Abstract
本发明涉及生物技术领域,具体涉及一种益生元组合物、合生元组合物及其应用。所述的益生元组合物,含有固态的唾液酸和pH调节剂,1%(w/v)的益生元组合物水溶液pH终值为5.0‑7.5。所述的合生元组合物含有益生菌和所述的益生元组合物,其中,所述益生菌为干菌体和/或经过包埋的干菌体复合物。本发明既能保证益生菌的存活率,又能体现唾液酸的功效,有利于指导与推广唾液酸的固态应用。同时,方案简单方便,原料损耗小,有利于控制成本,并提高产品的市场竞争力。

技术领域
本发明涉及生物技术领域,具体涉及一种益生元组合物、合生元组合物及其应用。
背景技术
合生元一般由益生菌和益生元组成。其中,益生菌(Probiotics)是一类对宿主有益的活性微生物,是定植于人体肠道、生殖系统内,能产生确切健康功效从而改善宿主微生态平衡、发挥有益作用的活性有益微生物的总称。它是一类口服的有益的活菌制剂,但是在生产、运输、保存等过程中,益生菌将受到诸多因素的的影响而导致活菌数大幅下降。而益生元是一种可通过调节微生物菌群而有益于宿主健康的无生命的食品成分,是一种不被人体消化但可以选择性刺激肠道内有益菌的生长或增强有益菌活性的益菌制品。据研究,益生元的主要功效有:增殖有益菌,抑制有害菌;调节肠道菌群;增强免疫力;润肠通便,排毒美容抗衰老:促进钙、铁、镁、锌等矿物质吸收;改善脂质代谢,降低血脂和胆固醇;预防与调理肝脏疾病等。近年的研究发现,益生元在控制体重、预防与调理糖尿病、提高中药的生物利用率及改善抑郁和焦虑情绪方面也有重大价值。
N-乙酰神经氨酸也称唾液酸、燕窝酸,是一种九碳单糖,它以各种形式广泛存在于许多生物体内,随生物进化程度的增加,N-乙酰神经氨酸在生物体内的含量及其衍生物类型亦增加。哺乳动物中的N-乙酰神经氨酸集中存在于脑脊液和黏液,N-乙酰神经氨酸为神经氨酸,是细胞膜蛋白的重要组分,参与细胞表面多种生理功能,在调节人体生理、生化功能方面起到非常重要的作用,能促进婴儿、胎儿、儿童神经系统发育、提高记忆力和智力水平,减缓脑细胞衰老,减慢记忆力衰减,预防老年痴呆。同时,还可作为抵抗肠道致病菌的保护因子、提供益生元功能、激活免疫系统。唾液酸是燕窝分级的唯一明确标示的功效成分,常食能护肤养颜,令皮肤滑润洁白,减少脸部皱纹。
本领域公知,唾液酸在溶液中使用时,0.1%的浓度会使溶液pH值降至2-3左右,此环境必然不利于大多数益生菌的存活。但大多数的关于益生菌的研究方向集中于固体粉末状态的制剂,唾液酸是益生菌可以利用的一种优秀的益生元,并且一定剂量唾液酸的补充对于人体营养也是具有积极意义的,但是,本申请人在研究中发现,将唾液酸直接同益生菌混合时,会对益生菌产生杀伤作用,使益生菌的存活率受到较大损害,这种现象既影响产品的成本也影响产品在存放过程中的稳定性,这成为唾液酸作为益生元应用时的一大障碍。目前,亟需一种既能保障唾液酸摄入又能够降低唾液酸对益生菌杀伤作用,且工艺简便的方法,以拓宽唾液酸的固态应用。
发明内容
为了解决上述技术问题,本发明首先提供了一种益生元组合物,含有固态的唾液酸和pH调节剂,1%(w/v)的益生元组合物水溶液pH终值为5.0-7.5。
通常,pH的概念,都是基于溶液中氢离子的总数和总物质的量的比来理解,所以关于pH调节剂的作用,众所周知都是在水溶液中发挥作用。固体粉末冲剂领域中pH调节剂通常所起的作用在于,调节口感、增加崩解作用,或者在制备及冲调的过程中增强溶液体系的稳定性,可见,通常pH调节剂解决的都是固体粉末冲剂溶于水之后所产生的问题。本发明在研究中发现在固体粉末状态的食品或饮料冲剂中,pH调节剂的加入能够降低唾液酸带给益生菌的杀伤作用,保障含有益生菌的冲剂在存放过程中活菌的数量维持在令人能够接受的范围内。通过本申请的方案按照一定配比形成固体粉末益生元体系,其1%(w/v)的水溶液pH值范围在5.0-7.5。
其中,以所述益生元组合物的总重为基准,所述唾液酸的含量为1-70wt%,所述pH调节剂的含量为0.1-50wt%;
优选的所述唾液酸与pH调节剂的重量比为(1:1)-(16:1)。
作为优选,所述唾液酸的含量为10-70wt%,所述pH调节剂的含量为1-50wt%;
优选的,所述唾液酸与pH调节剂的重量比为(1:1)-(8:1);
优选的,所述唾液酸与pH调节剂的摩尔比为0.5-2。
此比例下的益生元组合物能够广泛应用于益生菌配伍,并且能够保障益生菌的活度。
作为优选,本发明对其所适用的pH调节剂进行了研究,优选所述pH调节剂为有机酸、无机酸、盐、碱中的一种或其混合物;
其中,所述有机酸为富马酸、偏酒石酸、柠檬酸、乳酸、苹果酸、L(+)-酒石酸和酒石酸、冰乙酸和乙酸、己二酸、牛磺酸、叶酸、泛酸中的一种或其混合物;
所述无机酸为磷酸和/或盐酸;
所述盐为富马酸一钠、柠檬酸钠、柠檬酸钾、柠檬酸一钠、磷酸盐、硫酸钙、乳酸钙、乙酸钠、山梨酸钾中的一种或其混合物;
所述碱为碳酸钠、碳酸钾、碳酸氢钠、碳酸氢钾、氢氧化钙、氢氧化钾、氢氧化钠、碳酸钙中的一种或其混合物。
更优选所述pH调节剂为氢氧化钠、氢氧化钾、碳酸钠、碳酸钾、碳酸氢钠、碳酸氢钾、乳酸钙、碳酸钙、山梨酸钾中的一种或其混合物。

其中,所述其他益生元单糖优选为L-鼠李糖、阿拉伯糖、果糖、赤藓糖、木糖、甘露糖、半乳糖中的一种或其混合物;
所述益生元寡糖为菊粉、低聚果糖、低聚半乳糖、葡聚糖、壳寡糖、水苏糖、棉籽糖、乳果糖、葡萄糖、岩藻糖、木糖、麦芽糖、蔗糖、乳糖、半纤维素、岩藻糖基乳糖、唾液酸乳糖、乳糖-N-四糖、乳糖-N-新四糖中的一种或其混合物。
本发明中所述固态,可以是粉末状、颗粒状等其他具有流动性的固体形态,组合物可以通过干法混合或者湿法混合造粒等简便操作得到,优选的,固态成分均为能够通过20目筛的粒径大小。
本发明进一步提供一种合生元组合物,含有益生菌和所述的益生元组合物,所述益生菌为干菌体和/或经过包埋的干菌体复合物。

具体实施方式
以下实施例用于说明本发明,但不用来限制本发明的范围。
在本发明的具体实施方式中,益生元组合物的制备方法具体为:将配方中的唾液酸、pH调节剂以及其他益生元的固态原料进行混合,其中实施例2为湿法混合,其他实施例为干法混合,混合均匀度CV值在10%以内。
合生元组合物的制备方法具体为:将混合均匀的益生元与益生菌干菌体按照配方中的比例进行干混,水活度在0.25aw左右。
实施例1
一种益生元组合物,包括以下重量百分比的成分:20wt%的唾液酸,碳酸钠3.5wt%,50wt%的低聚果糖,其余部分为麦芽糖。

每g合生元中含有,益生菌菌剂的活菌数为1.9×1010cfu/g,添加100mg,益生元添加量750mg,其中唾液酸含量为150mg,其余150mg采用麦芽糊精进行填充。
其他实施例可按照此方法配置成所需要的组合物。
对比例
对比例中SA和活菌数与实施例的对应关系见下表2,其中,与对应实施例相比,对比例1-9中无pH调节剂,pH调节剂的成分全部由填充剂替代。

试验例取实施例及对比例中的合生元组合物作为样品,进行货架期实验,在分装后采用铝箔袋密封包装,室温、湿度75%的条件下,存放30天后检测菌活度。实验结果见下表3。

可以从实施例看到,添加有pH调节剂的益生元组合物在于益生菌混合后,益生菌的活度稳定性有较大的提升。
虽然,上文中已经用一般性说明、具体实施方式及试验,对本发明作了详尽的描述,但在本发明基础上,可以对之作一些修改或改进,这对本领域技术人员而言是显而易见的。因此,在不偏离本发明精神的基础上所做的这些修改或改进,均属于本发明要求保护的范围。
Claims (10)
1.一种益生元组合物,其特征在于,含有固态的唾液酸和pH调节剂,1%(w/v)的益生元组合物水溶液pH终值为5.0-7.5。
2.根据权利要求1所述的益生元组合物,其特征在于,以所述益生元组合物的总重为基准,所述唾液酸的含量为1-70wt%,所述pH调节剂的含量为0.1-50wt%,优选的,所述唾液酸与pH调节剂的重量比为(1:1)-(16:1)。
3.根据权利要求2所述的益生元组合物,所述唾液酸的含量为10-70wt%,所述pH调节剂的含量为1-50wt%,所述唾液酸与pH调节剂的重量比为(1:1)-(8:1)。
4.根据权利要求1~3中任一项所述的益生元组合物,其特征在于,所述pH调节剂为有机酸、无机酸、盐、碱中的一种或其混合物;
其中,所述有机酸为富马酸、偏酒石酸、柠檬酸、乳酸、苹果酸、L(+)-酒石酸和酒石酸、冰乙酸和乙酸、己二酸、牛磺酸、叶酸、泛酸中的一种或其混合物;
所述无机酸为磷酸和/或盐酸;
所述盐为富马酸一钠、柠檬酸钠、柠檬酸钾、柠檬酸一钠、磷酸盐、硫酸钙、乳酸钙、乙酸钠、山梨酸钾中的一种或其混合物;
所述碱为碳酸钠、碳酸钾、碳酸氢钠、碳酸氢钾、氢氧化钙、氢氧化钾、氢氧化钠、碳酸钙中的一种或其混合物。
5.根据权利要求4所述的益生元组合物,其特征在于,所述pH调节剂为氢氧化钠、氢氧化钾、碳酸钠、碳酸钾、碳酸氢钠、碳酸氢钾、碳酸钙、乳酸钙、山梨酸钾中的一种或其混合物。
6.根据权利要求1~5中任一项所述的益生元组合物,其特征在于,所述益生元组合物还含有固态的其他益生元单糖和/或益生元寡糖;
所述其他益生元单糖优选为L-鼠李糖、阿拉伯糖、果糖、赤藓糖、木糖、甘露糖、半乳糖中的一种或其混合物;
所述益生元寡糖优选为菊粉、低聚果糖、低聚半乳糖、葡聚糖、壳寡糖、水苏糖、棉籽糖、乳果糖、葡萄糖、岩藻糖、木糖、麦芽糖、蔗糖、乳糖、半纤维素、岩藻糖基乳糖、唾液酸乳糖、乳糖-N-四糖、乳糖-N-新四糖中的一种或其混合物。
7.一种合生元组合物,其特征在于,含有益生菌和权利要求1~6中任一项所述的益生元组合物,所述益生菌为干菌体和/或经过包埋的干菌体复合物。
8.根据权利要求7所述的合生元组合物,其特征在于,所述益生菌与唾液酸的用量比为(1×107cfu-1×1012cfu):(50mg-200mg);或,所述益生菌与唾液酸的用量比为(1×102cfu-1×107cfu):(1mg-50mg)。
9.根据权利要求7或8所述的合生元组合物,其特征在于,所述益生菌为有益细菌和/或有益真菌;
所述有益细菌选自乳杆菌属、双歧杆菌属、链球菌属、乳球菌属、丙酸杆菌属、明串球菌属、片球菌属、芽孢杆菌属、葡萄球菌属中的一种或其混合物;
所述有益真菌选自酿酒酵母、产朊假丝酵母、乳酸克鲁维酵母、卡氏酵母、蝙蝠蛾拟青霉、蝙蝠蛾被毛孢灵芝、紫汁、红曲霉、紫红曲霉中的一种或其混合物。
10.一种食品或药物组合物,其特征在于,含有权利要求1~6中任一项所述的益生元组合物或权利要求7~9中任一项所述的合生元组合物。
