Abstract
The invention discloses a preparation method of a ginsenoside extract and application of the ginsenoside extract in skin care products. The preparation method of the ginsenoside extract comprises the following steps: taking fresh ginseng as a raw material, crushing the fresh ginseng, performing enzymolysis on the ginseng powder by adopting compound biological enzyme, then adding water for extraction to obtain water extract and filter residue, soaking the filter residue in ethanol solution, passing the extract through a macroporous resin adsorption column, eluting with ethanol, concentrating the eluent and the water extract under reduced pressure, and drying to obtain the ginseng extract. The ginsenoside extract obtained by the method has high content of effective components, the extraction rate is greatly improved, and the method has mild extraction conditions, short time, simple operation and lower cost. The ginsenoside extract has strong antioxidant, antiaging, and moisturizing effects on skin, and can be used for preparing skin care products with skin whitening, moisturizing, antiaging, etc.
Description
Technical Field
The invention relates to a preparation process of a ginsenoside extract, and also relates to an application of the ginsenoside extract in a skin care product, belonging to the fields of traditional Chinese medicine technology and skin care product application.
Background
The ginseng is a traditional rare traditional Chinese medicine, has the effects of building up the body and prolonging life after long-term use, and has a long medicinal history in China. With the development of economy in China and the continuous improvement of the living standard of people, people pay more and more attention to the skin health of people, and people do not meet the medicinal effect of using ginseng alone. The ginseng contains various bioactive components such as ginsenoside, ginseng polysaccharide, protein, polyynol, volatile oil, amino acid and derivatives thereof, polypeptide, adenosine, vitamin, trace elements and the like, wherein the ginsenoside is one of the main active components, and researches show that the ginsenoside has a repairing effect on skin cells and has a very remarkable effect on the aspect of skin aging resistance. The ginsenoside extract can inhibit UVB-induced matrix metalloprotease MMP-1 and MMP-13 generation by regulating MAPKs and NF-kB channels in human fibroblasts, thereby achieving the effect of resisting photoaging. The ginsenoside extract can effectively improve the SOD activity of heart and brain tissues, reduce the contents of lipid peroxide and lipofuscin, resist peroxidation damage and have obvious effect of delaying senility. The ginseng extract can be added into skin care products to delay skin aging, prevent skin from drying and dehydrating, and increase skin elasticity, thereby playing the roles of protecting skin luster and tenderness, and preventing and reducing skin wrinkles, and the ginseng active substance also has the performance of inhibiting melanin generation, so that the skin is white and smooth. At present, the ginsenoside serving as an antioxidant active substance has a huge application prospect in daily chemicals.
At present, most of ginsenoside preparation methods are complex in process, long in time consumption, high in cost and environment pollution due to the use of a large amount of organic solvents, and the obtained extracting solution is strong in irritation and cannot be directly used in daily chemicals. In addition, the ginseng extract obtained by the existing technological means has low ginsenoside content and low extraction rate, so that the overall yield of the ginseng extract prepared from ginseng stem and leaf medicinal materials is low, and the large-scale application of industrial production is difficult.
Therefore, the invention provides a preparation method of a ginsenoside extract and application of the ginsenoside extract in skin care products, and at least one of the problems is solved.
Disclosure of Invention
The first purpose of the invention is to provide a preparation method of a ginsenoside extract, which has the advantages of simple operation, short time, high extraction efficiency, ginsenoside extraction rate of over 90 percent and ginsenoside content of over 99 percent in the extract.
The second purpose of the invention is to provide the application of the ginsenoside extract prepared by the method in skin care products. The skin care product is added with saponin extracted from ginseng and other components on the basis, so that the product has the functions of whitening, moisturizing, resisting wrinkles and aging, inhibiting the generation of melanin, making the skin glittering and translucent, promoting collagen proliferation, regulating skin metabolism, protecting skin barrier, assisting wound healing, effectively increasing the low temperature resistance, high temperature resistance, radiation resistance and oxidation resistance of the skin, and further delaying the aging process of the skin.
In order to achieve the first purpose, the invention adopts the following technical scheme:
a method for preparing ginsenoside extract comprises the following steps:
(1) taking Changbai mountain ginseng, and cleaning for later use;
(2) draining the ginseng under forest, drying, crushing and sieving to obtain 70-100 mesh ginseng under forest powder;
(3) carrying out enzymolysis on the ginseng powder under forest by adopting compound biological enzyme, carrying out ultrasonic treatment while carrying out enzymolysis, and filtering after enzymolysis to obtain an enzymolysis material; the composite biological enzyme consists of cellulase, protease, pectinase, hemicellulase and amylase in a mass ratio of (1-10) to (1-10);
(4) adding water to the enzymolysis material for extraction, and filtering to obtain filter residue and a water extract, and reserving the water extract;
(5) soaking the filter residue obtained in the step (4) in an ethanol solution, filtering to obtain a first soaking solution, and soaking the filter residue in the same method and filtering to obtain a second soaking solution;
(6) mixing the water extract obtained in the step (4), the first extract and the second extract obtained in the step (5) to obtain a mixed solution, passing the mixed solution through an adsorption column filled with macroporous resin at a speed of 2-3 times of column volume per hour until the mixed solution is saturated in adsorption, wherein the adsorption temperature is 30-50 ℃, eluting with ethanol, and collecting eluent; concentrating the eluate under reduced pressure to obtain ginsenoside concentrate, and drying the concentrate.
Preferably, in the step (1), the ginseng under Changbai mountain forest is obtained, which has glossy color, fine and smooth skin lines, full texture and clean section and is clear.
Preferably, in the step (2), the drying time is 30-40 minutes, and the drying temperature is 50-65 ℃; the rotating speed of the pulverizer is controlled at 120-180 r/min, and the pulverizing time is 12-15 min.
Preferably, in the step (3), the composite biological enzyme accounts for 0.1-0.3% of the weight of the ginseng powder under forest; the ultrasonic treatment time is 0.5-1.0 hour; the enzymolysis time is 3-4 hours, and the enzymolysis temperature is 30-40 ℃.
Furthermore, the method for extracting ginsenoside by enzymolysis adds a specific enzyme preparation in the step of enzyme extraction, carries out enzymolysis on natural raw material ginseng, can hydrolyze cellulose, hemicellulose, pectin and the like in plants, breaks plant cell walls, converts the cellulose and the like into micromolecular soluble saccharides, is beneficial to dissolving out effective components in cells, enables the extraction of the effective components to be more complete, and can effectively avoid the precipitation phenomenon generated in the traditional alcohol extraction process through the step of enzyme extraction, thereby not only improving the clarity of liquid medicine, but also increasing the content of nutrient components.
Preferably, in the step (4), the enzymolysis material is extracted by water with the mass of 8-14 times of that of the enzymolysis material for 0.5-2 hours.
Preferably, in the step (5), the mass ratio of the filter residue to the ethanol solution is 1:2-1: 4; the soaking conditions are as follows: soaking in 60-80 vol% ethanol solution at 30-40 deg.C for 2-4 hr.
Preferably, in the step (6), the ethanol elution conditions are as follows: eluting with 50-70 vol% ethanol solution at a flow rate of 0.5-0.6 column volume/hr.
Preferably, in the step (6), the macroporous resin is a D101 nonpolar macroporous adsorption resin.
Furthermore, the macroporous adsorption resin selectively adsorbs organic substances from the solution through physical adsorption, thereby achieving the purpose of separation and purification. The physical and chemical properties of the product are stable, the product is not dissolved in acid, alkali and organic solvent, the selectivity to organic matter is good, the product is not affected by the existence of inorganic salt, strong ion and low molecular compound, and the product can absorb solvent in water and organic solvent to swell. The D101 macroporous adsorption resin can be applied to separation and purification of chemical components of Chinese herbal medicines, so that a large amount of impurities can be removed, and effective components can be enriched.
Preferably, in the step (6), the temperature of reduced pressure concentration is 45-60 ℃, and the vacuum degree is-0.08 to-1 MPa; the drying temperature is 90-170 ℃.
The 'under forest ginseng' is a common Chinese medicinal finished product and can be directly purchased.
The crushing and drying are carried out by adopting conventional crushing and drying methods and equipment in the field of traditional Chinese medicine extraction.
In order to achieve the second purpose, the invention adopts the following technical scheme:
an application of the ginsenoside extract in skin care product is provided.
The skin care product comprises the following components in percentage by weight: ginsenoside extract 0.5-10%, palmitoyl pentapeptide 0.001-2%, acetyl hexapeptide 0.001-2%, panthenol 0.5-3%, glycerol 1-5%, vitamin C0.1-0.5%, cysteine 0.1-0.5%, glutathione 0.1-2%, SymSave H (p-hydroxyacetophenone) 0.1-1%, and Hydrolite 6O (1, 2-hexanediol) 0.1-1%.
Further, palmitoyl pentapeptide and acetyl hexapeptide are small molecule cosmetic agents. The palmitoyl pentapeptide can promote the synthesis of basic protein, increase the hyperplasia of collagen, elastic fiber and hyaluronic acid, improve the moisture content of skin, resist wrinkles and tighten skin. The acetyl hexapeptide can prevent wrinkle, improve aged skin, and reduce skin wrinkle.
Further, panthenol (previtamin B5) is a component of coenzyme a, which plays an important role in cellular metabolism. Such as the delivery of acyl groups during fatty acid biosynthesis and carbohydrate neogenesis. An increase in skin lipid synthesis will lead to an improvement in the barrier, facilitating the recovery of damaged skin.
Further, panthenol also promotes fibroblast proliferation and re-epithelialization of the epidermis, and promotes wound healing. Meanwhile, panthenol is beneficial to increasing the permeability of the skin, is a good absorption enhancer and can effectively promote the absorption of other components. Panthenol is also an effective humectant, has moisturizing and hydrating effects on the stratum corneum of the skin, thus improving the problems of rough skin, skin exfoliation, skin elasticity and the like, and has better combined effect with glycerin in the formula.
Further, vitamin C is a naturally occurring antioxidant and can be used for preventing and treating sunburn of the skin. The topical vitamin C can enhance the collagen production function of skin and reduce the degradation of collagen. In vitro studies have shown that elastin synthesis by fibroblasts is inhibited by vitamin C, which is beneficial in reducing elastin accumulation which is characteristic of photoaged skin. Vitamin C can also inhibit melanin formation by tyrosinase and melanoma cells, thereby whitening skin.
Furthermore, the components such as cysteine, glutathione and the like are good antioxidants, and can well protect effective components such as polypeptide, vitamin C and the like in the formula from being degraded, so as to fully exert the efficacy and the function of the components.
In addition, unless otherwise specified, any range recited herein includes any value between the endpoints and any sub-range defined by any value between the endpoints or any value between the endpoints.
The invention has the following beneficial effects:
(1) the preparation method of the ginsenoside extract has the advantages of simple operation, short time, high extraction efficiency, and the extraction rate of the ginsenoside can reach more than 90 percent, and the content of the ginsenoside in the extract can reach more than 99 percent.
(2) The ginsenoside extract of the invention contains a large amount of ginsenoside natural surfactant and a small amount of amino acid and other moisturizing factors which can well play a role in moisturizing. Wherein the ginsenoside has surfactant and antioxidant effects, and the amino acid has moisture keeping effect. The fresh ginseng ginsenoside extract provided by the invention can be used for a single dosage form or a special skin protection formula.
(3) The invention integrates the saponin extract of the ginseng with other components in the skin care field, so that the skin care product has the functions of whitening, moisturizing, resisting wrinkles and aging, inhibiting the generation of melanin, making the skin glittering and translucent, promoting the collagen proliferation, regulating the skin metabolism, protecting the skin barrier and assisting the wound healing, and effectively increases the low temperature resistance, high temperature resistance, radiation resistance and oxidation resistance of the skin, thereby delaying the aging process of the skin.
Detailed Description
In order to more clearly illustrate the invention, the invention is further described below in connection with preferred embodiments. It is to be understood by persons skilled in the art that the following detailed description is illustrative and not restrictive, and is not to be taken as limiting the scope of the invention.
The preparation process in the present invention is a conventional one unless otherwise specified, and the raw materials used are commercially available from publicly available sources or prepared according to the prior art unless otherwise specified, and the percentages are mass percentages unless otherwise specified.
Example 1
A method for preparing ginsenoside extract comprises the following steps:
1. taking 5g of Changbai mountain ginseng with glossy color, fine skin lines, full and firm texture and clean section, cleaning and then placing for one hour for later use;
2. drying the ginseng under forest obtained in the step 1 in a dryer at 50 ℃ for 30 minutes after water is drained off, crushing the dried ginseng under forest in a crusher at the rotation speed of 120 revolutions per minute for 12 minutes, and sieving to obtain 70-mesh ginseng under forest powder;
3. preparing 0.005g of composite biological enzyme comprising cellulase, protease, pectinase, hemicellulase and amylase, wherein the mass ratio of the cellulase, the protease, the pectinase, the hemicellulase and the amylase is 1:1:1:1, carrying out enzymolysis on the ginseng powder obtained in the step 2 at 30 ℃ for 3 hours, carrying out ultrasonic treatment simultaneously for 0.5 hour, and filtering to obtain an enzymolysis material;
4. extracting the enzymolysis material obtained in the step 3 with 8 times of water by mass for 0.5 hour, filtering to obtain filter residue, and retaining the water extract;
5. soaking the filter residue obtained in the step 4 in an ethanol solution with the volume concentration of 60% at 30 ℃ for 2 hours, filtering to obtain a first soaking solution, and soaking and filtering the obtained filter residue by the same method to obtain a second soaking solution; wherein the mass ratio of the filter residue to the ethanol solution is 1: 2;
6. mixing the water extract obtained in the step 4, the first extract and the second extract obtained in the step 5 to obtain a mixed solution, allowing the mixed solution to pass through an adsorption column filled with D101 nonpolar macroporous adsorption resin at the speed of 2 times of column volume/hour until the mixed solution is saturated in adsorption, wherein the adsorption temperature is 30 ℃, eluting the mixed solution with an ethanol solution with the volume concentration of 50% at the flow rate of 0.5 times of column volume/hour, and collecting eluent; concentrating the eluate under reduced pressure at 45 deg.C and vacuum degree of-0.08 MPa to obtain ginsenoside concentrate, and drying the concentrate.
Through detection, the extraction rate of the ginsenoside can reach 90.1%, and the content of the ginsenoside can reach 99.2%.
The invention also provides an application of the ginsenoside extract in a skin care product, and the skin care product is prepared from the following components in percentage by weight: ginsenoside extract 0.5%, palmitoyl pentapeptide 0.001%, acetyl hexapeptide 0.001%, panthenol 0.5%, glycerol 1%, vitamin C0.1%, cysteine 0.1%, glutathione 0.1%, symsuve H0.1%, Hydrolite 6O 0.1%, and deionized water 97.5%.
Example 2
A method for preparing ginsenoside extract comprises the following steps:
1. taking 5g of Changbai mountain ginseng with glossy color, fine skin lines, full and firm texture and clean section, cleaning and then placing for one hour for later use;
2. drying the ginseng under forest obtained in the step 1 in a dryer at 65 ℃ for 40 minutes, crushing the dried ginseng under forest in a crusher at the rotation speed of 180 revolutions per minute for 15 minutes, and sieving to obtain 100-mesh ginseng under forest powder;
3. preparing 0.015g of compound biological enzyme comprising cellulase, protease, pectinase, hemicellulase and amylase, wherein the mass ratio of the cellulase, the protease, the pectinase, the hemicellulase and the amylase is 1:10:1:10:1, carrying out enzymolysis on the ginseng powder obtained in the step 2 at 40 ℃ for 4 hours, simultaneously carrying out ultrasonic treatment for 1 hour, and filtering to obtain an enzymolysis material;
4. extracting the enzymolysis material obtained in the step 3 with 14 times of water by mass for 2 hours, filtering to obtain filter residue, and reserving water extract;
5. soaking the filter residue obtained in the step 4 in an ethanol solution with the volume concentration of 80% at 40 ℃ for 4 hours, filtering to obtain a first soaking solution, and soaking and filtering the obtained filter residue by the same method to obtain a second soaking solution; wherein the mass ratio of the filter residue to the ethanol solution is 1: 4;
6. mixing the water extract obtained in the step 4 and the first and second extracts obtained in the step 5 to obtain a mixed solution, allowing the mixed solution to pass through an adsorption column filled with D101 nonpolar macroporous adsorption resin at the speed of 3 times of column volume/hour until the mixed solution is saturated in adsorption, wherein the adsorption temperature is 50 ℃, eluting the mixed solution with an ethanol solution with the volume concentration of 70% at the flow rate of 0.6 times of column volume/hour, and collecting eluent; concentrating the eluate under reduced pressure at 60 deg.C and vacuum degree of-1 MPa to obtain ginsenoside concentrate, and drying the concentrate.
Through detection, the extraction rate of the ginsenoside can reach 90.4%, and the content of the ginsenoside can reach 99.5%.
The invention also provides an application of the ginsenoside extract in a skin care product, and the skin care product is prepared from the following components in percentage by weight: 10% of ginsenoside extract, 2% of palmitoyl pentapeptide, 2% of acetyl hexapeptide, 3% of panthenol, 5% of glycerol, 0.5% of vitamin C, 0.5% of cysteine, 2% of glutathione, SymSave H1%, 1% of Hydrolite 6O 1% and 73.0% of deionized water.
It should be understood that the above-mentioned embodiments of the present invention are only examples for clearly illustrating the present invention, and are not intended to limit the embodiments of the present invention, and it will be obvious to those skilled in the art that other variations or modifications may be made on the basis of the above description, and all embodiments may not be exhaustive, and all obvious variations or modifications may be included within the scope of the present invention.
Claims (10)
1. A method for preparing a ginsenoside extract is characterized by comprising the following steps:
(1) taking Changbai mountain ginseng, and cleaning for later use;
(2) draining the ginseng under forest, drying, crushing and sieving to obtain 70-100 mesh ginseng under forest powder;
(3) carrying out enzymolysis on the ginseng powder under forest by adopting compound biological enzyme, carrying out ultrasonic treatment while carrying out enzymolysis, and filtering after enzymolysis to obtain an enzymolysis material; the composite biological enzyme consists of cellulase, protease, pectinase, hemicellulase and amylase in a mass ratio of (1-10) to (1-10);
(4) adding water to the enzymolysis material for extraction, and filtering to obtain filter residue and a water extract, and reserving the water extract;
(5) soaking the filter residue obtained in the step (4) in an ethanol solution, filtering to obtain a first soaking solution, and soaking the filter residue in the same method and filtering to obtain a second soaking solution;
(6) mixing the water extract obtained in the step (4), the first extract and the second extract obtained in the step (5) to obtain a mixed solution, passing the mixed solution through an adsorption column filled with macroporous resin at a speed of 2-3 times of column volume per hour until the mixed solution is saturated in adsorption, wherein the adsorption temperature is 30-50 ℃, eluting with ethanol, and collecting eluent; concentrating the eluate under reduced pressure to obtain ginsenoside concentrate, and drying the concentrate.
2. The preparation method according to claim 1, wherein in the step (2), the drying time is 30-40 minutes, and the drying temperature is 50-65 ℃; the rotating speed of the pulverizer is controlled at 120-180 r/min, and the pulverizing time is 12-15 min.
3. The preparation method according to claim 1, wherein in the step (3), the composite biological enzyme accounts for 0.1-0.3% of the mass fraction of the under-forest ginseng powder; the ultrasonic treatment time is 0.5-1.0 hour; the enzymolysis time is 3-4 hours, and the enzymolysis temperature is 30-40 ℃.
4. The preparation method according to claim 1, wherein in the step (4), the enzymolysis material is extracted with water with the mass of 8-14 times of that of the enzymolysis material for 0.5-2 hours.
5. The preparation method according to claim 1, wherein in the step (5), the mass ratio of the filter residue to the ethanol solution is 1:2-1: 4; the soaking conditions are as follows: soaking in 60-80 vol% ethanol solution at 30-40 deg.C for 2-4 hr.
6. The method according to claim 1, wherein in the step (6), the ethanol is eluted under the following conditions: eluting with 50-70 vol% ethanol solution at a flow rate of 0.5-0.6 column volume/hr.
7. The process according to claim 1, wherein in the step (6), the temperature of the concentration under reduced pressure is 45 to 60 ℃ and the degree of vacuum is-0.08 to-1 MPa; the drying temperature is 90-170 ℃.
8. A ginsenoside extract obtained by the preparation method of claims 1-7.
9. A ginsenoside extract of claim 8, used in a skin care product.

The use according to claim 9, wherein the skin care product comprises the following components in percentage by weight: ginsenoside extract 0.5-10%, palmitoyl pentapeptide 0.001-2%, acetyl hexapeptide 0.001-2%, panthenol 0.5-3%, glycerol 1-5%, vitamin C0.1-0.5%, cysteine 0.1-0.5%, glutathione 0.1-2%, SymSave H0.1-1%, and Hydrolite 6O 0.1-1%.
