Description
Technical Field
The invention relates to a pre-incubation method for intermittent short-time heating of goose eggs, belonging to the technical field of poultry breeding.
Background
The goose has low egg laying performance and high egg laying value, and the improvement of the hatching rate of the breeding eggs is always an urgent need production technology for breeding goose production enterprises. In production, a batch of hatching eggs are often hatched every 7 days, which can ensure high hatching rate, but the method not only increases labor workload, but also is difficult to cope with seasonal demand fluctuation of seedlings and uncertainty of orders, so that the hatching egg preservation time is often required in production. However, hatching eggs, once stored for more than a week, begin to decline in their hatching capacity, and when they are stored for more than 14 days, hatching losses can be substantial. In goose breeding, the 7-day reserving of breeding eggs can not meet the requirement of basic group breeding, the breeding conditions of multiple batches of reserving offspring are difficult to be consistent, and the multiple male parent line reserving of breeding causes unclear blood relation. Therefore, effectively extending the shelf life of hatching eggs will play a decisive role in solving the above problems.
Long-term egg storage has an impact on both resting embryos and air cells. With extended storage time, both the internal protein quality of the egg and the integrity of the yolk membrane are compromised. There is evidence that strategic application of heat treatment during egg storage can promote the developmental stages of the embryo, making it more viable in storage, and these short incubation periods can induce repair/regeneration of embryonic cells, thereby minimizing damage from long term storage. Although Chenpeng et al have performed a short-term heat treatment on goose eggs preserved for 10 days, the hatching rate of fertilized eggs is only 74.1%, which is difficult to satisfy the production requirement, so establishing a better pre-hatching technique for a short period of time during the preservation of the eggs will help to improve the hatching rate of the goose eggs preserved for a long time.
Disclosure of Invention
Aiming at the problems in the prior art and the problem that the hatching rate of goose hatching eggs is reduced after long-time storage, the invention fully applies short-term heat treatment to induce the repair/regeneration of embryonic cells, reduces the damage caused by long-term storage, and provides a pre-hatching method for intermittently heating the goose hatching eggs for a short time so as to improve the hatching rate of the goose hatching eggs stored for a long time.
The invention aims to realize the following technical scheme that a pre-incubation method for intermittently heating goose eggs in a short time is characterized by comprising the following steps of:
(1) on the 1 st day, the egg storage chamber is fumigated for 20-30 minutes in a sealed manner by using 30 milliliters of formaldehyde solution and 15 grams of potassium permanganate in each cubic meter of space, the eggs on the same day are collected and placed on an egg rack, the blunt ends of the eggs are upward, and the eggs are turned by 120 degrees every 12 hours;
(2) turning over eggs and storing the eggs on the 2 nd to 3 rd days;
(3) on the 4 th day, directly moving the hatching eggs into an incubator, starting the power supply of the incubator, setting the program to be 37.5-38 ℃ and 3.5h, closing the power supply of the incubator after the treatment is finished, slowly cooling for 1h, opening the door of the incubator, and returning the hatching eggs to an egg storage chamber;
(4) turning over eggs and storing the eggs on the 5 th to 8 th days;
(5) on day 9, preheating treatment is carried out for a short time again;
(6) turning over eggs and storing the eggs on day 10-13;
(7) on day 14, preheating treatment is carried out for a short time again;
or collecting all the hatching eggs, and sending the hatching eggs to a hatchery for hatching in the morning of the next day;
(8) day 15-17: carrying out conventional egg turning preservation;
(9) day 18: carrying out short-time preheating treatment again;
(10) day 19-20: carrying out conventional egg turning preservation;
(11) day 21: all hatching eggs are gathered and sent to a hatchery for hatching in the morning of the next day.
In the step (5), the step (7) and the step (9), the short-time preheating treatment method comprises the following steps: directly moving the hatching eggs into an incubator, starting the power supply of the incubator, setting the program to be 37.5-38 ℃ and 3.5h, closing the power supply of the incubator after the treatment is finished, opening the incubator door after slowly cooling for 1h, and returning the hatching eggs to an egg storage chamber for storage and waiting for incubation.
The method is advanced and scientific, effectively solves the actual technical problem that the hatching rate is reduced due to long preservation time of goose eggs, provides technical support for reducing labor cost, reasonably arranging hatching period and avoiding seedling market risk, can meet the requirement of one-time seed reservation during breeding of breeding geese, and has good application prospect.

Detailed Description
The technical solutions of the present invention are further described in detail by the following specific examples, but it should be noted that the following examples are only used for describing the content of the present invention and should not be construed as limiting the scope of the present invention.
(1) Fumigating an egg storage chamber (with the temperature of 10 ℃ and the relative humidity of 60%) with 30 ml of formaldehyde solution and 15 g of potassium permanganate in a sealed manner for 20-30 minutes in each cubic meter space; collecting 300 qualified hatching eggs of 1000 Yangzhou geese (48 weeks old) in the peak period of egg laying on the same day, placing the 300 qualified hatching eggs in an egg tray of an egg rack, enabling blunt ends of the eggs to face upwards, and turning the hatching eggs by 120 degrees every 12 hours.
(2) Group 1 was incubated at room temperature for 3 weeks, and groups 2 and 3 were heated at 37.5 ℃ and 38 ℃ for intermittent short periods (3.5h), respectively. The specific scheme is as follows:
first, in the 4d th, 9d th, 14d th, 18d th: directly moving the hatching eggs into an incubator, starting the power supply of the incubator, setting the program to be 37.5 ℃ or 38 ℃ for 3.5 hours, closing the power supply of the incubator after the treatment is finished, opening the incubator door after slowly cooling for 1 hour, and returning the hatching eggs to an egg storage chamber.
② the egg turning and other routine preservation are carried out in the rest time
And thirdly, collecting all the hatching eggs at 21d, and waiting for the hatching eggs to be sent to a hatching place for hatching in the morning of the next day.
(3) Hatching 3 groups of hatching eggs using a manual machine, hatching period (days 1 to 27): turning 3-5 times every day at the temperature of 37.5 ℃ and the humidity of 85%; hatching out of the crust breaking period (starting from day 28): the temperature is 37.0 ℃, and the humidity is 95%; after the crust breaking, the temperature was lowered to 36.5 ℃ and the humidity was 70%. Performing first egg lighting on 8 days, removing unfertilized hatching eggs and recording; and (5) performing secondary egg lighting when the eggs are incubated for 28 days, and removing and recording dead embryo eggs. The fertilization rate and the hatching rate of fertilized eggs of each group of hatching eggs are respectively counted (Table 1).
As can be seen from Table 1, after the heat treatment is carried out for 3 weeks at 37.5 ℃ and 38 ℃ in a discontinuous short time, the hatchability of the fertilized eggs can reach more than 95 percent, which is 5.17 percent higher than that of the control group and 6.35 percent higher than that of the control group. The pre-incubation method for intermittently heating the goose eggs in a short time, which is established by the invention, achieves the incubation effect of short-term preservation of the produced hatching eggs, effectively solves the actual technical problem that the incubation rate is reduced due to long preservation time of the goose eggs, provides technical support for reducing the labor cost, reasonably arranging the incubation period and avoiding the seedling market risk, can also meet the requirement of one-time seed reservation during breeding of the breeding geese, and has good application prospect.
