Abstract
The invention discloses a glabridin micro-molecule based nano whitening emulsion preparation and a preparation method thereof, wherein the preparation consists of 0.1-3% of glabridin, 0.1-10% of surfactant, 0.1-20% of cosurfactant, 0.1-10% of solvent, 0.5-20% of oil phase, 0.01-1% of preservative, 0.01-1% of spice and 54-95% of distilled water. The invention utilizes the transdermal drug delivery technology to artificially wrap the glabridin, the novel inclusion body changes the traditional permeation mode, effectively prevents the aggregation state of macromolecules, enables the molecules to be smaller and to permeate more quickly, and simultaneously improves the biocompatibility of the inclusion body, so that more effective components can be remained in skin tissues for a longer time. The effectiveness and utilization rate of the glabridin are improved, and the using amount of the glabridin is greatly reduced, so that the manufacturing cost is reduced.
Description
Technical Field
The invention relates to the technical field of cosmetics, in particular to a glabridin micro molecule-based nano whitening emulsion preparation and a preparation method thereof.
Background
Glabridin, a natural ingredient extracted from Glycyrrhiza glabra, originated from Indian herbal medicine, was first used as a natural anti-inflammatory, anti-allergic ingredient. Subsequently, it was scientifically found that glabridin can effectively inhibit the formation of melanin in the skin, and is the most safe inhibitor of tyrosinase recognized so far (IC50 value, 0.0045 for light sensation grass, 9.14 for stearic acid, and 12.2 for vitamin C). Preferably, the surfactant is any one or combination of more of Tween-80, Tween-60, triglyceride, sodium lauryl sulfate, polyoxyethylene lauryl ether and polyoxyethylene hydrogenated castor oil.
Preferably, the cosurfactant is any one or combination of sorbitan monostearate, propylene glycol monostearate, glyceryl monostearate, span 20, span 60 and span 80.
Preferably, the oil is any one or combination of edible tricaprylin, caprylic/capric triglyceride, soybean oil, medicinal liquid paraffin, edible salad oil, castor oil, isopropyl myristate and ethyl oleate.
Preferably, the solvent is any one or combination of anhydrous ethanol, pentanediol, n-butanol, glycerol and 1, 3-butanediol.
Preferably, the preservative is any one or combination of phenoxyethanol, ethylhexyl glycerol, benzoic acid, sodium benzoate, sorbic acid, potassium sorbate and calcium propionate.
Preferably, the perfume is one or more combination of daily chemical perfumes.
More preferably, the emulsion preparation is any one of essence, toner and spray.
The preparation method of the glabridin nanoemulsion preparation comprises the following steps:
(1) firstly, dissolving glabridin by redistilled acetone;
(2) mixing the oil phase, the surfactant, the cosurfactant and the solvent, adding absolute ethyl alcohol which is equal to the total amount of the oil phase, the surfactant, the cosurfactant and the solvent, mixing according to a ratio of 1:1, magnetically stirring for 4 hours, and cooling to below 45 ℃;
(3) slowly adding the glabridin acetone solution obtained in the step (1) into the solution obtained in the step (2), uniformly stirring the glabridin acetone solution by using a high-pressure homogenizer with 1.38X 108 Pa-4.13X 108Pa, then reducing the pressure to 1.0X 10-2 Pa-1.0X 104Pa, distilling the glabridin acetone solution, removing acetone and ethanol, and cooling the glabridin acetone solution to room temperature for later use;
(4) and (4) adding water, a preservative and a spice into the solution obtained in the step (3) to obtain a finished product of the glabridin nanoemulsion preparation.
The invention has the beneficial effects that: the glabridin is wrapped manually by a transdermal drug delivery technology. The novel inclusion changes the traditional mode of permeation. Effectively prevents the aggregation state of macromolecules, and enables the molecules to be smaller and to permeate more quickly. Meanwhile, the biocompatibility of the inclusion is improved, so that the effective components can be more and stay in skin tissues for a longer time instead of being lost along with the blood circulation system. The effectiveness and utilization rate of the glabridin are improved, and the using amount of the glabridin is greatly reduced, so that the manufacturing cost is reduced.

The technical solutions of the present invention will be described clearly and completely in the following embodiments of the present invention, and it should be understood that the described embodiments are only a part of the embodiments of the present invention, and not all of the embodiments. All other embodiments, which can be derived by a person skilled in the art from the embodiments given herein without making any creative effort, shall fall within the protection scope of the present invention.
Example 1
A nanometer whitening emulsion based on glabridin micro-molecule comprises glabridin 0.1%, surfactant 0.4%, cosurfactant 0.5%, solvent 1.99%, oil phase 1.99%, antiseptic 0.01%, perfume 0.01%, and distilled water 95%.
The preparation method of the glabridin nanoemulsion preparation comprises the following steps:
(1) firstly, dissolving glabridin by redistilled acetone;
(2) mixing the oil phase, the surfactant, the cosurfactant and the solvent, adding absolute ethyl alcohol which is equal to the total amount of the oil phase, the surfactant, the cosurfactant and the solvent, mixing according to a ratio of 1:1, magnetically stirring for 4 hours, and cooling to below 45 ℃;
(3) slowly adding the glabridin acetone solution obtained in the step (1) into the solution obtained in the step (2), uniformly stirring the glabridin acetone solution by using a high-pressure homogenizer with 1.38X 108 Pa-4.13X 108Pa, then reducing the pressure to 1.0X 10-2 Pa-1.0X 104Pa, distilling the glabridin acetone solution, removing acetone and ethanol, and cooling the glabridin acetone solution to room temperature for later use;
(4) and (4) adding water, a preservative and a spice into the solution obtained in the step (3) to obtain a finished product of the glabridin nanoemulsion preparation.
Example 2
A nanometer whitening emulsion based on glabridin micro-molecule comprises 1% of glabridin, 3% of surfactant, 8% of cosurfactant, 3% of solvent, 7% of oil phase, 0.03% of preservative, 0.03% of perfume, and 77.94% of distilled water.
The preparation method is the same as that of example 1.
Example 3
A nanometer whitening emulsion based on glabridin micro-molecule comprises 2% of glabridin, 7% of surfactant, 12% of cosurfactant, 8% of solvent, 13% of oil phase, 0.05% of preservative, 0.05% of perfume and 57.9% of distilled water.
The preparation method is the same as that of example 1.
Example 4
A nanometer whitening emulsion based on glabridin micro-molecule comprises 3% of glabridin, 10% of surfactant, 15% of cosurfactant, 4% of solvent, 12% of oil phase, 1% of preservative, 1% of perfume and 54% of distilled water.
The preparation method is the same as that of example 1.
Firstly, an experimental object:
for example: 200 subjects were searched and screened, and divided into 5 groups of 40 persons, wherein 4 groups were each prepared with the milk preparation of examples 1 to 4 of the present invention, and 1 group was prepared with a commercially available whitening milk preparation.
Second, Experimental methods
The experimental group 1-4 and the control group 1 were respectively applied with the milk preparation according to the embodiment of the present invention and a commercially available general whitening milk preparation containing glabridin as an effective ingredient, the test time was 7 days, and the skin condition of the subject was recorded at the time of 2 days, 5 days and 7 days of use, respectively. A square experiment area of 2cm by 2cm is selected on the left face of a subject, after face washing every day, water is wiped off, 1-3ml of sample is smeared on the experiment area, and the using effect is checked after the sample is continuously used for 30 days. When the effect is evaluated, a visia skin detector of the American CANFIELD company is adopted to compare four aspects of skin color spots, brown areas, red areas and pores respectively, and comprehensively judge the whitening effect.
Thirdly, judging the standard:
obviously improving: compared with the right face, the skin whiteness of the experimental area is increased, the dark skin color is obviously lightened or solved, the skin is smooth and new, ruddy and bright, pores are reduced, or the color of the pores is lightened.
Secondly, invalidation: compared with the right face, the whiteness of the experimental area is not increased or is not obviously increased, the skin color is dark, has no obvious change or deepening, pores are not reduced, or the color of the pores is lightened.
Fourth, test results
The test effect of the test subjects was counted and the effect is detailed in table 1.

2. The emulsion formulation of claim 1, wherein the surfactant is any one or a combination of tween-80, tween-60, triglycerides, sodium lauryl sulfate, polyoxyethylene lauryl ether, polyoxyethylene hydrogenated castor oil.
3. The milk formulation of claim 1, wherein the co-surfactant is any one or a combination of sorbitan monostearate, propylene glycol monostearate, glyceryl monostearate, span 20, span 60 and span 80.
4. The emulsion formulation of claim 1, wherein the oil phase is any one or more of edible tricaprylin, caprylic/capric triglyceride, soybean oil, medicinal liquid paraffin, edible salad oil, castor oil, isopropyl myristate, and ethyl oleate.
5. The emulsion formulation of claim 1, wherein the solvent is any one or a combination of anhydrous ethanol, pentane diol, n-butanol, glycerol, 1, 3-butanediol.
6. The milk preparation of claim 1, wherein the preservative is any one or a combination of phenoxyethanol, ethylhexyl glycerol, benzoic acid, sodium benzoate, sorbic acid, potassium sorbate, calcium propionate.
7. The milk preparation of claim 1, wherein the flavor is one or more combinations of flavors commonly used in daily use.
8. The emulsion according to any one of claims 1 to 7, wherein the emulsion is a liquid preparation selected from the group consisting of a serum, a toner, and a spray.
9. A process for the preparation of a milk preparation as claimed in claim 8, comprising the steps of:
(1) firstly, dissolving glabridin by redistilled acetone;
(2) mixing the oil phase, the surfactant, the cosurfactant and the solvent, adding absolute ethyl alcohol which is equal to the total amount of the oil phase, the surfactant, the cosurfactant and the solvent, mixing according to a ratio of 1:1, magnetically stirring for 4 hours, and cooling to below 45 ℃;
(3) slowly adding the glabridin acetone solution obtained in the step (1) into the solution obtained in the step (2), uniformly stirring the glabridin acetone solution by using a high-pressure homogenizer with 1.38X 108 Pa-4.13X 108Pa, then reducing the pressure to 1.0X 10-2 Pa-1.0X 104Pa, distilling the glabridin acetone solution, removing acetone and ethanol, and cooling the glabridin acetone solution to room temperature for later use;
(4) and (4) adding water, a preservative and a spice into the solution obtained in the step (3) to obtain a finished product of the glabridin nanoemulsion preparation.
