Abstract

技术领域

背景技术
多年以来已知,当皮肤暴露在阳光下时,会在皮肤中产生脂质过氧化物(也称为脂质氢过氧化物或脂过氧化物)。这不仅是因为曾经想到的UVB波长,还归因于UVA、红外和可见光波长。最近的研究发现,几种大气污染物(臭氧、二氧化氮、颗粒物和多环碳氢化合物)和香烟烟雾也会在皮肤中产生脂质过氧化物。
脂质过氧化物被认为是衰老过程中在皮肤中发生的变化的重要原因。对于美容效果(例如皱纹、粗糙、变薄和褐斑)和所有三种主要类型的皮肤癌(它们在世界范围内的发病率在增加,并且共同地是最常见的癌症形式)都是如此。
脂质过氧化物是通过活性氧(ROS)氧化脂质而产生的。通过在皮肤中产生ROS,太阳辐射(例如阳光)、大气污染物和香烟烟雾诱导脂质过氧化物的形成。脂质过氧化物及其衍生的某些分子(例如MDA、4-HNE)对细胞膜有毒、破坏细胞之间的结构蛋白(例如胶原蛋白和弹性蛋白)、抑制某些抗氧化酶(例如超氧化物歧化酶)、增加细胞的基质金属蛋白酶(其消化胶原蛋白)的分泌并损伤DNA。所有这些作用被认为促成衰老过程中皮肤的美容变化,而对DNA的损伤则是所有类型癌症的原因。在严重暴露于阳光下后,脂质过氧化物还促成皮肤的发红、酸痛和疼痛(晒伤)。脂质过氧化物具有急性作用(例如炎症、疼痛和晒伤)和慢性作用(例如对细胞、蛋白质、DNA的损伤;酶活性改变;皮肤老化;和癌症)。

上述肽或用于皮肤护理产品的任何其他肽均未已知螯合脂质过氧化物。
已经讨论了脂质过氧化物的促炎性质对疾病特别是心血管系统的疾病的发病机理的潜在贡献(Navab M.Circ Res 2005;97:524-32)。但是,尚未考虑它们在皮肤的年龄相关变化中的作用及其美容效果,其不是炎症的结果,而是DNA完整性、胶原蛋白分解、弹性蛋白合成和酶活性的变化的结果。组织的老化,包括皮肤的老化,不是疾病,而是不可避免的转变(Rattan SIS.Aging and Disease 2014;5:196-202),尽管其速率可能受环境因素的影响。
原则上,可以通过以下三种机制中的一种或多种来防止脂质过氧化物的积累,以防止它们对皮肤的组成和结构的长期影响:降低其产生,增加其分解,或以消除它们的生物活性的方式来螯合它们。防晒霜和抗氧化剂(例如维生素E)可部分减少产量(RinnerthalerM.Biomolecules 2015;5,545-589)。从理论上讲,通过施加适当的酶可以增强分解,尽管考虑到脂质过氧化物的可变性质,仍需要几种酶。此外,当局部施加时它们不太可能进入皮肤,并且所得的终产物的作用尚不清楚。另一方面,分子螯合具有几个吸引力。
脂质过氧化物的有效螯合剂在皮肤中的基本特性是:该螯合剂(a)对它们具有高结合亲和力,以及(b)可以进入表皮和浅表真皮。渗透到皮肤中的两个主要决定因素是分子尺寸和亲脂性。分子的尺寸越小和亲脂性越大,渗透性就越大。不可逆地结合脂质过氧化物的酰化四肽理想地适合此目的。此外,这样的分子将具有与细胞的表面膜结合的附加性质(Resh MD.Current Biology 2013;23:R431-5),从而防止其进入血液并确保肽-过氧化物复合物最终在角质形成细胞的正常脱屑过程中脱落。该机制还避免了随之而来的其急性和慢性作用未知的代谢产物的产生。理想地,这样的试剂还应该是可生物降解的,在长期使用期间不太可能引起过敏,并且不引起皮肤中维生素D产生的减少。
进入皮肤的连续过程、脂质过氧化物的螯合、与细胞表面的结合以及通过脱屑的最终损失提供了一种机制,通过该机制使这样的分子在物理上“清洁”皮肤而不会产生有害的局部或全身性生化作用。
载脂蛋白模拟肽(AMP)是短链氨基酸,其模拟载脂蛋白的一些特性,载脂蛋白是血浆脂蛋白(在血液中携带脂肪的颗粒)的蛋白质成分。由于它们具有结合胆固醇和脂质过氧化物的能力,它们首先引起了心血管疾病研究人员的兴趣,胆固醇和脂质过氧化物在动脉壁上的局部积聚促成动脉粥样硬化(冠状动脉疾病的病因)的发展。当通过AMP螯合胆固醇和脂质过氧化物时,它们变得无害。
AMP在体外对脂质过氧化物的结合亲和力是载脂蛋白的亲和力的数千倍(VanLenten等人,J Lipid Res 2008;49:2302)。AMP与非氧化脂质的结合不会干扰脂质过氧化物的结合。AMP不是天然存在的,并且其还没有商业化的开发。
大多数关于AMP的学术研究是使用模拟特定载脂蛋白(称为apo A1)的活性的肽完成的,载脂蛋白apo A1是高密度脂蛋白(所谓的“好”胆固醇的载体)的主要蛋白成分。由于apo A1似乎从动脉壁除去胆固醇并将其带到肝脏,因此希望模拟其作用的AMP的静脉输注会逆转动脉粥样硬化。但是,还没有成功开发用于此用途或任何其他用途的AMP。
最初在心血管研究人员中引起最大兴趣的AMP是具有以下氨基酸序列的18个氨基酸链,其被命名为18A:DWLKAFYDKVAEKLKEAF(Anantharamaiah等人,J Biol Chem 1985;260:10248)。
学术团体随后的工作产生了逐渐减小的具有相似生物活性的仅几个氨基酸的AMP,包括具有四个氨基酸的两种肽:Lys-Arg-Glu-Ser(KRES)和Phe-Arg-Glu-Leu(FREL)(Navab等人,Circ Res 2005;97:524)。
在制药行业对AMP逆转冠状动脉疾病的潜力感兴趣的时候,在1期临床研究中将与18A肽紧密相似的18个氨基酸的AMP静脉内输注到人体内,但没有进行进一步的研究。当发现它很快从血液中清除时(可能是通过肾脏,这是消除大多数天然肽的生理途径)兴趣下降。没有报道不良反应。
尽管AMP共有载脂蛋白的一些物理化学和生物学特性,但它们并不共有其氨基酸序列的任何部分(即它们不是载脂蛋白的小片段的等同物)。它们是人工分子,与它们共有的性质对应的蛋白质没有结构相似性。

实施例1

实施例2

Claims (20)
1.一种组合物,其包含(a)式(I)的酰化肽
R-X1-X2-X3-X4(I)
其皮肤病学上可接受的盐、溶剂化物和/或对映异构体,其中R代表具有适合于帮助式(I)的化合物渗透皮肤的链长度的脂肪酸残基,X1代表碱性氨基酸或疏水性芳族氨基酸,X2代表碱性氨基酸,X3代表酸性氨基酸,和X4代表疏水性脂族氨基酸或极性中性氨基酸;和(b)皮肤病学上可接受的载体。
2.权利要求1所述的组合物,其中X1代表苯丙氨酸、酪氨酸、色氨酸、组氨酸、赖氨酸或精氨酸;X2代表组氨酸、赖氨酸或精氨酸;X3代表天冬氨酸或谷氨酸;和/或X4代表丙氨酸、缬氨酸、甲硫氨酸、亮氨酸、异亮氨酸、苏氨酸、谷氨酰胺、半胱氨酸、天冬酰胺或丝氨酸。
3.权利要求1或2所述的组合物,其中X1具有疏水性芳族侧链,X4具有疏水性脂族侧链。
4.前述权利要求中任一项所述的组合物,其中X1是碱性氨基酸,X4具有极性中性侧链。
5.前述权利要求中任一项所述的组合物,其中R代表式R2C(O)-的脂肪酸残基,其中R2代表具有12至24个碳原子的饱和或不饱和直链或支链烷基;优选地,R代表棕榈酰基或肉豆蔻酰基。
6.前述权利要求中任一项所述的组合物,其中式(I)的酰化肽可以是式(IA)或(IB)的酰化肽

8.前述权利要求中的任一项所述的组合物,其包含另外的皮肤护理剂;优选地,所述另外的皮肤护理剂是以下的一种或多种:脱屑剂、抗痤疮剂、皱纹修复剂、抗氧化剂、自由基清除剂、螯合剂、抗炎剂、局部麻醉剂、抗脂肪团剂、类黄酮、抗微生物剂、抗真菌剂、防晒剂和/或调理剂。
9.权利要求8所述的组合物,其中所述另外的皮肤护理剂包括防晒剂。
10.前述权利要求中任一项所述的组合物,其是皮肤护理组合物,其包含(a)安全且有效量的式(I)的肽,和(b)一种或多种皮肤病学上可接受的载体。
11.前述权利要求中任一项所述的组合物,其包含浓度为约0.1mg/ml至约1000mg/ml的式(I)的肽。
12.式(II)的酰化肽
R1-X1-X2-X3-X4(II)
其皮肤病学上可接受的盐、溶剂化物和/或对映异构体,其中R1代表具有适合于帮助式(II)的化合物渗透皮肤的链长度的脂肪酸残基,并且其中X1、X2、X3和X4是如关于式(I)的肽的权利要求1-4中的任一项所定义的。
13.权利要求12所述的酰化肽,其是式(IIA)或(IIB)的酰化肽
