技术领域
本发明涉及化妆品技术领域,具体涉及一种美白组合物及其制备工艺与应用。
背景技术
皮肤是人体的重要组成部分,更是直接表达人体健康与衰老的标志。当皮肤代谢旺盛,血液循环畅通,营养丰富时,则皮肤细腻、光滑、柔嫩,极富弹性,且不易出现皱纹和色斑。当皮肤代谢减弱,血液循环不畅,营养不良时,皮肤会灰暗、衰老,且易产生皱纹及色斑。
黑色素是决定人体肤色的重要因素,它在表皮基底层黑色素细胞中的黑色素体中生物合成。正常生理状态下,皮肤中黑色素可以保护皮肤免受紫外线伤害。当外部侵害、激素失调、老化或其他因素造成黑色素合成代谢机制障碍时,易使表皮基底层的黑色素增多,部分或全身皮肤色泽加深,引起雀斑、黄褐斑、妊娠斑、老年斑、黑色素瘤等色素沉着类皮肤病,影响了人的容貌。
色素沉着及色斑产生的原因比较复杂,如遗传因素、内分泌失调、环境污染、紫外线照射等,无论是哪一种原因引起的色素沉着和色斑,归根结底均是由黑色素沉着造成的,因此皮肤美白祛斑的主要途径是抑制黑色素的产生。抑制黑色素的产生有多种方法,如抑制酪氨酸酶的活力、清除活性氧或自由基、减少紫外线照射、阻止氧化反应发生等,均是减少黑色素形成的有效方法。
人们通过在化妆品中添加各种美白祛斑类物质,减轻或消除色斑,从而达到美白祛斑的效果。较早的美白祛斑成分含有汞及其化合物或氢醌,由于毒性、刺激性较大,目前已经被国家禁用。
目前,公知的美白祛斑原料的机理有:1.抑制黑色素基因表达,如美白肽等;2.络合铜离子,如植酸、曲酸衍生物等;3.抑制黑色素体的转移到角质层,如烟酰胺等;4.抑制黑色素生成,如乳酸、乳酸钠、甘醇酸等;5.加速酪氨酸酶降解,如亚油酸及乙酯等;6.还原性及竞争性酪氨酸酶活性,如苯乙基间苯二酚、光甘草啶等;7.还原性黑色素,如维生素C及其衍生物等。
大部份原料都是在某一单一的或几种机理结合上去实现美白淡斑的目的,往往把它运用到化妆品中的功能效果就不尽人意,要不然就是原料自身就会有这样或那样的缺点,像原料不稳定,或者很难加入到配方体系中,给配方师带来极大困惑,或者容易变色,或者对皮肤刺激性偏大,导致过敏率偏高,或者是消费者使用下来,没有效果等。
中国专利申请CN110721104A公开了一种美白霜及其制备方法。该发明美白霜包括以下制备原料:烟酰胺、白藜芦醇、3-o-乙基抗坏血酸、牡丹提取物、二葡糖基棓酸、鲟鱼子酱提取物、磷脂酰胆碱、神经酰胺3、对羟基苯乙酮、生育酚乙酸酯、透明质酸钠、保湿剂7-10%、润肤剂、乳化剂、增稠剂、中和剂、香精和水。该发明美白霜可从黑色素生成诱因、黑色素生成过程、黑色素转移到最后黑色素消化排出过程等多途径起到作用,实现快速美白祛斑的目的。但是该专利申请的产品美白效果有待进一步提高,且原料众多,制备成本较高。
中国专利申请CN110917051A一种美白精华液及其制备方法。该发明美白精华液包括以下重量份数的制备原料:尿囊素、透明质酸钠、九肽-1、烟酰胺、3-o-乙基抗坏血酸、甘草酸二钾、曲酸、肌肽、光果甘草提取物、葡萄叶提取物、水解胶原、马齿苋提取物、七叶树提取物、D-泛醇、多元醇、增稠剂、防腐剂和水。本发明美白精华液可通过多种作用机理实现高效、全面的美白效果,特别是抑制由太阳光照射引起的黑色素沉着,同时也为皮肤提供保湿、抗衰、修复等多种护肤功效。但是该专利申请含有多种植物提取物,原料众多,制备成本较高,且各组分之间容易相互作用造成过敏情况的发生。
因此,利用开发一种能解决上述技术问题的美白组合物及其制备工艺与应用是非常必要的。
发明内容
本发明的目的是克服现有技术的不足而提供一种美白祛斑效果显著、能够改善皮肤耐受性且制备工艺简单、生产成本较低的美白组合物及其制备工艺与应用。
本发明是通过以下技术方案予以实现的:
一种美白组合物,包括如下组分:二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、欧囊链藻提取物、肌肽、光果甘草根提取物、生物糖胶和玉米谷蛋白氨基酸类。
优选地,所述的美白组合物,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物1-10份、1-甲基乙内酰脲-2-酰亚胺2-8份、烟酰胺1-8份、欧囊链藻提取物2-8份、肌肽1-6份、光果甘草根提取物2-8份、生物糖胶2-10份和玉米谷蛋白氨基酸类3-10份。
更优选地,所述的美白组合物,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物2-8份、1-甲基乙内酰脲-2-酰亚胺3-6份、烟酰胺2-6份、欧囊链藻提取物3-7份、肌肽2-5份、光果甘草根提取物2-8份、生物糖胶3-8份和玉米谷蛋白氨基酸类4-8份。
更优选地,所述的美白组合物,按照重量份数计,还包括泛醇0.5-2份、甘油2-10份、黄原胶0.1-1份和去离子水100份。
更优选地,所述欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类的质量比为1:1.2-2:2.5-4。
本发明还涉及上述的美白组合物的制备工艺,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,搅拌至完全溶解,得到混合物1;
(2)在混合物1中加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、欧囊链藻提取物、肌肽、光果甘草根提取物、生物糖胶、玉米谷蛋白氨基酸类和泛醇,即得。
优选地,步骤(1)中加入去离子水、甘油和黄原胶后,升温至80-90℃,搅拌至完全溶解,得到混合物1。
优选地,步骤(2)中将混合物1降温至40-55℃后,先加入欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类,再加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、肌肽、生物糖胶和泛醇,搅拌均匀后得到透明料体,即得。
本发明还涉及一种美白精华液,包括上述的美白组合物或上述的制备工艺制备得到的美白组合物。
本发明还涉及上述的美白组合物或上述的制备工艺制备得到的美白组合物在制备化妆品中的应用。
本发明的有益效果是:
本发明通过抑制α-MSH的前体“POMC”的合成,从而减少黑色素的合成;激活AGRP基因表达,结合α-MSH受体,从而阻止由于α-MSH导致的黑色素生成。
本发明抑制黑素体成熟相关基因“PMEL前黑素体蛋白”的表达,及刺激“酪氨酸酶抑制剂”的表达,从而减少黑色素的合成。
本发明通过降低“角质层胰蛋白酶类酶”的表达,限制黑素体的转移。
本发明通过激活“组织蛋白酶”,促进黑素体降解;针对已经形成的黑色素沉着、固有色素暗沉、顽固黑斑可高效瓦解,预防及击退黑斑玉色素暗沉,从而改善皮肤色素沉着度和肤色均匀性。
本发明优化了组合物中各组分的组成及其用量配比,各组分协同作用,美白祛斑效果显著,对皮肤无刺激,且稳定性较好。
本发明组合物中欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类协同作用,美白祛斑效果进一步提高。

一种美白精华液,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物1份、1-甲基乙内酰脲-2-酰亚胺2份、烟酰胺1份、欧囊链藻提取物2份、肌肽1份、光果甘草根提取物2份、生物糖胶2份、玉米谷蛋白氨基酸类3份、泛醇0.5份、甘油2份、黄原胶0.1份和去离子水100份。
美白精华液的制备工艺,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,升温至80℃,搅拌至完全溶解,得到混合物1;
(2)将混合物1降温至40℃后,先加入欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类,再加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、肌肽、生物糖胶和泛醇,搅拌均匀后得到透明料体,即得。

一种美白精华液,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物10份、1-甲基乙内酰脲-2-酰亚胺8份、烟酰胺8份、欧囊链藻提取物8份、肌肽6份、光果甘草根提取物8份、生物糖胶10份、玉米谷蛋白氨基酸类10份、泛醇2份、甘油10份、黄原胶1份和去离子水100份。
美白精华液的制备工艺,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,升温至90℃,搅拌至完全溶解,得到混合物1;
(2)将混合物1降温至55℃后,先加入欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类,再加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、肌肽、生物糖胶和泛醇,搅拌均匀后得到透明料体,即得。
实施例3
一种美白精华液,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物5份、1-甲基乙内酰脲-2-酰亚胺5份、烟酰胺4份、欧囊链藻提取物5份、肌肽3份、光果甘草根提取物4份、生物糖胶6份、玉米谷蛋白氨基酸类7份、泛醇1份、甘油6份、黄原胶0.5份和去离子水100份。
美白精华液的制备工艺,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,升温至85℃,搅拌至完全溶解,得到混合物1;
(2)将混合物1降温至45℃后,先加入欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类,再加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、肌肽、生物糖胶和泛醇,搅拌均匀后得到透明料体,即得。
实施例4
一种美白精华液,与实施例3的区别仅在于欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类的质量比不同,其余条件均相同,具体如下:
一种美白精华液,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物5份、1-甲基乙内酰脲-2-酰亚胺5份、烟酰胺4份、欧囊链藻提取物3.4份、肌肽3份、光果甘草根提取物4.08份、生物糖胶6份、玉米谷蛋白氨基酸类8.52份、泛醇1份、甘油6份、黄原胶0.5份和去离子水100份。
实施例5
一种美白精华液,与实施例3的区别仅在于欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类的质量比不同,其余条件均相同,具体如下:
一种美白精华液,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物5份、1-甲基乙内酰脲-2-酰亚胺5份、烟酰胺4份、欧囊链藻提取物2.3份、肌肽3份、光果甘草根提取物4.6份、生物糖胶6份、玉米谷蛋白氨基酸类9.1份、泛醇1份、甘油6份、黄原胶0.5份和去离子水100份。
对比例1
与实施例3的区别仅在于美白精华液的组成不同,其余条件均相同,具体如下:

与实施例3的区别仅在于烟酰胺替换为等质量的维生素C,其余条件均相同。
对比例3
与实施例3的区别仅在于美白精华液制备过程中,原料的加入顺序不同,具体如下:
美白精华液的制备工艺,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,升温至85℃,搅拌至完全溶解,得到混合物1;
(2)将混合物1降温至45℃后,先加入欧囊链藻提取物、烟酰胺和玉米谷蛋白氨基酸类,再加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、光果甘草根提取物、肌肽、生物糖胶和泛醇,搅拌均匀后得到透明料体,即得。
测试例1
1、多次/急性皮肤刺激试验
检测方法:参照(化妆品卫生规范)2007卫生部卫生法制与监督司规定检测;
实验动物:豚鼠(雄/雌),体重250-300g,采用实施例1-5和对比例1-3的产品进行试验,每组5只;
温度:20-24℃,湿度:60-70%;
7天后,均无红肿、水肿现象,说明实施例1-5和对比例1-3的产品安全有效,无任何刺激。
2、急性经口毒性试验
试验方法:昆明小鼠,体重18-22g,雌雄各半,实施例1-5和对比例1-3每组10只,试验温度20-24℃,湿度60-70%,禁食时间,小鼠给药前禁食16小时,动物一次经口服5000mg/kg美白精华液后,小鼠活动及饮食基本正常,连续观察7天,体重正常,未出现明显毒性反应及动物死亡,说明本发明制备的产品安全,无毒性。
3、美白效果研究
3.1细胞增值率测定(MTT法)
培养小鼠黑色素瘤K111细胞,待细胞生长到对数期,用0.25%胰酶消化,调整细胞浓度为2×105个/mL,接种于96孔板中,每孔100μL。置于培养箱中培养24h后,弃上清液,添加含药培养基,样品终浓度为0.15wt%,设3个复孔,对照组不加药物,代之以相应的溶酶。孵育48小时后,每孔加入5g/L的MTT溶液20μL,4h后弃上清液,每孔加入细胞裂解液100μL,继续培养过夜,在酶标仪上检测各孔吸光度值,波长为570nm。细胞增殖抑制率%=(1-药物组平均吸光度值/对照组平均吸光度值)×100%,计算细胞增殖抑制率%。
3.2细胞内酪氨酸酶活力测定
细胞培养方法同上,样品作用终浓度样品浓度为0.15wt%;药物作用3天,弃去上清液,用pH7.0的PBS冲洗两遍,每孔加90μL含1%Triton X-100的PBS。冰浴中超声破碎振荡5min溶解细胞,经37℃预温后每孔加入1%的L-DOPA 10μL,37℃孵育60min,测量490nm各孔吸光度值。酪氨酸合成抑制率%=(1-药物组平均吸光度/对照组平均吸光度)×100%。
检测结果如表1所示。

4、皮肤耐受性试验
测试方法:随机选取60为女性志愿者,年龄22-35岁,分为6组每组10人,分别对实施例3和实施例4-5以及对比例1-3制备的精华液进行试用,试用面积200-250cm2,试用量0.3-0.5mL,试用部位:脸部,并且对相关指标进行打分,最高10分,结果如表2所示。

5、皮肤亮度和皮肤亮白度实验
采用色彩色差计CR-400(广东七彩仪器设备有限公司)分析使用实施例3的精华液的志愿者皮肤亮度L*值、皮肤亮白度ITA°值的变化情况,试用面积200-250cm2,试用量0.3-0.5mL,试用部位:脸部,实验组为涂抹精华液的志愿者皮肤,空白组为未涂抹精华液的志愿者皮肤,结果分别如图1和图2所示。另外,实施例4和5与实施例3相比,皮肤亮度和亮白度显著提高,对比例1-3与实施例3相比,皮肤亮度和亮白度显著降低。

Claims (7)
1.一种美白组合物,其特征在于,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物1-10份、1-甲基乙内酰脲-2-酰亚胺2-8份、烟酰胺1-8份、欧囊链藻提取物2-8份、肌肽1-6份、光果甘草根提取物2-8份、生物糖胶2-10份和玉米谷蛋白氨基酸类3-10份;
还包括泛醇0.5-2份、甘油2-10份、黄原胶0.1-1份和去离子水100份;
所述欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类的质量比为1:1.2-2:2.5-4;
所述的美白组合物的制备工艺,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,搅拌至完全溶解,得到混合物1;
(2)在混合物1中加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、欧囊链藻提取物、肌肽、光果甘草根提取物、生物糖胶、玉米谷蛋白氨基酸类和泛醇,即得。
2.根据权利要求1所述的美白组合物,其特征在于,按照重量份数计,包括如下组分:二裂酵母发酵产物溶胞物2-8份、1-甲基乙内酰脲-2-酰亚胺3-6份、烟酰胺2-6份、欧囊链藻提取物3-7份、肌肽2-5份、光果甘草根提取物2-8份、生物糖胶3-8份和玉米谷蛋白氨基酸类4-8份。
3.一种权利要求1所述的美白组合物的制备工艺,其特征在于,包括如下步骤:
(1)在乳化锅中加入去离子水、甘油和黄原胶,搅拌至完全溶解,得到混合物1;
(2)在混合物1中加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、欧囊链藻提取物、肌肽、光果甘草根提取物、生物糖胶、玉米谷蛋白氨基酸类和泛醇,即得。
4.根据权利要求3所述的制备工艺,其特征在于,步骤(1)中加入去离子水、甘油和黄原胶后,升温至80-90℃,搅拌至完全溶解,得到混合物1。
5.根据权利要求3所述的制备工艺,其特征在于,步骤(2)中将混合物1降温至40-55℃后,先加入欧囊链藻提取物、光果甘草根提取物和玉米谷蛋白氨基酸类,再加入二裂酵母发酵产物溶胞物、1-甲基乙内酰脲-2-酰亚胺、烟酰胺、肌肽、生物糖胶和泛醇,即得。
6.一种美白精华液,其特征在于,包括权利要求1-2任一所述的美白组合物或权利要求3-5任一所述的制备工艺制备得到的美白组合物。
7.权利要求1-2任一所述的美白组合物或权利要求3-5任一所述的制备工艺制备得到的美白组合物在制备化妆品中的应用。
