背景技术

发明详述
本公开内容一般地涉及能够调节NMDA受体的化合物,例如,NMDA受体拮抗剂、激动剂或部分激动剂以及使用公开的化合物的组合物和/或方法。在某些实施方案中,本文描述的化合物结合表达某些NR2亚型的NMDA受体。在某些实施方案中,本文所述化合物结合一种NR2亚型且不结合另一种。应当理解,公开的化合物可以调节其它蛋白靶标和/或特定NMDA受体亚型。

剂量单位形式的规格取决于并且直接依赖于:(a)所选化合物的独特特性和要实现的特定治疗效果,以及(b)配制这样的活性化合物以治疗个体的敏感性的技术上的固有限制。

提供了通过施用治疗有效量的本文所述的化合物或包括这样的化合物的组合物来治疗有此需要的患者的病症的方法。在某些实施方案中,所述病症可以是精神病症。例如,可以治疗精神疾病。在某些实施方案中,可以治疗神经系统病症。例如,可以治疗影响中枢神经系统、周围神经系统和/或眼睛的病症。在某些实施方案中,可以治疗神经变性疾病。

本公开内容的其它方法包括在有此需要的患者中治疗孤独症和/或孤独症谱群障碍的方法,所述方法包括向患者施用有效量的化合物。在一个实施方案中,提供了用于减轻有此需要的患者的孤独症症状的方法,所述方法包括向所述患者施用有效量的公开的化合物。例如,在施用后,所述化合物可以减少孤独症的一种或多种症状的发生率,诸如避免眼神接触、社交失败、注意力缺陷、情绪低落、活动过度、声音异常灵敏、不适当的言语、睡眠中断和持续言语。可以相对于该未治疗的个体或其它未治疗的个体中的发病率来测量这种降低的发病率。
本文还提供了调节细胞中孤独症靶基因表达的方法,其包括使细胞与有效量的本文所述的化合物接触。孤独症基因表达可以例如选自ABAT、APOE、CHRNA4、GABRA5、GFAP、GRIN2A、PDYN和PENK。在某些实施方案中,提供了在患有突触可塑性相关障碍的患者中调节突触可塑性的方法,所述方法包括向所述患者施用有效量的化合物。

在0℃在氮气氛下向化合物5(50mg,0.177mmol)在CH2Cl2(1mL)中的搅拌溶液中加入HCl(4M的在1,4-二烷中的溶液,1mL)。将反应混合物在室温搅拌16h。起始原料耗尽以后(通过TLC),在减压下蒸发挥发物。在圆底烧瓶中将粗制物用饱和的NaHCO3水溶液(5mL)稀释并进行低压冻干48h。将得到的固体产物先后与5%MeOH/CH2Cl2和乙醚一起研磨并在真空下干燥,得到作为灰白色固体的AG(32mg)。

B.NMDAR激动剂测定
如Moskal等人,“GLYX-13:a monoclonal antibody-derived peptide that actsas an N-methyl-D-aspartate receptor modulator,”Neuropharmacology,49,1077-87,2005所述进行测定。设计这些研究来确定试验化合物是否在表达NMDAR2A、NMDAR2B、NMDAR2C或NMDAR2D的HEK细胞膜中发挥促进NMDAR活化的作用,这通过[3H]MK-801结合的增加来测量。
在该测定中,在25℃在有饱和浓度的谷氨酸(50μM)和变化浓度的试验化合物(1x10-15M-1x10-7M)或1mM甘氨酸存在下将300μg表达NMDAR的HEK细胞膜提取物蛋白质预温育15分钟。在加入0.3μCi的[3H]MK-801(22.5Ci/mmol)之后,将反应物再次在25℃(非平衡条件)温育15分钟。使用Brandel设备经由快速过滤将结合的和游离的[3H]MK-801分离。
在分析数据中,对于试验化合物的各浓度或对于1mM甘氨酸,测量在滤器上剩余的[3H]MK-801的DPM(每分钟分解)。将配体的每个浓度(N=2)的DPM值取平均。从DPM值的最佳拟合曲线确定基线值(该曲线用GraphPad程序和log(激动剂)相对于应答(3参数)算法建模),然后从数据集的全部点扣除。然后相对于1mM甘氨酸计算%最大[3H]MK-801结合:将全部已扣除基线的DPM值除以对于1mM甘氨酸的平均值。然后从%最大[3H]MK-801结合数据的最佳拟合曲线获得EC50和%最大活性,该曲线用GraphPad程序和log(激动剂)相对于应答(3参数)算法建模。

C.药代动力学测定

D:PORSOLT测定

G.BENNETT神经损伤测定
如通过爪撤回阈值所测量的,用机械止痛的Bennett模型来评估化合物的镇痛作用。Bennett GJ,Xie YK,“A peripheral mononeuropathy in rat that producesdisorders of pain sensation like those seen in man,”Pain 33:87-107,1988。对动物进行坐骨神经慢性收缩神经损伤手术,一旦动物从手术中恢复但在应用von Frey细丝后仍表现出较低的爪撤回阈值,就测试镇痛应答。媒介物动物接受手术,然后接受媒介物而不是试验化合物。在试验化合物或媒介物施用后1小时、24小时和1周,对动物进行试验。

在试验过程中,使大鼠适应悬吊式金属丝网(1cm X 1cm,金属丝直径为0.3cm)的表面15-20分钟。从最小开始,将每根Von Frey细丝垂直压在受影响的(同侧的)后爪的足底表面直至稍微弯曲,然后保持6秒钟。如果在撤回细丝后未立即观察到明显的后爪撤回或退缩行为,则以相同方式使用下一个更大的细丝。如果有应答,则使用更低的细丝。重复进行直到收集到六个应答。
对于所有研究,在开始测试痛觉超敏(定义为低于5的爪撤回阈值)的研究之前,将动物作基线。用加巴喷丁(150mg/kg,PO)测试了动物的亚组,以确保至少50%镇痛。一旦确认动物已准备好研究开始,便在各组之间平衡动物。所有的研究人员都对处理条件不知情。经由经口管饲法(PO)给动物施用0.1、1或10mg/kg的试验化合物,给对照组的动物施用加巴喷丁(150mg/kg,PO)或媒介物(0.5%Na-CMC在0.9%无菌盐水中,PO)。在施用后1小时进行测试,在施用后24小时和1周对动物进行重新测试。使用以下方程式计算每只动物的镇痛百分比:镇痛百分比=[(log(x)-y)/((log(z)-y)]*100,其中x=药物治疗的动物的爪撤回阈值,以克为单位,y=媒介物治疗组的log(x)值的平均值,z=原初(naive)动物的爪撤回阈值,以克为单位(使用15的历史值)。化合物AD的结果如下,其中在化合物施用后1小时、24小时和1周测量镇痛百分比:对于0.1mg/kg剂量:在1h为38.7%,在24h为34.5%,和在1周为19.7%;对于1mg/kg剂量:在1h为24.1%,在24h为26.5%,和在1周为13.2%;和对于10mg/kg剂量:在1h为61.2%,在24h为45.1%,和在1周为39.5%。该研究具有加巴喷丁对照组,其中150mg/kg剂量的示例(典型)加巴喷丁对照值在1h为77%,在24h为17%,和在1周为0%。对于该研究,确认加巴喷丁有效(在施用后1h表现出至少50%镇痛)。加巴喷丁并未不同于媒介物,并在施用后24h和1周没有产生镇痛(<5%)。
