末也能制成其他剂型,如胶囊、颗粒剂、丸剂、冲剂、口服液体剂等,相对而言,片剂制作、携带和服用最为简单方便,故制成片剂为佳。
沙芥,十字花科植物Pugionium cornutum Gaertn,以全草、根入药。别名沙萝卜、沙白菜、沙芥菜、山萝沙卜。味辛,性温。归心、肺、胃经。行气止痛,消食解毒。用于治胃肠胀气,开胃通肠,消化不良,止咳,清肺热,食物中毒。《内蒙古中草药》载:“行气,止痛,消食,解毒。治消化不良,胸胁胀满,食物中毒。”
木香,菊科植物木香Aucklandia lappa Decne.的干燥根。味辛、苦,性温。归脾、胃、大肠、三焦、胆经。行气止痛,健脾消食。用于胸脘胀痛,泻痢后重,食积不消,不思饮食。
山楂,蔷薇科植物山里红Crataegus pinnatifida Bge.var.major N.E.Br.或山楂Crataegus pinnatifida Bunge的干燥成熟果实。酸、甘,微温。归脾、胃、肝经。功效为消食健胃,行气散瘀,化浊降脂。临床上用于肉食积滞,胃脘胀满,泻痢腹痛,瘀血经闭,产后瘀阻,心腹刺痛,胸痹心痛,疝气疼痛,高脂血症。现代药理研究所含脂肪酸可促进脂肪消化,增加胃消化酶的分泌,并对胃肠动力有一定调整作用。
麦芽,禾本科植物大麦Hordeum Vulgare L.的成熟果实经发芽干燥的炮制加工品。甘,平。归脾、胃经。功能为行气消食,健脾开胃,回乳消胀。临床用于食积不消,治疗米面薯芋类饮食积滞,脘腹胀痛,脾虚食少,乳汁郁积,乳房胀痛等。现代药理研究麦芽可以促进胃酸及胃蛋白酶的分泌。
沙芥长于消一切饮食积滞,行气止痛,消食解毒,开胃通肠;山楂酸温消肉食之积,化瘀行滞,化浊降脂;麦芽消薯蓣米面食积,消食健脾开胃;木香健脾养胃,消补兼施。诸料配伍合用,消食化滞,健脾和胃,治疗或预防消化不良,使积滞得以运化,胃气得以益养,则诸症自除。所制消食片于蒙医中又称“星格格其”,临床实验数据优异,排异性小,治疗效果理想,能促进胃功能的恢复,降低用药后病情复发率,实用价值较高。
为了丰富口味种类,进一步的改进,本发明的消食片在制备过程中,根据需要向其中添加其他类型的矫味剂,娇味剂的添加量为片剂重量的0-10%。上述娇味剂包括但不限于甜味剂、芳香剂等,其实例包括但不限于蔗糖、糖浆、甘油、山梨醇、甘露醇、甜菊苷、植物芳香性挥发油和果味香精等。
本发明提供的消食片及其制备方法的有益效果如下:
1)该消食片具有消食化滞、健脾和胃的功效,兼以补脾健胃、润肠通便、止痛解毒,能调节胃动力,促进胃部排空,改善肠胃消化吸收功能,治疗或预防饮食积滞、消化不良、脘腹胀闷等症状,且能避免对脾胃的负面损伤,安全可靠,无不良反应;
2)该制备方法将原料直接制成粉末再进行压片,工艺简单易行,能稳定产品质量,避免传统提取、浓缩等工艺的高温高热条件对有效成分的破坏和损伤,有效提升了原料利用率和成品疗效,提高了人体生物利用度,减少了生产损耗,节约了生产成本;
3)该制备方法制得的消食片口感细腻滑润,安全无副作用,疗效和顺应性好,人体生物利用度高;产品能明显改善因肉食积滞导致的食欲不振和消化不良症状,还具有明显的增强免疫功能作用,更有利于消化不良的体弱患者,还对饱食、受寒等引起的消化不良具有良好的功效。

实施例1:
一种消食片的制备方法,包括以下步骤:
1)将新鲜沙芥菜炮制晾干后,按照以下重量份称取各原料:沙芥菜6份,木香2份,山楂2.5份,麦芽2.5份;
2)将上述各原料分别研磨成细粉,过100目的筛后,统一放进混合机里混合均匀,得到原料粉末;
3)向混合均匀的原料粉末中加入植物淀粉,原料粉末的添加量为0.92g/g,植物淀粉的添加量为0.08g/g,混合后进行压片,压制成0.3g/片的规格,即得消食片;
上述压片采用湿法制粒压片:将原料粉末和植物淀粉混合,加入适量水湿混,制粒,压片,干燥至含水量为6%,即得消食片。
本实施例所得消食片,其片剂表面光泽度差,咀嚼有较粗糙的口感。
实施例2:
一种消食片的制备方法,包括以下步骤:
1)将新鲜沙芥菜炮制晾干后,按照以下重量份称取各原料:沙芥菜3份,木香1.5份,山楂2.5份,麦芽2份;
2)将上述各原料分别研磨成细粉,过90目的筛后,统一放进混合机里混合均匀,得到原料粉末;
3)向混合均匀的原料粉末中加入植物淀粉,原料粉末的添加量为0.95g/g,植物淀粉的添加量为0.05g/g,混合后进行压片,压制成0.4g/片的规格,即得消食片;
上述压片采用干法制粒压片:将原料粉末和植物淀粉混合,于超微粉碎机中混合30min,制粒,压片,即得消食片。
本实施例所得消食片,其片剂片面均匀,咀嚼无粗糙感,细腻滑润。
实施例3:
一种消食片的制备方法,包括以下步骤:
1)将新鲜沙芥菜炮制晾干后,按照以下重量份称取各原料:沙芥菜4.5份,木香3份,山楂1.5份,麦芽2份;
2)将上述各原料分别研磨成细粉,过70目的筛后,统一放进混合机里混合均匀,得到原料粉末;
3)向混合均匀的原料粉末中加入植物淀粉,原料粉末的添加量为0.94g/g,植物淀粉的添加量为0.06g/g,混合后进行压片,压制成0.5g/片的规格,即得消食片;
上述压片采用全粉末压片:将原料粉末和植物淀粉混合,于超微粉碎机中混合20min,直接全粉末压片,即得消食片。
本实施例所得消食片,其片剂片面均匀,咀嚼时细腻滑润。
综合实施例1-3对比发现,干法制粒压片、全粉末压片均能获得咀嚼无粗糙感、口感较舒适细腻的消食片,消除了湿法制粒造成的粗糙感、砂砾感,更具有实用价值和推广价值。
实施例4:
一种消食片的制备方法,包括以下步骤:
1)将新鲜沙芥菜炮制晾干后,按照以下重量份称取各原料:沙芥菜5份,山楂2份,木香1.5份,麦芽1.5份;
2)将上述各原料分别研磨成细粉,过80目的筛后,统一放进混合机里混合均匀,得到原料粉末;
3)向混合均匀的原料粉末中加入植物淀粉,原料粉末的添加量为0.95g/g,植物淀粉的添加量为0.05g/g,混合后进行压片,压制成0.2g/片的规格,即得消食片;
上述压片采用全粉末压片:将原料粉末和植物淀粉混合,于超微粉碎机中混合30min,直接全粉末压片,即得消食片。
实施例5:
一种消食片的制备方法,与实施例4的不同之处仅在于:
步骤3)中,向原料粉末和植物淀粉中还添加了重量占比为0.5%的娇味剂甜菊苷,混合后进行全粉末压片,压制成0.2g/片的规格;其他步骤和参数与实施例4中一致,制得消食片。
实验例1:
毒性试验
1)蓄积毒性试验
实验动物:选用30只健康小白鼠,雌雄各半,体重20-25g,进行蓄积毒性试验。
实验药物:将本发明实施例4制得的消食片研磨成粉末,临用前用蒸馏水配制成不同剂量的溶液备用。
实验方法:将小鼠随机分为两组,每组15只,即对照组和给药组,实验前禁食12h。剂量设计每7d为一期,每期剂量分别为0.10LD50、0.20LD50、0.25LD50、0.30LD50、0.40LD50,此样品的雌性、雄性小鼠的LD50均为6785mg/(kg·bw)。给药量按0.5mL/只经口灌胃。每组设有3个平行。
实验结果:与对照组比较,给药后小鼠未见明显差异,连续给药观察35天后,未见任何明显毒性反应,动物无死亡情况发生。因此本发明实施例制得的消食片为实际低毒,临床用药安全。
2)急性毒性试验
实验动物:选用昆明种小白鼠40只,雌雄各半,体重15-20g,进行急性毒性试验。
实验药物:将本发明实施例4制得的消食片研磨成粉末,临用前用蒸馏水配成1.6g/mL(生药)的溶液备用。
实验方法:将小鼠随机分为两组,每组20只,即对照组和给药组,实验前禁食12h。给药组小鼠按0.8mL/只灌胃,对照组给予等量生理盐水,一天给药3次,给药间隔时间8h,给药后连续观察14d,并记录小鼠的行为表现、毒性反应及死亡情况等,试验结束时称取体重。每组设有3个平行。
实验结果:与对照组比较,给药后小鼠未见明显差异,实验连续观察14d,未见任何明显毒性反应,小鼠皮肤、粘膜、进食、呼吸、自主活动、行为表现未见异常,小鼠全身状况、饮食、饮水、体重增长均正常。因此本发明实施例制得的消食片的急性毒性低,临床用药安全。
实验例2:
药效学实验
实验动物:选用昆明种小白鼠,雌雄各半,体重15-20g。
实验药物:本发明实施例4制得的消食片。
1)对小鼠胃排空运动的影响
实验方法:取昆明种小白鼠40只,随机分为4组,雌雄各半。设有实验组(消食片50mg/kg)、阳性对照组(多潘立酮15mg/kg)、阴性对照组(植物淀粉50mg/kg)、空白对照组(生理盐水0.1mL/10g)的剂量给药,每日一次,连续7d。末次给药前禁食不禁水12h。末次给药30min后用0.1%甲基橙以0.2mL/只的剂量灌胃,20min后处死,取胃于试管中,加入5mL蒸馏水,剪开胃并将内容物充分洗入蒸馏水中,用5%NaHCO3调pH至6.0-6.5,2000rpm离心10min后,取上清液于波长420nm比色,用蒸馏水调零,所测吸光度值为“胃中甲基橙光密度”。以0.2mL 0.1%甲基橙溶液加5mL蒸馏水测吸光度值作为“基数甲基橙光密度”。按下式计算甲基橙胃残留率。甲基橙残留率(%)=胃中甲基橙光密度/基数甲基橙光密度×100%。每组设有3个平行。结果如表1所示。

胃排空延迟是胃肠运动功能障碍性疾病的主要病理生理基础,增强胃肠动力是治疗的主要措施之一。结果显示,与空白对照组比,实验组和阳性对照组能明显促进小鼠胃排空,同时消食片组的功效还显著强于多潘立酮组。说明,本发明消食片能降低小鼠胃中甲基橙残留率,通过促进小鼠胃排空,起到增加胃肠动力、健胃消食的作用。可见,本发明实施例制得的消食片中各原料组份发挥协同作用,能达到调节胃动力、促进胃部排空、改善肠胃消化吸收功能的作用,有利于改善因饮食(尤其是肉食)积滞导致的食欲不振和消化不良症状。
2)对小鼠免疫功能的影响
实验方法:取昆明种小白鼠40只,随机分为4组,雌雄各半。设有实验组(消食片60mg/kg)、阳性对照组(盐酸左旋咪唑10mg/kg)、阴性对照组(植物淀粉60mg/kg)、空白对照组(生理盐水0.1mL/10g)的剂量给药,每日1次,连续5d。末次给药60min后,每只小鼠腹腔注射5%鸡红细胞0.5mL,30min后处死,腹腔注射生理盐水2mL后按摩小鼠腹部,使腹腔液混匀。然后剪开腹膜,吸取腹腔渗液均匀涂于载玻片上,放入垫有湿纱布的搪瓷盘内,移至37℃恒温箱内孵育30min后,用水洗去未贴片的细胞及杂质,晾干。然后用1:3的醋酸-甲醇溶液固定,晾干后用1:10的Giemsa染色液原液与pH值6.8的磷酸缓冲液染色20min,在油镜下每片玻片计数200个巨噬细胞,并计数巨噬细胞吞噬鸡红细胞数,计算吞噬百分率及吞噬指数。吞噬百分率(%)=吞噬鸡红细胞的巨噬细胞数/200个巨噬细胞×100%。吞噬指数=被吞噬鸡红细胞数/200个巨噬细胞。每组设有3个平行。结果如表2所示。

结果显示,与空白对照组比,实验组和阳性对照组能明显增强小鼠腹腔巨噬细胞的吞噬能力,对小鼠免疫功能有显著的保护和增强作用。可见,本发明实施例制得的消食片中各原料组份发挥协同作用,能明显增强免疫功能,更能适用于消化不良的体弱患者,还对饱食、受寒等引起的消化不良具有良好的功效。
3)对小鼠体重、采食量的影响
实验方法:将30只小鼠随机分成3组,每组雌雄各半,分别设为A、B、C组,各组单独饲喂,自由采食,试验时间为30天,观察采食频率和活动情况,记录每组的日采食量,并于实验结束12h后检测血糖含量,试验结束时测量并计算体重增重、日均采食量。A、B、C三组的饲料中,A组添加消食片,B组添加植物淀粉,C组不添加其他设为空白对照组,AB组的添加量以质量计,为饲料质量的1%。日采食量=(日投食量-剩余食量)/小白鼠数量,日均采食量为30天的日采食量平均值。每组设有3个平行。结果如下表3所示。

结果显示,A组小鼠的进食量明显增加,采食频率也有提升,说明消食片加快了小白鼠的消化速度;从活动情况看,小白鼠的精神状态和体力都比较好,可能是由于脾胃功能和消化功能的增强,使得小白鼠精神饱满,精力充足。最后,A组实验结束后血糖测量与空白组无明显差异,说明本发明的消食片对小鼠的健康没有负面影响。
实验例3:
临床病例研究
采用本发明实施例4的片剂为试样,采用多中心对照研究,观察本发明的片剂治疗脾胃虚弱、消化不良的临床疗效及安全性,共观察235例,病例均来源于乌审旗蒙医综合医院门诊。
1)对象和方法:全部病人年龄3-65岁,性别、职业不限。
A、中医诊断标准:自觉胃脘部痞塞、胀满或腹痛不适;起病缓慢,时轻时重,反复发作2月以上;发病常与饮食、情志、起居、寒温等诱因有关。凡具备前两2项,参考第三项及其他症状、舌苔、脉象即可诊断。
B、中医证候辨证标准:脾胃虚弱证:食后脘腹胀满,食少纳呆、大便溏,嗳气、面色无华、倦怠乏力、恶心呕吐,舌质淡或有齿印,苔薄白,脉沉细。
C、纳入病例标准:诊断为食欲降低,消化不良,而又符合西医、中医辨证脾胃虚弱证者均可纳入观察对象;年龄在3-65岁。
D、排除病例标准:①未能排除其他系统疾病所致厌食者;②合并消化性溃疡、胃粘膜有重度异常增生,或病理诊断疑有恶变者;③心、肝、肾及其相关检查发现异常者及精神病史患者;④不符合中医脾胃虚弱证辨证标准者;不符合其他入选标准者。
2)治疗方法:口服给药,连续服药7d为1个疗程。观察时间为1个疗程。成人每日12片,儿童每日5片。观察期间,除采用实验用药外,不用其他脱敏、消炎等任何药物。
3)疗效判断标准:
痊愈:食欲和食量恢复到正常水平,临床证候“计分和”减少90%以上,异常理化指标恢复正常。
显效:食欲明显恢复,食量恢复到正常水平的四分之三,临床证候“计分和”减少60-90%,异常理化指标明显改善。
有效:食欲有改善,食量有所恢复,临床证候“计分和”减少30-60%,异常理化指标有所改善。
无效:不符合以上标准者。
4)结果:
表4 消食片的临床疗效数据
其中,病例中包括57例:因经常吃肉类食物导致,表现出胃部疼、胀满食少、食欲不振和消化不良的症状,服用本发明的消食片后,用药3-5d后,所有症状均消失,其后一个月内未有再复发。
5)结论:结果表明,本发明的消食片总有效率达到96%以上,是一种预防和治疗脾胃虚弱、消化不良的疗效显著、安全性高的产品。
最后应说明的是,以上各实施例仅用以说明本发明的技术方案,而非对其限制;尽管参照前述各实施例对本发明进行了详细的说明,本领域的普通技术人员应当理解;其依然可以对前述各实施例所记载的技术方案进行修改,或者对其中部分或者全部技术特征进行等同替换;而这些修改或者替换,并不使相应技术方案的本质脱离本发明各实施例技术方案的范围。
Claims (10)
1.一种消食片,其特征在于:包括以下原料及重量份:沙芥菜3-6份,木香1-3份,山楂1-3份,麦芽1-3份;所述消食片的片剂的规格为0.2-0.5g/片。
2.根据权利要求1所述的消食片,其特征在于:所述原料及重量份如下:沙芥菜4-6份,木香1-2份,山楂2-3份,麦芽1-2份。
3.根据权利要求1所述的消食片,其特征在于:所述原料及重量份如下:沙芥菜5份,山楂2份,木香1.5份,麦芽1.5份。
4.根据权利要求1所述的消食片,其特征在于:所述消食片中还包括植物淀粉,上述植物淀粉在片剂中含量为0.05-0.08g/g。
5.根据权利要求1-4任一项所述的消食片,其特征在于:所述消食片的用量如下:成人每日10-15片,儿童每日4-8片。
6.根据权利要求1-3任一项所述的消食片,其特征在于:所述消食片能用于制备消食药物和/或消食保健品。
7.一种权利要求1-6任一项所述的消食片的制备方法,其特征在于,包括:
S1.按照以下重量份称取各原料:沙芥菜3-6份,木香1-3份,山楂1-3份,麦芽1-3份;
S2.将所述各原料分别研磨成细粉,过70-100目的筛后,统一放进混合机里混合均匀,得到原料粉末;
S3.向所述原料粉末中加入植物淀粉,混合后进行压片,即得消食片;
所述消食片中,原料粉末含量为0.92-0.95g/g,植物淀粉含量为0.05-0.08g/g。
8.根据权利要求7所述的制备方法,其特征在于:所述压片的成型工艺为湿法制粒压片、干法制粒压片或全粉末压片。
9.根据权利要求8所述的制备方法,其特征在于:所述湿法制粒压片步骤如下:将原料粉末和植物淀粉混合,加入水湿混后,制粒,压片,再干燥至含水量为4-6%,即得消食片。
10.根据权利要求8所述的制备方法,其特征在于:所述全粉末压片步骤如下:将原料粉末和植物淀粉混合,于超微粉碎机中混合15-30min,直接全粉末压片,即得消食片。
