Abstract

The present invention belongs to the field of cosmetics, and more particularly, to a cosmetic composition for moisturizing skin and improving skin barrier comprising an electrolyte material, which comprises sodium chloride, potassium chloride, sodium bicarbonate and calcium chloride. The technical problem of how to provide a cosmetic composition for moisturizing skin or improving skin barrier, which increases the synthesis of ceramide, thereby not only improving skin barrier but also having excellent effects on skin moisturizing, is solved by the technical scheme of the present invention.

Description

Cosmetic composition for moisturizing skin and improving skin barrier comprising electrolyte material
Technical Field
The present invention relates to a cosmetic composition comprising an electrolyte material.
Background
The skin acts as an organ, which is constantly in contact with the environment outside the body, and mainly acts as a physical barrier. The skin is roughly divided into three layers: epidermis, dermis and subcutaneous fat, wherein the stratum corneum of the epidermis inhibits the loss of moisture and electrolytes to the exterior, thereby preventing the skin from drying out and providing an environment for the normal biochemical metabolism of the skin. And, it can also protect the human body from humidity and temperature changes, ultraviolet rays, other physical or chemical external stimuli.
When the moisture content is maintained at about 10% to 30%, the layer of keratinocytes that constitutes the normal skin barrier appears to be smooth, moist and healthy skin. However, when the moisture content of the keratinocyte layer is equal to or less than 10%, the skin becomes dry, cracked and rough, and looks older. Thus, if certain moisture and electrolytes of the skin are not maintained, the acidity and osmotic pressure of the skin are reduced, the skin becomes dry and thus is sensitive to even a small stimulus, thereby possibly causing itching, stinging, inflammation, and the like. To improve this impaired skin barrier, lost oil and moisture may be provided to the skin by a cosmetic composition. So far, research into cosmetics for providing oil and moisture to the skin has been actively conducted, but research into cosmetics including electrolytes contributing to immediate and rapid recovery upon application to the skin and compositions thereof is still required.
When the electrolyte balance of the skin is disrupted, drying may continue even after the cosmetic is used. When the osmotic pressure of the cosmetic is higher than that of the skin, the skin becomes dry because moisture in skin cells escapes to the outside due to the osmotic pressure. In contrast, when the osmotic pressure of the cosmetic is lower than that of the skin, swelling and destruction of cells may be caused due to excessive entry of water into the cells. Therefore, there is a need to research and develop a cosmetic composition having similar osmotic pressure to that of electrolytes present in the human body to help promptly and rapidly restore the skin when it is applied to the skin, thereby maintaining skin homeostasis and improving skin barrier and moisture retention.
Disclosure of Invention
Technical problem
One aspect of the present invention provides a cosmetic composition for moisturizing skin or improving skin barrier, comprising sodium chloride, potassium chloride, sodium bicarbonate, and calcium chloride.
Technical scheme
One aspect of the present invention provides a cosmetic composition for moisturizing skin or improving skin barrier, comprising sodium chloride, potassium chloride, sodium bicarbonate, and calcium chloride. In one example, it was confirmed that the expression level of the CerS3 gene was significantly higher in the composition prepared by containing all of sodium chloride, potassium chloride, sodium bicarbonate, and calcium chloride, compared to the composition prepared by containing sodium chloride, potassium chloride, sodium bicarbonate, or calcium chloride. In addition, according to one aspect, it was confirmed that the skin moisture level of the composition comprising sodium chloride, potassium chloride, sodium bicarbonate and calcium chloride in a certain content ratio was significantly improved immediately as compared with the comparative example. Therefore, since the composition according to an aspect entirely contains sodium chloride, potassium chloride, sodium bicarbonate, and calcium chloride, the effects of effectively promoting the synthesis of ceramide, improving skin barrier weakened by aging or external stimuli, and instantly enhancing skin moisturizing power and moisturizing duration power are excellent, and thus may be used to improve skin barrier or skin moisturizing. In this case, the "skin moisturizing" may refer to any action of maintaining skin moisture or preventing moisture loss.
In a specific embodiment, the sodium chloride is 0.5 to 1.5 weight% (w/v), the potassium chloride is 0.005 to 0.1 weight% (w/v), the sodium bicarbonate is 0.01 to 0.1 weight% (w/v), and the calcium chloride is 0.005 to 0.1 weight% (w/v), based on the total weight of the composition.
In one embodiment, the sodium chloride, potassium chloride, sodium bicarbonate, and calcium chloride may be mixed in a weight ratio of 85 to 97: 0.5 to 5: 1 to 5: 0.5 to 5. The sodium chloride, potassium chloride, sodium bicarbonate and calcium chloride may be mixed, for example, in a weight ratio of 90 to 97: 0.5 to 2: 1 to 5: 0.5 to 3, 90 to 97: 0.5 to 1.5: 1 to 5: 0.5 to 3.5, 90 to 97: 0.5 to 1.5, 2 to 5: 0.5 to 3, 90 to 97: 0.5 to 1.5: 2 to 4: 0.5 to 4, 90 to 97: 0.5 to 1.5: 0.5 to 3.5: 0.5 to 3, 90 to 97: 1 to 2: 1 to 5: 1 to 3, 90 to 97: 1 to 2: 1 to 4: 1 to 5, 90 to 97: 1 to 1.5: 1 to 5: 1.5 to 3, 92 to 96: 0.5: 2 to 4: 1 to 3, 93 to 97: 0.5 to 2: 2 to 3, 90 to 5: 1 to 5: 1.5: 1 to 3, 90 to 5: 1.5: 1 to 3, and 5: 0.5 to 5: 1 to 5: 1.5: 3, 95: 1 to 5: 1.5: 1 to 5: 3, 93 to 97: 0.5: 1 to 5: 1 to 5: 1.5: 1 to 5: 3, and. At this time, when the mixing ratio of sodium chloride, potassium chloride, sodium bicarbonate and calcium chloride is less than the range, there is a problem that the skin moisturizing and barrier improving effects are negligible; when it exceeds the range, there is a problem of affecting the stability of the dosage form or precipitating in the cosmetic composition, etc.
In one embodiment, the cosmetic composition may have an osmolality of 150 to 300 mOsm/L. For example, it may be 150 to 250mOsm/L, 150 to 200mOsm/L, 200 to 300mOsm/L, 200 to 250mOsm/L, or 250 to 300 mOsm/L. When the osmotic pressure of the cosmetic composition is less than 150mOsm/L, since the osmotic pressure concentration of the cosmetic composition is lower than that of the skin, moisture excessively enters into skin cells, thereby possibly causing cell swelling and destruction; when it exceeds 300mOsm/L, since the osmolality of the cosmetic composition is higher than that of the skin, moisture in the skin cells escapes to the outside, and thus there may occur a problem that the skin becomes dry. According to an aspect, since the cosmetic composition comprising 0.5 to 1.5 parts by weight of sodium chloride, 0.005 to 0.1 parts by weight of potassium chloride, 0.01 to 0.1 parts by weight of sodium bicarbonate, and 0.005 to 0.1 parts by weight of calcium chloride has an osmotic pressure similar to that of an electrolyte present in a human body, based on 100 parts by weight of the total composition, it may help to promptly recover skin upon its application to the skin, and improve skin barrier and moisturization.
The cosmetic composition according to an aspect may be prepared into a specific formulation by a method well known in the art of cosmetics, and may further comprise a cosmetically acceptable additive. Such additives include stabilizers, solubilizers, opacifiers, vitamins, pigments, nutrients or combinations thereof, and the like, and may vary depending on the formulation to be prepared.

Examples include solutions, suspensions, emulsions, pastes, gels, creams, lotions, shampoos, surfactant-containing cleansers, foundation lotions, foundation creams, and the like, and more particularly, can be prepared in the form of pliable lotions, nourishing emulsions, nourishing creams, massage creams, essences, eye creams, cleansing foams, cleansing lotions, facial masks, and the like.
Another aspect of the present invention provides a composition for external preparation for skin comprising sodium chloride, potassium chloride, sodium bicarbonate and calcium chloride. The specific contents of the sodium chloride, potassium chloride, sodium bicarbonate and calcium chloride are as described above.
The external preparation composition according to one embodiment may be prepared in a formulation generally prepared in the art. For example, it can be prepared in the form of cream, gel, patch, spray, ointment, plaster, emulsion, liniment, paste, cataplasm, and the like.

Cellular assessment to improve the efficacy of moisturization and barrier
As skin ages, the amount of ceramide, an important component that accounts for 40% of the lipid constituting the skin barrier, is significantly reduced, and moisture retention and the skin barrier are changed. In this experiment, the expression level of CerS3, which is an enzyme that synthesizes ceramide, was measured at the mRNA level using Human keratinocytes (Human keratincyte) to measure the moisturizing and barrier-improving effects of skin.
(1) Preparation of sample
The samples prepared in example 1 and comparative examples 1 to 5 were filtered with a 0.2 μm Syringe filter (Syringe filter). This was added to the cell culture broth so that the final concentration was 10% (v/v). Pure distilled water was added as a control.
(2) Culture of human keratinocytes
HaCaT cells, which are keratinocytes, were cultured in Dulbecco Modified Eagle's Medium (DMEM), which is the most widely used medium for culturing skin cells. After mixing 10% Fetal Bovine Serum (FBS) and 1% penicillin in a 100mm culture dish, cells were cultured. Maintaining the culture solution containing the sample at 37 deg.C and 5% CO2The culture was carried out in an incubator at a concentration and humidity of 95% for 24 hours.
(3) Test of skin moisturizing Effect
The cells cultured in the medium of (1) were recovered, washed with cooled Phosphate Buffered Saline (PBS), and total RNA was extracted using PureLink RNA miniprep kit (seemer feishel technology). The specific RNA extraction procedure followed the manual attached to the product. The gene expression change was measured by quantitative real-time PCR (qRT-PCR) which binds a fluorescent substance to a DNA product amplified in Polymerase Chain Reaction (PCR) and continuously detects the fluorescent substance. To quantify the expression of ceramide synthase CerS3 gene, the fluorescence emitted by the PCR product was measured by sibogerin (SYBR green, siemer fly technologies). A reaction solution was prepared by mixing 0.2. mu.M of a primer, 50mM potassium chloride, 20mM Tris-HCl (pH8.4), 0.8mM deoxyribonucleoside triphosphate (dNTP), 0.5U Extaq DNA polymerase, 3mM magnesium chloride and 1 XSaibogelin in a PCR tube. A total of 40 cycles of denaturation (30 seconds), annealing (annealing) and polymerization (polymerization) were performed using a PCR instrument at 94 ℃ for 3 minutes, 30 seconds at 58 ℃ and 30 seconds at 72 ℃ for 30 seconds, and the fluorescence intensity was measured after the end of each cycle. The fluorescence intensity of the CerS3 gene was normalized to GAPDH value and 100% of the control group was converted to compare the difference. The primer sequences used in the experiments are shown in table 2 below.
