Abstract
本发明公开了一种含有锌螯合物的水凝胶材料,本发明亦提供这种水凝胶的制造方法。本发明提供的水凝胶具有亲水性和生物相容性,实现锌元素在体表的长效停留吸收,可应用于多种皮肤和毛发护理产品中。

技术领域
本发明涉及一种可用于口腔、皮肤、毛发护理产品的水凝胶,特别涉及一种聚乙二醇(PEG)修饰的赖氨酸锌螯合物及其制备方法。
背景技术
锌是人体所必需的微量元素。已知锌在口腔、皮肤、毛发护理领域有抗菌消炎、控油、祛头屑皮屑等功效。现有的护理产品中锌往往以盐类或螯合物、络合物的形式添加,这类添加物易于随水洗、排汗流失,难以长期作用。
本发明提供了一种具有生物亲和性的水凝胶材料,该材料搭载了锌的螯合物,可以实现锌元素在体表的长效停留吸收,与日化产品中各种常用成分配伍性好,可应用于多种口腔、皮肤和毛发护理产品中。
发明内容
本发明的技术方案如下:
1. 一种含有锌螯合物的水凝胶材料,其特征在于具有以下结构:
其中n=3-11。
2. 所述螯合物中,锌的配位数为4,参与配位的基团为赖氨酸中的α氨基和氧。
3. 所述水凝胶材料的合成方法为:

具体步骤为:(I)A与B在水中反应,得到C的水溶液,C的水溶液经过滤后浓缩结晶,过滤收集结晶,经无水乙醇洗涤后真空干燥得到固体C。(II)将聚乙二醇(PEG)加热溶解于溶剂,氯化亚砜(SOCl2)和吡啶按1:1的摩尔比溶于相同溶剂,在氮气保护下按PEG:SOCl2=0.48:1的摩尔比缓慢滴加PEG溶液,50℃下回流反应4h,加碱中和后抽滤、旋蒸,得到化合物E。(III)C与E溶解于二甲基亚砜(DMSO)中,60℃下反应3h,得到絮状不溶物,抽滤后分别用DMSO和无水乙醇清洗沉淀,真空干燥后得到产物F,F经吸水溶胀后得到水凝胶。
4.所述反应(I)中,化合物A为赖氨酸或赖氨酸的盐酸盐,化合物B为氧化锌、氯化锌、氢氧化锌、醋酸锌或其组合物,A与B的摩尔比为3:1-2:1。
5.所述反应(I)中,反应温度为40-60℃,pH为6-8,pH调节剂为稀盐酸或氨水。
6.所述反应(II)中,溶剂为二氯甲烷、四氢呋喃或其组合物。
7.所述反应(II)中,加入的碱为碳酸钠、碳酸钾或其组合物。
具体实施方式
以下通过实施例进一步说明本发明
实施例1:
步骤(I),赖氨酸盐酸盐与氯化锌按2:1的摩尔比溶于去离子水,加入氨水调节pH=8,60℃下反应1h,抽滤后滤液加热浓缩至有晶膜出现,冷却静置后抽滤,固体经无水乙醇洗涤后真空干燥得到产物C。(收率89.5%)。步骤(II),0.48当量的PEG(聚合度=6)加热溶解于20倍质量的二氯甲烷并冷却至室温,1当量的氯化亚砜和1当量的无水吡啶溶解于20倍氯化亚砜质量的二氯甲烷,氮气保护下将PEG溶液逐滴加入氯化亚砜溶液中,加料完成后在50℃下回流反应4h,加入0.02当量的碳酸钾,抽滤后滤液经旋蒸得到产物E(收率92.1%)。步骤(III)1当量的C与1当量的E溶解于20倍C质量的DMSO中,60℃下反应3h,C与E溶解于二甲基亚砜(DMSO)中,60℃下反应3h,得到絮状不溶物,抽滤后分别用DMSO和无水乙醇清洗沉淀,真空干燥后得到产物F(收率80.7%),F经吸水溶胀后得到水凝胶。
实施例2:
步骤(I)氢氧化锌粉体加入去离子水中,持续搅拌,滴加赖氨酸盐酸盐的水溶液,同时滴加氨水保持pH=7-8,待粉体接近完全溶解后停止加料,抽滤后滤液真空干燥得到产物C(收率93.3%)。步骤(II),0.48当量的PEG(聚合度=8)加热溶解于20倍质量的二氯甲烷并冷却至室温,1当量的氯化亚砜和1当量的无水吡啶溶解于20倍氯化亚砜质量的二氯甲烷,氮气保护下将PEG溶液逐滴加入氯化亚砜溶液中,加料完成后在60℃下回流反应2h,加入0.02当量的碳酸钾,抽滤后滤液经旋蒸得到产物E(收率82.7%)。步骤(III)1当量的C与1当量的E溶解于30倍C质量的DMSO中,60℃下反应3h,得到絮状不溶物,抽滤后分别用DMSO和无水乙醇清洗沉淀,真空干燥后得到产物F(收率84.0%),F经吸水溶胀后得到水凝胶。
实施例3:
赖氨酸与氯化锌按2:1的摩尔比溶于去离子水, 60℃下反应1h,抽滤后滤液加热浓缩至有晶膜出现,冷却静置后抽滤,固体经无水乙醇洗涤后真空干燥得到产物C。(收率81.0%)。步骤(II),0.48当量的PEG(聚合度=6)加热溶解于20倍质量的二氯甲烷并冷却至室温,1当量的氯化亚砜和1当量的无水吡啶溶解于20倍氯化亚砜质量的二氯甲烷,氮气保护下将PEG溶液逐滴加入氯化亚砜溶液中,加料完成后在40℃下回流反应8h,加入0.02当量的碳酸钾,抽滤后滤液经旋蒸得到产物E(收率84.0%)。步骤(III)1当量的C与1当量的E溶解于40倍C质量的DMSO中,60℃下反应3h,得到絮状不溶物,抽滤后分别用DMSO和无水乙醇清洗沉淀,真空干燥后得到产物F(收率72.9%),F经吸水溶胀后得到水凝胶。

4.所述反应(I)中,化合物A为赖氨酸或赖氨酸的盐酸盐,化合物B为氧化锌、氯化锌、氢氧化锌、醋酸锌或其组合物,A与B的摩尔比为3:1-2:1。
