Preparation method

Pharmaceutical composition
The invention provides a pharmaceutical composition for eliminating dampness and strengthening spleen, which comprises the above composition.
The pharmaceutical composition further comprises pharmaceutically acceptable excipients including any solvent, solid excipient, diluent, binder, disintegrant, or other liquid excipient, dispersant, flavoring or suspending agent, surfactant, isotonic agent, thickening agent, emulsifier, preservative, solid binder, glidant or lubricant, and the like, suitable for the particular target dosage form. Except insofar as any conventional adjuvant is incompatible with the pharmaceutical compositions of the invention, e.g., any adverse biological effect produced or interaction with any other component of a pharmaceutically acceptable composition in a deleterious manner, its use is contemplated by the present invention.
Substances that may serve as pharmaceutically acceptable excipients include, but are not limited to, ion exchangers; aluminum; aluminum stearate; lecithin; serum proteins, such as human serum albumin; buffer substances such as phosphates; glycine; sorbic acid; potassium sorbate; partial glyceride mixtures of saturated vegetable fatty acids; buffering agents such as magnesium hydroxide and aluminum hydroxide; glycols, such as propylene glycol and polyethylene glycol; esters such as ethyl oleate and ethyl laurate; agar; alginic acid; pyrogen-free water; isotonic salt; ringer's solution; ethanol; phosphoric acid buffer solution; and other non-toxic suitable lubricants such as sodium lauryl sulfate and magnesium stearate; a colorant; a release agent; coating the coating material; a sweetener; a flavoring agent; a fragrance; preservatives and antioxidants.
The pharmaceutical compositions of the present invention may be administered in unit dosage form. The administration dosage form can be liquid dosage form or solid dosage form. The liquid dosage forms can be true solutions, colloids, microparticles, and suspensions. Other dosage forms such as tablet, capsule, dripping pill, aerosol, pill, powder, solution, suspension, emulsion, granule, etc.
Oral tablets and capsules may contain excipients such as binding agents, for example syrup, acacia, sorbitol, tragacanth or polyvinylpyrrolidone; fillers, such as lactose, sucrose, corn starch, calcium phosphate, sorbitol, glycine; lubricants, such as magnesium stearate, talc, polyethylene glycol, silica; disintegrants, such as potato starch; or acceptable humectants such as sodium lauryl sulfate. The tablets may be coated by methods known in the art of pharmacy.
Oral liquids may be in the form of suspensions, solutions, emulsions, syrups or elixirs containing hydrated oils, or may be presented as a dry product for constitution with water or other suitable vehicle before use. Such liquid preparations may contain conventional additives such as suspending agents, sorbitol, cellulose methyl ether, glucose syrup, gelatin, hydroxyethyl cellulose, carboxymethyl cellulose, aluminium stearate gelatin, hydrogenated edible fats and oils, emulsifying agents, such as lecithin, sorbitan monooleate, acacia; or a non-aqueous carrier (which may comprise an edible oil), such as almond oil, an oil such as glycerol, ethylene glycol, or ethanol; preservatives, e.g. methyl or propyl p-hydroxybenzoate, sorbic acid. Flavoring or coloring agents may be added if desired.
The invention will now be described with reference to specific examples, which are intended to be illustrative only and not to be limiting in any way.

200 patients (20-55 years old) with phlegm-dampness constitution were collected, and specifically, the patients were divided into 4 groups of 50 patients each. Respectively taking the dampness-eliminating and spleen-invigorating tea prepared by the schemes 1-4 for 8 weeks from the first group to the fourth group, and brewing one bag per person every day; the effect was then observed. The population to be excluded when collecting volunteers includes pregnant and pregnancies, lactating women; those who are allergic to the ingredients of the present compositions; patients who are participating in other drug clinical trials; the patients in the disease state are discovered through comprehensive physical examination; suspected or certain abuse of alcohol, past history of drugs, etc.
The curative effect results are as follows:

Example 2 toxicity test
Selecting 50 SPF-grade Wistar mice (25 male mice and female mice respectively) with the weight of 20-25g, dividing the mice into 5 groups, and dividing each group of the mice with the weight of 5 male mice and female mice into 5 groups, wherein the spleen-tonifying and dampness-eliminating tea prepared according to the formula in the scheme 1 in the embodiment 1 is brewed by adopting boiled water, and the adding amount of the boiled water is calculated according to the water consumption of normal tea brewing. The dose was administered at the maximum allowable volume of mice, 0.4ml/10g, continuously drenched for 30 days. During the period of tea infusion and 1 week after the withdrawal, the white mouse was observed to have normal appetite, activity, hair color and mental status. Mice develop normally by identifying hematology, blood biochemistry, organ tissue structure, and urinary convention. All mice survive healthily without any toxic and side effect; liver function and organ tissue state were observed by dissection, and there was no difference compared with normal mice.
Experiments prove that the spleen-tonifying dampness-eliminating tea provided by the invention has no toxic or side effect.
Example 3 intervention experiment on Damp-Heat syndrome rat behaviourology
Laboratory animal
60 SPF-level Wistar rats, half male and female, with the body mass of 180-220 g, purchased from Liaoning Biotechnology Limited, and the license number of the experimental animal: SCXK (jew) 2015-:

2011007) at 21-23 deg.C and humidity of 55-58%. The clean-grade feed for experimental animals was purchased from Beijing, Australian cooperative feed Co., Ltd (license number: SCXK (Jing) 2014-0010).
Test article
Example 1 the spleen-invigorating and dampness-eliminating tea prepared by the formula of scheme 1 is brewed with boiled water, and the amount of the boiled water is calculated by the water consumption of normal tea brewing.
Animal grouping and molding method
The method comprises the following steps of randomly dividing 60 Wistar rats with half male and female into a blank group, a model group, a dampness-eliminating and spleen-tonifying tea high-dose group (a dampness-eliminating and spleen-tonifying group), a dampness-eliminating and spleen-tonifying tea medium-dose group (a dampness-eliminating and spleen-tonifying group) and a dampness-eliminating and spleen-tonifying tea low-dose group (a dampness-eliminating and spleen-tonifying group), wherein each group comprises 12 Wistar rats. The blank group is fed with common mixed feed at the temperature of 21-23 ℃ and the relative humidity of 55-58% in a room temperature environment at 8:30 a.m. every day. Preparing a rat model with spleen and stomach damp-heat syndrome, and feeding the rat model with high-sugar and high-fat feed for 10 days. 9: 00-11: 00 a.m. each day, placing the artificial climate chamber in which the temperature is 33 +/-2 ℃ and the relative humidity is 75 +/-5% for 8 days. Day 9, 8 am: and (3) performing intragastric administration 30 times, performing intragastric administration 2mL of escherichia coli for 1 time per 200g, performing intensive infection for 1 time after 24 hours, performing intragastric administration 1mL per 200g, removing, and placing in a natural environment. The damp clearing decoction is prepared by taking out the damp clearing decoction from the 4 th day on the basis of preparing a rat model with spleen and stomach damp-heat syndrome, and filling low-dose, medium-dose and high-dose damp clearing and spleen tonifying tea into a natural environment for 12 hours, wherein each 200g of damp clearing and spleen tonifying tea is filled into a stomach for 2mL, and the dose is 1 time per day.
Observation indicator and detection method
The general condition of the animals including behavior, symptoms, signs and other indicators were observed and recorded every day. After gavage for 1h on days 3 and 11 of the experiment, the behavioral performance of the animals was observed by Open-field behavioral measurement. The small animal behavioural recording and analyzing system is used for recording, and the camera system is installed and fixed above an open field for recording. A single rat is placed in the center of a square experimental box, the peripheral wall of the experimental box is a black open field, the bottom of the experimental box is divided into 25 grids with equal size, and the middle 9 grids are called as central grids. Pinching the tail of the rat and slightly placing the tail of the rat into the central grid from the root 1/3, enabling the animal to adapt in the experimental box for more than 10min before each experiment, recording the action performance of the rat within 3min, including the staying time in the central grid, the number of movement grids (which is the score of horizontal movement and the number of grids where four claws enter), the number of standing times (which is the score of vertical movement and the score of two front claws leave the ground), and removing the excrement of the animal after each measurement, wherein each measurement is only performed for 1 time.
Statistical method
The data analysis uses SPSS 20.0 statistical package to process the data statistical treatment, the measurement data is expressed by mean + -standard deviation (' x + -s), the comparison between each group uses one-factor variance analysis, the difference P <0.05 is statistical significance.
Results
Influence of dampness-eliminating and spleen-strengthening tea on behavior, symptoms and physical signs of rats
The rats in each group have no obvious abnormal behavior. Most of the rats in the model group show lassitude, sleepiness and laziness, slow reaction, fluffy hair, withered and luggy color, reduced diet, increased defecation times and sticky and greasy stool along with the lapse of time, the body temperature of the rats is obviously increased after the rats are drenched with escherichia coli, and the spleen and stomach damp-heat syndrome is well simulated. The damp-clearing spleen-tonifying tea group with high dose, medium dose and low dose also shows symptoms of activity, diet frequency reduction, sticky stool and the like before the damp-clearing spleen-tonifying tea is infused, which indicates that the molding is successful; after administration, the damp-clearing spleen-tonifying tea has the advantages that the activity times of animals in high-dose, medium-dose and low-dose groups are gradually increased, the food consumption is increased, the defecation is gradually recovered to be normal, the reaction is recovered to be normal, the fur is glossy, and the mental state is recovered to be normal. See table 2.

Compared with a model group, the dampness-eliminating and spleen-tonifying tea can improve the behavioral performance of rats in each dose group, and the dampness-eliminating and spleen-tonifying tea has the function of regulating the behavioral performance of the rats, which is probably related to the effects of dampness elimination, spleen strengthening and heat clearing of the medicines in the formula and the function of improving the environment in a body.
Example 4 clinical cases
1. preferably, the composition comprises:
1.5 parts of phaseolus calcaratus, 1 part of semen coicis, 0.5 part of poria cocos, 1 part of Chinese yam, 1 part of tartary buckwheat, 1 part of white hyacinth bean, 1.5 parts of red date, 1.5 parts of orange peel, 0.2 part of hawthorn, 0.5 part of liquorice and 1 part of cape jasmine.
4. A method for preparing a composition according to any one of claims 1 to 3, comprising:
cooking red bean, coix seed and tartary buckwheat to obtain the cooked red bean, the cooked coix seed and the cooked tartary buckwheat;
pulverizing and mixing the cooked adzuki beans, the cooked adlay and the cooked tartary buckwheat, poria cocos, Chinese yam, white hyacinth beans, red dates, orange peel, hawthorn, liquorice and gardenia to obtain the composition;
optionally, treating the phaseolus calcaratus and the coix seeds at 80 ℃ for 20-30 minutes so as to obtain the cured phaseolus calcaratus and the cured coix seeds;
