背景技术
医疗诊断过程中内镜的使用是一种侵入性诊疗方法,近年来,内镜已从原来的单纯诊断拓展至外科治疗,内镜的应用越来越广泛,如果内镜清洗消毒不彻底,将导致医源性感染的发生。有报道称内镜导致的感染率为0.8%。日本胃镜检查过程中,幽门螺杆菌感染率高达1.1%,且丙肝病毒、梅毒、结核菌、人类免疫缺陷病毒等也可经内镜感染。
内镜消毒是指消灭内镜上的病原微生物的方法,但是由于内镜结构复杂、材料特殊,许多部位不耐高温、高压或易腐蚀,给临床使用后的消毒带来了挑战。目前针对内镜高水平消毒所选用的消毒剂一般为戊二醛、邻苯二甲醛和过氧乙酸。但是由于戊二醛已经产生分枝杆菌抗力株,龟分枝杆菌脓肿亚种菌株对戊二醛产生耐药性。另外,在使用过程中,很多因素会造成药效浓度降低,需每天检测有效浓度,使用比较繁琐,且戊二醛对人体有毒性,伴随有刺激气味, 可引起局部皮肤黏膜过敏,要求保持室内通风或者穿戴防护设备,目前已经慢慢淘汰。过氧乙酸虽然消毒效果较好,但是过氧乙酸稳定性差的问题,器械腐蚀性问题,导致内镜消毒领域没有获得广泛应用。
OPA 是在戊二醛之后出现的一种高水平醛类消毒剂, 其杀菌机理与戊二醛类似,都是通过与氨基酸发生交联作用,使蛋白质变性,达到杀死细菌的目的。不同之处是OPA具有更好的脂溶性,更易穿透含脂质较多的分枝杆菌细胞膜,进入菌体内部起作用,因此对分枝杆菌的杀灭作用强于戊二醛。1999年, Walsh 等人率先提出将OPA代替戊二醛应用于高水平消毒,商业化的OPA产品由强生公司推出并通过FDA认证,如今已在美国、欧洲和日本获得广泛应用,常规使用浓度为 0.5% ~ 0.6%。
目前常见的复方邻苯二甲醛消毒剂都是基于季铵盐复配邻苯二甲醛而成,利用季铵盐的杀菌原理,改变细菌膜的通透性,继而发生溶胞作用,破坏细胞结构,引起细胞的溶解和死亡。邻苯二甲醛和季铵盐的复配过程中,季铵盐尤其是双链季铵盐由于其丰富的泡沫,加上其需要邻苯二甲醛的含量依然不低0.5% ~ 0.6%,复配而成的产品对人的刺激性依然很大,且邻苯二甲醛的价格成本居高不下,对比于戊二醛及其低廉的价格,也对产品的成本和市场竞争造成很大的压力。
发明内容
本发明的目的在于克服现有技术中的不足,提供一种消毒剂,具有良好的医疗器械消毒效果及器械保护功能,提供一种消毒剂制备方法,简单可靠,更加安全环保。
为达到上述目的,本发明所采用的技术方案是:
第一方面,本发明提供一种消毒剂,包括以下质量百分比的成分:
两性杀菌型表面活性剂 0.1~30%
辅助表面活性剂 0.1~10%
邻苯二甲醛 0.1~20%
螯合剂 0.01~5%
缓蚀剂 0.1~10%
溶剂 1~50%
pH调节剂 0.1~20%
助剂 0.1-20%
去离子水 余量。
结合第一方面,进一步的,所述两性杀菌型表面活性剂为烷基甜菜碱、烷基酰胺丙基甜菜碱、烷基酰胺丙基甜菜碱酰胺、烷基磺基甜菜碱、氧化胺、两亲性羧基乙酸酯、羧基丙酸酯、氨基酸型表面活性剂中的任意一种或多种。
进一步的,所述两性杀菌型表面活性剂至少包括有一种氧化胺和一种氨基酸型表面活性剂;
所述氧化胺包括月桂基二甲基氧化胺、肉豆蔻基二甲基氧化胺、二甲基椰子基氧化胺、椰油酰胺丙基二甲基氧化胺或牛油酰胺丙基二甲基氧化胺;
所述氨基酸型表面活性剂包括N-酰基氨基酸表面活性剂,包括月桂基氨酸盐、肉豆蔻酰基氨酸盐、椰油酰基肌氨酸盐或油酰基肌氨酸盐和N-烷基丙胺基甘氨酸。
进一步的,所述辅助表面活性剂为阴离子表面活性剂或非离子表面活性剂。
进一步的,所述辅助表面活性剂为烷基硫酸盐、烷基磷酸盐、脂肪醇聚氧乙烯醚羧酸盐、烯基磺酸盐、琥珀酸酯磺酸盐、烷基甘油醚硫酸盐、月桂醇磺酸盐、十二醇硫酸钠、烷基苯磺酸盐、脂肪醇聚氧乙烯醚、脂肪酸甲酯乙氧基化物、异构醇聚氧乙烯醚中的任意一种或多种。
进一步的,所述螯合剂为羟基乙叉二膦酸、乙二胺四乙酸及其钠盐、二乙烯三胺五甲叉膦酸、乙二胺四甲叉膦酸钠、磷酸盐类、硅酸盐类、葡庚糖酸钠、谷氨酸二乙酸四钠、甲基甘氨酸二乙酸三钠盐、柠檬酸及柠檬酸盐、马来酸、聚丙烯酸及其钠盐,其中任意一种或多种。
进一步的,所述缓蚀剂为苯丙三氮唑、钼酸钠、磺化木质素、聚乙烯类、POCA、聚天冬氨酸、铬酸盐、钼酸盐、钨酸盐、钒酸盐、亚硝酸盐、硼酸盐、磷酸盐、含磷有机化合物,任意一种或多种混合物。
进一步的,所述溶剂为甘油、丙二醇、乙醇、异丙醇、二乙二醇甲醚、三乙二醇甲醚、二乙二醇乙醚、三乙二醇乙醚、乙二醇丙醚、乙二醇丁醚、二乙二醇丁醚、三乙二醇丁醚、乙二醇己醚、二乙二醇己醚、乙二醇丁醚醋酸酯、二乙二醇丁醚醋酸酯、乙二醇苯醚、丙二醇甲醚、二丙二醇甲醚、三丙二醇甲醚、丙二醇丙醚、二丙二醇丙醚、丙二醇丁醚、二丙二醇丁醚、三丙二醇丁醚、丙二醇甲醚醋酸酯、二丙二醇甲醚醋酸酯、丙二醇二醋酸酯、丙二醇苯醚、二丙二醇二甲醚中的任意一种或多种混合。
进一步的,所述助剂包含至少一种抗氧剂和至少一种增效剂;
所述抗氧剂包括没食子酸酯、三羟基苯丁酮、丁基羟基茴香醚、二丁基羟基甲苯、特丁基对苯二酚、维生素E、抗坏血酸、D-异抗坏血酸、抗坏血酸棕榈酯;
所述增效剂包括乙醇酸、苯甲醇、水杨酸、糠酸、苯甲酸、乳酸。
进一步的,所述pH调节剂包括氢氧化钠、氢氧化钾、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾、磷酸氢二钠、磷酸和柠檬酸中一种或者多种混合。
第二方面,本发明提供一种消毒剂的制备方法,具体包括如下步骤:
按照上述质量百分比称取各组分;
将邻苯二甲醛、溶剂和助剂混合后加热到35~65℃,再加入不少于30%去离子混合,溶解完成后停止加热,得到第一预混液;
将不少于30%去离子水与两性杀菌型表面活性剂、辅助表面活性剂、pH调节剂、螯合剂和缓蚀剂混合均匀,得到第二预混液;
将第一预混液和第二预混液和剩余的去离子水混合,制得基于两性杀菌剂的消毒剂。
与现有技术相比,本发明所达到的有益效果:
本发明提供的一种消毒剂,通过采用两性杀菌型表面活性剂,复配助剂,能有效降低邻苯二甲醛在消毒剂中的含量,且杀菌效果良好;
产品稳定性好,对微生物杀灭作用强,毒性低且环保,降低对阳极氧化铝、黄铜、碳钢、不锈钢等金属器械的腐蚀性,是一种理想的医疗器械消毒剂;
本发明提供的一种消毒剂制备方法,制备过程简单,性能可靠,绿色环保。
具体实施方式
以下实施例仅用于更加清楚地说明本发明的技术方案,而不能以此来限制本发明的保护范围。
本发明提供一种消毒剂配方,包括以下质量百分比的成分:
两性杀菌型表面活性剂 0.1~30%
辅助表面活性剂 0.1~10%
邻苯二甲醛 0.1~20%
螯合剂 0.01~5%
缓蚀剂 0.1~10%
溶剂 1~50%
pH调节剂 0.1~20%
助剂 0.1-20%
去离子水 余量
一种情况:两性杀菌型表面活性剂为烷基甜菜碱、烷基酰胺丙基甜菜碱、烷基酰胺丙基甜菜碱酰胺、烷基磺基甜菜碱、氧化胺、两亲性羧基乙酸酯、羧基丙酸酯、氨基酸型表面活性剂中的任意一种或多种。
另一种情况:两性杀菌型表面活性剂至少包括有一种氧化胺和一种氨基酸型表面活性剂;氧化胺包括月桂基二甲基氧化胺、肉豆蔻基二甲基氧化胺、二甲基椰子基氧化胺、椰油酰胺丙基二甲基氧化胺或牛油酰胺丙基二甲基氧化胺;氨基酸型表面活性剂包括N-酰基氨基酸表面活性剂,包括月桂基氨酸盐、肉豆蔻酰基氨酸盐、椰油酰基肌氨酸盐或油酰基肌氨酸盐和N-烷基丙胺基甘氨酸。
本发明选用的辅助表面活性剂为阴离子表面活性剂或非离子表面活性剂,为烷基硫酸盐、烷基磷酸盐、脂肪醇聚氧乙烯醚羧酸盐、烯基磺酸盐、琥珀酸酯磺酸盐、烷基甘油醚硫酸盐、月桂醇磺酸盐、十二醇硫酸钠、烷基苯磺酸盐、脂肪醇聚氧乙烯醚、脂肪酸甲酯乙氧基化物、异构醇聚氧乙烯醚中的任意一种或多种。
本发明选用的螯合剂为羟基乙叉二膦酸、乙二胺四乙酸及其钠盐、二乙烯三胺五甲叉膦酸、乙二胺四甲叉膦酸钠、磷酸盐类、硅酸盐类、葡庚糖酸钠、谷氨酸二乙酸四钠、甲基甘氨酸二乙酸三钠盐、柠檬酸及柠檬酸盐、马来酸、聚丙烯酸及其钠盐,其中任意一种或多种。
本发明选用的缓蚀剂为苯丙三氮唑、钼酸钠、磺化木质素、聚乙烯类、POCA、聚天冬氨酸、铬酸盐、钼酸盐、钨酸盐、钒酸盐、亚硝酸盐、硼酸盐、磷酸盐、含磷有机化合物,任意一种或多种混合物。
本发明选用的溶剂为甘油、丙二醇、乙醇、异丙醇、二乙二醇甲醚、三乙二醇甲醚、二乙二醇乙醚、三乙二醇乙醚、乙二醇丙醚、乙二醇丁醚、二乙二醇丁醚、三乙二醇丁醚、乙二醇己醚、二乙二醇己醚、乙二醇丁醚醋酸酯、二乙二醇丁醚醋酸酯、乙二醇苯醚、丙二醇甲醚、二丙二醇甲醚、三丙二醇甲醚、丙二醇丙醚、二丙二醇丙醚、丙二醇丁醚、二丙二醇丁醚、三丙二醇丁醚、丙二醇甲醚醋酸酯、二丙二醇甲醚醋酸酯、丙二醇二醋酸酯、丙二醇苯醚、二丙二醇二甲醚中的任意一种或多种混合。
本发明选用的助剂包含至少一种抗氧剂和至少一种增效剂;其中抗氧剂包括没食子酸酯、三羟基苯丁酮、丁基羟基茴香醚、二丁基羟基甲苯、特丁基对苯二酚、维生素E、抗坏血酸、D-异抗坏血酸、抗坏血酸棕榈酯;增效剂包括乙醇酸、苯甲醇、水杨酸、糠酸、苯甲酸、乳酸。
本发明选用的pH调节剂包括氢氧化钠、氢氧化钾、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾、磷酸氢二钠、磷酸和柠檬酸中一种或者多种混合。
本发明还提供了一种消毒剂的制备方法,包括如下步骤:
按照上述质量百分比称取各组分;
将邻苯二甲醛、溶剂、助剂和不少于30%去离子混合后加热到35~65℃,溶解完成后停止加热,得到第一预混液;
将不少于30%去离子水与两性杀菌型表面活性剂、辅助表面活性剂、pH调节剂、螯合剂和缓蚀剂混合均匀,得到第二预混液;
将第一预混液和第二预混液和剩余的去离子水混合,制得基于两性杀菌剂的消毒剂。

实施例一

本实施例中,消毒剂的制备方法包括如下步骤:
S1将30%的去离子水与0.3%邻苯二甲醛、30%丙二醇和助剂1%糠酸混合,加热到35℃,溶解完成后停止加热,得到第一预混液;
S2将0%的去离子水与0.1%Rewocid WK30和辅助表面活性剂0.5%AEO-9,3%柠檬酸、3%螯合剂STPP和缓蚀剂BTA0.1%混合,混合均匀后得到第二预混液;
S3将第一预混液和第二预混液和剩余的去离子水混合,得到基于两性杀菌剂的高水平消毒剂。
实施例二
Crodasinic LS30 3%
LAS 0.2%
邻苯二甲醛 0.25%
Crodaquest A300 0.5%
钼酸钠 0.1%
乙醇 50%
柠檬酸 1%
苯甲醇 1%
去离子水 余量
本实施例中,消毒剂的制备方法包括如下步骤:
S1将30%的去离子水与0.25%邻苯二甲醛、50%乙醇和助剂1%苯甲醇混合,加热到45℃,溶解完成后停止加热,得到第一预混液。
S2将30%的去离子水与3% Crodasinic LS30和辅助表面活性剂0.2%LAS,1%柠檬酸、0.5%螯合剂Crodaquest A300和缓蚀剂钼酸钠0.1%混合,混合均匀后得到第二预混液。
S3将第一预混液和第二预混液和剩余的去离子水混合,得到基于两性杀菌剂的高水平消毒剂。
实施例三
Rewocid WK30 30%
LF 221 10%
邻苯二甲醛 0.3%
STPP 3%
BTA 0.1%
PEG200 30%
柠檬酸 3%
糠酸 1%

本实施例中,消毒剂的制备方法,包括如下步骤:
S1将30%的去离子水与0.3%邻苯二甲醛、30%PEG200和助剂1%糠酸混合,加热到65℃,溶解完成后停止加热,得到第一预混液。
S2将30%的去离子水与30%Rewocid WK30和辅助表面活性剂10% LF221,3%柠檬酸、3%螯合剂STPP和缓蚀剂BTA0.1%混合,混合均匀后得到第二预混液。
S3将第一预混液和第二预混液和剩余的去离子水混合,得到基于两性杀菌剂的高水平消毒剂。
将实施例一、二与市面上的消毒剂对消毒效果和对金属的腐蚀性进行评价,其中,金属腐蚀性评价方法按照消毒技术规范进行,结果见下表1。

从表1可以得出:本实施例的腐蚀性较低,对金属的兼容性较好。

从表3可以得出: 实施例1通过灭菌模拟现场试验,可以用于医疗器械灭菌。

本发明设计一种基于两性杀菌剂的消毒剂配比,及其制备方法,采用两性杀菌型表面活性剂复配邻苯二甲醛等制成超低金属腐蚀性、高水平消毒的消毒剂,由实验结果可知,制得的消毒剂产品稳定性好,对微生物杀灭作用强,毒性低且环保,对医疗复用器械消毒时,环境污染小,是一种理想的液体消毒剂。且本发明提供的消毒剂制备方法简单,性能可靠,制备过程绿色环保,无毒无害无污染,能够推广应用。
以上所述仅是本发明可选择的实施方式,应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明技术原理的前提下,还可以做出若干改进和变形,这些改进和变形也应视为本发明的保护范围。
Claims (10)
1.一种消毒剂,其特征在于,包括以下质量百分比的成分:
两性杀菌型表面活性剂 0.1~30%
辅助表面活性剂 0.1~10%
邻苯二甲醛 0.1~20%
螯合剂 0.01~5%
缓蚀剂 0.1~10%
溶剂 1~50%
pH调节剂 0.1~20%
助剂 0.1-20%
去离子水 余量。
2.根据权利要求1所述的一种消毒剂,其特征在于,所述两性杀菌型表面活性剂为烷基甜菜碱、烷基酰胺丙基甜菜碱、烷基酰胺丙基甜菜碱酰胺、烷基磺基甜菜碱、氧化胺、两亲性羧基乙酸酯、羧基丙酸酯、氨基酸型表面活性剂中的任意一种或多种。
3.根据权利要求1所述的一种消毒剂,其特征在于,所述两性杀菌型表面活性剂至少包括有一种氧化胺和一种氨基酸型表面活性剂;
所述氧化胺包括月桂基二甲基氧化胺、肉豆蔻基二甲基氧化胺、二甲基椰子基氧化胺、椰油酰胺丙基二甲基氧化胺或牛油酰胺丙基二甲基氧化胺;
所述氨基酸型表面活性剂包括N-酰基氨基酸表面活性剂,包括月桂基氨酸盐、肉豆蔻酰基氨酸盐、椰油酰基肌氨酸盐或油酰基肌氨酸盐和N-烷基丙胺基甘氨酸。
4.根据权利要求1所述的一种消毒剂,其特征在于,所述辅助表面活性剂为阴离子表面活性剂或非离子表面活性剂;所述辅助表面活性剂为烷基硫酸盐、烷基磷酸盐、脂肪醇聚氧乙烯醚羧酸盐、烯基磺酸盐、琥珀酸酯磺酸盐、烷基甘油醚硫酸盐、月桂醇磺酸盐、十二醇硫酸钠、烷基苯磺酸盐、脂肪醇聚氧乙烯醚、脂肪酸甲酯乙氧基化物、异构醇聚氧乙烯醚中的任意一种或多种。
5.根据权利要求1所述的一种消毒剂,其特征在于,所述螯合剂为羟基乙叉二膦酸、乙二胺四乙酸及其钠盐、二乙烯三胺五甲叉膦酸、乙二胺四甲叉膦酸钠、磷酸盐类、硅酸盐类、葡庚糖酸钠、谷氨酸二乙酸四钠、甲基甘氨酸二乙酸三钠盐、柠檬酸及柠檬酸盐、马来酸、聚丙烯酸及其钠盐,其中任意一种或多种。
6.根据权利要求1所述的一种消毒剂,其特征在于,所述缓蚀剂为苯丙三氮唑、钼酸钠、磺化木质素、聚乙烯类、POCA、聚天冬氨酸、铬酸盐、钼酸盐、钨酸盐、钒酸盐、亚硝酸盐、硼酸盐、磷酸盐、含磷有机化合物,任意一种或多种混合物。
7.根据权利要求1所述的一种消毒剂,其特征在于,所述溶剂为甘油、丙二醇、乙醇、异丙醇、二乙二醇甲醚、三乙二醇甲醚、二乙二醇乙醚、三乙二醇乙醚、乙二醇丙醚、乙二醇丁醚、二乙二醇丁醚、三乙二醇丁醚、乙二醇己醚、二乙二醇己醚、乙二醇丁醚醋酸酯、二乙二醇丁醚醋酸酯、乙二醇苯醚、丙二醇甲醚、二丙二醇甲醚、三丙二醇甲醚、丙二醇丙醚、二丙二醇丙醚、丙二醇丁醚、二丙二醇丁醚、三丙二醇丁醚、丙二醇甲醚醋酸酯、二丙二醇甲醚醋酸酯、丙二醇二醋酸酯、丙二醇苯醚、二丙二醇二甲醚中的任意一种或多种混合。
8.根据权利要求1所述的一种消毒剂,其特征在于,所述助剂包含至少一种抗氧剂和至少一种增效剂;
所述抗氧剂包括没食子酸酯、三羟基苯丁酮、丁基羟基茴香醚、二丁基羟基甲苯、特丁基对苯二酚、维生素E、抗坏血酸、D-异抗坏血酸、抗坏血酸棕榈酯;
所述增效剂包括乙醇酸、苯甲醇、水杨酸、糠酸、苯甲酸、乳酸。
9.根据权利要求1所述的一种消毒剂,其特征在于,所述pH调节剂包括氢氧化钠、氢氧化钾、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾、磷酸氢二钠、磷酸和柠檬酸中一种或者多种混合。
10.根据权利要求1-9任一所述的一种消毒剂的制备方法,其特征在于,包括如下步骤:
按照上述质量百分比称取各组分;
将邻苯二甲醛、溶剂、助剂和不少于30%去离子混合后加热到35~65℃,溶解完成后停止加热,得到第一预混液;
将不少于30%去离子水与两性杀菌型表面活性剂、辅助表面活性剂、pH调节剂、螯合剂和缓蚀剂混合均匀,得到第二预混液;
将第一预混液和第二预混液和剩余的去离子水混合,制得基于两性杀菌剂的消毒剂。
