Abstract
The invention belongs to the technical field of health-care food, and relates to a composition for treating insomnia, a preparation method and application thereof, wherein the composition comprises the following raw materials in parts by weight: 100-200 parts of spina date seed, 30-70 parts of Chinese yam, 30-50 parts of fingered citron, 70-130 parts of poria cocos, 30-90 parts of acanthopanax, 10-30 parts of dendrobe, 10-40 parts of cassia twig, 5-20 parts of liquorice and 20-100 parts of gamma-aminobutyric acid. The composition has remarkable effects in treating insomnia and relieving various anxiety, and has no toxic and side effects.
Description
Technical Field
The invention relates to the technical field of health-care food, in particular to a composition for treating insomnia and a preparation method and application thereof.
Background
Insomnia is a healthy predator of human beings, and long-term insomnia can affect normal work and life, easily cause mental asthenia, mental disorder and mental disorder, induce physical diseases such as heart and brain, and cause accidents. The long-term insomnia is also easy to cause the reduction of immunity, so that the aging of the human body is accelerated. Preferably, the feed additive comprises the following raw materials in parts by weight: 180 parts of spina date seed, 35-65 parts of Chinese yam, 35-45 parts of finger citron, 75-125 parts of poria cocos, 35-85 parts of acanthopanax, 13-27 parts of dendrobe, 13-27 parts of cassia twig, 7-20 parts of liquorice and 30-70 parts of gamma-aminobutyric acid.
Preferably, the feed additive comprises the following raw materials in parts by weight: 130 parts of spina date seed, 170 parts of wild yam, 40-60 parts of Chinese yam, 37.5-42.5 parts of finger citron, 80-120 parts of tuckahoe, 40-80 parts of acanthopanax, 16-24 parts of dendrobium, 16-24 parts of cassia twig, 8-15 parts of liquorice and 35-65 parts of gamma-aminobutyric acid.
Preferably, the feed additive comprises the following raw materials in parts by weight: 160 parts of spina date seed 140, 45-55 parts of Chinese yam, 37.5-42.5 parts of fingered citron, 90-110 parts of poria cocos, 50-70 parts of acanthopanax, 18-22 parts of dendrobium, 18-22 parts of cassia twig, 8-12 parts of liquorice and 40-60 parts of gamma-aminobutyric acid.
Preferably, the feed additive comprises the following raw materials in parts by weight: 150 parts of spina date seed, 50 parts of Chinese yam, 40 parts of fingered citron, 100 parts of poria cocos, 60 parts of acanthopanax, 20 parts of dendrobium, 20 parts of cassia twig, 10 parts of liquorice and 50 parts of gamma-aminobutyric acid.
The composition can be used for preparing medicine for treating insomnia caused by heart yang deficiency and heart spirit malnutrition.
The invention also provides a preparation method of the composition for treating insomnia, which comprises the following steps:
s1, weighing semen Ziziphi Spinosae, rhizoma Dioscoreae, fructus Citri Sarcodactylis, Poria, radix Acanthopanacis Senticosi, herba Dendrobii, ramulus Cinnamomi, Glycyrrhrizae radix, and gamma-aminobutyric acid;
s2, parching semen Ziziphi Spinosae, and squeezing to obtain oil;
s3, mixing the spina date seed oil obtained in the S2 with dendrobium, Chinese yam and liquorice, and carrying out superfine grinding at a low temperature of less than or equal to-5 ℃ to obtain a mixture A;
s4, mixing fructus Citri Sarcodactylis, Poria, radix Acanthopanacis Senticosi, and ramulus Cinnamomi, adding water 8-10 times of the total weight of the four materials, decocting for 1.5-2h, and collecting the first decoction; adding 6-8 times of water into the residue, decocting for 1.0-1.5 hr, and collecting the second decoction; adding water 6-8 times the total weight of the four medicinal materials into the obtained residue, decocting for 0.5-1.0h, collecting the third decoction, mixing with the first decoction and the second decoction, filtering, and concentrating to relative density of 1.1 at 40 deg.C to obtain extract;
s5, crushing gamma-aminobutyric acid, sieving the crushed gamma-aminobutyric acid with a 100-mesh sieve, uniformly mixing the crushed gamma-aminobutyric acid and the mixture A, adding the extract, granulating and drying to obtain the composition.
The invention also provides a pharmaceutical preparation which takes the composition as an active ingredient and is supplemented with pharmaceutically acceptable auxiliary materials.
Preferably, the pharmaceutical formulation is a solid formulation.
Preferably, the solid preparation is a tablet, capsule or granule.
The composition provided by the invention aims at improving heart yang deficiency and heart spirit malnutrition, and the spina date seed is used as a monarch drug for nourishing liver yin, tonifying heart blood, tranquilizing and allaying excitement; the Chinese yam has the effects of tonifying spleen and nourishing stomach, the fingered citron has the effects of soothing liver and regulating qi, reducing phlegm and relieving chest stuffiness, and harmonizing spleen and stomach, the two medicines cause the spleen and stomach to be vigorous, the vital energy of stomach elements is vigorous, the vital essence of food is upwards injected into the lung, the spleen and stomach calm, the qi movement is regulated, wood can generate fire to calm the heart and mind, and the two medicines are ministerial medicines; poria has effects in invigorating spleen, strengthening middle warmer, promoting diuresis, removing dampness, calming heart, tranquilizing mind, invigorating qi and spleen, tranquilizing mind, invigorating kidney, strengthening body constitution, and is used as adjuvant drug; the dendrobium has sweet and slightly cold nature and flavor, enters stomach and kidney channels, has the effects of benefiting stomach, promoting fluid production, nourishing yin and removing heat, has the effects of warming and activating yang, can nourish heart yin by steaming kidney water and nourish heart yin by filling heart yin, and is also used as an adjuvant drug; the liquorice has the effects of tonifying qi, strengthening the middle warmer and harmonizing the other drugs, and can also be sweet, slow and nourish yin, and is matched with the spina date seed to increase the effect of nourishing yin; being compatible with Gui Zhi, pungent, sweet and pungent in flavor and capable of activating yang, it is a guiding drug to tonify deficiency of heart yang. Animal experiment results also show that the sedative and hypnotic effects of the traditional Chinese medicine formula and the gamma-aminobutyric acid are obviously better than those of the gamma-aminobutyric acid singly applied.
Compared with the prior art, the invention has the beneficial effects that:
the invention takes the ancient famous medical science Lidongyuan 'the theory of spleen and stomach' of gold yuan, four everybody as a guide, starts from the importance of nutrition in maintaining mental health, takes medicinal and edible medicinal materials as raw materials under the guidance of the theory of traditional Chinese medicine, is supplemented with a food additive gamma-aminobutyric acid, makes up the deficiency of the gamma-aminobutyric acid in the traditional Chinese medicine components, provides a medicinal composition which has the functions of nourishing the heart, soothing the nerves, relieving restlessness and resolving depression and has the functions of having less medicinal flavor and simple preparation method, and better playing the role of external aid nutrient components, and is suitable for the insomnia crowds caused by insufficient heart yang and malnutrition of heart spirit.

A composition for treating insomnia is prepared from the following raw materials: 100g of spina date seed, 30g of Chinese yam, 30g of fingered citron, 70g of poria cocos, 30g of acanthopanax, 10g of dendrobium, 10g of cassia twig, 5g of liquorice and 20g of gamma-aminobutyric acid.
Example 2
A composition for treating insomnia is prepared from the following raw materials: 200g of spina date seed, 70g of Chinese yam, 50g of fingered citron, 130g of poria cocos, 90g of acanthopanax, 30g of dendrobium, 30g of cassia twig, 25g of liquorice and 80g of gamma-aminobutyric acid
Example 3
A composition for treating insomnia is prepared from the following raw materials: 120g of spina date seed, 35g of Chinese yam, 35g of fingered citron, 75g of poria cocos, 35g of acanthopanax, 13g of dendrobium, 13g of cassia twig, 7g of liquorice and 30g of gamma-aminobutyric acid.
Example 4
A composition for treating insomnia is prepared from the following raw materials: 180g of spina date seed, 65g of Chinese yam, 45g of fingered citron, 125g of poria cocos, 85g of acanthopanax, 27g of dendrobium, 27g of cassia twig, 20g of liquorice and 70g of gamma-aminobutyric acid.
Example 5
A composition for treating insomnia is prepared from the following raw materials: 130g of spina date seed, 40g of Chinese yam, 37.5g of fingered citron, 80g of poria cocos, 40g of acanthopanax, 16g of dendrobium, 16g of cassia twig, 8g of liquorice and 35g of gamma-aminobutyric acid.
Example 6
A composition for treating insomnia is prepared from the following raw materials: 170g of spina date seed, 60g of Chinese yam, 42.5g of fingered citron, 120g of poria cocos, 80g of acanthopanax, 24g of dendrobium, 24g of cassia twig, 15g of liquorice and 65g of gamma-aminobutyric acid.
Example 7
A composition for treating insomnia is prepared from the following raw materials: 160g of spina date seed, 55g of Chinese yam, 42.5g of fingered citron, 110g of poria cocos, 70g of acanthopanax, 22g of dendrobium, 22g of cassia twig, 12g of liquorice and 60g of gamma-aminobutyric acid.
Example 8
A composition for treating insomnia is prepared from the following raw materials: 140g of spina date seed, 45g of Chinese yam, 37.5g of fingered citron, 90g of poria cocos, 50g of acanthopanax, 18g of dendrobium, 18g of cassia twig, 8g of liquorice and 40g of gamma-aminobutyric acid.
Example 9
A composition for treating insomnia is prepared from the following raw materials: 150g of spina date seed, 50g of Chinese yam, 40g of fingered citron, 100g of poria cocos, 60g of acanthopanax, 20g of dendrobium, 20g of cassia twig, 10g of liquorice and 50g of gamma-aminobutyric acid.
The methods for preparing the compositions of examples 1-9 above, each comprising the steps of:
s1, weighing semen Ziziphi Spinosae, rhizoma Dioscoreae, fructus Citri Sarcodactylis, Poria, radix Acanthopanacis Senticosi, herba Dendrobii, ramulus Cinnamomi, Glycyrrhrizae radix, and gamma-aminobutyric acid;
s2, parching semen Ziziphi Spinosae, and squeezing to obtain oil;
s3, mixing the spina date seed oil obtained in the S2 with dendrobium, Chinese yam and liquorice, and carrying out superfine grinding at a low temperature of less than or equal to-5 ℃ to obtain a mixture A;

The composition can be further processed into tablet, granule or capsule according to pharmaceutically acceptable method.
Example 10
The composition for treating insomnia is different from the preparation method of the embodiment 9, and specifically comprises the following steps:
s1, weighing semen Ziziphi Spinosae, rhizoma Dioscoreae, fructus Citri Sarcodactylis, Poria, radix Acanthopanacis Senticosi, herba Dendrobii, ramulus Cinnamomi, Glycyrrhrizae radix, and gamma-aminobutyric acid;
s2, parching semen Ziziphi Spinosae, and squeezing to obtain oil;
s3, mixing the spina date seed oil obtained in the S2 with dendrobium, Chinese yam and liquorice, and carrying out superfine grinding at a low temperature of less than or equal to-5 ℃ to obtain a mixture A;
s4, mixing fructus Citri Sarcodactylis, Poria, radix Acanthopanacis Senticosi, and ramulus Cinnamomi, adding water 10 times of the total weight of the four materials, decocting for 2h, and collecting the first decoction; adding water 8 times the total weight of the four medicinal materials into the filter residue, decocting for 1.5h, and collecting the second decoction; adding water 8 times the total weight of the four medicinal materials into the obtained residue, decocting for 1.0h, collecting the third decoction, mixing with the first decoction and the second decoction, filtering, and concentrating to relative density of 1.1 at 40 deg.C to obtain extract;
s5, crushing the gamma-aminobutyric acid, sieving the crushed gamma-aminobutyric acid with a 100-mesh sieve, uniformly mixing the crushed gamma-aminobutyric acid and the mixture A, adding the extract, granulating and drying to obtain the composition.
Since the therapeutic effects of the compositions prepared in examples 1 to 10 on insomnia are substantially the same, the effects of the present invention will be described below by taking example 9 as an example only.
1. Sedative effect on mice
1.1 Experimental materials
The clean-grade Kunming mice, which are 18-22g in weight and half in sex, are provided by the animal testing center of Shanxi Chinese medicine university, and all groups of mice are fed in cages and are fed with drinking water freely.
1.2, grouping and administration
100 healthy Kunming mice are taken and randomly divided into a blank control group, a high dose group, a medium dose group, a low dose group and 5 groups of gamma-aminobutyric acid, wherein each group comprises 20 mice. Wherein the composition of example 9 was administered in the high dose group at 8 g/kg-d-1The composition of example 9 was administered in a medium dose group at 4 g/kg. d-1In the low dose group, the composition of example 9 was administered at 2 g/kg. d-1The gamma-aminobutyric acid group is given with the same dose of gamma-aminobutyric acid as the medium dose group, the blank control group is given with the same volume of normal saline as the medium dose group, and the administration is performed by drenching once a day for 3 consecutive days. Mice were placed in an activity box for 3 minutes before dosing, and the number of spontaneous activity in each group of animals for 3 minutes was recorded, and the mean value was taken as the pre-dose normal value. The rate of inhibition of activity was measured 1 hour after the last administration, and = (mean of activity in blank group-mean of administration group)/mean of blank group × 100%.
1.3 results
The results are shown in Table 1.

The results show that the inhibition rates of high, medium and low doses on the number of the autonomic activities of the mice are all higher than those of the gamma-aminobutyric acid group, and the combination of the composition provided by the invention can play a remarkable role in sedation.
2. Effect on sleep time in mice with threshold dose of sodium pentobarbital
2.1 method
100 male mice weighing 18-22g were randomly divided into 5 groups (20 per group): blank control group (giving equal volume of distilled water), low dose group (3.1023g crude drug/Kg), middle dose group (6.1324g crude drug/Kg), high dose group (12.2217g crude drug/Kg), positive control group (estazolam, 5mg/Kg), low, middle, high dose component respectively equivalent to clinical dose (0.6g crude drug/Kg) 5, 10, 20 times. The administration is performed by gavage 1 time per day, 0.4ml each time, and is continued for 3 days. Pentobarbital sodium (45mg/Kg) is intraperitoneally injected 1 hour after the last administration, the sleep latency period is from the injection of the pentobarbital sodium to the disappearance of the mouse righting reflex, and the sleep duration period is from the disappearance of the righting reflex to the recovery time. Animals whose data deviated 3-fold (3s) from the standard deviation were statistically excluded, and the sleep latency and sleep time between the administered group and the control group were compared, and the results are shown in table 2.

the results show that the sleep latency time of the low, medium and high dose groups is lower than that of the blank control group, and the sleep time is higher than that of the blank control group, which indicates that the composition provided by the invention has the effect of obviously prolonging the sleep time.
3. Safety analysis and adverse reaction observation
No death cases exist during the clinical observation period, and vital signs (body temperature, blood pressure, respiration and heart rate), blood routine, urine routine + microscopic examination, stool routine + occult blood test, liver function, kidney function, electrocardiogram examination and the like of two groups of patients before and after treatment are in a normal range. No adverse events occurred during the observation period.
Although the invention has been described in detail in the foregoing by way of general description, specific embodiments and experiments, it will be apparent to those skilled in the art that various modifications and improvements can be made thereto without departing from the spirit of the invention.

The composition for treating insomnia is characterized by comprising the following raw materials in parts by weight: 100-200 parts of spina date seed, 30-70 parts of Chinese yam, 30-50 parts of fingered citron, 70-130 parts of poria cocos, 30-90 parts of acanthopanax, 10-30 parts of dendrobe, 10-30 parts of cassia twig, 5-25 parts of liquorice and 20-80 parts of gamma-aminobutyric acid.

The composition for treating insomnia according to claim 1, which is prepared from the following raw materials in parts by weight: 180 parts of spina date seed, 35-65 parts of Chinese yam, 35-45 parts of finger citron, 75-125 parts of poria cocos, 35-85 parts of acanthopanax, 13-27 parts of dendrobe, 13-27 parts of cassia twig, 7-20 parts of liquorice and 30-70 parts of gamma-aminobutyric acid.
3. The composition for treating insomnia according to claim 1, which is prepared from the following raw materials in parts by weight: 160 parts of spina date seed 140, 45-55 parts of Chinese yam, 37.5-42.5 parts of fingered citron, 90-110 parts of poria cocos, 50-70 parts of acanthopanax, 18-22 parts of dendrobium, 18-22 parts of cassia twig, 8-12 parts of liquorice and 40-60 parts of gamma-aminobutyric acid.

The composition for treating insomnia according to claim 1, which is prepared from the following raw materials in parts by weight: 150 parts of spina date seed, 50 parts of Chinese yam, 40 parts of fingered citron, 100 parts of poria cocos, 60 parts of acanthopanax, 20 parts of dendrobium, 20 parts of cassia twig, 10 parts of liquorice and 50 parts of gamma-aminobutyric acid.
6. Use of the composition according to any one of claims 1 to 5 for the preparation of a medicament for the treatment of insomnia caused by insufficient heart-yang and malnutrition of the heart-mind.
7. The process for preparing a composition for the treatment of insomnia according to any one of claims 1 to 5, comprising the steps of:
s1, weighing semen Ziziphi Spinosae, rhizoma Dioscoreae, fructus Citri Sarcodactylis, Poria, radix Acanthopanacis Senticosi, herba Dendrobii, ramulus Cinnamomi, Glycyrrhrizae radix, and gamma-aminobutyric acid;
s2, parching semen Ziziphi Spinosae, and squeezing to obtain oil;
s3, mixing the spina date seed oil obtained in the S2 with dendrobium, Chinese yam and liquorice, and carrying out superfine grinding at a low temperature of less than or equal to-5 ℃ to obtain a mixture A;
