肉毒神经毒素(BoNT)是由肉毒梭菌产生的致死性毒素。这些毒素可特异性地靶向多种脊椎动物的神经元末端,阻断神经元递质释放,并引起通常称为“肉毒中毒(botulism)”的松弛性瘫痪。在另一方面,所述毒素的这种特性可用于治疗性目的,尤其是用于神经元相关疾病。自20世纪60年代开始,已探究了BoNT在治疗神经元疾病中的效力,并且BoNT现已广泛用于治疗多种神经元疾病,包括但不限于睑痉挛、斜视、上运动神经元综合征、发汗、颈肌张力障碍和慢性偏头痛。BoNT还广泛用于化妆品工业。在1989年12月,BOTOX(首个BoNT产品)被美国食品与药品监督管理局(Food and Drug Administration,FDA)批准用于临床治疗。

在一些实施方案中,编码BoNT的核苷酸序列与启动子可操作地连接。“启动子”是指核苷酸序列的控制区,在该区域核苷酸序列的其余部分的转录的起始和速率被控制。启动子驱动表达或驱动其调节的核苷酸序列的转录。启动子还可包含亚区域(sub-region),在该区域调节蛋白和调节分子(例如RNA聚合酶和其他转录因子)可结合。启动子可以是组成型的、诱导型的、激活型的、阻遏型的、组织特异性的或其任意组合。当启动子相对于其调节的核苷酸序列处于正确的功能位置和取向以控制(“驱动”)该序列的转录起始和/或表达时,该启动子被认为是“可操作地连接的”。
启动子可以是与基因或序列天然缔合的启动子,如可通过分离位于给定基因或序列的编码区段上游的5’非编码序列来获得。这样的启动子可被称为“内源性的”。
在一些实施方案中,编码核苷酸序列可置于重组或异源启动子的控制下,所述重组或异源启动子是指在其天然环境中通常与所编码的序列无关的启动子。这样的启动子可包括其他基因的启动子;从任何其他细胞分离的启动子;以及非“天然存在的”合成启动子或增强子,例如如包含不同转录调节区的不同元件和/或通过本领域已知的遗传工程方法改变表达的突变的启动子或增强子。除合成产生启动子和增强子的核酸序列之外,还可使用重组克隆和/或核酸扩增技术,包括聚合酶链式反应(polymerase chain reaction,PCR)来产生序列(参见美国专利No.4,683,202和美国专利No.5,928,906)。
在一些实施方案中,启动子是“诱导型启动子”,其是指特征在于当存在诱导物信号时、受诱导物信号影响时或与诱导物信号接触时调节(例如,起始或激活)转录活性的启动子。诱导物信号可以是以某种方式接触诱导型启动子以在调节诱导型启动子的转录活性中具有活性的内源性或正常的外源性条件(例如,光)、化合物(例如,化学或非化学化合物)或蛋白质。因此,核酸的“调节转录的信号”是指作用于诱导型启动子的诱导物信号。调节转录的信号可激活转录或使其失活,这取决于所使用的调节系统。转录的激活可包括直接作用于启动子以驱动转录或通过使阻止启动子驱动转录的阻遏物失活来间接作用于启动子。相反地,转录的失活可包括直接作用于启动子以阻止转录,或通过激活随后作用于启动子的阻遏物来间接作用于所述启动子。在一些实施方案中,在细胞状态分类器的遗传回路中使用诱导型启动子导致基因产物的条件性表达或“延迟”表达。
诱导物信号的施用或去除导致可操作连接的核苷酸序列转录的激活与失活之间的转换。因此,与核苷酸序列可操作地连接的启动子的激活状态是指当启动子主动调节核苷酸序列的转录(即,表达所连接的核苷酸序列)时的状态。相反地,与核苷酸序列可操作地连接的启动子的失活状态是指当启动子不主动调节核苷酸序列的转录(即,不表达所连接的核苷酸序列)时的状态。
本公开内容的诱导型启动子可通过一种或更多种生理条件,例如光、pH、温度、辐射、渗透压、盐梯度、细胞表面结合的变化,和一种或更多种外在或内在诱导剂的浓度来诱导(或抑制)。外在诱导物信号或诱导剂可包括但不限于氨基酸和氨基酸类似物、糖类和多糖、核酸、蛋白质转录激活物和阻遏物、细胞因子、毒素、基于石油的化合物、含金属的化合物、盐、离子、酶底物类似物、激素、或其组合。

本文中所述的产生BoNT的方法包括在适合于表达BoNT的条件下培养包含编码BoNT的核苷酸序列的芽孢杆菌细胞。在一些实施方案中,该方法还包括将编码BoNT的核苷酸序列递送到芽孢杆菌细胞。使用标准分子生物学技术来制备和递送重组表达载体,并培养芽孢杆菌细胞。可通过常规技术(例如,电穿孔、转化、转导或缀合)将包含编码BoNT的核苷酸序列的表达载体转移至宿主细胞,并随后通过常规技术培养所得芽孢杆菌细胞以产生本文中所述的BoNT。
芽孢杆菌细胞可在适当的温度(例如,16℃至42℃)下培养适当量的时间(例如,4至72小时)。在一些实施方案中,芽孢杆菌细胞在16、18、20、25、30、35、37、40或42℃下培养。在一些实施方案中,芽孢杆菌细胞培养4、5、6、7、8、9、10、11、12、13、14、15、16、17、18、19、20、21、22、23、24、36、48、60、72小时或更长时间。可使用适合于芽孢杆菌细胞的任何标准培养基(例如,卢里亚-贝尔塔尼(Luria-Bertani,LB)培养基)。如果BoNT的表达由诱导型启动子驱动,则培养基还可包含激活诱导型启动子的适当浓度的诱导物。
一旦BoNT已重组表达,则其可通过本领域已知的任何方法进行纯化,例如,通过色谱法(例如,亲和色谱法、离子交换色谱法、尺寸排阻色谱法、或其组合)、离心、差别溶解度或者通过用于纯化多肽的任何其他标准技术。
使用本文中所述方法产生的BoNT基本上不含(例如,至少80%、90%、95%、97%、99%或99.5%不含)其他蛋白质和/或其他多肽(例如,其他芽孢杆菌蛋白质)。在一些实施方案中,分离的多肽100%不含其他蛋白质和/或其他多肽(例如,芽孢杆菌蛋白质)。本文中所述的方法提供了高产量的完整BoNT。“完整”意指BoNT产物基本上不含截短的产物(例如,由于翻译中止或蛋白酶切割而产生的那些)。如本文中所示,在一些实施方案中,可从1升经LB培养的枯草芽孢杆菌中获得约5至10mg蛋白质。

实施例
肉毒梭菌是革兰氏阳性菌,其在进化上接近于模型细菌枯草芽孢杆菌,但在遗传上远离大肠杆菌(图1)。在本研究中,探索了在芽孢杆菌中表达BoNT的可能性。使用芽孢杆菌作为表达宿主存在如下数个优点:(1)芽孢杆菌物种在进化上更接近于肉毒梭菌(BoNT的天然宿主)得多。因此,芽孢杆菌物种可为这些毒素的蛋白质翻译和偶联折叠提供更好的内部环境。一个已知的实例是TcdA/B毒素可在巨大芽孢杆菌中而不在大肠杆菌中良好地表达。(2)枯草芽孢杆菌是经充分研究的细菌模型生物体;在该细菌物种中,遗传操作是高度可修改的。因此,当使用该细菌作为宿主时,蛋白质工程将是可行的。(3)使用枯草芽孢杆菌时的细胞培养和蛋白质纯化的成本相对较低。因此,该系统非常适合于大规模蛋白质生产。(4)枯草芽孢杆菌被认为是人中常见的肠共生体,并且通常被认为是GRAS(公认安全(general recognized as safe))生物体。食品与药品监督管理局(FDA)表示,来自枯草芽孢杆菌的非产毒性和非致病性菌株的蛋白质产物可广泛获得并已安全用于多种食品应用中。在芽孢杆菌细菌中已成功产生的产物包括淀粉酶、透明质酸、聚羟基链烷酸酯和许多抗生素。由于重组蛋白表达技术的不可预测性,是否可在芽孢杆菌中以高产量产生完整且具有活性的BoNT尚待检验。
结果:
先前已解决了全长BoNT/A、BoNT/B和BoNT/E的晶体结构,给出了这些分子的整体图。未在这些150kd的分子内发现许多二硫键,这表明氧化的环境或另外的氧化辅因子对于BoNT的产生不是必不可少的;这是可以预期的,因为肉毒梭菌是厌氧细菌。在这些毒素(例如BoNT/A和BoNT/B)中,疏水性和亲水性残基的表面分布均相对均匀(图2A至2B)。
使用iBoNT/B(“i”是指非活性形式,所述形式在其酶结构域处包含R370A/Y373F突变)作为BoNT的一个实例,并且分析了其在大肠杆菌中的表达模式。通常,尽管改变了表达条件,包括生长温度、诱导物浓度和培养基,但使用大肠杆菌均获得了非常少的产物。因此,iBoNT/B在大肠杆菌中的低产量可能主要是因为宿主的固有缺陷。为了验证这一点,在大肠杆菌中表达与N端或C端His-标签融合的iBoNT/B。无论标签在何处连接,细胞裂解物中大量的iBoNT/B产物均以较短片段存在。对于C端经标记的iBoNT/B,通过Ni-NTA珠进行的纯化去除了大多数的较短片段。相比之下,在IMAC纯化之后,保留了N端经标记的iBoNT/B的所有片段(图3)。这些结果指示iBoNT/B在大肠杆菌中的低产量主要是由翻译期间非预期的提前终止所致。
接下来,通过Western印迹在枯草芽孢杆菌中分析了iBoNT/B的表达模式。与在大肠杆菌中不同,大多数所表达的iBoNT/B作为完整的全长蛋白质存在(图4A至4B)。粗略估计,可从1升经LB培养的枯草芽孢杆菌中获得约5至10mg蛋白质;而在大肠杆菌中表达时,通常产量为<0.5mg/L培养物。当在大肠杆菌或枯草芽孢杆菌中测试包含iBoNT/A催化结构域、BoNT/A易位结构域和BoNT/B结合结构域的嵌合iBoNT/AB的表达时,也获得了相同的表型(数据未示出)。这些结果表明对于BoNT表达,枯草芽孢杆菌总体上是更好的表达宿主。
接下来,创建另一些肉毒神经毒素(包括iBoNT/A、iBoNT/C、iBoNT/D)的表达质粒,并在枯草芽孢杆菌中测试这些蛋白质的表达。如所预期地,所有这些蛋白质均能够在枯草芽孢杆菌中以不错的产量产生(图5)。
总结:

20.权利要求1至19中任一项所述的方法,其中所述芽孢杆菌细胞选自:枯草芽孢杆菌(Bacillus subtilis)、巨大芽孢杆菌(Bacillus megaterium)、炭疽芽孢杆菌(Bacillusanthracis)和短芽孢杆菌(Bacillus brevis)。
21.权利要求20所述的方法,其中所述芽孢杆菌细胞是野生型细胞。
22.权利要求20所述的方法,其中所述芽孢杆菌细胞是经改造的细胞。
23.权利要求22所述的方法,其中所述芽孢杆菌是蛋白酶缺陷型芽孢杆菌细胞。
24.芽孢杆菌细胞,其包含编码肉毒神经毒素(BoNT)的核苷酸序列。
25.权利要求24所述的芽孢杆菌细胞,其中所述编码BoNT的核苷酸序列与启动子可操作地连接。
