Abstract
The invention provides a composition for eliminating dampness and strengthening spleen. The composition comprises: the red bean food comprises 1-2 parts by mass of red bean, 0.5-1.5 parts by mass of coix seed and 0.2-0.8 part by mass of poria cocos. The composition provided by the invention has the effects of clearing damp and tonifying spleen, can effectively relieve the symptoms of abdominal fullness and distention, inappetence, dry and sticky mouth, halitosis, loose stool, body sleepiness and hypodynamia, heavy limbs and excessive phlegm, and is reasonable in Chinese medicine compatibility, convenient to use, safe and nontoxic.
Description
Technical Field
The invention belongs to the field of food and medicine, and particularly relates to a composition, a method for preparing the composition, application of the composition in preparing food or medicine, a beverage and a pharmaceutical composition.
Background
Food and water enter the human body and are transformed into fine substances such as body fluids through the transportation and transformation of the spleen and stomach. If food and water are not normally transported and absorbed, the symptom of water-wet in traditional Chinese medicine can be caused. Whether food and water eaten by human body can be transformed into fine substances required by human body is critical to whether the spleen and stomach function is strong, and the spleen and stomach are the acquired root and mainly transport and resolve water dampness. If the spleen and stomach are fatigued, the food grain can not be transformed into the useful delicate substance after entering the human body, but into the pathological product-water-dampness. The traditional Chinese medicine holds that the dampness is accumulated as water, the accumulated water is accumulated into water and the water is condensed into phlegm, the water is accumulated too much and becomes water, and the water is accumulated too long and becomes phlegm.
On the other hand, due to climate change, people's living habits are bad, and factors such as air conditioning are excessively depended on, so that wind-cold-dampness pathogen easily invades the human body, and the moisture of the human body is aggravated.
Phlegm and water-dampness which has not yet been coagulated into phlegm are gathered together to form phlegm-dampness, and the phlegm-dampness flows everywhere along with qi in the body: stay in the liver, so that fatty liver is formed; if the waist is retained between the waist, the abdomen and the bucket waist are formed; edema and bloated due to overflow of the skin, muscles, face and limbs; it is also prone to cause irregular menstruation, leukorrhagia, lumbago, headache, and cervical spondylosis.
At present, dampness-eliminating and spleen-tonifying products on the market mainly adopt meal replacement powder, the effect is not obvious, and dampness-eliminating and spleen-tonifying medicines are inconvenient to take and have poor taste and cannot meet the requirements of users.
In view of the above, it is necessary to provide a composition for eliminating dampness and strengthening spleen to solve the above technical problems.
Disclosure of Invention
The present invention is directed to solving, at least to some extent, one of the technical problems in the related art.
To this end, in a first aspect of the invention, the invention proposes a composition for eliminating dampness and strengthening the spleen. According to an embodiment of the invention, the composition comprises: the red bean food comprises 1-2 parts by mass of red bean, 0.5-1.5 parts by mass of coix seed and 0.2-0.8 part by mass of poria cocos. The medicines with the effects of eliminating dampness and tonifying spleen disclosed in the prior art are numerous, and the inventor optimizes the formula through a large amount of theoretical analysis and experimental verification, changes one or more components in the formula, and obtains the formula with a less effect than the formula. In addition, the inventor finds that the components in the formula have good effects of clearing damp and tonifying spleen through a large number of experiments, and other mixture ratios are not as good as the formula.
At present, food materials/medicinal materials adopted for eliminating dampness and strengthening spleen are numerous in types, and meticulous screening finds that the food materials/medicinal materials can have the effects of eliminating dampness and strengthening spleen, and inducing diuresis and excreting dampness by compounding the red phaseolus bean, the coix seed and the poria cocos. Further, deep research and analysis show that the compounding ratio of the three components can obviously influence the drug effect, wherein the effects of clearing damp and tonifying spleen, and promoting diuresis and excreting dampness are better under the compounding ratio, so that the symptoms of digestive function reduction such as inappetence, loose stool, abdominal distension, sticky and greasy taste, diarrhea, halitosis and the like can be effectively improved, and the body is strong and energetic.

According to an embodiment of the present invention, the food or pharmaceutical product is used for treating abdominal fullness and distention, anorexia, dry and sticky mouth, halitosis, loose stool, tiredness and weakness, heavy limbs and excessive phlegm.

According to embodiments of the present invention, the beverage may further include food acceptable adjuvants such as preservatives, flavoring agents, excipients, coloring agents, sweetening agents, and the like. The food acceptable auxiliary materials include but are not limited to: water; salts or electrolytes, such as protamine sulfate, disodium hydrogen phosphate, potassium hydrogen phosphate, sodium chloride, zinc salts; lanolin; sugars such as lactose, glucose and sucrose; starches such as corn starch and potato starch; cellulose and its derivatives such as sodium carboxymethyl cellulose, ethyl cellulose and cellulose acetate; gum powder; malt; gelatin; talc powder; adjuvants such as cocoa butter; oils such as peanut oil, cottonseed oil, safflower oil, sesame oil, olive oil, corn oil and soybean oil.
Pharmaceutical composition
The invention provides a pharmaceutical composition for eliminating dampness and strengthening spleen, which comprises the above composition.
The pharmaceutical composition further comprises pharmaceutically acceptable excipients including any solvent, solid excipient, diluent, binder, disintegrant, or other liquid excipient, dispersant, flavoring or suspending agent, surfactant, isotonic agent, thickening agent, emulsifier, preservative, solid binder, glidant or lubricant, and the like, suitable for the particular target dosage form. Except insofar as any conventional adjuvant is incompatible with the pharmaceutical compositions of the invention, e.g., any adverse biological effect produced or interaction with any other component of a pharmaceutically acceptable composition in a deleterious manner, its use is contemplated by the present invention.
Substances that may serve as pharmaceutically acceptable excipients include, but are not limited to, ion exchangers; aluminum; aluminum stearate; lecithin; serum proteins, such as human serum albumin; buffer substances such as phosphates; glycine; sorbic acid; potassium sorbate; partial glyceride mixtures of saturated vegetable fatty acids; buffering agents such as magnesium hydroxide and aluminum hydroxide; glycols, such as propylene glycol and polyethylene glycol; esters such as ethyl oleate and ethyl laurate; agar; alginic acid; pyrogen-free water; isotonic salt; ringer's solution; ethanol; phosphoric acid buffer solution; and other non-toxic suitable lubricants such as sodium lauryl sulfate and magnesium stearate; a colorant; a release agent; coating the coating material; a sweetener; a flavoring agent; a fragrance; preservatives and antioxidants.
The pharmaceutical compositions of the present invention may be administered in unit dosage form. The administration dosage form can be liquid dosage form or solid dosage form. The liquid dosage forms can be true solutions, colloids, microparticles, and suspensions. Other dosage forms such as tablet, capsule, dripping pill, aerosol, pill, powder, solution, suspension, emulsion, granule, etc.
Oral tablets and capsules may contain excipients such as binding agents, for example syrup, acacia, sorbitol, tragacanth or polyvinylpyrrolidone; fillers, such as lactose, sucrose, corn starch, calcium phosphate, sorbitol, glycine; lubricants, such as magnesium stearate, talc, polyethylene glycol, silica; disintegrants, such as potato starch; or acceptable humectants such as sodium lauryl sulfate. The tablets may be coated by methods known in the art of pharmacy.
Oral liquids may be in the form of suspensions, solutions, emulsions, syrups or elixirs containing hydrated oils, or may be presented as a dry product for constitution with water or other suitable vehicle before use. Such liquid preparations may contain conventional additives such as suspending agents, sorbitol, cellulose methyl ether, glucose syrup, gelatin, hydroxyethyl cellulose, carboxymethyl cellulose, aluminium stearate gelatin, hydrogenated edible fats and oils, emulsifying agents, such as lecithin, sorbitan monooleate, acacia; or a non-aqueous carrier (which may comprise an edible oil), such as almond oil, an oil such as glycerol, ethylene glycol, or ethanol; preservatives, e.g. methyl or propyl p-hydroxybenzoate, sorbic acid. Flavoring or coloring agents may be added if desired.

the main symptoms of phlegm-dampness constitution include: abdominal distention, inappetence, dry and sticky mouth, halitosis, loose stool, asthenia, heavy limbs, and excessive phlegm. The curative effect is evaluated by comparing the relevant symptoms of the patients before and after treatment, wherein the effect is shown as follows: most symptoms disappear after treatment; the method has the following advantages: partial symptoms are improved after treatment; and (4) invalidation: the symptoms are not obvious after treatment. Specific efficacy statistics are shown in table 1.

The results show that: the formula proportion of the scheme 1 is superior to that of other schemes.
Example 2 toxicity test
Selecting 50 SPF-grade Wistar mice (25 male mice and female mice respectively) with the weight of 20-25g, dividing the mice into 5 groups, and dividing each group of the mice with the weight of 5 male mice and female mice into 5 groups, wherein the spleen-tonifying and dampness-eliminating tea prepared according to the formula in the scheme 1 in the embodiment 1 is brewed by adopting boiled water, and the adding amount of the boiled water is calculated according to the water consumption of normal tea brewing. The dose was administered at the maximum allowable volume of mice, 0.4ml/10g, continuously drenched for 30 days. During the period of tea infusion and 1 week after the withdrawal, the white mouse was observed to have normal appetite, activity, hair color and mental status. Mice develop normally by identifying hematology, blood biochemistry, organ tissue structure, and urinary convention. All mice survive healthily without any toxic and side effect; liver function and organ tissue state were observed by dissection, and there was no difference compared with normal mice.
Experiments prove that the spleen-tonifying dampness-eliminating tea provided by the invention has no toxic or side effect.
Example 3 intervention experiment on Damp-Heat syndrome rat behaviourology
Laboratory animal
60 SPF-level Wistar rats, half male and female, with the body mass of 180-220 g, purchased from Liaoning Biotechnology Limited, and the license number of the experimental animal: SCXK (jew) 2015-:2011007) at 21-23 deg.C and humidity of 55-58%. The clean-grade feed for experimental animals was purchased from Beijing, Australian cooperative feed Co., Ltd (license number: SCXK (Jing) 2014-0010).
Test article
Example 1 the spleen-invigorating and dampness-eliminating tea prepared by the formula of scheme 1 is brewed with boiled water, and the amount of the boiled water is calculated by the water consumption of normal tea brewing.
Animal grouping and molding method
The method comprises the following steps of randomly dividing 60 Wistar rats with half male and female into a blank group, a model group, a dampness-eliminating and spleen-tonifying tea high-dose group (a dampness-eliminating and spleen-tonifying group), a dampness-eliminating and spleen-tonifying tea medium-dose group (a dampness-eliminating and spleen-tonifying group) and a dampness-eliminating and spleen-tonifying tea low-dose group (a dampness-eliminating and spleen-tonifying group), wherein each group comprises 12 Wistar rats. The blank group is fed with common mixed feed at the temperature of 21-23 ℃ and the relative humidity of 55-58% in a room temperature environment at 8:30 a.m. every day. Preparing a rat model with spleen and stomach damp-heat syndrome, and feeding the rat model with high-sugar and high-fat feed for 10 days. 9: 00-11: 00 a.m. each day, placing the artificial climate chamber in which the temperature is 33 +/-2 ℃ and the relative humidity is 75 +/-5% for 8 days. Day 9, 8 am: and (3) performing intragastric administration 30 times, performing intragastric administration 2mL of escherichia coli for 1 time per 200g, performing intensive infection for 1 time after 24 hours, performing intragastric administration 1mL per 200g, removing, and placing in a natural environment. The damp clearing decoction is prepared by taking out the damp clearing decoction from the 4 th day on the basis of preparing a rat model with spleen and stomach damp-heat syndrome, and filling low-dose, medium-dose and high-dose damp clearing and spleen tonifying tea into a natural environment for 12 hours, wherein each 200g of damp clearing and spleen tonifying tea is filled into a stomach for 2mL, and the dose is 1 time per day.
Observation indicator and detection method
The general condition of the animals including behavior, symptoms, signs and other indicators were observed and recorded every day. After gavage for 1h on days 3 and 11 of the experiment, the behavioral performance of the animals was observed by Open-field behavioral measurement. The small animal behavioural recording and analyzing system is used for recording, and the camera system is installed and fixed above an open field for recording. A single rat is placed in the center of a square experimental box, the peripheral wall of the experimental box is a black open field, the bottom of the experimental box is divided into 25 grids with equal size, and the middle 9 grids are called as central grids. Pinching the tail of the rat and slightly placing the tail of the rat into the central grid from the root 1/3, enabling the animal to adapt in the experimental box for more than 10min before each experiment, recording the action performance of the rat within 3min, including the staying time in the central grid, the number of movement grids (which is the score of horizontal movement and the number of grids where four claws enter), the number of standing times (which is the score of vertical movement and the score of two front claws leave the ground), and removing the excrement of the animal after each measurement, wherein each measurement is only performed for 1 time.
Statistical method
The data analysis uses SPSS 20.0 statistical package to process the data statistical treatment, the measurement data is expressed by mean + -standard deviation (' x + -s), the comparison between each group uses one-factor variance analysis, the difference P <0.05 is statistical significance.
Results
Influence of dampness-eliminating and spleen-strengthening tea on behavior, symptoms and physical signs of rats
The rats in each group have no obvious abnormal behavior. Most of the rats in the model group show lassitude, sleepiness and laziness, slow reaction, fluffy hair, withered and luggy color, reduced diet, increased defecation times and sticky and greasy stool along with the lapse of time, the body temperature of the rats is obviously increased after the rats are drenched with escherichia coli, and the spleen and stomach damp-heat syndrome is well simulated. The damp-clearing spleen-tonifying tea group with high dose, medium dose and low dose also shows symptoms of activity, diet frequency reduction, sticky stool and the like before the damp-clearing spleen-tonifying tea is infused, which indicates that the molding is successful; after administration, the damp-clearing spleen-tonifying tea has the advantages that the activity times of animals in high-dose, medium-dose and low-dose groups are gradually increased, the food consumption is increased, the defecation is gradually recovered to be normal, the reaction is recovered to be normal, the fur is glossy, and the mental state is recovered to be normal. See table 2.

The effect of dampness-eliminating and spleen-invigorating tea on the residence time of the rat central compartment is shown in table 3.

The experiment result shows that on the 3 rd day of the experiment, compared with the blank group, the residence time of the central lattices of each dose group of the model group and the dampness-eliminating and spleen-tonifying tea is obviously prolonged, which indicates that the space cognitive ability of rats of each dose group of the model group and the administration group is reduced; the exercise lattice number and the upright times are obviously reduced, which shows that the excitability of rats in each dosage group of the model group and the dampness-eliminating and spleen-tonifying tea is reduced, the adaptability to a new environment is weakened, the autonomous activity and the exploration behavior of the rats in a strange environment are reduced, and the success of model building is indicated. Compared with a model group, the dampness-eliminating and spleen-tonifying tea can improve the behavioral performance of rats in each dose group, and the dampness-eliminating and spleen-tonifying tea has the function of regulating the behavioral performance of the rats, which is probably related to the effects of dampness elimination, spleen strengthening and heat clearing of the medicines in the formula and the function of improving the environment in a body.
Example 4 clinical cases
1. Li Shi, male, age 44, white and greasy tongue coating, sticky and greasy mouth, loose stool, loose body, lack of strength, fat body and fat abdomen, which belongs to the phlegm-damp constitution according to the differentiation of the traditional Chinese medicine. After the dampness-eliminating and spleen-tonifying tea is taken for 6 months, clinical symptoms are fully recovered, and the physique is basically recovered to be normal.
2. Zhangzhi, female, age 22, greasy tongue coating, sticky mouth with breath, poor appetite, lassitude and hypodynamia, which is dialectical from phlegm-dampness constitution in traditional Chinese medicine. After the dampness-eliminating and spleen-tonifying tea is taken for 3 months, clinical symptoms are healed, and the physique is basically recovered to be normal.
3. Qin in, female, 38 years old, greasy tongue coating, drowsiness and hypodynamia, dreaminess, dizziness, body weight, which is the syndrome of phlegm-dampness according to the traditional Chinese medicine. After the dampness-eliminating and spleen-tonifying tea disclosed by the invention is taken for 4 months, clinical symptoms are healed, and the physique is basically recovered to be normal.
4. Lu Yi, male, age 42, white and greasy tongue coating, profuse sputum, fullness and oppression in chest and epigastrium, profuse and sticky sweat, heavy body, easy drowsiness, lethargy and sticky stool, which are distinguished by the traditional Chinese medicine to belong to phlegm-damp physique. After the dampness-eliminating and spleen-tonifying tea is taken for 4 months, clinical symptoms are healed, and the physique is basically recovered to be normal.

2. The composition as claimed in claim 1, further comprising: chinese yam, tartary buckwheat, white hyacinth bean, red date, orange peel, hawthorn, liquorice and gardenia.
3. The composition as claimed in claim 1, comprising:
1.2-1.8 parts by mass of phaseolus calcaratus, 0.5-1.5 parts by mass of coix seeds, 0.2-0.8 part by mass of poria cocos, 0.5-1.5 parts by mass of Chinese yam, 0.5-1.5 parts by mass of tartary buckwheat, 0.5-1.5 parts by mass of white hyacinth beans, 1-2 parts by mass of red dates, 1-2 parts by mass of orange peel, 0.1-0.3 part by mass of hawthorn, 0.2-0.8 part by mass of liquorice and 0.5-1.5 parts by mass of gardenia;
preferably, the composition comprises:
1.5 parts of phaseolus calcaratus, 1 part of semen coicis, 0.5 part of poria cocos, 1 part of Chinese yam, 1 part of tartary buckwheat, 1 part of white hyacinth bean, 1.5 parts of red date, 1.5 parts of orange peel, 0.2 part of hawthorn, 0.5 part of liquorice and 1 part of cape jasmine.
4. A method for preparing a composition according to any one of claims 1 to 3, comprising:
cooking red bean, coix seed and tartary buckwheat to obtain the cooked red bean, the cooked coix seed and the cooked tartary buckwheat;
pulverizing and mixing the cooked adzuki beans, the cooked adlay and the cooked tartary buckwheat, poria cocos, Chinese yam, white hyacinth beans, red dates, orange peel, hawthorn, liquorice and gardenia to obtain the composition;
optionally, treating the phaseolus calcaratus and the coix seeds at 80 ℃ for 20-30 minutes so as to obtain the cured phaseolus calcaratus and the cured coix seeds;
optionally, treating the tartary buckwheat at 140 ℃ for 20-30 minutes so as to obtain the aged tartary buckwheat;
optionally, the crushing is to crush the material to 20-80 meshes.
5. A beverage for eliminating dampness and strengthening spleen, comprising the composition of any one of claims 1 to 3.
6. The beverage according to claim 5, wherein the beverage is in the form of a solid, gel or liquid.
7. A pharmaceutical composition for eliminating dampness and strengthening spleen, comprising the composition of any one of claims 1 to 3.
8. The pharmaceutical composition of claim 7, wherein the pharmaceutical composition is in the form of a capsule, tablet or granule.
9. Use of the composition of any one of claims 1 to 3 for the preparation of a food or pharmaceutical product for eliminating dampness and strengthening the spleen.
10. The use according to claim 9, wherein the food or pharmaceutical product is for the treatment of abdominal fullness and distention, loss of appetite, dry and sticky mouth, bad breath, loose stools, poor health, heavy limbs and profuse sputum.
