背景技术
研究表明,益生菌对机体的健康生理具有多种作用,能够抑制肠内有害菌促进肠内有益菌的增殖, 或者减少病原体繁殖生长的营养物质, 在肠道中形成“竞争排斥”的生态平衡环境,同时,产生发酵非消化淀粉的酶, 而这些非消化淀粉是肠道微生物群落的主要能源物质,另外机体可以利用肠道益生菌产生的短链脂肪酸防止胆汁对 DNA的损害,清除自由基,并调节自身细胞活性 。
随着社会的发展进步,工作节奏的加快,越来越多的人忽视自身营养问题,再加上膳食的不均衡,结构单一,导致很多潜在疾病的爆发,如:肠道菌群失衡造成的腹泻、便秘、消化不良,蛋白质的缺乏造成的机体代谢失常、免疫力低下等。壳寡糖作为一种功能性低聚糖,经国内外大量研究验证,能够活化肠内益生菌,抑制肠道中致病菌的生长,促进人体对矿物质的吸收,提高机体免疫力,因此,开发壳寡糖功能性系列牦牛乳制品必将有广阔的市场前景。
发明内容
本发明的目的在于提供一种壳寡糖益生菌牦牛乳多肽配方奶粉,以解决现有功能性乳制品品种单一的问题。
本发明另一个目的在于提供一种壳寡糖益生菌牦牛乳多肽配方奶粉的制备方法,具有成本低,能有效解决酶解过程中产生苦味肽的问题。
本发明所采用的技术方案为:
一种壳寡糖益生菌牦牛乳多肽配方奶粉,该配方奶粉由如下重量份的原料制成:全脂乳粉50-65份、脱盐乳清粉10-30份、全脂牦牛乳粉5-15份、牦牛乳蛋白多肽粉5-15份、壳寡糖5-15份、维生素C 0.005-0.01份、磷脂酰丝氨酸0.90-0.95份、乳双歧杆菌,HN-019,0.01-0.02份、嗜酸乳杆菌,NCMF,0.01-0.02份、保加利亚乳杆菌0.01-0.02份、嗜热链球菌0.01-0.02份。
一种壳寡糖益生菌牦牛乳多肽配方奶粉的制备方法,步骤如下:
A、发酵:将枯草芽孢杆菌活化,振荡培养6-12h,得到菌液,再以新鲜牦牛乳为底物混合菌液发酵,结束后经高压灭菌得到混合产物;
B、离心:将灭菌后的混合产物在6 000 r/min离心机中离心10-15 min,得到上清液;
C、超滤:将得到的上清液经 0.45 μm 滤膜过滤,去除菌体及残渣;采用截留分子量为10 kDa、的超滤离心管对膜过滤液进行分离,再经冷冻干燥得到牦牛乳蛋白多肽产品。
干混 :将冷冻干燥好的牦牛乳蛋白多肽和全脂乳粉、脱盐乳清粉、全脂牦牛乳粉、牦牛乳蛋白多肽粉、壳寡糖、维生素C、磷脂酰丝氨酸在紫外线强度≥70μw/cm2条件下,杀菌30-50s,最后加入益生菌干混120-900s,既得成品。
本发明的优点具体如下:
(1)本发明添加的壳寡糖是一种功能性低聚糖,能够补充足够的膳食纤维以及充足菌群生长因子,具有提高身体免疫、降血脂、调节血糖、改善肝脏和心肺功能、抗疲劳、美容及其它多种生理功能外,其与保加利亚乳杆菌、乳双歧杆菌(HN-019)等配伍,能够增殖有益菌,增强有益菌的数量和活性;与牦牛乳蛋白多肽、全脂乳粉、脱盐乳清粉、全脂牦牛乳粉结合,能促进人体对乳品中钙镁等矿物质的吸收。
(2)本发明添加的全脂牦牛乳粉,具有多种活性物质,能够在为人体补充充足蛋白质的同时能提高人体自身免疫力。
(3)本发明添加的益生菌与全脂乳粉、脱盐乳清粉以及全脂牦牛乳粉相互配伍,能有效地提高益生菌的存活率,并为益生菌的生长提供必需的氨基酸。
(4)本发明添加的益生菌与牦牛乳多肽相结合,增强产品的免疫力功效,且多肽类物质提供的矿物质与维生素能有效的促进肠道益生菌的生长,二者协同作用,相互促进。

实施例1,一种壳寡糖益生菌牦牛乳多肽配方奶粉,该配方奶粉由如下重量份的原料制成:全脂乳粉65份、脱盐乳清粉10份、全脂牦牛乳粉5份、牦牛乳蛋白多肽粉8份、壳寡糖5份、磷脂酰丝氨酸0.90份、维生素C 0.01份、乳双歧杆菌(HN-019)0.02份、嗜酸乳杆菌(NCMF)0.02份、保加利亚乳杆菌0.01份、嗜热链球菌0.01份。
实施例2,一种壳寡糖益生菌牦牛乳多肽配方奶粉,该配方奶粉由如下重量份的原料制成:全脂乳粉50份、脱盐乳清粉25份、全脂牦牛乳粉10份、牦牛乳蛋白多肽粉5份、壳寡糖9份、维生素C 0.005份、磷脂酰丝氨酸0.94份、乳双歧杆菌(HN-019)0.01份、嗜酸乳杆菌(NCMF)0.01份、保加利亚乳杆菌0.02份、嗜热链球菌0.02份。
实施例3,一种壳寡糖益生菌牦牛乳多肽配方奶粉,该配方奶粉由如下重量份的原料制成:全脂乳粉60份、脱盐乳清粉30份、全脂牦牛乳粉15份、牦牛乳蛋白多肽粉15份、壳寡糖15份、维生素C 0.008份、磷脂酰丝氨酸0.95份、乳双歧杆菌(HN-019)0.015份、嗜酸乳杆菌(NCMF)0.015份、保加利亚乳杆菌0.015份、嗜热链球菌0.015份。
实施例1-3所述的壳寡糖益生菌牦牛乳多肽配方奶粉的制备工艺,包括以下步骤:
A、发酵:将枯草芽孢杆菌活化,振荡培养6-12h,得到菌液,再以新鲜牦牛乳为底物混合菌液发酵,结束后经高压灭菌得到混合产物。所得菌液是1×109 cfu/mL,底物质量浓度为80 份/mL ,新鲜牦牛乳接种体积分数为 2.7%菌液,30-36 ℃,135 r/min 振荡培养发酵60-70 h,发酵结束后120 ℃高压灭5-10min 。
B、离心:将灭菌后的混合产物在6 000 r/min离心机中离心10-15 min,得到上清液。
C、超滤:将得到的上清液经 0.45 μm 滤膜过滤,去除菌体及残渣;采用截留分子量为 10 kDa、的超滤离心管对膜过滤液进行分离(4 000 r/min,10 min),再经冷冻干燥得到牦牛乳蛋白多肽产品。
D、干混 :将冷冻干燥好的牦牛乳蛋白多肽和除去益生菌的各原料在紫外线强度≥70μw/cm2条件下,杀菌30-50s,最后加入益生菌干混120-900s,既得成品。
本发明的动物性试验
1、抗疲劳试验:20只健康雄性小鼠,随机分为两组,每组10只,空白对照组灌胃5ml生理盐水,另一组每天灌胃5ml实施例1配方奶粉奶液1次,连续灌胃30d,灌胃期间各组小鼠喂食与饮水量一致。最后一次灌胃结束1h后,各组随机选取5只小鼠,开展负重游泳试验,记录小鼠自入水开始游泳至沉没超过10s的时间,即为动物负重游泳时间。
运动耐力可反映机体抗运动性疲劳的程度,而负重游泳是常见评估身体抗疲劳能力的运动实验模型,因此考察配方奶粉对小鼠的负重游泳时间长短的影响,结果如下表。
配方奶粉对小鼠运动时间的影响
从上表可知,与空白对照组相比,阳性对照组小鼠的力竭游泳时间延长率为14.7%,由此可知壳寡糖益生菌牦牛乳多肽配方奶粉有助于延长运动时间,具有抗疲劳的功效。
2、本发明四种菌种组合组比其他单一菌种效果试验:选取60只雄性小白鼠,先进行适应性饲养一周,然后随机分成六组,分别为乳双歧杆菌(HN-019)组、嗜酸乳杆菌(NCMF)组、保加利亚乳杆菌组、嗜热链球菌组、四种菌种组合组及对照组,对照组喂养生理盐水,实验组喂养(1.5x109CFU/ml)菌悬液0.2ml,连续三天注射环磷酰胺(80m份/k份),干预四周,每周称两次体重,求取平均值。小白鼠体重及偏差结果如下:
第一周给小白鼠注射环磷酰胺后免疫力降低,实验组小白鼠体重均有所下降,第二周实验组小白鼠体重回升,通过四周的观察和称重,记录数据,并进行分析,结果表明,四种菌群组合组标准偏差小于其他四组实验组,体重波动较小。可以说明,四种菌种组合组比其他单一菌种实验组更能有效的维持小白鼠体重,并且能够提高小白鼠免疫力。
Claims (5)
1.一种壳寡糖益生菌牦牛乳多肽配方奶粉,其特征在于,该配方奶粉由如下重量份的原料制成:全脂乳粉50-65份、脱盐乳清粉10-30份、全脂牦牛乳粉5-15份、牦牛乳蛋白多肽粉5-15份、壳寡糖5-15份、维生素C 0.005-0.01份、磷脂酰丝氨酸0.90-0.95份、乳双歧杆菌,HN-019,0.01-0.02份、嗜酸乳杆菌,NCMF,0.01-0.02份、保加利亚乳杆菌0.01-0.02份、嗜热链球菌0.01-0.02份。
2.根据权利要求1所述的一种壳寡糖益生菌牦牛乳多肽配方奶粉,其特征在于,该配方奶粉由如下重量份的原料制成:全脂乳粉65份、脱盐乳清粉10份、全脂牦牛乳粉5份、牦牛乳蛋白多肽粉8份、壳寡糖5份、磷脂酰丝氨酸0.90份、维生素C 0.01份、乳双歧杆菌,HN-019,0.02份、嗜酸乳杆菌,NCMF,0.02份、保加利亚乳杆菌0.01份、嗜热链球菌0.01份。
3.根据权利要求1或2所述的一种壳寡糖益生菌牦牛乳多肽配方奶粉的制备方法,其特征如下:
发酵:将枯草芽孢杆菌活化,振荡培养6-12h,得到菌液,再以新鲜牦牛乳为底物混合菌液发酵,结束后经高压灭菌得到混合产物;
离心:将灭菌后的混合产物在6 000 r/min离心机中离心10-15 min,得到上清液;
超滤:将得到的上清液经 0.45 μm 滤膜过滤,去除菌体及残渣;采用截留分子量为 10kDa、的超滤离心管对膜过滤液进行分离,再经冷冻干燥得到牦牛乳蛋白多肽产品。
4.干混 :将冷冻干燥好的牦牛乳蛋白多肽和全脂乳粉、脱盐乳清粉、全脂牦牛乳粉、牦牛乳蛋白多肽粉、壳寡糖、维生素C、磷脂酰丝氨酸在紫外线强度≥70μw/cm2条件下,杀菌30-50s,最后加入益生菌干混120-900s,既得成品。
5.根据权利要求3所述的一种壳寡糖益生菌牦牛乳多肽配方奶粉的制备方法,其特征在于:所得菌液是1×109 cfu/mL,底物质量浓度为 80 份/mL ,新鲜牦牛乳接种体积分数为 2.7%菌液,30-36 ℃,135 r/min 振荡培养发酵 60-70 h,发酵结束后120 ℃高压灭5-10min 。
