Example two: effectiveness test of the Chinese medicinal composition applied to acupuncture points for treating yang qi deficiency and meridian stasis.
Case (sample) selection correlation criteria and selection grouping method.
1. Diagnostic criteria:
the diagnosis standard of the pattern of yang-qi deficiency and venation stasis is formulated according to the definition of the pattern of yang-qi deficiency and venation stasis in TCM syndrome:
(1) yang qi deficiency manifested by lassitude, fatigue, qi deficiency, disinclination to talk, aversion to heat, cold, loose stool, etc.;
(2) the manifestations of meridian stasis, such as heavy limbs, numbness of limbs, and muscular pain;
(3) a pale tongue with petechia and ecchymosis, and a thready and unsmooth or deep and thready pulse.
2. Inclusion (selection) criteria.
The patients who meet the diagnosis standard and are 18-80 years old and willing to receive acupuncture point application treatment.
3. Exclusion criteria.
Patients accompanying the following should be excluded from the subject: (1) allergic constitution and people allergic to various drugs; (2) the patient is not suitable for the acupoint application due to the self reasons (such as skin damage or inflammation at the acupoint part); (3) patients with serious diseases such as cardiovascular and cerebrovascular diseases, gastrointestinal tract, liver, kidney, hematopoietic system and endocrine system; (4) patients with diabetic peripheral neuropathy or limb venous thrombosis; (5) patients with mental illness or other conditions that lead to poor compliance; (6) the study panel did not scale to the number of patients undergoing the study.

Rejection standard: patients who did not follow the study protocol during treatment.
Shedding standard: patients who did not complete the entire course of treatment and follow-up observations.

5. A grouping situation.
All the cases are patients with yang qi deficiency and vein stasis syndrome, which are diagnosed in traditional Chinese medicine hospitals in 2016 (1) to 2020 (12), and the total number of the cases is 179. Subjects were randomized into treatment and control groups using randomization software. The patients in the treatment group are divided into a treatment group 1, a treatment group 2 and a treatment group 3 according to the sequence of the treatment group 1:1: 1.

After selecting the acupuncture points, taking a certain amount of the fine powder of the traditional Chinese medicine composition and purified water with the temperature of 60-80 ℃, and mixing the following traditional Chinese medicine compositions in percentage by mass: purified water =2 g: 3ml of the mixture is fully and uniformly mixed to prepare medicinal mud; taking a proper amount of medicinal mud, rubbing into medicinal mud balls with the diameter of 1.0-1.5 cm, placing the medicinal mud balls in a plaster, and directly sticking the medicinal mud balls on the selected acupuncture points, wherein one plaster is stuck on each acupuncture point.
2. The Chinese medicinal composition components of different treatment groups and control groups are compared and tested.
The traditional Chinese medicine composition of the treatment group 1 comprises 12 parts of white mustard seed, 12 parts of corydalis tuber, 10 parts of euphorbia kansui, 3 parts of asarum, 6 parts of evodia rutaecarpa, 6 parts of pepper, 6 parts of clove, 12 parts of rhizoma cyperi, 6 parts of pepper, 12 parts of dried ginger, 15 parts of rhizoma typhonii, 12 parts of rhizoma atractylodis, 15 parts of eucommia bark, 15 parts of sharpleaf galangal fruit, 10 parts of angelica, 30 parts of astragalus root, 15 parts of teasel root, 12 parts of cynomorium songaricum, 12 parts of phellodendron bark, 15 parts of radix angelicae pubescentis, 12 parts of loranthus. The acupoint application is carried out for 4-6 h/time, every other day is 1 time, 5 times are 1 treatment course, and 2 treatment courses are total.
The traditional Chinese medicine composition of the treatment group 2 comprises 10 parts of white mustard seed, 12 parts of corydalis tuber, 10 parts of euphorbia kansui, 3 parts of asarum, 6 parts of evodia rutaecarpa, 4 parts of pepper, 6 parts of clove, 12 parts of rhizoma cyperi, 4 parts of pepper, 10 parts of dried ginger, 15 parts of rhizoma typhonii, 12 parts of rhizoma atractylodis, 12 parts of eucommia bark, 15 parts of sharpleaf galangal fruit, 10 parts of angelica, 30 parts of astragalus root, 15 parts of teasel root, 10 parts of cynomorium songaricum, 12 parts of phellodendron bark, 15 parts of radix angelicae pubescentis, 12 parts of loranthus. The acupoint application is carried out for 4-6 h/time, every other day is 1 time, 5 times are 1 treatment course, and 2 treatment courses are total.
The traditional Chinese medicine composition of the treatment group 3 comprises 10 parts of white mustard seed, 12 parts of corydalis tuber, 10 parts of euphorbia kansui, 3 parts of asarum, 6 parts of evodia rutaecarpa, 6 parts of pepper, 6 parts of clove, 12 parts of rhizoma cyperi, 6 parts of pepper, 12 parts of dried ginger, 15 parts of rhizoma typhonii, 12 parts of rhizoma atractylodis, 15 parts of eucommia bark, 15 parts of sharpleaf galangal fruit, 10 parts of angelica, 30 parts of astragalus root, 15 parts of teasel root, 12 parts of cynomorium songaricum, 12 parts of phellodendron bark, 12 parts of radix angelicae pubescentis, 10 parts of loranthus. The acupoint application is carried out for 4-6 h/time, every other day is 1 time, 5 times are 1 treatment course, and 2 treatment courses are total.
The composition in the control group comprises a mixture of food pigment, dextrin and starch, and is applied to acupuncture points for 4-6 h/time, 1 time every other day, and 5 times of the composition are 1 treatment course for 2 treatment courses.
3. The evaluation standard of the curative effect of the syndrome of yang-qi deficiency and vein blood stasis.
The traditional Chinese medicine is formulated according to the traditional Chinese medicine syndrome curative effect standard in the clinical research guiding principle of new traditional Chinese medicines issued by the ministry of health of China.
(1) And (3) healing: the original symptoms completely disappear, and no relapse occurs within 6 months after the treatment is stopped;
(2) the effect is shown: the original symptoms are obviously improved, and only slight numbness, pain or discomfort is left;
(3) the method has the following advantages: the original symptoms are slightly relieved;
(4) and (4) invalidation: the original symptoms are not obviously improved or are aggravated.
And thirdly, testing results.
1. Clinical curative effect
The total effective rate of the treatment group is 84.44% (76 cases/90 cases), the total effective rate of the control group is 5.62% (5 cases/90 cases), the comparison difference of the two groups has statistical significance (P is less than 0.01), namely, the curative effect of the treatment group is obviously better than that of the control group, and detailed results are shown in table 1.

2. Adverse reaction
No obvious adverse reaction is seen in the control group; in the treatment group, only 2 patients showed local skin redness after 6h of application, and all of the redness resolved within 24 hours after drug withdrawal without other adverse effects.

A traditional Chinese medicine composition for acupoint application for yang-qi deficiency and vein stasis syndrome is characterized by comprising the following traditional Chinese medicine components: semen Sinapis Albae, rhizoma corydalis, radix kansui, herba asari, fructus evodiae, fructus Piperis, flos Caryophylli, rhizoma Cyperi, fructus Zanthoxyli, Zingiberis rhizoma, radix Aconiti lateralis Preparata, rhizoma Atractylodis, Eucommiae cortex, fructus Alpinae Oxyphyllae, radix Angelicae sinensis, radix astragali, radix Dipsaci, herba Cynomorii, cortex Phellodendri, radix Angelicae Pubescentis, herba Taxilli, fructus Ligustri Lucidi and Ecliptae herba.
2. The traditional Chinese medicine composition for acupoint application for yang qi deficiency and vein stasis syndrome according to claim 1, which comprises the following traditional Chinese medicine components in parts by weight:
10-14 parts of semen brassicae, 10-14 parts of rhizoma corydalis, 8-12 parts of euphorbia kansui, 1-5 parts of asarum, 4-8 parts of fructus evodiae, 4-8 parts of pepper, 4-8 parts of clove, 10-14 parts of rhizoma cyperi, 4-8 parts of pepper, 10-14 parts of rhizoma zingiberis, 13-17 parts of rhizoma typhonii, 10-14 parts of rhizoma atractylodis, 12-17 parts of eucommia ulmoides, 13-17 parts of fructus alpiniae oxyphyllae, 8-12 parts of angelica sinensis, 25-35 parts of astragalus membranaceus, 13-17 parts of radix dipsaci, 10-14 parts of cynomorium songaricum, 10-14 parts of cortex phellodendri, 13-17 parts of radix angelicae pubescentis, 10-14 parts of parasitic loranthus, 13-.
