Abstract
桃金娘提取物与表面活性物质的组合物,涉及天然提取物的提取和应用领域。本发明公开了一种由桃金娘(Rhodomyrtus tomentosa)提取物及其提纯物与表面活性物质混合溶解形成的稳定均一组合物及其在美白、抑菌等日化领域和食品添加剂领域的用途。组合物具有非常出色的配方易用性,同时表现出优良的抑制黑色素和抑菌的双重功效。组合物还兼具表面活性剂带来的去污力。

技术领域
本发明涉及天然提取物的提取和应用领域,尤其涉及一种水可溶解或分散的桃金娘(Rhodomyrtus tomentosa)提取物与表面活性剂的组合物的制备方法和应用。
背景技术
桃金娘(Rhodomyrtus tomentosa)是桃金娘科桃金娘属植物,又称捻子、山捻、岗捻等,主要分布于中国的南部和东南部,尤其盛产于岭南地区,为酸性土指示植物。桃金娘(Rhodomyrtus tomentosa)是我国民间传统药用植物,根、茎、叶、果均可入药或食用,具有清热、解毒、活血、消炎、止痛、消食、健胃等作用,其根可治疗虚咳,茎和叶可治牙疼,嫩尖主治痈䵸、毒伧和结核病。其花玫瑰红色,果实味美,可酿酒,果仁可供榨油食用。
桃金娘(Rhodomyrtus tomentosa)的根、茎、叶的提取物,均含有抗菌成分桃金娘酮类物质。这些抗菌成分主要为桃金娘酮(Rhodomyrtone、Tomentosone、Rhodomyrtosone)等酰基间苯三酚类化合物。通过溶剂萃取和分子蒸馏或层析分离的方法,已经从桃金娘植物的组织中提取并分离出多种酰基间苯三酚化合物:Rhodomyrtone A (C26H34O6)、Tomentosone A (C41H52O9)、Tomentosone B(C41H52O9)、Tomentosone C(C38H48O8)、Tomentodione S(C28H32O5)、Tomentodione T(C28H32O5)、Rhodomyrtosone A(C26H32O7)、Rhodomyrtosone B(C26H34O6) 、Rhodomyrtosone C(C41H54O8)、Rhodomyrtosone E(C30H34O6)Rhodomyrtosone F (C27H36O6) 、Rhodomyrtosone G (C26H32O7) 、Rhodomyrtosone H (C26H34O6) 、Rhodomyrtosone I (C28H30O6) 、Rhodomyrtosone C(C41H54O8);多种黄酮类、萜类、鞣花酸衍生物类等化合物。其中,酰基间苯三酚系列化合物具有非常突出的抗菌活性。由于发现桃金娘植物的叶子具有良好的抗菌性能,民间有使用桃金娘植物治疗腹泻和伤口的传统,甚至用于治疗多种妇科疾病。研究发现,桃金娘叶的醇提物对多种常见细菌具有很强的抑制活性,具有广谱、高效、低耐药的突出效果。
从植物学分类的角度看,桃金娘科是一个很大的科类。包括了桉属、番樱桃属、红千层属、桃金娘属、香桃木属、玫瑰木属、番石榴属等132个属类,涉及6000余物种。该科常见的植物有:桉树、番石榴、白千层、桃金娘、香桃木等。
中国专利ZL200980113237.8 “桃金娘提取物作为脱色剂的用途”公开了一种包含桃金娘科植物香桃木Myrtus communis的提取物的组合物,其所述的“桃金娘”不是本发明涉及的桃金娘科桃金娘属植物桃金娘(Rhodomyrtus tomentosa),而是属于桃金娘科香桃木属植物香桃木(Myrtus communis),所述桃金娘科植物香桃木(Myrtus communisMyrtuscommunis),所述桃金娘科植物香桃木(Myrtus communis)的花为白色,主产自地中海沿岸国家,花朵为白色。而本发明涉及的桃金娘科桃金娘属植物桃金娘(Rhodomyrtustomentosa),主产于中国南方和东南亚国家,花朵为玫瑰红色。
中国专利 ZL200980123433.3公开了一种从桃金娘科桉属植物中提取大果桉醛的方法。另一篇中国专利CN201080018128.0公开了一种桃金娘科植物桉树提取物作为舌苔去除和口臭去除效果的口腔用组合物。这两个专利都是关于桃金娘科桉属植物的发明,与本发明涉及的桃金娘科桃金娘属植物桃金娘并非同一物种。
中国专利CN104761565B从桃金娘属植物桃金娘(Rhodomyrtus tomentosa)中分离出两种Tomentosone C、 Rhodomyrtosone B。发现他们具有非常特出的抗菌性能,对多种常见致病菌的最低抑制浓度MIC甚至可以与万古霉素媲美。该专利通过证明这两个化合物的抑菌性能,确立了将这两个化合物用于抗菌药物中的权利。该专利同时也从侧面证明,含有上述间苯三酚类化合物的桃金娘提取物具有良好的抗菌性能。而本发明将桃金娘提取物与特定的表面活性物质互溶,组合成均一组合物,其特征是将有抗菌性能的提取物充分溶解于适当的表面活性物质中,从而使之具有与天然提取物截然不同的水溶性,从水难溶物质转变成水溶或者水分散性的物质,并因此带来了该组合物在食品、化妆品、皮肤护理、口腔护理等诸多领域的广泛用途。
专利CN201510958420.5公开了一种天然乳液、制备方法及应用。该文件公开了一种含有桃金娘果实乳酸菌发酵提取物、珍珠绣线菊叶提取物、芦笋提取物等天然提取物和天然大分子聚合物增稠剂、多元醇制备的一种乳液,宣称具有清除自由基,抑制酪氨酸酶活性等作用。该公开文件使用的桃金娘提取物是桃金娘果实去皮后,用乳酸菌发酵制得的发酵液。桃金娘果实以多糖为主,其多酚类物质和间苯三酚化合物含量非常低。因此,该公开专利文件所提到的“乳液”也不具备抑菌的功效。而本发明涉及的植物部位是富含抗菌物质的桃金娘属植物桃金娘叶子、根茎部位。专利CN201510958420.5是以桃金娘果实发酵提取物与多种其他天然提取物混合,再与天然大分子聚合物增稠剂、多元醇制备的一种“乳液”。所谓的乳液不具备本发明所具有的特征,即所使用的桃金娘提取物是桃金娘叶和桃金娘根茎部位的提取物,提取物中富含酰基间苯三酚化合物,并且使之溶解到表面活性剂中形成均一透明的溶液,具备专利CN201510958420.5所公开的混合物 “乳液”所不具备的热力学稳定性。
桃金娘(Rhodomyrtus tomentosa)根、茎、叶提取物,尤其是叶的提取物有很高的抗菌抗炎活性,但是这些提取物的使用受到它们溶解度的限制。因为它们是不溶于水的,所以,在很多日用品、化妆品、食品、口腔护理品中难以应用,为了提高桃金娘提取物的生物利用度和使用方便性,有必要将桃金娘提取物,包括水蒸汽蒸馏所得精油、醇提或亚临界提取或超临界提取所得浸膏(粗提物)及其进一步纯化得到的间苯三酚系列化合物组合物、或再进一步纯化得到的单个间苯三酚化合物纯品,制备成水溶性的制剂,以方便在日化及食品等领域的应用。
本发明将桃金娘提取物与表面活性物质互溶,组合成均一组合物,其特征是将具备抗菌性能的桃金娘叶、根或(和)茎提取物充分溶解于表面活性物质中,从而使之具有提取物本身截然不同的水溶性,从水难溶物质转变成水溶或者水分散性的制剂,并因此带来了该组合物制剂在食品、化妆品、皮肤护理、口腔护理等诸多领域的广泛用途。
事实上,不是所有的表面活性剂都能够良好地增溶桃金娘多酚或其它桃金娘提取物组分,所以进行表面活性物质的筛选试验也是本发明很重要的前期工作。
发明内容
本发明的目的是提供一种溶解于表面活性物质或其水溶液中的桃金娘(Rhodomyrtus tomentosa)提取物组合物,这种组合物是水溶性的或者是水分散性的。所述的溶解于表面活性物质的桃金娘提取物组合物,是以常压溶剂提取、亚临界提取或超临界提取获得的桃金娘(Rhodomyrtus tomentosa)根、茎或叶提取物为原料,或以上述提取物预先通过分子蒸馏、层析分离进一步提纯得到的单一间苯三酚类物质或多种间苯三酚类物质的组合物为原料,与适当的表面活性物质混合溶解所制备的均一的水溶性制剂。所述的常压溶剂提取包括但不限于使用C1-C4的低分子量醇、酮、酯、氯代烃在常压下的浸提、釜式回流提取、超声波辅助提取、连续逆流提取。所述的亚临界提取包括但不限于亚临界丁烷提取、亚临界水提取、亚临界卤代烃提取。所述的超临界提取包括但不限于超临界二氧化碳提取。所述的表面活性物质为乙醇、乙二醇、丙二醇、丙三醇、丙二醇脂肪酸酯、蔗糖脂肪酸酯、烷基糖苷(APG)、水溶性淀粉、单双甘油脂肪酸酯(油酸、亚油酸、棕榈酸、山嵛酸、硬脂酸、月桂酸、亚麻酸)、D-甘露糖醇、海藻酸丙二醇酯、琥珀酸单甘油酯、聚甘油蓖麻醇酸酯(PGPR)、聚甘油脂肪酸酯、聚乙二醇、大豆磷脂、乙酸及酒石酸一及二甘油酯、二乙酰酒石酸一及二甘油酯、聚氧乙烯(20)及(40)脂肪酸酯、聚氧乙烯木糖醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单月桂酸酯、聚氧乙烯(20)山梨醇酐单棕榈酸酯、聚氧乙烯(20)山梨醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单油酸酯、山梨醇酐单月桂酸酯、山梨醇酐单棕榈酸酯、山梨醇酐单硬脂酸酯、山梨醇酐单油酸酯、双乙酰酒石酸单双甘油酯、辛酸甘油酯、辛烯基琥珀酸淀粉钠、酪蛋白酸钠、羟丙基淀粉、乙酰化单双甘油脂肪酸酯、癸酸甘油酯、磷脂、木糖醇酐单硬脂酸酯、柠檬酸脂肪酸甘油酯、乳酸脂肪酸甘油酯、乳糖醇、海藻酸钠、壳聚糖、果胶、卡拉胶、果胶酸钠、氢化蓖麻油聚氧乙烯醚、茶皂素、无患子皂素中的一种或多种。这些表面活性物质也可含有一定量的作为溶剂的水。
本发明还提供一种桃金娘(Rhodomyrtus tomentosa)精油溶解于表面活性物质得到的组合物,这种组合物是以水蒸汽蒸馏所得到的桃金娘精油为原料,与适当的表面活性物质混合溶解所得的均一水溶性制剂,所述的表面活性物质为乙醇、乙二醇、丙二醇、丙三醇、丙二醇脂肪酸酯、蔗糖脂肪酸酯、烷基糖苷(APG)、水溶性淀粉、单双甘油脂肪酸酯(油酸、亚油酸、棕榈酸、山嵛酸、硬脂酸、月桂酸、亚麻酸)、D-甘露糖醇、海藻酸丙二醇酯、琥珀酸单甘油酯、聚甘油蓖麻醇酸酯(PGPR)、聚甘油脂肪酸酯、聚乙二醇、大豆磷脂、乙酸及酒石酸一及二甘油酯、二乙酰酒石酸一及二甘油酯、聚氧乙烯(20)及(40)脂肪酸酯、聚氧乙烯木糖醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单月桂酸酯、聚氧乙烯单月桂酸酯、山梨醇酐单棕榈酸酯、山梨醇酐单硬脂酸酯、山梨醇酐单油酸酯、双乙酰酒石酸单双甘油酯、辛酸甘油酯、辛烯基琥珀酸淀粉钠、酪蛋白酸钠、羟丙基淀粉、乙酰化单双甘油脂肪酸酯、癸酸甘油酯、磷脂、木糖醇酐单硬脂酸酯、柠檬酸脂肪酸甘油酯、乳酸脂肪酸甘油酯、乳糖醇、海藻酸钠、壳聚糖、果胶、卡拉胶、果胶酸钠、氢化蓖麻油聚氧乙烯醚、茶皂素、无患子皂素中的一种或多种。该组合物也可含有一定量的作为溶剂的水。
使用单一的表面活性剂溶解桃金娘提取物是一种优选的方式。
当使用聚氧乙烯失水山梨醇脂肪酸酯系列表面活性剂时,部分表面活性剂可以溶解多至10%的桃金娘精油,如使用聚氧乙烯(20)失水山梨醇油酸酯作为表面活性物质时,可以溶入100克/升桃金娘精油,从而配制较高桃金娘提取物含量的透明组合物。
使用对桃金娘提取物溶解度低的表面活性物质,将形成不透明的分散体。
使用需加热才能熔融成液态的固态表面活性物质,最终将获得块状、粒状或粉末状的固体制剂的形态。
当组合物中含有水分时,可以形成乳液状态。
本发明使用乙醇、乙二醇、丙二醇、丙三醇、丙二醇脂肪酸酯、蔗糖脂肪酸酯、烷基糖苷(APG)、水溶性淀粉、单双甘油脂肪酸酯(油酸、亚油酸、棕榈酸、山嵛酸、硬脂酸、月桂酸、亚麻酸)、D-甘露糖醇、海藻酸丙二醇酯、琥珀酸单甘油酯、聚甘油蓖麻醇酸酯(PGPR)、聚甘油脂肪酸酯、聚乙二醇、大豆磷脂、乙酸及酒石酸一及二甘油酯、二乙酰酒石酸一及二甘油酯、聚氧乙烯(20)及(40)脂肪酸酯、聚氧乙烯木糖醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单月桂酸酯、聚氧乙烯(20)山梨醇酐单棕榈酸酯、聚氧乙烯(20)山梨醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单油酸酯、山梨醇酐单月桂酸酯、山梨醇酐单棕榈酸酯、山梨醇酐单硬脂酸酯、山梨醇酐单油酸酯、双乙酰酒石酸单双甘油酯、辛酸甘油酯、辛烯基琥珀酸淀粉钠、酪蛋白酸钠、羟丙基淀粉、乙酰化单双甘油脂肪酸酯、癸酸甘油酯、磷脂、木糖醇酐单硬脂酸酯、柠檬酸脂肪酸甘油酯、乳酸脂肪酸甘油酯、乳糖醇、海藻酸钠、壳聚糖、果胶、卡拉胶、果胶酸钠、氢化蓖麻油聚氧乙烯醚、茶皂素、无患子皂素中的一种或多种与通过已知的提取方式提取得到的桃金娘多酚粗提取物进行混合溶解,得到水溶性的桃金娘提取物与表面活性物质的组合物。所述的已知的提取方式是常压提取、亚临界提取或超临界提取。所述的常压-提取包括但不限于使用C1-C4的低分子量醇、酮、酯、氯代烷烃在常压下的浸提、回流提取、超声波辅助提取和连续逆流提取。所述的亚临界提取包括但不限于亚临界丁烷提取、亚临界水提取、亚临界卤代烃提取。所述的超临界提取包括但不限于超临界二氧化碳提取。所述的桃金娘提取物是桃金娘科桃金娘属植物桃金娘的根、茎或叶三个部位中的一种或多种的提取物。
本发明使用乙醇、乙二醇、丙二醇、丙三醇、丙二醇脂肪酸酯、蔗糖脂肪酸酯、烷基糖苷(APG)、水溶性淀粉、单双甘油脂肪酸酯(油酸、亚油酸、棕榈酸、山嵛酸、硬脂酸、月桂酸、亚麻酸)、D-甘露糖醇、海藻酸丙二醇酯、琥珀酸单甘油酯、聚甘油蓖麻醇酸酯(PGPR)、聚甘油脂肪酸酯、聚乙二醇、大豆磷脂、乙酸及酒石酸一及二甘油酯、二乙酰酒石酸一及二甘油酯、聚氧乙烯(20)及(40)脂肪酸酯、聚氧乙烯木糖醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单月桂酸酯、聚氧乙烯(20)山梨醇酐单棕榈酸酯、聚氧乙烯(20)山梨醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单油酸酯、山梨醇酐单月桂酸酯、山梨醇酐单棕榈酸酯、山梨醇酐单硬脂酸酯、山梨醇酐单油酸酯、双乙酰酒石酸单双甘油酯、辛酸甘油酯、辛烯基琥珀酸淀粉钠、酪蛋白酸钠、羟丙基淀粉、乙酰化单双甘油脂肪酸酯、癸酸甘油酯、磷脂、木糖醇酐单硬脂酸酯、柠檬酸脂肪酸甘油酯、乳酸脂肪酸甘油酯、乳糖醇、海藻酸钠、壳聚糖、果胶、卡拉胶、果胶酸钠、氢化蓖麻油聚氧乙烯醚、茶皂素、无患子皂素中的一种或多种与桃金娘粗提取物经过进一步分子蒸馏、层析分离、溶剂萃取分离提纯得到的单个桃金娘酮间苯三酚化合物或它们的组合物进行混合溶解,得到水溶性或水分散性的桃金娘多酚与表面活性物质的组合物。这些间苯三酚化合物为从桃金娘植物中提取的桃金娘酮、桃金娘异酮,包括但不限于:Rhodomyrtone A (C26H34O6)、Tomentosone A (C41H52O9)、Tomentosone B(C41H52O9)、Tomentosone C(C38H48O8)、Tomentodione S(C28H32O5)、Tomentodione T(C28H32O5)、Rhodomyrtosone A(C26H32O7)、Rhodomyrtosone B(C26H34O6) 、Rhodomyrtosone C(C41H54O8)、Rhodomyrtosone E (C30H34O6)、Rhodomyrtosone F (C27H36O6) 、Rhodomyrtosone G (C26H32O7) 、Rhodomyrtosone H (C26H34O6) 、Rhodomyrtosone I(C28H30O6) 、Rhodomyrtosone C(C41H54O8)。
通过将桃金娘根、茎或叶的粗提取物或桃金娘根、茎或叶的粗提物经已知的提纯方法进一步分离得到的酚、类黄酮、间苯三酚类化合物与适当的表面活性物质混合溶解,可得到均一、稳定的水溶性或水分散性的桃金娘提取物的组合物。当表面活性物质中含有水分,可形成油包水或水包油的乳液状制剂。
由于桃金娘根、茎或叶的提取物与表面活性物质的组合物改变了桃金娘提取物的难溶属性,使之容易应到食品、化妆品、药品配方中,大大拓宽了桃金娘根、茎或叶提取物的应用范围。
进一步实验发现,经过表面活性物质的增效,本发明所提供的桃金娘提取物与表面活性剂的组合物比相同总酚浓度的桃金娘提取物具有更强的抑菌能力,同时具有更好的抑制皮肤色素形成与沉着的效果。由此,本发明也带来了桃金娘提取物与表面活性物质的组合物的多种应用。
本发明首次研究并发现桃金娘提取物与表面活性物质的组合物对舌苔具有抑制作用,并提供一种桃金娘提取物与表面活性物质的组合物在口臭预防领域的应用。所述的桃金娘是指桃金娘科桃金娘属植物桃金娘(Rhodomyrtus tomentosa)。这种抑制舌苔的作用在桃金娘科桉属植物上也有类似的表现。
舌苔是指食物残渣、细菌及细胞等堆积于舌背部、呈苔状的苔状物。已知舌苔的蓄积是引起口臭的原因。本发明通过试验证明了掺有桃金娘提取物的表面活性物质组合物的口香糖、口腔护理液、糖果、果冻、口腔喷剂等可以使受试者舌苔量减少,由此减轻口臭,并且减轻的程度十分明显。系列研究的结果充分体现了桃金娘提取物抗菌性能在日用品、化妆品、食品上的潜在用途。
本发明公开了溶解于表面活性物质的桃金娘根、茎或叶的提取物在口臭预防作用的口腔用制剂上的用途,其特征在于,溶解于表面活性物质的桃金娘提取物是其中必须的有效成分。
本发明公开了溶解于表面活性物质的桃金娘提取物在口臭预防作用的口腔用制剂上的用途,其特征在于,溶解于表面活性物质的桃金娘提取物是桃金娘属植物桃金娘的根、茎或叶的提取物。
本发明还发现使用溶解于表面活性物质的桃金娘根、茎或叶的提取物,能够抑制酪氨酸酶,对黑素细胞形成黑色素的过程具有明显的抑制作用。通过实验证明,溶解于表面活性物质的桃金娘粗提取物和溶解于表面活性剂的桃金娘提取物经进一步纯化得到的间苯三酚类化合物(桃金娘酮)都可以明显抑制黑色素的合成和向皮肤外部输送的过程。这种作用表现在桃金娘提取物能抑制黑素细胞的树突形成,同时具备抑制黑素合成的能力。由此,本发明将桃金娘提取物与表面活性活性物质组合物的应用范围拓展到皮肤美容领域的淡化色斑和美白,方法是向皮肤涂抹桃金娘提取物与表面活性活性物质组合物。
在化妆品领域、洗护产品或医学美容领域应用时,溶解于表面活性物质的桃金娘提取物可以是以下的形式:
乙醇溶液,乙醇水溶液,水凝胶,水包油或油包水乳液,以清液、乳液、凝胶、洗剂、糊剂、膏霜或固体的外观呈现。
应用于祛斑,减少黑素用途的溶解于表面活性物质的桃金娘提取物,还可以以胶囊、片剂、粉末、饮用悬浮液的口服制剂形式呈现。 所使用的表面活性物质可以是固态的表面活性剂,固态的其它表面活性物质,例如高分子量聚乙二醇,以固体共融物或者固体分散体的形式形成稳定的组合物。
与同属桃金娘科的香桃木属植物香桃木(Myrtus communis)的提取物相比,桃金娘属植物桃金娘同样含有多种酚类物质,因此,有可能具有类似的美白、淡斑机理。试验证明,桃金娘属植物桃金娘提取物兼有抗菌消炎的功效和美白淡斑的效果,而香桃木提取物的抗菌消炎效果较弱,美白效果则基本类似。
本发明还发现具备卓越抑菌能力的桃金娘提取物与表面活性物质的组合物能够用于洗发剂,起到抑菌去屑的作用。将桃金娘提取物与适当的表面活性剂形成的组合物可以直接用于洗发,无需加入其它抑菌防腐物质;或者可以加入到普通洗发配方中,起到抑菌,控油,控屑的作用。尤其是桃金娘提取物与天然表面活性剂茶皂素或(和)无患子皂素组成的组合物,无需加入其它抗菌防腐物质,即可形成长期稳定,并具有良好去污能力,具备抑菌,控油,控屑作用的全天然洗发剂。
具体实施例
以下实施例是对本发明的进一步说明,而不是对本发明的限制。
实施例1:一种溶解于聚氧乙烯(20)失水山梨醇单油酸酯的桃金娘精油的制备:将10g通过水蒸气蒸馏桃金娘(桃金娘科桃金娘属)的鲜叶获得的精油与90g聚氧乙烯(20)失水山梨醇单油酸酯混合,用常规搅拌的方式搅拌溶解成透明均一的溶液状态,得到溶解于聚氧乙烯(20)失水山梨醇单油酸酯溶解的桃金娘精油。该组合物的桃金娘精油含量为10%,具有良好的水溶性(水分散性),可用于美容护肤品,头皮护理品,口腔护理品,具有抑菌、抗炎、美白、淡斑的功效。用作口香糖添加剂,可以起到减少舌苔,从而起到减轻口臭的作用。也可用作食品添加剂,具有防腐的功效。
实施例2:一种溶解于聚氧乙烯(20)失水山梨醇单油酸酯的桃金娘酮(混合)的制备:将100.0克桃金娘叶的乙醇提取物(总酚26.2%)通过一步分子蒸馏的方法提纯获得较高纯度的混合桃金娘酮(以总酚计为72.1%) 31.7克,取10克总酚含量72.1%的桃金娘提取物与90克聚氧乙烯(20)失水山梨醇单油酸酯混合,用常规搅拌的方式搅拌溶解成透明均一的溶液状态,得到完全溶解于聚氧乙烯(20)失水山梨醇单油酸酯溶解的桃金娘酮(混合)。该组合物的桃金娘提取物含量为10%,总酚含量约为7.2%。该组合物具有良好的水溶性(水分散性),可用于美容护肤品,头皮护理品,口腔护理品,具有非常抑菌功效。用作口香糖添加剂,可以起到减少舌苔,从而起到减轻口臭的作用。也可用作食品添加剂,具有防腐功效。
实施例3:测定总酚含量7.2%的聚氧乙烯(20)失水山梨醇单油酸酯-桃金娘酮(混合)组合物对黑素细胞、酪氨酸酶的抑制作用和对黑色素合成的影响:
于含8%FCS的DMEM培养液中,以104/3ml, 3ml/孔(6孔板),体外培养B16黑素瘤细胞,分别加入0 μg/ ml 、10μg/ ml 、50μg/ ml 、200μg/ ml(以总酚含量计,每组4复孔)上述总酚含量7.2%的聚氧乙烯(20)失水山梨醇单油酸酯-桃金娘酮(混合)制剂,在37℃,5%CO2条件下培养72h,分别测定B16黑素瘤细胞生长数量和细胞内黑素含量。
黑素瘤细胞生长数量测定:在待测样中加入5mg/mlS四甲基偶氮唑蓝(MTT)10μl/ml,继续培养72h后弃去上清液,加入二甲基亚砜2ml,混合均匀后在570nm处测定吸光值。以(1-OD/OD对照)×100%计算桃金娘提取物(桃金娘酮)对黑素瘤细胞生长抑制率。
黑素瘤细胞内黑素含量的测定:在每孔待测样品中,加入浓度0.3%胰酶0.5ml,细胞全部消化脱落后,37℃水浴中保温2h,2000rpm离心后取上清液,在470nm处测定吸光值A。以(1-A/A对照)×100%计算桃金娘提取物(桃金娘酮)对黑色素的抑制率。
黑素瘤细胞内酪氨酸酶含量的测定:3.7%福尔马林液固定生长在载玻片上的B16黑素瘤细胞后,用10mmol PBS洗涤3次,用含1% BSA的10mmol PBS封闭10min,再用10mmolPBS洗涤3次;加入鼠抗人酪氨酸酶一抗,37℃,置湿盒中2h, 10mmol PBS洗涤3次;加HRP-羊抗鼠二抗,置湿盒中,37℃,30min,10mmol PBS洗涤3次,用DAB显色液显色,用彩色图文病理扫描仪测定细胞中酪氨酸酶吸光度B,以(1-B/B对照)×100%计算桃金娘提取物(桃金娘酮)对黑素瘤细胞内酪氨酸酶的抑制率。
结果如下:

由此可见, 桃金娘酮在0-200μg/ ml浓度范围对黑素细胞生长本身并无明显抑制作用,随着桃金娘酮的浓度增加,影响并没有明显增长。在此浓度范围,桃金娘酮对黑素细胞内酪氨酸酶具有一定的抑制作用,但仍然不算显著;而相同条件下桃金娘酮对于黑素合成的综合抑制作用则十分明显,在上述实验中,200μg/ ml可将黑素合成率降低接近一半。
实施例3:一种溶解于单硬脂酸甘油酯的桃金娘提取物的组合物的制备:将50g桃金娘叶的超临界提取物与950g单硬脂酸甘油酯加热至70摄氏度混合搅拌,溶解成透明均一的溶液状态后冷却,得到溶解于单硬脂酸甘油酯的天然桃金娘提取物组合物,最后以固体形态呈现。可进一步制成粒状、条状或块状。该固态桃金娘提取物组合物具有良好的水分散性,可用于美容护肤品,头皮护理品,口腔护理品,具有抑菌、美白的功效。用作口香糖添加剂,可以起到减少舌苔,从而起到减轻口臭的作用。也可用作食品添加剂,具有防腐的功效。
实施例4:一种用作洗发剂的溶解于无患子皂素的桃金娘提取物的制备:
取桃金娘醇提物10g,与990g皂苷含量40%的无患子皂素(水提浓缩液)混合溶解,得到一种天然表面活性物质无患子皂素与桃金娘提取物的组合物(含水)。该组合物具有优异的清洁能力和抑菌能力,无需添加其它成分即可用作具有抑菌去头屑功效的洗发剂。经过12名志愿者的使用,证明该洗发剂能够大大延长头发的清洁时长。这是具有抑菌消炎功能的桃金娘提取物为组合物带来的重要功能。
Claims (11)
1.一种桃金娘(Rhodomyrtus tomentosa)根、茎或(和)叶的提取物与表面活性物质的组合物,该组合物包含选自桃金娘(Rhodomyrtus tomentosa)的根、茎或叶的提取物和选自乙醇、乙二醇、丙二醇、丙三醇、丙二醇脂肪酸酯、蔗糖脂肪酸酯、烷基糖苷、水溶性淀粉、单双甘油脂肪酸酯(油酸、亚油酸、棕榈酸、山嵛酸、硬脂酸、月桂酸、亚麻酸)、D-甘露糖醇、海藻酸丙二醇酯、琥珀酸单甘油酯、聚甘油蓖麻醇酸酯、聚甘油脂肪酸酯、聚乙二醇、大豆磷脂、乙酸及酒石酸一及二甘油酯、二乙酰酒石酸一及二甘油酯、聚氧乙烯(20)及(40)脂肪酸酯、聚氧乙烯木糖醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单月桂酸酯、聚氧乙烯(20)山梨醇酐单棕榈酸酯、聚氧乙烯(20)山梨醇酐单硬脂酸酯、聚氧乙烯(20)山梨醇酐单油酸酯、山梨醇酐单月桂酸酯、山梨醇酐单棕榈酸酯、山梨醇酐单硬脂酸酯、山梨醇酐单油酸酯、双乙酰酒石酸单双甘油酯、辛酸甘油酯、辛烯基琥珀酸淀粉钠、酪蛋白酸钠、羟丙基淀粉、乙酰化单双甘油脂肪酸酯、癸酸甘油酯、磷脂、木糖醇酐单硬脂酸酯、柠檬酸脂肪酸甘油酯、乳酸脂肪酸甘油酯、乳糖醇、海藻酸钠、壳聚糖、果胶、卡拉胶、果胶酸钠、氢化蓖麻油聚氧乙烯醚、茶皂素、无患子皂素的表面活性物质。
2.如权利要求1所述的组合物,其桃金娘提取物的含量以总酚计为0.01至20%。
3.如权利要求1所述的组合物,其桃金娘提取物是以常规提取方法获得的根、茎或(和)叶的提取物。
4. 如权利要求1所述的组合物,其桃金娘提取物是以常规提取方法提取得到的粗提物再经过分子蒸馏、层析分离等常规提纯工艺进一步提纯得到的单一间苯三酚类物质或多种间苯三酚类物质的组合物,或是以间苯三酚化合物为有效物的桃金娘提取物的浓缩物,这些间苯三酚化合物包含但不限于:Rhodomyrtone A (C26H34O6)、Tomentosone A(C41H52O9)、Tomentosone B(C41H52O9)、Tomentosone C(C38H48O8)、Tomentodione S(C28H32O5)、Tomentodione T(C28H32O5)、Rhodomyrtosone A(C26H32O7)、RhodomyrtosoneB(C26H34O6) 、Rhodomyrtosone C(C41H54O8)、Rhodomyrtosone E (C30H34O6)、Rhodomyrtosone F (C27H36O6) 、Rhodomyrtosone G (C26H32O7) 、Rhodomyrtosone H(C26H34O6) 、Rhodomyrtosone I (C28H30O6) 、Rhodomyrtosone C(C41H54O8)。
5.如权利要求1所述的组合物,所述的表面活性物质不含水。
6.如权利要求1所述的组合物,所述的表面活性物质含有一定量的作为溶剂的水。
7.如权利要求1所述的组合物,这种组合物是水溶性的或者是水分散性的。
8.如权利要求1所述的组合物,宣称具有抑菌,预防口臭功效,用于口香糖、口腔护理液、糖果、果冻、口腔喷剂、牙膏。
9.如权利要求1所述的组合物,宣称具有美白、淡斑、抗氧化、护肤功效,使用方法是向皮肤涂抹该组合物。
10.如权利要求1所述的组合物,尤其是桃金娘提取物与天然表面活性剂茶皂素或(和)无患子皂素组成的组合物,无需加入其它抗菌防腐物质,即可形成长期稳定,并具有良好去污能力,具备抑菌,控油,控屑作用的全天然洗发剂。
11.如权利要求1所述的组合物,作为饲料添加剂或混合型饲料添加剂的应用,起到预防细菌感染和提升免疫力的作用。
