背景技术
对脑病症进行准确和快速检测可以使得对受试者进行治疗的过程更加快速和合适。例如,准确且快速地检测头部损伤或脑疾病或异常的发作可以允许在早期应用合适的治疗和/或预防性措施来为受试者提供最大利益。
然而,当前用于脑病症的检测方法具有各种缺点。有些检测方法可能要求受试者出现在诊所或其他护理设施以进行抽血、成像和诊断。有些检测方法可能不提供有用的信息,直到个体已经出现显著伤害和/或损伤以后。

具体实施方式
头部损伤或脑疾病或异常的发作可能导致受试者的呼气内的各种挥发性有机化合物(VOC)增加和/或减少。因此,检测受试者的呼气内的VOC和/或与其相关的模式可以具有很高诊断价值,以在受试者遭受头部损伤或者疾病状态发作后向该受试者提供合适的护理和/或治疗。在一些情形中,VOC和/或与其相关的模式可以在发生损伤后数分钟内被检测到。
根据本文中的实施例,可以检测受试者呼气内的各种挥发性有机化合物(VOC)以帮助诊断脑病症(比如脑损伤、疾病或异常)和/或作为治疗或护理该脑病症的方法的一部分。在各个实施例中,可以快速地现场(而不仅仅是在护理设施内)进行VOC的分析。因此,本文中的各个实施例可以提供以可能的最快速方式对头部损伤或疾病状态进行检测的机会。
在一些实施例中,可以使用在损伤或疾病发作之后一段时间内的VOC和/或与其相关的模式进行的检测来监测响应于治疗的进展或根据需要改变治疗过程。
现在参考图1,示出了根据本文中的各个实施例用于检测受试者的脑病症的方法100的示意图。用于检测脑病症的方法100可以包括在102处从受试者获得呼出气样本以及在104处使该呼出气样本与化学传感器元件接触。该化学传感器元件可以包括多个分立的石墨烯变容二极管(这将在下文中参见图5至图10说明)。在一些实施例中,从受试者获得呼出气样本的步骤可以包括在所述脑损伤之后10分钟内获得呼气样本。在一些实施例中,从受试者获得呼出气样本的步骤包括在脑损伤发作之后24小时的时段内至少两次获得呼气样本。在一些实施例中,受试者是人类。在其他实施例中,受试者是动物,包括但不限于肉牛、野牛、猪、绵羊、山羊、马、狗、猫和鸡。

方法100还可以包括在106处感测和存储分立的石墨烯变容二极管的电容以获得样本数据集。方法100可以包括在108处将样本数据集分类为一个或多个预先建立的脑病症类别。将在下文更详细地说明该一个或多个预先建立的脑病症类别。

当在一定偏置电压范围内以步进方式感测和存储石墨烯变容二极管的电容时,可以在每个偏置电压下获得每个分立的石墨烯变容二极管的样本数据集。在一定偏置电压范围内感测和存储石墨烯变容二极管的电容以获得样本数据集可以包括在该偏置电压范围内为每个石墨烯变容二极管存储至少10个、20个、30个、40个、50个、60个、70个、80个、90个、100个、125个、150个、175个、200个、250个、300个、350个、400个、450个或500个离散电容值(或落入前述任意数量之间的范围内的数量的离散电容值)。
本文中的方法还可以包括收集和/或存储关于该受试者的、超出所感测的电容的附加数据作为被分类的样本数据集的一部分。该附加数据可以包括但不限于:受试者的先前创伤性脑损伤;自已经发生导致受试者损伤的事件以后所经过的时间;受试者的年龄;神经检查的结果;受试者所经历的症状;以及关于脑病症的具体生物标志物的数据。该附加数据还可以包括有关过去治疗方案以及过去治疗方案的成功或失败的信息。
将理解的是,来自受试者的呼出气样本的挥发性有机化合物(VOC)可以与分立的石墨烯变容二极管相接以影响所感测的电容。头部损伤之前受试者呼出气中的VOC可能不同于损伤之后受试者呼出气中的VOC。在训练方案期间,可以在受试者头部损伤之前从该受试者获得一个或多个呼出气样本。根据未受损伤的受试者的呼出气利用感测和存储电容而获得的数据可以用作未受损伤状态的基线值。从处于未受损伤状态的受试者获得呼出气样本的示例可以包括但不应限于:在赛季前运动训练计划期间、在基本军事训练计划期间、或在就业培训计划期间每天、每周或每月获得呼气样本。在一些实施例中,来自呼出气的数据可以在临床设置中作为常规身体检查的一部分从受试者获得,并且如果在将来某个时间发生头部损伤或脑异常,可以作为该患者的呼气中的VOC含量的基线。
在运动赛事、军事部署、工作环境等中的身体活动期间,受试者可能会经历头部损伤,该头部损伤可能导致受试者的脑病症。具有在损伤现场对受试者进行评估的能力可能是在发生损伤事件之后尽快提供快速的诊断和合适的护理的关键。举例而言,现在参考图2,示出了根据本文中的各个实施例用于检测经历头部损伤的受试者的脑病症的方法200的示意图。在图2中,在202处示出了参加体育赛事的受试者正经历头部损伤。作为头部损伤的结果,受试者在204处由于该损伤而丧失行为能力。在206处,从受试者获得呼出气样本并使该呼出气样本与包含多个分立的石墨烯变容二极管的化学传感器元件接触。在208处示出了感测和存储分立的石墨烯变容二极管的电容以获得样本数据集。
脑病症的类型可能在类型、严重性和持续时间方面不同,并且可以被分类为预先建立的脑病症类别。现在参考图3,示出了根据本文中各个实施例的预先建立的脑病症类别302的示意图。在一些实施例中,脑病症可以是脑损伤304,比如,创伤性脑损伤308或缺血性脑损伤310。在其他实施例中,脑病症可以包括脑疾病或脑疾病306。如本文中所用,术语“脑疾病”和“脑异常”可互换使用。脑损伤304的类别可以进一步包括创伤性脑损伤308、缺血性脑损伤310,并且脑疾病306的类别可以进一步包括神经退行性脑异常312、自身免疫性脑异常314、精神异常316、发育异常318等。

石墨烯变容二极管900可以包括设置在所述石墨烯层908a和908b的一部分上的一个或多个接触电极910。接触电极910可以由导电材料,比如铬、铜、金、银、钨、铝、钛、钯、铂、铱以及其任何组合或合金形成。示例性石墨烯变容二极管的另外方面可以在美国专利号9,513,244中找到,该专利的全部内容通过援引用并入本文。
可以通过在特定电压和/或在电压范围内输送激励电流来测量石墨烯变容二极管的电容。测量电容提供了反映分析物与(多个)石墨烯变容二极管的结合状态的数据。可以使用各种测量电路系统来测量(多个)石墨烯变容二极管的电容。
现在参考图10,示出了根据本文中各个实施例的用于测量多个石墨烯传感器的电容的电路系统的示意图。该电路系统可以包括与多路复用器1004电通信的电容到数字转换器(CDC)1002。多路复用器1004可以提供与多个石墨烯变容二极管1006的选择性电通信。与石墨烯变容二极管1006的另一侧的连接可以由开关1003控制(如由CDC控制),并且可以提供与第一数模转换器(DAC)1005和第二数模转换器(DAC)1006的选择性电通信。DAC 1005、1007的另一侧可以连接至总线装置1010,或者在某些情况下,连接至CDC 1002。该电路系统可以进一步包括微控制器1012,其将在下面更详细地讨论。

可以基于电容数据来计算许多不同的方面。例如,可以计算的方面包括电容对电压的最大斜率、电容对电压的最大斜率在基线值上的变化、电容对电压的最小斜率、电容对电压的最小斜率在基线值上的变化、最小电容、最小电容在基线值上的变化、最小电容处的电压(狄拉克点)、最小电容处的电压变化、最大电容、最大电容的变化、最大电容与最小电容之比、响应时间常数、以及在不同的石墨烯传感器之间(特别是在对不同的分析物具有特异性的不同的石墨烯传感器之间)前述任何方面之比。
以上计算的方面可以用于各种诊断目的。在一些情况下,以上计算的方面可以指示气体样本中特定挥发性有机组分的身份和/或浓度。因此,以上的每个计算值都可以作为形成给定受试者和/或给定气体样本的模式的一部分的独特数据片。另外如本文其他地方所描述的,然后可以将该模式对照现有模式、实时识别的模式、或通过比如机器学习或其他技术等技术从较大的存储数据集导出的模式进行匹配,其中,将这些模式确定为表征各种病症或疾病状态。以上计算的方面也可以用于其他目的(诊断和其他)。
在一些实施例中,比如上述的那些计算可以由控制器电路执行。控制器电路可以被配置为接收反映石墨烯变容二极管的电容的电信号。在一些实施例中,控制器电路可以包括微控制器以执行这些计算。在一些实施例中,控制器电路可以包括与测量电路电通信的微处理器。微处理器系统可以包括比如地址总线、数据总线、控制总线、时钟、CPU、处理装置、地址解码器、RAM、ROM等的部件。在一些实施例中,控制器电路可以包括与测量电路电通信的计算电路(比如专用集成电路——ASIC)。
另外,在一些实施例中,该系统可以包括存储有特定传感器的灵敏度校准信息的非易失性存储器。举例来说,该传感器可以在生产设施中进行测试,在该处可以确定该传感器对各种分析物(比如VOC)的灵敏度,然后将该灵敏度存储在EPROM或类似部件中。另外或可替代地,在将受试者数据发送到中心位置供分析和诊断时,灵敏度校准信息可以存储在中央数据库中,并附有传感器序列号以供参考。这些部件可以包括在本文所述的任何硬件中。
在本文中的一些实施例中,部件可以被配置为通过比如互联网或类似网络等网络进行通信。在各个实施例中,可以包括中央存储设备和数据处理设施。在一些实施例中,在存在受试者(本地)的情况下从传感器收集的数据可以通过互联网或类似网络发送至中央处理设施(远程),并且来自被评估的特定受试者的模式可以与成千上万的其他受试者的模式进行比较,其他受试者中的许多受试者先前已被诊断有各种病症,并且其中这些病症数据已经被存储。模式匹配算法可用于查找与当前受试者的模式最相似的其他受试者或受试者类(例如疾病或病症特定类)。每一类受试者可以包括具有给定病症或疾病状态的预定可能性。以这种方式,在模式匹配之后,可以通过数据网络将具有给定病症或疾病状态的可能性返回提供给受试者当前所在的设施。
在一些实施例中,电路系统可以包括有源和无源感测电路系统。这样的电路系统可以实施有线(直接电接触)或无线感测技术。
本文所述的呼气感测系统可以包括用于从分立的石墨烯变容二极管生成信号的各种电路系统。这样的电路系统可以包括有源和无源感测电路系统。这样的电路系统可以实施有线(直接电接触)或无线感测技术。现在参考图11,示出了根据本文中各个方面的无源传感器电路1102以及一部分读取电路1122的示意图。在一些实施例中,无源传感器电路1102可以包括耦合到感应器1110的金属氧化物石墨烯变容二极管1104(其中RS表示串联电阻并且CG表示变容二极管电容器)。在一些实施例中,读取电路1122可以包括具有电阻1124和电感1126的读取线圈。然而,将理解的是,图10和图11中所示的电路仅仅是示例性方法。本文设想了许多不同的方法。在共同待决的美国申请号62/533,916中描述了另外的用于生理气体样本中的分析物感测的系统和方法,该申请通过援引以其全文并入本文。
现在参考图12,示出了根据本文各个实施例的示出石墨烯变容二极管的电容与DC偏置电压的曲线图。可以通过在将化学传感器暴露于受试者的呼出气时使用LCR仪表在一定范围的偏置电压内测量电容来建立图12所示的电容与电压曲线。在一些实施例中,偏置电压的范围可以包括从-3V至3V。在一些实施例中,DC偏置电压的范围可以是从-2V至2V,或者从-1.5V至1.5V,或者从-1V至1V,或者从-0.5V至0.5V。
类别和模式匹配
可以根据许多不同的机器学习技术(比如模式识别)将样本数据集分类为一个或多个预先建立的脑病症类别。分类可以包括使用模式匹配或模式识别算法将样本数据集与先前确定的一个或多个模式进行比较以确定为最佳匹配的模式,其中,作为最佳匹配的先前确定的特定模式指示患者的脑病症。
举例来说,最初可以通过机器学习分析或另一类似的算法技术来标识大量患者数据集中的模式。与特定的脑病症类别相关的模式可以从标记的“训练”数据(监督学习)或者在没有标记数据的情况下(无监督学习)导出。
本文中使用的用于模式匹配的算法可以包括但不限于分类算法(预测分类标签的监督算法)、聚类算法(预测分类标签的无监督算法)、集成学习算法(用于将多个学习算法组合在一起的监督元算法)、用于预测任意结构标签集的通用算法、多线性子空间学习算法(使用张量表示来预测多维数据的标签)、实值序列标签算法(预测实值标签的序列)、回归算法(预测实值标签)、以及序列标签算法(预测分类标签的序列)。
分类算法可以包括参数算法(比如线性判别分析、二次判别分析和最大熵分类器)和非参数算法(比如决策树、核估计、朴素贝叶斯分类器、神经网络、感知器和支持向量机)。聚类算法可以包括分类混合模型、深度学习方法、分层聚类、K均值聚类、相关聚类和核主成分分析。集成学习算法可以包括增强、自举聚合、集合平均和专家混合。用于预测任意结构标签集的通用算法可以包括贝叶斯网络和马尔可夫随机场。多线性子空间学习算法可以包括多线性主成分分析(MPCA)。实值序列标签算法可以包括卡尔曼滤波器和粒子滤波器。回归算法可以包括监督(比如高斯过程回归、线性回归、神经网络和深度学习方法)和无监督(比如独立成分分析和主成分分析)方法。序列标签算法可以包括监督(比如条件随机字段、隐马尔可夫模型、最大熵马尔可夫模型和递归神经网络)方法和无监督(隐马尔可夫模型和动态时间规整)方法。
许多不同的类别可以用于本文中讨论的病症。本文中的类别可以包括但不限于给定病症的程度(比如不存在、轻度、严重)。本文中的类别可以包括但不限于存在给定病症的概率(比如,无迹象、低概率、高概率)。
治疗方法
本文的实施例可以具体包括治疗受试者的脑病症的方法。该方法可以包括从受试者获得呼出气样本并使该呼气样本与化学传感器元件接触,该化学传感器元件包括多个分立的石墨烯变容二极管。该方法可以进一步包括感测和存储这些分立的石墨烯变容二极管的电容以获得样本数据集。该方法可以进一步包括将样本数据集分类为一个或多个预先建立的脑病症类别。该方法可以进一步包括基于脑病症类别来治疗患者。举例来说,下表1提供了一组示例性的类别以及针对创伤性脑损伤的可能治疗。

Claims (15)
1.一种用于检测受试者的脑病症的方法,包括:
从所述受试者获得呼气样本并且使所述呼气样本与化学传感器元件接触,所述化学传感器元件包括多个分立的石墨烯变容二极管;
感测和存储所述分立的石墨烯变容二极管的电容以获得样本数据集;以及
将所述样本数据集分类为一个或多个预先建立的脑病症类别。
2.如权利要求1和权利要求3至14中任一项所述的方法,其中,所述脑病症包括脑损伤。
3.如权利要求1至2和4至14中任一项所述的方法,其中,所述脑损伤是创伤性脑损伤或缺血性脑损伤。
4.如权利要求1至3和5至14中任一项所述的方法,其中,从所述受试者获得呼气样本包括在所述脑损伤之后10分钟内获得呼气样本。
5.如权利要求1至4和6至14中任一项所述的方法,其中,从所述受试者获得呼气样本包括在所述脑损伤发作之后24小时的时段内至少两次获得呼气样本。
6.如权利要求1至5和7至14中任一项所述的方法,其中,对所述样本数据集进行进一步分析以确定所述受试者的脑病症在24小时内的改善或恶化。
7.如权利要求1至6和8至14中任一项所述的方法,其中,从所述受试者获得呼气样本在所述受试者参加运动赛事之前执行。
8.如权利要求1至7和9至14中任一项所述的方法,其中,从所述受试者获得呼气样本在所述受试者参加运动赛事之前执行,并且在所述运动赛事开始或结束之后至少一个加法时间执行。
9.如权利要求1至8和10至14中任一项所述的方法,其中,感测和存储所述石墨烯变容二极管的电容以获得样本数据集是在一定偏置电压范围内执行的。
10.如权利要求1至9和11至14中任一项所述的方法,其中,所述偏置电压是从-3V至3V。
11.如权利要求1至10和12至14中任一项所述的方法,其中,在所述偏置电压范围内为每个石墨烯变容二极管存储至少40个离散的电容值。
12.如权利要求1至11和13至14中任一项所述的方法,其中,所述多个分立的石墨烯变容二极管被使用具有从1.5D至5D的偶极矩的极性化合物来官能化。
13.如权利要求1至12和14中任一项所述的方法,进一步存储关于所述受试者的、超出所感测的电容的附加数据作为被分类的所述样本数据集的一部分,所述附加数据包括一项各项中的至少一项:
所述受试者的先前创伤性脑损伤;
自已经发生导致所述受试者损伤的事件以后所经过的时间;
所述受试者的年龄;
神经检查的结果;
所述受试者所经历的症状;以及
关于脑病症的具体生物标志物的数据。
14.如权利要求1至13中任一项所述的方法,所述一个或多个预先建立的脑病症类别包括创伤性脑损伤、缺血性脑损伤、神经退行性脑异常、自身免疫性脑异常、精神异常、或发育异常。
15.一种治疗受试者的脑病症的方法,包括:
从所述受试者获得呼出气样本并且使所述呼出气样本与化学传感器元件接触,所述化学传感器元件包括多个分立的石墨烯变容二极管;
感测和存储所述分立的石墨烯变容二极管的电容以获得样本数据集;以及
将所述样本数据集分类为一个或多个预先建立的脑病症类别;
基于所述脑病症类别治疗所述患者。
