本发明属于消毒剂制品技术领域,具体涉及一种基于两性杀菌剂的高水平消毒剂及其制备方法。
背景技术
随着现代化医学技术的发展,内镜的应用越来越广泛,但作为一种侵入性诊疗方法,如果内镜清洗消毒不彻底,将导致医源性感染的发生。由于内镜结构复杂、材料特殊,许多部位不耐高温、高压或易腐蚀,给临床使用后的消毒带来了挑战。有报道,内镜导致的感染率为0.8%。日本胃镜检查过程中,幽门螺杆菌(HP)感染率高达1.1%,且丙肝病毒(HCV)、梅毒、结核菌、人类免疫缺陷病毒(HIV)等也可经内镜感染。近年来,内镜已从原来的单纯诊断拓展至外科治疗。目前,在我国进行内镜检查人数不断增加,而由此引发的内镜消毒不彻底、交叉感染问题愈发严重,因此内镜消毒技术越来越引起重视。
内镜消毒是指消灭内镜上的病原微生物的方法。目前针对内镜高水平消毒所选用的消毒剂一般为戊二醛、邻苯二甲醛和过氧乙酸。但是由于戊二醛已经产生分枝杆菌抗力株,龟分枝杆菌脓肿亚种菌株对戊二醛产生耐药性。另外,在使用过程中,很多因素会造成药效浓度降低,需每天检测有效浓度,使用比较繁琐,且戊二醛对人体有毒性,伴随有刺激气味,可引起局部皮肤黏膜过敏,要求保持室内通风或者穿戴防护设备,目前已经慢慢淘汰。过氧乙酸虽然消毒效果较好,但是过氧乙酸稳定性差的问题,器械腐蚀性问题严重,导致在内镜消毒领域没有获得广泛应用。
邻苯二甲醛(OPA)是在戊二醛之后出现的一种高水平醛类消毒剂,其杀菌机理与戊二醛类似,都是通过与氨基酸发生交联作用,使蛋白质变性,达到杀死细菌的目的。不同之处是OPA具有更好的脂溶性,更易穿透含脂质较多的分枝杆菌细胞膜,进入菌体内部起作用,因此对分枝杆菌的杀灭作用强于戊二醛。1999年,Walsh等人率先提出将OPA代替戊二醛应用于高水平消毒,商业化的OPA产品由强生公司推出并通过FDA认证,如今已在美国、欧洲和日本获得广泛应用,常规使用浓度为0.5%~0.6%。
目前常见的复方邻苯二甲醛消毒剂都是基于季铵盐复配邻苯二甲醛而成,利用季铵盐的杀菌原理,改变细菌膜的通透性,继而发生溶胞作用,破坏细胞结构,引起细胞的溶解和死亡。邻苯二甲醛和季铵盐的复配达到比较好的杀菌效果。但是季铵盐尤其是双链季铵盐由于其丰富的泡沫,加上其需要邻苯二甲醛的含量依然不低(0.5%~0.6%),复配而成的产品对人的刺激性依然很大,且邻苯二甲醛的价格成本居高不下,对比于戊二醛及其低廉的价格,也对产品的成本和市场竞争造成很大的压力。
发明内容
针对现有技术的不足,本发明的目的在于提供一种基于两性杀菌剂的高水平消毒剂,不仅具有良好的消毒效果,还具有良好的器械保护功能,同时进一步降低邻苯二甲醛的含量,减轻对人的刺激性,更绿色环保。
为实现上述目的,本发明采用如下技术方案:
一种基于两性杀菌剂的高水平消毒剂,按质量百分比计,由以下原料组成:
两性杀菌型表面活性剂0.1~30%,辅助表面活性剂0.1~10%,邻苯二甲醛0.1~20%,螯合剂0.01~5%,缓蚀剂0.1~10%,溶剂1~50%,pH调节剂0.1~20%,其它助剂0.1-20%,去离子水余量;
所述两性杀菌型表面活性剂为甜菜碱型、氧化胺型、羧酸盐型、氨基酸型表面活性剂中的至少一种;
所述辅助表面活性剂为阴离子表面活性剂或非离子表面活性剂中的一种;
所述溶剂为醇、醇醚、醚酯中的至少一种,包括甘油、丙二醇、乙醇、异丙醇、二乙二醇甲醚、三乙二醇甲醚、二乙二醇乙醚、三乙二醇乙醚、乙二醇丙醚、乙二醇丁醚、二乙二醇丁醚、三乙二醇丁醚、乙二醇己醚、二乙二醇己醚、乙二醇丁醚醋酸酯、二乙二醇丁醚醋酸酯、乙二醇苯醚、丙二醇甲醚、二丙二醇甲醚、三丙二醇甲醚、丙二醇丙醚、二丙二醇丙醚、丙二醇丁醚、二丙二醇丁醚、三丙二醇丁醚、丙二醇甲醚醋酸酯、二丙二醇甲醚醋酸酯、丙二醇二醋酸酯、丙二醇苯醚、二丙二醇二甲醚;
所述其它助剂包含至少一种抗氧剂和至少一种增效剂。
进一步的,所述氧化胺型表面活性剂为月桂基二甲基氧化胺、肉豆蔻基二甲基氧化胺、二甲基椰子基氧化胺、椰油酰胺丙基二甲基氧化胺、牛油酰胺丙基二甲基氧化胺中的至少一种。具体商品名包括TEGO Betain P 50C、Tomamine AO-455(EVONIK)、(Stepan)、(lonza)等。
进一步的,所述氨基酸型表面活性剂为月桂基氨酸盐、肉豆蔻酰基氨酸盐、椰油酰基肌氨酸盐、油酰基肌氨酸盐、N-烷基丙胺基甘氨酸中的至少一种,具体商品名包括(Stepan)、(R.T.Vanderbilt)、Crodasinic LS30(Croda)、Tego51/2000和Rewocid WK30(EVONIK)等。
进一步的,所述辅助表面活性剂为烷基硫酸盐、烷基磷酸盐、脂肪醇聚氧乙烯醚羧酸盐、烯基磺酸盐、琥珀酸酯磺酸盐、烷基甘油醚硫酸盐、月桂醇磺酸盐、十二醇硫酸钠、烷基苯磺酸盐、脂肪醇聚氧乙烯醚、脂肪酸甲酯乙氧基化物、异构醇聚氧乙烯醚中的至少一种。
进一步的,所述螯合剂为羟基乙叉二膦酸(HEDP)、乙二胺四乙酸及其钠盐(EDTA/EDTA-2Na/EDTA-4Na)、二乙烯三胺五甲叉膦酸(DTPMPA)、乙二胺四甲叉膦酸钠(EDTMPS)、磷酸盐类(如三聚磷酸钠/焦磷酸钾等)、硅酸盐类、葡庚糖酸钠(Crodaquest A300)、谷氨酸二乙酸四钠(GLDA 47)、甲基甘氨酸二乙酸三钠盐(Trilion M)、柠檬酸及柠檬酸盐、马来酸、聚丙烯酸及其钠盐中的至少一种。

进一步的,所述抗氧剂为没食子酸酯、三羟基苯丁酮、丁基羟基茴香醚、二丁基羟基甲苯、特丁基对苯二酚、维生素、抗坏血酸、D-异抗坏血酸、抗坏血酸棕榈酯中的至少一种。
进一步的,所述增效剂为乙醇酸、苯甲醇、水杨酸、糠酸、苯甲酸、乳酸中的至少一种。
进一步的,所述的pH调节剂为氢氧化钠、氢氧化钾、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾、磷酸氢二钠、磷酸和柠檬酸中的至少一种。
本发明的另一个目的是提供上述基于两性杀菌剂的高水平消毒剂的制备方法,包括以下步骤:
S1.将部分水与邻苯二甲醛、溶剂和其他助剂混合,加热至35~65℃,搅拌溶解后停止加热,得到第一预混液;
S2.将部分水与两性表面活性剂、辅助表面活性剂、pH调节剂、螯合剂和缓蚀剂混合,混合均匀后得到第二预混液;
S3.将将第一预混液、第二预混液和剩余的水混合,得到基于两性杀菌剂的高水平消毒剂。
与现有技术相比,本发明的有益效果在于:
本发明的基于两性杀菌剂的高水平消毒剂通过采用特有的两性表面活性剂,复配上特殊的助剂,能有效降低邻苯二甲醛在消毒剂中的含量,而且杀菌效果良好,消毒5min即能达到消毒技术规范的高水平消毒剂的要求,此外能显著降低对金属器械的腐蚀性,包括阳极氧化铝、黄铜、碳钢、不锈钢等材质。制备过程和方法简单,性能可靠,绿色环保,是一种理想的医疗器械尤其是软式内镜的消毒剂。

实施例1
一种基于两性杀菌剂的高水平消毒剂,按质量百分比计,由以下原料组成:
N-烷基丙胺基甘氨酸(Rewocid WK30)1%,脂肪醇聚氧乙烯醚(AEO-9)0.5%,邻苯二甲醛0.3%,三聚磷酸钠(STPP)3%,苯丙三氮唑(BTA)0.1%,丙二醇30%,柠檬酸3%,糠酸1%,余量为去离子水。
制备方法如下:
S1.将部分水与邻苯二甲醛、丙二醇和糠酸混合,加热至35~65℃,搅拌溶解后停止加热,得到第一预混液;
S2.将部分水与N-烷基丙胺基甘氨酸、脂肪醇聚氧乙烯醚、柠檬酸、三聚磷酸钠和苯丙三氮唑混合,混合均匀后得到第二预混液;
S3.将将第一预混液、第二预混液和剩余的水混合,得到基于两性杀菌剂的高水平消毒剂。
实施例2
一种基于两性杀菌剂的高水平消毒剂,按质量百分比计,由以下原料组成:
月桂酰基肌氨酸钠(Crodasinic LS30)3%,阴离子表面活性剂(LAS)0.2%,邻苯二甲醛0.25%,α-葡庚糖酸钠(Crodaquest A300)0.5%,钼酸钠0.1%,乙醇30%,柠檬酸1%,苯甲醇1%,余量为去离子水。
制备方法如下:
S1.将部分水与邻苯二甲醛、乙醇和苯甲醇混合,加热至35~65℃,搅拌溶解后停止加热,得到第一预混液;
S2.将部分水与月桂酰基肌氨酸钠、阴离子表面活性剂、柠檬酸、α-葡庚糖酸钠和钼酸钠混合,混合均匀后得到第二预混液;
S3.将将第一预混液、第二预混液和剩余的水混合,得到基于两性杀菌剂的高水平消毒剂。
实施例3
一种基于两性杀菌剂的高水平消毒剂,按质量百分比计,由以下原料组成:
活性氧化胺(Tomamine AO-455)1%,EO/PO共聚物(LFE 635)0.5%,邻苯二甲醛0.3%,焦磷酸钾5%,苯丙三氮唑(BTA)0.2%,乙醇20%,柠檬酸5%,水杨酸1%,余量为去离子水。
制备方法如下:
S1.将部分水与邻苯二甲醛、乙醇和水杨酸混合,加热至35~65℃,搅拌溶解后停止加热,得到第一预混液;
S2.将部分水与活性氧化胺、EO/PO共聚物、柠檬酸、焦磷酸钾和苯丙三氮唑混合,混合均匀后得到第二预混液;
S3.将将第一预混液、第二预混液和剩余的水混合,得到基于两性杀菌剂的高水平消毒剂。
将实施例与市面上的消毒剂对消毒效果和对金属的腐蚀性进行评价。
金属腐蚀性评价按照《消毒技术规范》2002版进行,结果如表1所示。

可以看到,本发明的实施例对金属的腐蚀性远远小于市面产品。
杀灭细菌能力评价,按照《消毒技术规范》2002版进行,结果如表2-5所示。

阴性对照无菌生长;分母为试验样片数,分子为阳性样片数。

上述结果表明本发明的基于两性杀菌剂的高水平消毒剂杀菌效果良好,消毒5min即能达到消毒技术规范的高水平消毒剂的要求。

Claims (10)
1.一种基于两性杀菌剂的高水平消毒剂,其特征在于,按质量百分比计,由以下原料组成:
两性杀菌型表面活性剂0.1~30%,辅助表面活性剂0.1~10%,邻苯二甲醛0.1~20%,螯合剂0.01~5%,缓蚀剂0.1~10%,溶剂1~50%,pH调节剂0.1~20%,其它助剂0.1-20%,去离子水余量;
所述两性杀菌型表面活性剂为甜菜碱型、氧化胺型、羧酸盐型、氨基酸型表面活性剂中的至少一种;
所述辅助表面活性剂为阴离子表面活性剂或非离子表面活性剂中的一种;
所述溶剂为醇、醇醚、醚酯中的至少一种;
所述其它助剂包含至少一种抗氧剂和至少一种增效剂。
2.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述氧化胺型表面活性剂为月桂基二甲基氧化胺、肉豆蔻基二甲基氧化胺、二甲基椰子基氧化胺、椰油酰胺丙基二甲基氧化胺、牛油酰胺丙基二甲基氧化胺中的至少一种。
3.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述氨基酸型表面活性剂为月桂基氨酸盐、肉豆蔻酰基氨酸盐、椰油酰基肌氨酸盐、油酰基肌氨酸盐、N-烷基丙胺基甘氨酸中的至少一种。
4.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述辅助表面活性剂为烷基硫酸盐、烷基磷酸盐、脂肪醇聚氧乙烯醚羧酸盐、烯基磺酸盐、琥珀酸酯磺酸盐、烷基甘油醚硫酸盐、月桂醇磺酸盐、十二醇硫酸钠、烷基苯磺酸盐、脂肪醇聚氧乙烯醚、脂肪酸甲酯乙氧基化物、异构醇聚氧乙烯醚中的至少一种。
5.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述螯合剂为羟基乙叉二膦酸、乙二胺四乙酸及其钠盐、二乙烯三胺五甲叉膦酸、乙二胺四甲叉膦酸钠、磷酸盐类、硅酸盐类、葡庚糖酸钠、谷氨酸二乙酸四钠、甲基甘氨酸二乙酸三钠盐、柠檬酸及柠檬酸盐、马来酸、聚丙烯酸及其钠盐中的至少一种。
6.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述缓蚀剂为苯丙三氮唑、钼酸钠、磺化木质素、聚乙烯类、POCA、聚天冬氨酸、铬酸盐、钼酸盐、钨酸盐、钒酸盐、亚硝酸盐、硼酸盐、磷酸盐、含磷的有机化合物中的至少一种。
7.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述抗氧剂为没食子酸酯、三羟基苯丁酮、丁基羟基茴香醚、二丁基羟基甲苯、特丁基对苯二酚、维生素、抗坏血酸、D-异抗坏血酸、抗坏血酸棕榈酯中的至少一种。
8.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述增效剂为乙醇酸、苯甲醇、水杨酸、糠酸、苯甲酸、乳酸中的至少一种。
9.根据权利要求1所述的基于两性杀菌剂的高水平消毒剂,其特征在于,所述的pH调节剂为氢氧化钠、氢氧化钾、碳酸盐、碳酸氢盐、硼酸、硼砂、磷酸二氢钾、磷酸氢二钠、磷酸和柠檬酸中的至少一种。
10.权利要求1-9任一项所述的基于两性杀菌剂的高水平消毒剂的制备方法,其特征在于,包括以下步骤:
S1.将部分水与邻苯二甲醛、溶剂和其他助剂混合,加热至35~65℃,搅拌溶解后停止加热,得到第一预混液;
S2.将部分水与两性表面活性剂、辅助表面活性剂、pH调节剂、螯合剂和缓蚀剂混合,混合均匀后得到第二预混液;
S3.将将第一预混液、第二预混液和剩余的水混合,得到基于两性杀菌剂的高水平消毒剂。
