Weighing the cinnamon with the formula amount, adding a 10% ethanol solution, carrying out heating reflux extraction at 100 ℃, carrying out extraction for three times, wherein the extraction time is 1h, the volumes of the added ethanol solution are respectively 8 times, 6 times and 6 times of the volume of the cinnamon, and soaking the medicinal materials for 2h in the first extraction. After extraction is finished, combining the three extracting solutions, and sequentially carrying out centrifugation, impurity removal and reduced pressure concentration to obtain the cinnamon extract with the relative density of 1.20-1.30(60 ℃).
(3) Accurately weighing the extracted superfine powder of the grassleaf sweelflag rhizome, the hypericum perforatum, the freeze-dried powder of the valeriana jatamansi jones and 10 parts of lactose, 40 parts of water-soluble starch and 10 parts of superfine silica gel powder as auxiliary materials, putting the mixture into a mixer, mixing for 20min, adding the cinnamon extract, stirring for 35min, mixing to prepare a soft material, sieving the obtained soft material with a 14-mesh sieve to prepare wet granules, further placing the wet granules into a reduced-pressure drying oven, drying for 2h at 50 ℃, and finally sieving the dried granules with the 14-mesh sieve to obtain finished granules.

firstly, crushing the grass-leaved sweetflag by using an ultrafine crusher, and sieving the grass-leaved sweetflag by using a 200-mesh sieve to obtain the ultrafine grass-leaved sweetflag powder;
② weighing the hypericum perforatum and the valeriana jatamansi jones according to the formula ratio, and ultrasonically extracting for three times at 60 ℃ by using 50 percent ethanol solution, wherein each extraction time is 10min, the ultrasonic power is 110W, and the ultrasonic frequency is 90 kHz. The volumes of the ethanol solutions added for three times are respectively 8 times, 6 times and 6 times of the amount of the medicinal materials, and the medicinal materials are soaked for 1.5 hours during the first extraction. Weighing the pomegranate rind according to the formula ratio, adding 10% ethanol solution, carrying out heating reflux extraction at 100 ℃, carrying out extraction for three times, wherein the extraction time is 1h, the volumes of the added ethanol solution are 10 times, 8 times and 8 times of the volume of the pomegranate rind respectively, and soaking the medicinal materials for 2h during the first extraction. After extraction, combining the three extracting solutions, and sequentially carrying out centrifugation, impurity removal and reduced pressure concentration to obtain the pomegranate bark extract with the relative density of 1.20-1.30(60 ℃).
(3) Accurately weighing the extracted superfine powder of the grassleaf sweelflag rhizome, the hypericum perforatum, the spider aroma freeze-dried powder and the auxiliary materials of 35 parts of cane sugar and 15 parts of dextrin, putting the materials into a mixer for mixing for 15min, adding the pomegranate rind extract, stirring for 30min, mixing to prepare a soft material, preparing the obtained soft material into wet granules by using a 14-mesh sieve, further placing the wet granules into a reduced-pressure drying oven, drying for 2h at 50 ℃, and finally sieving the dried granules by using the 14-mesh sieve to obtain finished granules.

firstly, crushing the grass-leaved sweetflag by using an ultrafine crusher, and sieving the grass-leaved sweetflag by using a 150-mesh sieve to obtain the ultrafine grass-leaved sweetflag powder;
② weighing the hypericum perforatum and the valeriana jatamansi jones according to the formula ratio, and ultrasonically extracting for three times at 55 ℃ by using 60 percent ethanol solution, wherein each extraction time is 15min, the ultrasonic power is 100W, and the ultrasonic frequency is 100 kHz. The volumes of the ethanol solutions added for three times are respectively 8 times, 6 times and 6 times of the amount of the medicinal materials, and the medicinal materials are soaked for 2 hours during the first extraction. The volumes of the ethanol solutions added for three times are respectively 8 times, 6 times and 6 times of the amount of the medicinal materials, and the medicinal materials are soaked for 2 hours during the first extraction. After extraction is finished, combining the three extracting solutions, sequentially carrying out centrifugation and impurity removal, decompression and concentration at 60 ℃, and freeze drying to obtain the hypericum perforatum and valeriana jatamansi freeze-dried powder.

Fourth, example of experiment
Experimental example 1
The experimental example evaluates the difference of the extraction effects of different extraction methods. Evaluating the extraction effects of different extraction methods of the rhizoma acori graminei by taking the extraction rate of the volatile oil as an evaluation index; the extraction efficiency of hyperin was used as an evaluation index to evaluate the extraction effect of different extraction methods of hyperin, and the results are shown in tables 1-2.
Wherein, the comparative example 1 refers to the prior art of CN106728579A, and the volatile oil in the rhizoma acori graminei is extracted by a distillation method. Wherein, the extraction rate (%) of the volatile oil is equal to the mass of the volatile oil/the mass of the traditional Chinese medicinal materials multiplied by 100.

As can be seen from Table 1, the method of the example is more advantageous in the extraction of volatile oil from Acorus gramineus soland.
Furthermore, the invention researches another common extraction mode of the effective components of the rhizoma acori graminei, and concretely, the rhizoma acori graminei and petroleum ether are mixed and extracted at the temperature of 60 ℃ to obtain the rhizoma acori graminei extracting solution; and (4) performing reduced pressure rotary evaporation to obtain a rhizoma acori graminei concentrated solution. Similarly, the volatile oil of rhizoma Acori Graminei is used as effective component, the content of effective component in rhizoma Acori Graminei extractive solution is 5.43 + -0.22 (ml/100g), and the content of effective component in rhizoma Acori Graminei concentrated solution is 0.45 + -0.34 (ml/100 g).
In the common dosage forms, the rhizoma acori graminei extract cannot be directly used, and is prepared into the rhizoma acori graminei concentrated solution through concentration, but after concentration, the loss of active ingredients is large, and the drug effect is also greatly influenced, namely the extraction-concentration form of the rhizoma acori graminei is not suitable for practical application.
In table 2, comparative example 2 refers to CN106728579A in the prior art, and extraction is performed by a heating reflux extraction method. Wherein, the extraction rate (%) of hyperoside is equal to the amount of hyperoside in the extract/amount of hyperoside in the raw medicinal material x 100.

As can be seen from tables 1 and 2, the methods of the examples have advantages in the extraction of volatile oil from acorus gramineus and the extraction of hyperin, thereby creating good conditions for further improving the drug effect.
Experimental example 2
The experimental example is a clinical test of the traditional Chinese medicine granules for treating piglet diarrhea, and the specific experimental process is as follows:
the usage and dosage are as follows: the administration was carried out in a batch of 0.25g/kg.w body weight for 3 consecutive days.
Test animals: 200 pigs which are confirmed to be diarrhea of piglets in a certain pig farm of Hebei Handan are randomly selected from the pig herd which is diagnosed as diarrhea of piglets according to clinical symptoms and epidemiology, and are divided into 8 groups.
Test drugs: the Chinese medicinal granules prepared in examples 13 to 18 were used in the test groups 1 to 6 in sequence for 3 days, and the blank control group was not administered. The drug control group has the same dosage as the traditional Chinese medicinal materials in example 13, and is different from the traditional Chinese medicinal materials in extraction modes, according to the method recorded in the prior art CN106728579A, the rhizoma acori graminei adopts a distillation method to extract volatile oil and is included by cyclodextrin; the herba Hyperici perforati and radix Pimpinellae Candolleanae are extracted by heating and refluxing, and then made into lyophilized powder in the same way.
The test method comprises the following steps: the medicines are used for 3 days together, the number of cure, death, invalidation and improvement after 3 days of use is observed and counted, the total effective rate is calculated, and the test result is shown in table 3.
And (3) curing: the diarrhea and diarrhea clinical symptoms disappear, and the feces, the ingestion and the mental status are recovered to be normal.
Improvement: diarrhea and diarrhea symptoms of sick pigs are reduced but not completely recovered to normal.

Claims (10)

1. A pharmaceutical composition for treating piglet diarrhea is characterized by mainly comprising the following components: low temperature extract of Hypericum perforatum, low temperature extract of Valeriana jatamansi Jones, and ultrafine powder of Acorus tatarinowii Schott; the dosages of the traditional Chinese medicinal materials including the hypericum perforatum, the valeriana jatamansi jones and the grassleaf sweelflag rhizome are respectively 20-60 parts, 20-60 parts and 5-30 parts by weight; the low temperature extract corresponds to an extraction temperature of not higher than 60 ℃.
2. The pharmaceutical composition for treating diarrhea in piglets according to claim 1, further comprising cinnamon extract; the dosage of the traditional Chinese medicine cinnamon is not more than 20 parts.
3. The pharmaceutical composition for treating diarrhea in piglets according to claim 1 or 2, further comprising pomegranate rind extract; the usage amount of the traditional Chinese medicine pomegranate rind is not more than 20 parts.
4. The pharmaceutical composition for treating piglet diarrhea according to claim 1 or 2, which is prepared from the following Chinese medicinal materials in parts by weight: 20-60 parts of hypericum perforatum, 20-60 parts of valeriana jatamansi jones, 5-30 parts of rhizoma acori graminei, 5-20 parts of cinnamon and 5-20 parts of pomegranate rind.
5. A traditional Chinese medicine granule for treating piglet diarrhea, which is characterized in that the medicine composition of any one of claims 1 to 4 is used as a main material and is prepared into granules with auxiliary materials.
6. A method for preparing the traditional Chinese medicine granule as claimed in claim 5, comprising the following steps:
1) micronizing rhizoma Acori Graminei to obtain superfine powder;
extracting herba Hyperici perforati and radix Pimpinellae Candolleanae with ethanol water at a temperature of not higher than 60 deg.C to obtain low temperature extract;
2) granulating the superfine pulverized material of rhizoma Acori Graminei, low temperature extracts of herba Hyperici perforati and radix Pimpinellae Candolleanae, and adjuvants.
7. The method for preparing traditional Chinese medicine granules according to claim 6, wherein in the step 1), the temperature of not higher than 60 ℃ is 50-60 ℃.
8. The method for preparing traditional Chinese medicine granules according to claim 6, wherein in the step 1), the low-temperature extract is lyophilized powder prepared by lyophilizing the extract.

The method for preparing Chinese medicinal granules according to any one of claims 6 to 8, wherein in step 1), the low-temperature extraction is ultrasonic extraction, the medicinal materials are subjected to ultrasonic extraction for multiple times by using an alcohol-water solvent, and the extracting solutions are combined.
10. The method for preparing a Chinese medicinal granule according to claim 6, wherein the Chinese medicinal material further comprises cinnamon and/or pomegranate rind, the cinnamon and/or pomegranate rind are/is extracted by heating and refluxing with an alcohol-water solvent, and the obtained extract is used after being made into an extract.
