Abstract
The invention discloses a moisturizing and antioxidant cordyceps mask emulsion and a preparation method thereof, wherein the cordyceps mask emulsion comprises the following raw materials in parts by weight: 70-80 parts of deionized water, 8-12 parts of cordyceps polysaccharide extract, 7-12 parts of glycerol, 2-3 parts of hyaluronic acid, 0.3-0.5 part of carbomer, 0.1-0.3 part of allantoin, 0.1-0.3 part of essence and 0.05-0.1 part of methylparaben, wherein the cordyceps polysaccharide extract is fine cordyceps polysaccharide extract, and the non-sexual type of the fine cordyceps is lobelina. The cordyceps mask emulsion prepared by the invention takes intracellular polysaccharide extracted from deep-fermented cordyceps sinensis as a main component in the mask emulsion, fully utilizes the biological activity of the cordyceps sinensis polysaccharide, and is matched with various auxiliary materials to achieve multiple effects of moisturizing, resisting oxidation, whitening, relieving and the like.
Description
Technical Field
The invention belongs to the field of cordyceps sinensis products, and particularly relates to a moisturizing antioxidant cordyceps sinensis mask emulsion and a preparation method thereof.
Background
With the improvement of living standard, people increasingly pursue beauty treatment, and skin which is wet and can be broken by blowing is advocated, so that the development of a novel, safe and efficient skin care product is particularly necessary. The mask is a relatively common skin care product, and the principle of the mask is that essence attached to the mask permeates into skin, so that the skin is improved. The existing cordyceps sinensis is mostly used for medicinal materials and food, has the main functions of tonifying lung and kidney, stopping cough, benefiting deficiency and nourishing essence and qi, and is not mature in the field of skin care, so that the cordyceps sinensis mask has a wide prospect as a novel mask in the current market. And according to the existing cordyceps mask, we find that part of potential problems exist, such as: the extraction method of the cordyceps extract is coarse and simple and is not fine enough, so that excessive impurities exist; the specific components and the structure of the cordyceps extract are not clear enough, so that the explanation of the effects of moisturizing, whitening and the like is not clear enough; the price is high, and particularly, the cordyceps mask has high extraction cost due to the troublesome extraction process of the cordyceps extract, so that the selling price of the cordyceps extract is influenced, and the cordyceps mask cannot be popularized to the public.
Disclosure of Invention
The purpose of the invention is as follows: aiming at the problems in the prior art, the invention provides the moisturizing and antioxidant cordyceps mask emulsion, which solves the problems that the cordyceps sinensis facial mask in the prior cordyceps sinensis facial mask on the market is not clear in extracted component and structure, uncertain in effect and high in price caused by the effect, and provides the moisturizing and antioxidant cordyceps mask emulsion which is clear in characterization, novel in component, safe, effective, high in quality, low in price and free of toxic and side effects for the market.

The technical scheme is as follows: in order to achieve the purpose, the moisturizing and antioxidant cordyceps mask emulsion provided by the invention comprises the following raw materials in parts by weight: 70-80 parts of deionized water, 8-12 parts of cordyceps polysaccharide extract, 7-12 parts of glycerol, 2-3 parts of hyaluronic acid, 0.3-0.5 part of carbomer, 0.1-0.3 part of essence, 0.1-0.3 part of allantoin and 0.05-0.1 part of methylparaben, wherein the cordyceps polysaccharide extract is fine cordyceps polysaccharide extract, and the non-sexual type of the fine cordyceps is lobelina leucotricha.
Preferably, the moisturizing and antioxidant cordyceps mask emulsion comprises the following raw materials in parts by weight: 12 parts of fine cordyceps polysaccharide extract, 7 parts of glycerol, 3 parts of hyaluronic acid, 0.5 part of carbomer, 0.3 part of allantoin, 0.1 part of methylparaben, 0.3 part of rose essence and 75 parts of deionized water.
Wherein the fine Cordyceps is mycelia of non-sexual type Leucocephala of fine Cordyceps cultured by liquid submerged fermentation.
Further, the fermentation medium comprises 20-30g/L of glucose, 10-15g/L of peptone, 0.3-0.5g/L of magnesium sulfate heptahydrate and 2-3g/L of potassium dihydrogen phosphate.
Further, the fermentation culture conditions comprise temperature of 20-25 deg.C, stirring speed of 100rpm-200rpm, ventilation amount of 50-100VVM, and pH of 5-7, and fermentation time of 14-20 days.
Further, the fine cordyceps polysaccharide extract is used for extracting purified cordyceps polysaccharide from cordyceps mycelia by a hot water extraction method.
Further, the hot water extraction method comprises the following specific processes:
grinding: fully grinding and crushing the dried mycelium of the amorphous lobelia of the fine cordyceps sinensis;
leaching: adding the ground mycelium of the fine cordyceps sinensis anamorphic type lobium leucotrichum into hot water, leaching in a constant-temperature water bath, filtering, reserving filtrate, repeatedly leaching filter residue, and combining the filtrate;
alcohol precipitation: the concentrated filtrate was placed in ethanol and allowed to stand overnight.
Deproteinization: standing overnight, centrifuging to remove supernatant, dissolving precipitate with hot water, centrifuging to remove precipitate, and precipitating supernatant with 95% ethanol; repeating the operation according to the first deproteinization, and performing second and third deproteinization;
crude polysaccharide: centrifuging the deproteinized solution to remove supernatant, dissolving the precipitate with hot water, and freeze drying to obtain crude Cordyceps polysaccharide;
separation and purification: dissolving the crude Cordyceps polysaccharide in water, and freeze drying the eluate collected by chromatographic column to obtain refined Cordyceps polysaccharide.
The preparation method of the moisturizing and antioxidant cordyceps mask emulsion comprises the following steps:
preparation of phase A: weighing refined Cordyceps polysaccharide according to formula, dissolving in hot deionized water, sequentially adding allantoin, glycerol, hyaluronic acid, methyl hydroxybenzoate and essence, and stirring to dissolve completely;
preparation of phase B: heating the rest deionized water, sequentially adding carbomer while stirring, stirring to make it fully swell, making into gel substance, and cooling.
(3) Phase a was slowly added to the stirred phase B, allowed to mix well, and the pH adjusted.
Wherein, the fine cordyceps polysaccharide extract in the step (1) is dissolved in hot water with the temperature of 60-80 ℃, and allantoin, glycerol, hyaluronic acid, methylparaben and essence are sequentially added and fully stirred to be completely dissolved.
Heating the rest deionized water in the step (2), sequentially adding carbomer while stirring, stirring for 30-60min to fully swell the carbomer, preparing into gel substance, and cooling to 30-40 deg.C.
Wherein, the pH value is adjusted to 5-6 by citric acid in the step (3).
The cordyceps polysaccharide extract is from fine cordyceps, the fine cordyceps is a complex of fungi parasitizing on the corpses of hepialus altaicus larvae, the anamorph is the lobelia leucotricha, and the cordyceps polysaccharide extract is mainly distributed in the region of the Aleptai mountain in Xinjiang. At present, the development and utilization of cordyceps are less, and only related products developed by taking cordyceps sinensis and cordyceps militaris as materials are seen in the market, so that the research and development of fine cordyceps sinensis produced in Xinjiang have wide market prospects.
The Xinjiang cordyceps sinensis selected by the lotion provided by the invention is a traditional Chinese medicine, has the effects of benefiting essence, protecting lung, tonifying kidney, stopping bleeding, reducing phlegm, stopping cough and the like, and also has the effects of bacteriostasis, antioxidation and anti-aging. The invention obtains the fine cordyceps mycelia through submerged fermentation, the cordyceps extract in the mask emulsion is taken as a main component of the intracellular polysaccharide extract of the fine cordyceps with certain characteristics and definite components, and the fine hot water extraction method is adopted to extract the intracellular polysaccharide from the mycelium powder, so that the uniform and fine intracellular polysaccharide is extracted, the efficiency is improved, the cost is reduced, the problems of high price, more impurities and unknown components of the same cordyceps mask on the market are solved, and the environmental damage caused by wild cordyceps is avoided. The facial mask liquid disclosed by the invention fully utilizes the biological activity of cordyceps polysaccharide, is matched with various auxiliary materials, is safe and free from toxic and side effects, can achieve multiple effects of moisturizing, resisting oxidation, relieving and the like, and has wide market prospect. The emulsion prepared by the invention is prepared by dissolving fine cordyceps sinensis polysaccharide with uniform texture obtained by refining in deionized water, slowly stirring and adding into a solution added with glycerol, hyaluronic acid, carbomer, allantoin and methylparaben, and then adjusting the pH value by citric acid. The prepared emulsion is milk white semitransparent colloid, has uniform texture, good sticky feeling, easy coating, fineness, smoothness, moist and refreshing skin feeling, slight faint scent, pH of about 5-6, and good oxidation resistance and moisture retention.
Has the advantages that: compared with the prior art, the invention has the following advantages:
1. the novel fine cordyceps sinensis polysaccharide is adopted in the mask emulsion, the antioxidant activity of the novel fine cordyceps sinensis polysaccharide is very high, the capability of removing DPPH and ABTS free radicals and the total reducing power of the novel fine cordyceps sinensis polysaccharide are obviously superior to those of cordyceps sinensis and cordyceps militaris, and meanwhile, fine cordyceps sinensis mycelia obtained through submerged fermentation are low in cost and low in price, and are more beneficial to market popularization.
2. The fine cordyceps sinensis polysaccharide obtained by separating and purifying through a hot water extraction method and a column chromatography method is high in purification efficiency, and the obtained polysaccharide is free of impurities and uniform in components, so that the problems of unclear active ingredients and high impurity content of cordyceps sinensis masks in the current market are effectively solved.
3. The formula of the cordyceps mask emulsion has two active effects of moisture retention and oxidation resistance, and the reasonability and effectiveness of the dispensing design are ensured by adding corresponding reagents to focus on the texture of the mask and the stimulation effect on the skin.
4. The invention will be better understood from the following examples. It is easily understood by those skilled in the art that the descriptions of the embodiments are only for illustrating the present invention and should not be construed as limiting the present invention as detailed in the claims. Materials, reagents and the like used in the following examples are commercially available unless otherwise specified. The experimental procedures, in which specific conditions are not indicated in the examples, are generally carried out under conventional conditions or conditions recommended by the manufacturer.
The idiotypic brevibacillus pinnata strain of the cordyceps sinensis is provided by food and pharmaceutical engineering colleges of south Beijing university of Sinkiang, and the effect of the idiotypic brevibacillus pinnata of any wild type cordyceps sinensis is basically the same.
Example 1
The moisture-retaining and antioxidant cordyceps mask emulsion in the embodiment is prepared from the following components in parts by weight: 12 parts of fine cordyceps polysaccharide extract, 7 parts of glycerol, 3 parts of hyaluronic acid, 0.5 part of carbomer, 0.3 part of allantoin, 0.1 part of methylparaben, 0.3 part of rose essence and 75 parts of deionized water.
Fermentation of cordyceps sinensis mycelia
Preparing a seed solution:
the seed liquid culture medium comprises: 20g/L glucose, 10g/L peptone, 0.5g/L magnesium sulfate heptahydrate and 2g/L potassium dihydrogen phosphate.
Inoculating the non-sexual type lobelia of the fine cordyceps on the test tube inclined plane into a shake flask for culturing for 7 days at the culture temperature of 20 ℃ and the rotating speed of a shaking table of 120rpm to obtain seed liquid.
Culturing in a fermentation tank:
inoculating the seed solution into a 5L fermentation tank filled with a fermentation tank culture medium according to the volume ratio of 5%, and performing fermentation culture; the culture conditions are as follows: temperature: 20 ℃, rotation speed: 150rpm, aeration: 50VVM, pH 6.5, fermentation time 14 days.
The culture medium of the fermentation tank is as follows: 20g/L glucose, 10g/L peptone, 0.5g/L magnesium sulfate heptahydrate and 2g/L potassium dihydrogen phosphate.
And (3) treating a fermentation product:
after the fermentation is finished, filtering with a 300-mesh filter screen, and keeping filter residues. Drying in a freeze drier for 2 days to obtain crude mycelia of Cordyceps sinensis Inonotus type Leucoporia.
The extraction method of the fine cordyceps polysaccharide extract comprises the following steps:
grinding: fully grinding and crushing the dried mycelium of the amorphous lobelia of the fine cordyceps sinensis.
Leaching: adding the ground cordyceps sinensis mycelium powder into hot water at 80 ℃ according to the material-liquid ratio (g: mL) of 1:20, and leaching for 3 hours in a constant-temperature water bath at 80 ℃. Centrifuging the solution at 8000rpm, 20 deg.C for 5min, removing precipitate, collecting supernatant, extracting the precipitate once again according to the same requirement, centrifuging under the same condition, and mixing the two supernatants.
Alcohol precipitation: the combined supernatants were concentrated to 1/3 volume and then placed in 95% ethanol at a volume ratio of 1: 4. The mixture was allowed to stand overnight at 4 ℃.
First deproteinization: after standing overnight, the supernatant was centrifuged at 9000rpm at 20 ℃ for 5min, and the precipitate was dissolved in 80 ℃ hot water. The dissolved liquid is centrifuged to remove the precipitate at 8000rpm, 20 ℃ and 3 min. Putting the supernatant into 95% ethanol according to the volume ratio of 1:4, and precipitating with ethanol at 4 ℃ for 2 h.
Second and third deproteinization: repeating the operation according to the first deproteinization requirement, and performing second and third deproteinization.
Crude polysaccharide: centrifuging the solution after the third deproteinization at 9000rpm at 20 deg.C for 5min to remove supernatant, dissolving the precipitate with 80 deg.C hot water, and freezing at-80 deg.C. Freeze drying to obtain coarse Cordyceps polysaccharide.
Separation and purification: dissolving crude Cordyceps polysaccharide in water, adding water into Sephadex G-200 gel, swelling, and filling into 1.6cm × 60cm chromatographic column, and controlling flow rate at constant flow pump to be 0.5 mL/min-1Then eluting with distilled water for collection. The absorbance was measured at 490nm by phenol-sulfuric acid method for detection, and the collected eluate was freeze-dried to obtain the refined cordyceps polysaccharide, i.e. the fine cordyceps polysaccharide extract used in this example.
Preparation of facial mask lotion
Preparation of phase A: weighing refined Cordyceps polysaccharide according to formula, dissolving in 80 deg.C deionized water, sequentially adding allantoin, glycerol, hyaluronic acid, methyl hydroxybenzoate, and flos Rosae Rugosae essence, stirring, and dissolving completely (small amount of deionized water can dissolve polysaccharide and other substances);
preparation of phase B: heating the rest deionized water to 80 deg.C, sequentially adding carbomer under stirring, stirring for 60min to make it fully swell, making into gel substance, and cooling to 30 deg.C.
Slowly adding the phase A into the stirred phase B, adjusting the pH value to 5 by using citric acid, and uniformly stirring to obtain the cordyceps mask emulsion.
Example 2
The moisture-retaining and antioxidant cordyceps mask emulsion in the embodiment is prepared from the following components in parts by weight: 8 parts of fine cordyceps polysaccharide extract, 7 parts of glycerol, 2 parts of hyaluronic acid, 0.3 part of carbomer, 0.1 part of allantoin, 0.1 part of rose essence, 0.05 part of methylparaben and 70 parts of deionized water.
Fermentation of cordyceps sinensis mycelia
The fermentation medium comprises 20g/L glucose, 10g/L peptone, 0.3g/L magnesium sulfate heptahydrate and 2g/L potassium dihydrogen phosphate.
The fermentation culture conditions are temperature of 20 deg.C, stirring speed of 200rpm, ventilation of 50VVM, pH 5, and fermentation time of 20 days.
Preparation of facial mask lotion
Preparation of phase A: weighing refined Cordyceps polysaccharide according to the above formula, dissolving in a certain amount of 60 deg.C deionized water, sequentially adding allantoin, glycerol, hyaluronic acid, methyl hydroxybenzoate, and flos Rosae Rugosae essence, and stirring to completely dissolve;
preparation of phase B: heating the rest amount of deionized water to 60 deg.C, sequentially adding carbomer under stirring, stirring for 30min to make it fully swell, making into gel substance, and cooling to 30 deg.C.
Slowly adding the phase A into the stirred phase B, adjusting the pH value to 6 by using citric acid, and uniformly stirring to obtain the cordyceps mask emulsion.
The other steps were the same as in example 1.
Example 3
The moisture-retaining and antioxidant cordyceps mask emulsion in the embodiment is prepared from the following components in parts by weight: 10 parts of fine cordyceps polysaccharide extract, 12 parts of glycerol, 3 parts of hyaluronic acid, 0.5 part of carbomer, 0.2 part of allantoin, 0.1 part of rose essence, 0.1 part of methylparaben and 80 parts of deionized water.
Fermentation of cordyceps sinensis mycelia
The fermentation medium comprises 30g/L glucose, 15g/L peptone, 0.5g/L magnesium sulfate heptahydrate and 3g/L potassium dihydrogen phosphate.
The fermentation culture conditions were 25 deg.C, stirring speed 100rpm, aeration 100VVM, pH 7, and fermentation time 14 days.
Preparation of facial mask lotion
Preparation of phase A: weighing refined Cordyceps polysaccharide according to the above formula, dissolving in a certain amount of 60 deg.C deionized water, sequentially adding allantoin, glycerol, hyaluronic acid, methyl hydroxybenzoate, and flos Rosae Rugosae essence, and stirring to completely dissolve;
preparation of phase B: heating the rest amount of deionized water to 80 deg.C, sequentially adding carbomer under stirring, stirring for 60min to make it fully swell, making into gel substance, and cooling to 30 deg.C.
Slowly adding the phase A into the stirred phase B, adjusting the pH value to 6 by using citric acid, and uniformly stirring to obtain the cordyceps mask emulsion.
The other steps were the same as in example 1.
Test example 1
1. ABTS free radical scavenging test
2.5ml of ABTS (7 mmol. multidot.L)-1) With 0.5mL of potassium persulfate (15 mmol. L)-1) Mixing, reacting at room temperature in dark for 12h, diluting with distilled water to give absorbance value of 0.70 at 734nm wavelength, transferring 0.2mL of the solution, and adding different concentrations (0.1-2.5 g.L)-1) The sample solution of the Leucocephala leucotricha polysaccharide (the fine Cordyceps polysaccharide extract prepared in example 1) and the sample solution of Cordyceps militaris polysaccharide (common Cordyceps militaris strain is adopted as the difference of the fermentation method in example 1), mixing uniformly, keeping the temperature at 25 deg.C for 15min, measuring the absorbance value at 734nm wavelength, using distilled water as blank group, and measuring each sample for 3 times in parallel.
Polysaccharide pairs ABTS+The calculated formula for the scavenging activity is:

By comparing the ABTS free radical clearance, it can be clearly seen that the clearance of the Leucospora leucotricha (anamorph of the fine Cordyceps) is significantly higher than that of the Cordyceps militaris as shown in FIG. 1.
2. Test for moisture retention
The test population selects 100 students of Nanjing university for trial, wherein the age of the volunteer is 19-22 years, and the symptoms are mainly manifested by dry peeling and dark skin on the face, and the 100 volunteers use the emulsion prepared in example 1. The test method comprises the following steps: after cleaning the face every morning and evening, the lotion is applied uniformly on the face until the lotion is absorbed, the application is continued for one month, the application result is observed, and the moisture content of the skin is tested; stipulating: an obvious effect is recorded when the moisture content of the skin is increased by more than or equal to 20 percent; an obvious effect is marked when the moisture content of the skin is increased by more than or equal to 10 percent; the effect is weak when the moisture content of the skin is increased by more than or equal to 5 percent; an increase of < 2% in skin moisture content is recorded as essentially no effect.
Note that: the test volunteers did not use any other brand and type of lotion or mask, etc. during the test; the results show that after one month, the obvious effect reaches 55 percent, the obvious effect reaches 32 percent, the effect is 8 percent weak, and the basically no effect is 5 percent; the obvious effect and the obvious effect reach 87 percent, which shows that the moisturizing effect is good.
Therefore, the emulsion prepared by the specific fine cordyceps polysaccharide extract is milk white semitransparent colloid, has uniform texture, good sticky feeling, easy coating, fineness, smoothness, moist and refreshing skin feeling and slight faint scent, has the pH value of about 5-6, and has good oxidation resistance and moisture retention.

2. The moisturizing and antioxidant cordyceps mask emulsion according to claim 1, wherein the fine cordyceps is a mycelium of cultured non-sexual type leucotrichum of the fine cordyceps by utilizing liquid submerged fermentation.
3. The moisture-preserving and antioxidant cordyceps mask emulsion according to claim 2, wherein the fermentation medium components preferably comprise 20-30g/L of glucose, 10-15g/L of peptone, 0.3-0.5g/L of magnesium sulfate heptahydrate and 2-3g/L of potassium dihydrogen phosphate.
4. The moisturizing and antioxidant cordyceps mask emulsion according to claim 2, wherein the fermentation culture conditions comprise a temperature of 20-25 ℃, a stirring speed of 100rpm-200rpm, a ventilation volume of 50-100VVM and a pH of 5-7, and the fermentation time is 14-20 days.

The moisturizing and antioxidant cordyceps mask emulsion according to any one of claims 1 to 4, wherein the fine cordyceps polysaccharide extract is prepared by extracting purified cordyceps polysaccharide from mycelia of nondestructive type leucotrichum of fine cordyceps by a hot water extraction method.
6. The moisturizing and antioxidant cordyceps mask emulsion according to claim 5, wherein the hot water extraction method comprises the following specific steps:
grinding: fully grinding and crushing the dried mycelium of the amorphous lobelia of the fine cordyceps sinensis;
leaching: adding the ground mycelium of the fine cordyceps sinensis anamorphic type lobium leucotrichum into hot water, leaching in a constant-temperature water bath, filtering, reserving filtrate, repeatedly leaching filter residue, and combining the filtrate;
alcohol precipitation: the concentrated filtrate was placed in ethanol and allowed to stand overnight.
Deproteinization: standing overnight, centrifuging to remove supernatant, dissolving precipitate with hot water, centrifuging to remove precipitate, and precipitating supernatant with ethanol; repeating the operation according to the first deproteinization, and performing second and third deproteinization;
crude polysaccharide: centrifuging the deproteinized solution to remove supernatant, dissolving the precipitate with hot water, and freeze drying to obtain crude Cordyceps polysaccharide;
separation and purification: dissolving the crude Cordyceps polysaccharide in water, and freeze drying the eluate collected by chromatographic column to obtain refined Cordyceps polysaccharide.
7. The preparation method of the moisturizing and antioxidant cordyceps mask emulsion as claimed in claim 1, which is characterized by comprising the following steps:
(1) preparation of phase A: weighing refined Cordyceps polysaccharide according to formula, dissolving in hot deionized water, sequentially adding allantoin, glycerol, hyaluronic acid, methyl hydroxybenzoate and essence, and stirring to dissolve completely;
(2) preparation of phase B: heating the rest deionized water, sequentially adding carbomer while stirring, stirring to make it fully swell, making into gel substance, and cooling.
(3) Phase a was slowly added to the stirred phase B, allowed to mix well, and the pH adjusted.

The moisturizing and antioxidant cordyceps mask emulsion as claimed in claim 7, wherein the fine cordyceps polysaccharide extract in step (1) is dissolved in hot water at 60-80 ℃, and allantoin, glycerin, hyaluronic acid, methylparaben and essence are sequentially added and fully stirred to be completely dissolved.
9. The moisturizing and antioxidant cordyceps mask emulsion according to claim 7, wherein the rest deionized water in the step (2) is heated to 60-80 ℃, carbomer is sequentially added while stirring, the stirring is carried out for 30-60min, the mixture is fully swelled, a gel substance is prepared, and the temperature is reduced by 30-40 ℃.
10. The moisturizing and antioxidant cordyceps mask lotion according to claim 7, wherein the pH of the solution obtained in step (3) is adjusted to 5 to 6 by citric acid.
