实施例1 GLP-1类似物表达载体和抑制蛋白酶sRNA载体的构建
重组融合基因HIS6-Ub-EK-Arg34GLP-1(7-37)、HIS6-Ub-EK-Arg34GLP-1(9-37)通过NcoI和EcoRI双酶切后,分别与PRSFDuet1经NcoI和EcoRI双酶切后回收的载体片段进行连接,筛选得到重组表达质粒PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)、PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37),再将重组融合基因HIS6-Ub-EK-Arg34GLP-1(7-37)、HIS6-Ub-EK-Arg34GLP-1(9-37)通过NdeI和XhoI双酶切后,分别与质粒PRSFDuet1- HIS6-Ub-EK-Arg34GLP-1(7-37)、PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)经NdeI和XhoI双酶切后回收的载体片段进行连接,筛选得到双表达质粒PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)、PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37),PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)结果见图1。
将基因片段NO:11-18通过酶SpeI和PstI双酶切后,与pBAD202经SpeI和PstI双酶切后回收的载体片段连接,筛选得到抑制蛋白酶表达的sRNA重组质粒pBAD-sRNA-clpA、pBAD-sRNA-clpP、pBAD-sRNA-clpQ、pBAD-sRNA-clpX、pBAD-sRNA-clpY、pBAD-sRNA-Dcp、pBAD-sRNA-PepD和pBAD-sRNA-hflB,pBAD-sRNA-clpA结果见图2,其他sRNA重组质粒略。
实施例2 表达GLP-1类似物重组工程菌的构建及高表达筛选
以BL21(DE3)为宿主菌,制备感受态细胞,将重组表达质粒PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)、PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和质粒pBAD-sRNA-clpA、pBAD-sRNA-clpP、pBAD-sRNA-clpQ、pBAD-sRNA-clpX、pBAD-sRNA-clpY、pBAD-sRNA-Dcp、pBAD-sRNA-PepD和pBAD-sRNA-hflB用氯化钙法转化至其感受态细胞,分别形成BL21(DE3)双质粒表达菌株1(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-clpA)、2(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-clpQ)、3(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-clpP)、4(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-clpX)、5(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-clpY)、6(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-Dcp)、7(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37)和pBAD-sRNA-PepD)、8(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-hflB),9(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-clpA)、10(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-clpQ)、11(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-clpP)、12(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-clpX)、13(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-clpY)、14(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-Dcp)、15(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-PepD)、16(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-hflB),转化方法按照《分子克隆实验指南》第四版的氯化钙法进行。将转化液涂布至添加了100μg/ml氨苄青霉素、100μg/ml卡那霉素的LB固体培养基,37℃倒置培养直到出现单菌落,即获得了重组表达菌株,挑选单克隆菌株测序鉴定正确。将上述菌株同等培养条件下诱导表达,经过高表达筛选,BL21(DE3)双质粒表达菌株3(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(7-37),和pBAD-sRNA-clpP)、BL21(DE3)双质粒表达菌株11(PRSFDuet1-HIS6-Ub-EK-Arg34GLP-1(9-37)和pBAD-sRNA-clpP),见图3,a双质粒表达菌株3与对照相比产量提高最多,可达200%,b双质粒表达菌株11与对照相比产量提高最多,可达185%。
实施例3重组GLP-1类似物的表达与纯化
在平板上挑取重组双质粒表达菌株3、双质粒表达菌株11单菌落,在试管培养8h后按照1:1000转入500mL三角瓶中37℃震荡培养,当OD值达到0.6-0.8时,用0.5 mM IPTG进行诱导,25℃诱导20h后离心弃上清液得到菌体沉淀。
对离心得到的菌体用30mL蛋白纯化的Buffer A悬浮,利用高压均质破碎机对悬浮细胞进行破碎,得到蛋白的粗样品液,离心收集上清液,利用亲和Ni进行蛋白的纯化,用不同比例的Buffer B进行梯度洗脱,将含有HIS6的蛋白洗脱下来,对洗脱液进行脱盐处理,置换到溶液Buffer C中。融合蛋白分离纯化见图4。10ug融合蛋白加入1U肠激酶,25℃酶切6h-12h。酶切后通过亲和层析或离子交换分离纯化获得GLP-1类似物。质谱图见图5、6。

实施例4重组工程菌株高密度发酵
接种实施例2所得的重组双质粒表达菌株3、双质粒表达菌株11阳性单克隆于3mL液体LB培养基中,37℃、250rpm震荡过夜培养后,按1%左右的比例接种于400mL液体LB培养基中,培养至OD600达到4时作为种子液,接入2L的发酵培养基中进行高密度发酵。初始温度37℃,搅拌速度300rpm,通气量1.5vvm/L/min,pH为6.8,之后不断提高搅拌转速最大至1000rpm。高密度发酵需氧量大,如氧气缺乏,会产生乙酸,甚至造成菌体裂解释放有害物质,对重组工程菌的表达产生不可逆的影响。因此,后期需补充一定的纯氧。发酵培养分为两个阶段,第一阶段接种后,培养4小时左右,培养基中的碳源消耗完全,按照DO进行反馈补料。补料后温度降为30℃,溶氧保持30%以上,补料8小时后加入异丙基硫代半乳糖苷(IPTG)进行诱导,诱导12小时后放罐。
重组双质粒表达菌株3、菌株11高密度发酵后分别进行分离纯化及酶切,通过BCA蛋白定量试剂盒定量测定,GLP-1类似物表达量分别可达2g/L、2.11g/L;表明采用本发明方法干扰获得的菌株实现了GLP类似物的高密度发酵表达。
通过液相色谱检测,酶切后分离纯化分别可获得800mg-1000mg的GLP-1类似物。通过电泳检测,菌株发酵获得的融合蛋白Arg34GLP-1(7-37)可溶占比83%,融合蛋白Arg34GLP-1(9-37)可溶占比80%,其中,全菌为细胞破碎后样品,上清为细胞破碎后样品离心的上清-即可溶部分,沉淀为细胞破碎后样品离心后的沉淀-即为不可溶部分。检测结果参见图7(其中1#是融合蛋白HIS6-Ub-EK-Arg34GLP-1(7-37),2#是融合蛋白HIS6-Ub-EK-Arg34GLP-1(9-37))。
