Abstract
本发明提供了一种快速响应细微酸碱度变化的指示剂,包括酸碱指示剂、等电点为3.0‑10.0、分子量为10kDa‑200kDa的稳定性蛋白质和缓冲溶液。本发明的指示剂的颜色突变区间可以控制在0.1‑0.2之间,变色明显,可以使用含有该指示剂的固态基质对不同人体代谢物进行快速检测。

技术领域
本发明涉及生物化学技术领域,具体涉及快速响应细微酸碱度变化的颜色指示剂及其制备方法和应用。
背景技术
酸碱指示剂是一类在特定pH范围内,通过颜色变化反应酸碱度变化的有机、无机化合物及其组合。目前常用的pH指示剂有:麝香草酚蓝指示剂,变色范围为pH1.2-2.8(红→黄);溴酚蓝指示剂,变色范围为pH2.8-4.6(黄→蓝绿);甲基橙指示剂,变色范围pH3.2-4.4(红→黄);刚果红指示剂,变色范围为pH3.5-5.2(蓝→红);溴甲酚绿指示液,变色范围为pH3.6-5.2(黄→蓝);茜素磺酸钠指示剂,变色范围pH3.7-5.2(黄→紫);甲基红指示剂,变色范围为pH4.2-6.3(红→黄);石蕊指示剂,变色范围为pH4.2-8.0(红→蓝);溴甲酚紫指示剂,变色范围pH5.2-6.8(黄→紫);硝氮黄指示剂,变色范围为pH6.0-7.0(黄→蓝紫色);溴麝香草酚蓝指示剂,变色范围为pH6.0-7.6(黄→蓝);中性红指示剂,变色范围为pH6.8-8.0(红→黄);甲酚红指示剂,变色范围pH7.2-8.8(黄→红);姜黄素指示剂,变色范围为pH7.8-9.2(黄→红棕色);酚酞指示液,变色范围为pH8.3-10.0(无色→红);孔雀石绿指示剂,变色范围为pH11.0-13.5(绿→无色)等。
pH检测应用在各行各业,例如在医学领域,人体的血液pH范围在7.35-7.45的范围内,当pH低于7.35时会发生酸中毒,pH低于7.0会发生严重酸中毒昏迷致死;血液的pH高于7.45会发生碱中毒,pH高于7.8就会发生严重碱中毒导致的手足抽搐致死的严重后果;在农业上,土壤的pH关系到农作物的生长,有的作物如芝麻、油菜、萝卜等可以生长在较大的pH范围内,有的却对土壤的pH反应非常敏感,如,茶树适宜在pH约为4.0~5.5的土壤中生长;在污水处理中,pH值也是一项重要指标。
人们也在不断的寻找新型指示剂组分,对指示剂的检测范围进行调整优化,多数指示剂最初来源于植物,例如石蕊指示剂、麝香草酚蓝指示剂、姜黄素指示剂等。同时对指示剂进行组合,扩大指示范围,突变范围变窄,例如一定比例的中性红乙醇溶液和百里酚蓝乙醇溶液混合,变色范围在pH7.0-7.4(玫瑰色→暗绿色);一定比例的溴甲酚绿指示剂-甲基橙指示剂混合,变色范围为pH3.5-4.3(黄色→蓝绿色)。然而细分领域的检测对于pH指示剂并不追求范围,而对具体的变色点更加关注,例如江苏耐雀生物工程技术有限公司(CN105784702A)利用姜黄素与紫草素按照一定比例混合,扩大了酸碱指示剂的指示范围,突变范围窄;四川大学(CN111896536A)公开发明了一种苯并噻唑类偶氮化合物酸碱指示剂,颜色的突变范围窄,颜色变化直观明显便于观察;太原跃鹏电子科技有限公司(CN108120796A)公开发明了一种检测铁路蓄电池CO3 2-时pH值突变点的测定计算方法。但以上发明中的配方是基于简单的指示剂组合和改变溶液的离子环境,而得到的近似于突变的显色,例如对于阴道分泌物检测的pH指示剂变色区间只能限制在4.5-5.0之间,范围较宽,不能有效指示变色区间,对于实际样本检测具有较大的限制。
发明内容
本发明要解决的技术问题是开发一种能够将变色范围区间缩小,快速响应细微酸碱度变化的指示剂。
基于上述问题,本发明提出的技术方案是一种快速响应细微酸碱度变化的指示剂,包括酸碱指示剂、等电点为3.0-10.0、分子量为10kDa-200kDa的蛋白质和缓冲溶液。
当蛋白质所处缓冲体系的pH≠pI时,蛋白质属于解离状态,并带有净电荷,利用缓冲溶液改变蛋白质所带电荷量,即利用缓冲溶液的pH值调节蛋白质的净电荷,进而与酸碱指示剂发生协同作用,调节其pH显色区间。理论上蛋白质所处的缓冲溶液pH偏离其pI越多时,所带净电荷越多,溶解度越大,反之净电荷越少溶解度越小,pH=pI时溶解度最小,但是偏离越大,电荷越多反而会影响离子平衡体系,使得酸碱指示剂偏离过大,变色范围难以控制,无法响应细微变化,因此所选蛋白质的等电点范围控制为3.0-10.0之间。蛋白质分子量过大或者过小都会影响到蛋白在缓冲溶液的稳定性,因此将分子量限定值10kDa-200kDa是为了保证其稳定性。
所述蛋白质为钙调蛋白、牛血清白蛋白、肌球蛋白、胶原蛋白或乳铁蛋白中的一种。优选为牛血清白蛋白,肌球蛋白、胶原蛋白、乳铁蛋白中的一种,最优选为肌球蛋白、乳铁蛋白、胶原蛋白的一种。
所述酸碱指示剂为溴甲酚绿、甲基橙、甲基红、硝氮黄、酚酞,溴甲酚绿-甲基橙、溴甲酚绿-甲基红、溴甲酚绿-硝氮黄、溴甲粉绿-酚酞、甲基橙-甲基红、甲基橙-硝氮黄、甲基橙-酚酞、甲基红-硝氮黄、甲基红-酚酞、硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红、溴甲酚绿-甲基橙-硝氮黄、溴甲酚绿-甲基橙-酚酞、溴甲酚绿-甲基红-硝氮黄、溴甲酚绿-甲基红-酚酞、甲基橙-甲基红-硝氮黄、甲基橙-甲基红-酚酞、甲基红-硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄、溴甲酚绿-甲基橙-甲基红-酚酞、溴甲酚绿-甲基红-硝氮黄-酚酞、甲基橙-甲基红-硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄-酚酞中的一种。优选为溴甲酚绿、硝氮黄、溴甲酚绿-甲基橙、溴甲酚绿-硝氮黄、甲基橙-甲基红、甲基红-硝氮黄、硝氮黄-酚酞、溴甲酚绿-甲基红-硝氮黄、溴甲酚绿-甲基红-酚酞、甲基橙-甲基红-酚酞、甲基红-硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄、溴甲酚绿-甲基红-硝氮黄-酚酞、甲基橙-甲基红-硝氮黄-酚酞或溴甲酚绿-甲基橙-甲基红-硝氮黄-酚酞中的一种,最优选为溴甲酚绿-硝氮黄、甲基橙-甲基红、硝氮黄-酚酞、溴甲酚绿-甲基红-硝氮黄、溴甲酚绿-甲基红-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄、甲基橙-甲基红-硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄-酚酞中的一种。
缓冲溶液包括柠檬酸-柠檬酸钠、柠檬酸-氢氧化钠、柠檬酸钠-盐酸、磷酸二氢钠-磷酸氢二钾、磷酸二氢钠-氢氧化钠、磷酸氢二钾-盐酸、磷酸氢二钾-磷酸、醋酸-醋酸钠、醋酸-醋酸钾。优选为柠檬酸-柠檬酸钠、柠檬酸-氢氧化钠、磷酸二氢钠-磷酸氢二钾、磷酸氢二钾-盐酸、磷酸氢二钾-磷酸、醋酸-醋酸钠、醋酸-醋酸钾。最优选为柠檬酸-柠檬酸钠、磷酸二氢钠-磷酸氢二钾、醋酸-醋酸钠中的一种。
一种制备权利要求1所述的快速响应细微酸碱度变化的指示剂的方法,其特征在于:包括以下步骤:
1)称取一定量蛋白质溶解于甘油中,制备质量分数为0.01%~0.05%的蛋白质母液;
2)称取一定量酸碱指示剂并使之溶解,制备质量分数为0.05%~0.5%的酸碱指示剂,溶剂可以为超纯水、乙醇或甲醇;
3)称取一定量缓冲溶液的各组分,制备0.1M的不同pH的缓冲溶液,pH范围为2.0-14.0;
4)将1)得到的蛋白质溶液与2)得到的酸碱指示剂以1:5~1:50的体积比混合;
5)将4)的混合液用3)得到的缓冲溶液定容。
将该指示剂附着于固体基质对人体代谢物,如唾液、血液、汗液、尿液、精液、羊水、乳汁、阴道黏液、宫颈黏液等进行检测,所述固体基质包括指示拭子、试纸、无尘布、纤维素敷料。
本发明的优点和有益效果:
本发明利用向常规指示剂中添加不同的等电点及分子量的蛋白质以及配套的缓冲溶液的组合,获得能够快速响应酸碱度变化的超敏感颜色指示剂,指示剂由于额外添加特殊的等电点和分子量的蛋白,使得指示剂的颜色突变区间可以控制在0.1-0.2之间,变色明显,可以使用含有该指示剂的固态基质对不同人体代谢物进行快速检测。

具体实施方式
下面结合具体实施例对本发明的具体实施方式作详细说明。
实施例1突变显色点为pH 6.2的超敏感指示剂的制备
配制0.015%乳铁蛋白溶液:称取1.5mg乳铁蛋白溶解并定容于10mL甘油中;配制0.2%硝氮黄指示剂:称取0.2g硝氮黄溶解并定容于100mL无水乙醇中;配制0.1mol/L柠檬酸溶液:取柠檬酸21.01g溶解并定容于1000mL纯水,配制0.1mol/L柠檬酸钠溶液:取柠檬酸钠29.41g溶解并定容于1000mL纯水;将19.0mL柠檬酸溶液和81.0mL柠檬酸钠溶液混合,制成pH6.0的SSC缓冲溶液;将上述100μL乳铁蛋白溶液和1.5mL硝氮黄溶液混合均匀,并用pH6.0的SSC溶液定容至10mL,制成突变显色点为pH6.2的超敏感指示剂。
如图1所示,将制成的突变点为pH 6.2的超敏感指示剂浸润滤纸或者无尘布块烘干后,滴加不同pH的检测溶液,当pH≥6.2时,指示滤纸和布块由黄色突变为绿色。
实施例2突变显色点为pH 4.4的超敏感指示剂的制备
配制0.025%肌球蛋白溶液:称取2.5mg肌球蛋白溶解并定容于100mL甘油中;配制0.1%溴甲酚绿-硝氮黄指示剂:同时称取0.02g溴甲酚绿和0.08g硝氮黄试剂,溶解并定容于100mL无水乙醇中;配制0.1mol/L柠檬酸溶液:取柠檬酸21.01g溶解并定容于1000mL纯水,配制0.1mol/L柠檬酸钠溶液:取柠檬酸钠29.41g溶解并定容于1000mL纯水;将65.5mL柠檬酸溶液和34.5mL柠檬酸钠溶液混合,制成pH 4.0的SSC缓冲溶液;将上述50μL肌球蛋白溶液和2mL溴甲酚绿-硝氮黄指示剂混合均匀,并用pH 4.0的SSC溶液定容至10mL,制成突变显色点为pH 4.4的超敏感指示剂。
实施例3突变显色点为pH 3.8的超敏感指示剂制备
配制0.02%牛血清白蛋白溶液:称取20mg乳铁蛋白溶解并定容于100mL甘油中;配制0.47%溴甲酚绿-甲基橙-甲基红指示剂:同时称取0.25g溴甲酚绿、0.1g甲基橙、0.12g甲基红试剂,溶解并定容于100mL无水乙醇中;配制0.1mol/L醋酸钠溶液:取醋酸钠8.2g溶解并定容于1000mL纯水;配制0.1mol/L醋酸溶液:2.86mL醋酸溶液用超纯水稀释至500mL;将15.0mL醋酸溶液和75.0mL醋酸钠溶液混合,制成pH 3.5的ABS缓冲溶液;将上述200μL牛血清白蛋白溶液和1.5mL溴甲酚绿-甲基橙-甲基红指示剂混合均匀,并用pH 3.5的ABS溶液定容至10mL,制成突变显色点为pH 3.8的超敏感指示剂。
如图2所示,将制成的突变点为pH 3.8的超敏感指示剂浸润滤纸或者无尘布块烘干后,滴加不同pH的检测溶液,当pH≥3.8时,指示滤纸和布块由黄色突变为绿色。
实施例4突变显色点为pH8.0的超敏感指示剂的制备
配制0.04%牛血清白蛋白溶液:称取4.0mg牛血清白蛋白溶解并定容于10mL甘油中;配制0.48%溴甲酚绿-甲基红-硝氮黄-酚酞指示剂:同时称取0.1g溴甲酚绿、0.12g甲基红、0.16g硝氮黄、0.1g酚酞溶解并定容于100mL甲醇中;配制0.1mol/L磷酸二氢钠溶液:取磷酸二氢钠12.0g溶解并定容于1000mL纯水;配制0.1mol/L磷酸氢二钾溶液:取磷酸氢二钾17.4g溶解并定容于1000mL纯水;将16.0mL磷酸二氢钠溶液和84.0mL磷酸氢二钾溶液混合,制成pH 7.5的PBS缓冲溶液;将上述30μL牛血清白蛋白溶液和1.0mL溴甲酚绿-甲基红-硝氮黄-酚酞混合均匀,并用pH 7.5的PBS溶液定容至10mL,制成突变显色点为pH 8.0的超敏感指示剂,即检测物质pH≥8.0时,溶液由灰色突变为蓝紫色。
实施例5pH突变显色的检测阴道粘液的超敏感指示拭子的制备
配制0.025%肌球蛋白溶液:称取2.5mg乳铁蛋白溶解并定容于10mL甘油中;配制0.26%溴甲酚绿-硝氮黄指示剂:同时称取0.06g溴甲酚绿和0.2g硝氮黄试剂,溶解并定容于100mL无水乙醇中;配制0.1mol/L柠檬酸溶液:取柠檬酸21.01g溶解并定容于1000mL纯水,配制0.1mol/L柠檬酸钠溶液:取柠檬酸钠29.41g溶解并定容于1000mL纯水;将65.5mL柠檬酸溶液和34.5mL柠檬酸钠溶液混合,制成pH 4.0的SSC缓冲溶液;将上述50μL肌球蛋白溶液和2mL溴甲酚绿-硝氮黄指示剂混合均匀,并用pH4.0的SSC溶液定容至10mL,得到突变显色点为pH 4.6的超敏感指示剂。将超敏感指示剂滴加在取样拭子上,使其完全浸湿之后置于真空干燥箱中,在30℃的条件下干燥1小时,制得超敏感检测阴道粘液的超敏感指示拭子。
阴道粘液的正产pH值为3.8~4.5,当pH值高于4.5时,是白带异常的表现之一。用超敏感检测阴道粘液的指示拭子沾取阴道分泌物样本,若拭子不变色为黄色,则说明分泌物pH正常;若拭子变色为绿色,则说明分泌物pH异常。如图3所示,将不同阴道粘液样本分别滴加在pH试纸和超敏感指示拭子上,根据pH试纸判断其实际pH值,其中pH≤4.6的拭子颜色为黄色,pH≥4.6的拭子为绿色。
实施例6pH突变显色的检测尿液用超敏感试纸的制备
配制0.015%乳铁蛋白溶液:称取1.5mg乳铁蛋白溶解并定容于10mL甘油中;配制0.2%硝氮黄-酚酞指示剂:称取0.12g硝氮黄指示剂和0.08g酚酞溶解并定容于100mL无水乙醇中;配制0.1mol/L柠檬酸溶液:取柠檬酸21.01g溶解并定容于1000mL纯水,配制0.1mol/L柠檬酸钠溶液:取柠檬酸钠29.41g溶解并定容于1000mL纯水;将19.0mL柠檬酸溶液和81.0mL柠檬酸钠溶液混合,制成pH6.0的SSC缓冲溶液;将上述280μL乳铁蛋白溶液和1.5mL硝氮黄-酚酞指示剂溶液混合均匀,并用pH6.0的SSC溶液定容至10mL,得到突变显色点为pH 6.5的超敏感指示剂。
若将超敏感指示剂滴加在滤纸上,使其完全浸湿之后置于真空干燥箱中在30℃的条件下干燥20分钟,制得检测尿液的超敏感试纸。
尿液的正常pH值为5.5~6.5,当pH值高于6.5时,是身体预警的表现之一。用超敏感尿液检测沾取尿液样本,若试纸不变色为黄色,则说明尿液pH正常;若试纸变色为蓝绿色,则说明尿液pH异常。图4所示,将不同尿液样本分别滴加在pH试纸和超敏感试纸上,根据pH试纸判断其实际pH值,其中pH≤6.5的拭子颜色为黄色,pH>6.5的拭子为蓝绿色。
实施例7 pH突变显色的检测精液的超敏感试纸的制备
配制0.04%牛血清白蛋白溶液:称取4.0mg牛血清白蛋白溶解并定容于10mL甘油中;配制0.48%溴甲酚绿-甲基红-硝氮黄-酚酞指示剂:同时称取0.1g溴甲酚绿、0.12g甲基红、0.16g硝氮黄、0.1g酚酞溶解并定容于100mL甲醇中;配制0.1mol/L磷酸二氢钠溶液:取磷酸二氢钠12.0g溶解并定容于1000mL纯水;配制0.1mol/L磷酸氢二钾溶液:取磷酸氢二钾17.4g溶解并定容于1000mL纯水;将16.0mL磷酸二氢钠溶液和84.0mL磷酸氢二钾溶液混合,制成pH 7.5的PBS缓冲溶液;将上述30μL牛血清白蛋白溶液和1.0mL溴甲酚绿-甲基红-硝氮黄-酚酞混合均匀,并用pH 7.5的PBS溶液定容至10mL,制成突变显色点为pH 8.0的超敏感指示剂。
将超敏感指示剂滴加在吸水试纸上,使其完全浸湿之后置于真空干燥箱中在30℃的条件下干燥20分钟,制得检测精液的超敏感试纸。
精液的正产pH值为7.2~7.8,当pH值高于8.0时,则可能出现炎症。用超敏感精液检测试纸沾取精液样本,若试纸不变色为灰色,则说明精液pH正常;若试纸变色为蓝紫色,则说明精液pH偏高。
实施例8皮肤表面pH指示用的超敏感创面酸性敷料的制备
将超敏感颜色指示剂与纤维素敷料混合,作为伤口愈合指示剂的使用,具体配方如下:
配制0.025%胶原蛋白溶液:称取2.5mg胶原蛋白溶解并定容于10mL甘油中;配制0.13%硝氮黄-酚酞指示剂:同时称取0.05g硝氮黄和0.08g酚酞试剂,溶解并定容于100mL无菌水中;配制0.1mol/L柠檬酸溶液:取柠檬酸21.01g溶解并定容于1000mL纯水;将27.5mL柠檬酸溶液和72.5mL柠檬酸钠溶液混合,制成pH 5.5的SSC缓冲溶液;将上述50μL胶原蛋白溶液和2mL硝氮黄-酚酞指示剂混合均匀,并用pH 5.5的SSC溶液定容至10mL,得到突变显色点为pH 6.5的超敏感指示剂。
将1mL超敏感指示剂与一片纤维素敷料混合,当其渗透均匀之后置于真空干燥箱中在25℃的条件下干燥10分钟,制得超敏感创面酸性敷料。
正常皮肤的pH值范围在4.0~6.0,慢性创面pH范围在6.5~8.5。当在病人的皮肤创面敷上此酸性敷料,敷料为浅蓝色;当伤口愈合后,敷料可显示为无色或者淡黄色。
以上对发明的几个实施例进行了详细说明,但所述内容仅为本发明的较佳实施例,不能被认为用于限定本发明的实施范围。凡依本发明申请范围所作的均等变化与改进等,均应仍归属于本发明的专利涵盖范围之内。

1.一种快速响应细微酸碱度变化的指示剂,包括酸碱指示剂,其特征在于:还包括等电点为3.0-10.0、分子量为10kDa-200kDa的稳定性蛋白质和缓冲溶液。
2.根据权利要求1所述的快速响应细微酸碱度变化的指示剂,其特征在于:所述蛋白质为钙调蛋白、牛血清白蛋白、肌球蛋白、胶原蛋白或乳铁蛋白中的一种。
3.根据权利要求2所述的快速响应细微酸碱度变化的指示剂,其特征在于:所述蛋白质优选为肌球蛋白、乳铁蛋白或胶原蛋白中的一种。
4.根据权利要求1所述的快速响应细微酸碱度变化的指示剂,其特征在于:所述酸碱指示剂为溴甲酚绿、甲基橙、甲基红、硝氮黄、酚酞,溴甲酚绿-甲基橙、溴甲酚绿-甲基红、溴甲酚绿-硝氮黄、溴甲粉绿-酚酞、甲基橙-甲基红、甲基橙-硝氮黄、甲基橙-酚酞、甲基红-硝氮黄、甲基红-酚酞、硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红、溴甲酚绿-甲基橙-硝氮黄、溴甲酚绿-甲基橙-酚酞、溴甲酚绿-甲基红-硝氮黄、溴甲酚绿-甲基红-酚酞、甲基橙-甲基红-硝氮黄、甲基橙-甲基红-酚酞、甲基红-硝氮黄-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄、溴甲酚绿-甲基橙-甲基红-酚酞、溴甲酚绿-甲基红-硝氮黄-酚酞、甲基橙-甲基红-硝氮黄-酚酞或溴甲酚绿-甲基橙-甲基红-硝氮黄-酚酞中的一种。
5.根据权利要求4所述的快速响应细微酸碱度变化的指示剂,其特征在于:酸碱指示剂优选为溴甲酚绿-硝氮黄、甲基橙-甲基红、硝氮黄-酚酞、溴甲酚绿-甲基红-硝氮黄、溴甲酚绿-甲基红-酚酞、溴甲酚绿-甲基橙-甲基红-硝氮黄、甲基橙-甲基红-硝氮黄-酚酞或溴甲酚绿-甲基橙-甲基红-硝氮黄-酚酞中的一种。
6.根据权利要求1所述的快速响应细微酸碱度变化的指示剂,其特征在于:缓冲溶液包括柠檬酸-柠檬酸钠、柠檬酸-氢氧化钠、柠檬酸钠-盐酸、磷酸二氢钠-磷酸氢二钾、磷酸二氢钠-氢氧化钠、磷酸氢二钾-盐酸、磷酸氢二钾-磷酸或醋酸-醋酸钠或醋酸-醋酸钾中的一种。
7.根据权利要求6所述的快速响应细微酸碱度变化的指示剂,其特征在于:缓冲溶液优选柠檬酸-柠檬酸钠、磷酸二氢钠-磷酸氢二钾或醋酸-醋酸钠中的一种。
8.一种制备权利要求1~7任一所述的快速响应细微酸碱度变化的指示剂的方法,其特征在于:包括以下步骤:
1)称取一定量蛋白质溶解于甘油中,制备质量分数为0.01%~0.05%的蛋白质母液;
2)称取一定量酸碱指示剂并使之溶解,制备质量分数为0.05%~0.5%的酸碱指示剂;
3)称取一定量缓冲溶液的各组分,制备0.1M的不同pH的缓冲溶液,pH为2.0~14.0;
4)将1)得到的蛋白质溶液与2)得到的酸碱指示剂以1:5~1:50的体积比混合;
5)将4)的混合液用3)得到的缓冲溶液定容。
9.权利要求1所述的一种快速响应细微酸碱度变化的指示剂在检测人体代谢物中的应用。
10.根据权利要求9所述的快速响应细微酸碱度变化的指示剂在检测人体代谢物中的应用,其特征在于:将指示剂附着于固体基质对人体代谢物进行检测,固体基质包括指示拭子、试纸、纤维素敷料。
