Abstract
The invention relates to the technical field of kits, in particular to a paraffin embedding pretreatment kit for micro tissues, which comprises a reagent box body, a cover plate and an operation pore plate, wherein the operation pore plate is provided with a reaction hole, a filter groove, a forceps groove, a water absorption paper using groove and a water absorption paper storage groove, and the reaction hole sequentially comprises a 40% polymethyl alcohol hole, a 70% alcohol hole, an 85% alcohol hole, a 95% alcohol hole, a 100% alcohol hole, a xylene hole and a liquid paraffin hole in an anticlockwise arrangement mode. The invention also provides a use method of the micro-tissue paraffin embedding pretreatment kit, the kit comprises various prepared soaking solutions, and the liquid preparation time and the reagent loss are saved; the method has the advantages of simple operation, few experimental steps, short time, good protection of the shape of the micro tissue, no influence on the concentration of the soak solution, more reliable data, market prospect and suitability for popularization.

Technical Field
The invention relates to the technical field of kits, in particular to a paraffin embedding pretreatment kit for a micro tissue and a using method thereof.
Background
The immunostaining technique is the most commonly used technique for studying tissue/cell antigens and can perform qualitative, positioning and quantitative determination on the antigens. Before immunostaining, tissues/cells must be fixed, dehydrated, cleared, paraffin embedded, sectioned, etc. For the experimental steps of fixing, dehydrating, transparentizing and the like of micro tissues (such as blastula, stem cell balls and the like, the volume is only 0.2-1.0 mm), the soaking of the micro tissues in different liquids is realized by repeatedly transferring liquid by a liquid transferring gun and centrifuging and removing the liquid by a centrifugal machine in the traditional method. The traditional operation method has the following disadvantages: 1. the immersion liquid of the micro tissue can not be completely removed by centrifugation and liquid transfer, so that the concentration deviation of the liquid is caused, and the fixing, dehydrating and transparent effects are poor; 2. repeated pipetting operations can lead to a reduction in the overall number by taking away parts of the micro-tissue; 3. repeated pipetting operations can cause minute tissue deformations; 4. the preparation of alcohols of different concentrations and the numerous operating steps are time-consuming and laborious. Therefore, a micro tissue paraffin embedding pretreatment kit and a use method thereof are provided to solve the problems.
Disclosure of Invention
The invention aims to solve the defects of poor effect and large experimental error of the existing experimental steps of fixing, dehydrating, transparentizing and the like of the micro tissue in the prior art, and provides a micro tissue paraffin embedding pretreatment kit and a using method thereof.

Detailed Description
