본 발명의 유자씨 및 망고씨 혼합오일을 함유한 화장료 조성물의 자극완화 효과 및 피부안전성을 확인하기 위하여 과거력상 피부자극에 과민 반응을 보인 적이 없으며, 현재 피부병 내지 피부 알러지 증상이 없는 20~30대 여성 20명을 대상으로 인체 피부첩포시험을 하였다. 피부 자극 물질인 락틱산 5.0%을 본 발명 실시예 2, 비교예 1 ~ 3에 첨가하여 시험에 사용하였다. 우선 시험 부위를 70% 에탄올로 닦아낸 뒤 건조시켰다. 준비된 시험 물질을 15㎍씩 핀챔버(Finn chamber, 100x 10, EPITEST, 핀란드) 내에 적하시킨 후, 시험대상자의 전박(forearm) 안쪽 부위에 밀폐 첩포하였다. 24시간 동안 첩포하고, 첩포를 제거한 후, 펜으로 시험부위를 표시하였다. 표시 후 각각 1시간 및 24시간 후에 확대경(8MC-150, DAZOR, 미합중국)을 이용하여 시험부위를 관찰하여 홍반 및 부종 유무를 관찰하였다. 피부 반응은 국제접촉피부염연구회(ICDRG, International Contact Dermatitis Research Group)의 규정에 따라 판정하고, 하기식에 따라 평균피부반응도(mean response rate)를 구하고, 그 결과를 표 8에 나타내었다. 피부반응의 평가기준 및 점수는 표 7에 나타내었다.
