Detailed Description
In order to make the objects, technical solutions and advantages of the present invention more apparent, the present invention is further described in detail with reference to the following embodiments. It should be understood that the specific embodiments described herein are merely illustrative of the invention and are not intended to limit the invention.
Aiming at the problems in the prior art, the invention provides a traditional Chinese medicine composite health product and a preparation process thereof, and the invention is described in detail with reference to the accompanying drawings.
As shown in fig. 1, the preparation process of the traditional Chinese medicine composite health product provided by the embodiment of the invention comprises the following steps:
s101, picking and cleaning dry roots of the pseudo-ginseng, and extracting the pseudo-ginseng in a secondary leaching mode by taking citric acid as a protective agent to obtain a pseudo-ginseng extract.
S102, pretreating rhizoma polygonati, and adding an ethanol solution for ultrasonic extraction; and (4) after elution, spin-drying by using a rotary evaporator to obtain a purified polygonatum sibiricum extract.
S103, soaking the angelica sinensis, the adenophora stricta, the schisandra chinensis, the medlar and the semen allii tuberosi for 12-15 hours, adding cold water to submerge the medicinal materials, heating for 4-5 hours, filtering to obtain filtrate and filter residues, and keeping the filtrate for later use.
S104, adding cold water into the filter residue obtained in the S103 to immerse the filter residue, heating for 2-5 hours, and taking a filtrate; and combining the filtrates obtained by the two times of decoction, standing for 20-30 min, and removing bottom precipitates to obtain a liquid medicine.
S105, putting the liquid medicine into a concentration pot for heating and concentrating, transferring the concentrated liquid medicine into a stirrer, sequentially adding the pseudo-ginseng extract, the sealwort extract and the starch drying agent, and uniformly stirring to obtain a viscous substance.
And S106, putting the viscous substance obtained in the step S105 into a constant-temperature reduced-pressure drying box, vacuumizing, drying under reduced pressure, crushing, and sieving with a 80-100-mesh sieve to obtain medicinal powder.
And S107, adding the wetting agent and the anti-sticking agent into the medicinal powder obtained in the step S106, uniformly mixing and stirring, placing the mixture into a mold, and pressing the mixture into tablets to obtain the traditional Chinese medicine composite health-care product.
The wetting agent provided by the embodiment of the invention is docusate sodium or honey.
The antisticking agent provided by the embodiment of the invention is a composition of one or more of talcum powder, glyceryl monostearate and magnesium stearate.
The traditional Chinese medicine composite health care product provided by the embodiment of the invention comprises, by mass, 20-25 parts of pseudo-ginseng, 18-20 parts of rhizoma polygonati, 10-12 parts of angelica sinensis, 10-12 parts of adenophora stricta, 3-6 parts of schisandra chinensis, 4-6 parts of medlar and 1-2 parts of semen allii tuberosi.
The preparation method of the pseudo-ginseng extract provided by the embodiment of the invention comprises the following steps:
(1) picking and cleaning dry roots of the pseudo-ginseng, drying and then putting the pseudo-ginseng into a crusher to be crushed into particles with the particle size of 2-3 mm; adding cold water and citric acid into the pseudo-ginseng particles, heating to 60-70 ℃, and leaching for 3-5 h.
(2) Filtering, collecting residue and filtrate, adding water and citric acid into the residue, heating and extracting, filtering to obtain filtrate, and mixing the filtrates to obtain Notoginseng radix extract.
(3) Adsorbing the obtained pseudo-ginseng extract by using resin, then performing dynamic cycle elution by using ethanol, heating and concentrating at the temperature of 60-70 ℃, and recovering the ethanol to obtain a concentrated solution.
(4) Adding ethyl acetate with the volume 3-5 times that of the concentrated solution into the obtained concentrated solution for extraction, and then respectively collecting an organic layer and a water layer; adding ethyl acetate into the water layer again for extraction, and collecting an organic layer; the two collected organic layers were then combined to give an extract.
(5) Concentrating and drying the obtained extract to obtain a pseudo-ginseng crude extract; adding water into the pseudo-ginseng crude extract, heating for dissolving, cooling for 36-48 h at 4-6 ℃, and crystallizing to obtain crystals; and under the condition of gas protection, drying and crushing the crystals, and then sieving the crystals with a 50-80-mesh sieve to obtain fine powder, namely the pseudo-ginseng extract.
The amount of the cold water added is 2-2.5 times of the weight of the pseudo-ginseng particles, and the amount of the citric acid is 2-3% of the mass of the added water.
The resin provided by the embodiment of the invention is AB-8, LX-8 or HPD826 resin.
The amount of ethanol added during dynamic circulation elution and concentration provided by the embodiment of the invention is 3-4 times of the mass of the pseudo-ginseng particles, the concentration of the ethanol is 65%, and the dynamic circulation extraction time is 20-24 h.
The main component of the panax notoginseng extract provided by the embodiment of the invention is panaxtriol saponins.
The preparation method of the polygonatum sibiricum extract provided by the embodiment of the invention comprises the following steps:
(I) preprocessing rhizoma polygonati: cleaning rhizoma polygonati, removing dust and granular impurities on the surface, cutting into thin slices with the thickness of 0.2-1 mm, and drying at a low temperature of 45-60 ℃.
(II) crushing the pretreated rhizoma polygonati slices into particles with the particle size of 2-3mm, adding 75-85% ethanol solution which is 15-30 times of the weight of the rhizoma polygonati particles into the rhizoma polygonati particles, carrying out ultrasonic extraction at 50-65 ℃ for 3-5 times for 0.5-2.5 h each time, combining ethanol extracting solutions, and filtering to obtain rhizoma polygonati filtrate I and rhizoma polygonati filter residues.
(III) adding cold water into the sealwort filter residue, heating for 2 hours, and filtering to obtain a filtrate II; adding cold water into the filter residue, heating for 2h, filtering to obtain filtrate III, mixing filtrate I, filtrate II and filtrate III, concentrating, and spin-drying with rotary evaporator to obtain crude rhizoma Polygonati extract.
(IV) diluting the obtained crude product of the rhizoma polygonati extract with a proper amount of distilled water, adding the diluted crude product of the rhizoma polygonati extract into prepared adsorption resin for adsorption, eluting with distilled water, then eluting with 70-85% ethanol until no extract flows out of the eluent, namely, the eluent is completely eluted, collecting the ethanol eluent, spin-drying with a rotary evaporator, and drying with a reduced-pressure dryer until the weight is constant, thus obtaining the purified rhizoma polygonati extract.
The rhizoma polygonati extract provided by the embodiment of the invention mainly contains rhizoma polygonati polysaccharide.
The water content of the concentrated liquid medicine provided by the embodiment of the invention is 20-30%.
The invention is further described with reference to specific examples.
Example 1
Case one
The insomnia problem of plum, male and 21 years old, which is caused by fatigue and poor energy before one year, can be relieved after 8 days of taking the product, symptoms disappear after 25 days of treatment, and the sleep is normal.
Case two
Zhangqi, male, age 39, smoking and drinking, poor immunity, and susceptibility to diseases for years, and no appetite in the last month due to overwork. The symptoms are relieved after the product is taken for three days, and the symptoms are eliminated after the product is taken for ten days.
Example 2
Picking and cleaning dried root of Notoginseng radix, drying, and pulverizing into 2-3mm granule; adding cold water 2 times the weight of the particles into the pseudo-ginseng particles, heating to 40 ℃, and keeping the temperature for 12 hours; adding ethanol with the mass 3 times of that of the pseudo-ginseng particles and the concentration of 65% for dynamic circulation extraction for 24 hours, recovering the ethanol, and concentrating to obtain an extract; drying the extract to obtain the pseudo-ginseng extract.
Example 3
Cleaning rhizoma Polygonati, drying, pulverizing into 2-3mm granules, adding cold water into rhizoma Polygonati granules, heating for 2 hr, and filtering; adding cold water into the filter residue, heating for 2h, and filtering; and adding cold water into the filter residue again, heating for 2h, filtering, combining the filtrates, concentrating to obtain an extract, and drying the extract to obtain the sealwort extract.
Example 4
4kg of rhizoma polygonati is used for extracting the rhizoma polygonati extract, and the extraction time is used for analyzing the yield of the rhizoma polygonati extract:

Soaking radix Angelicae sinensis, radix Adenophorae, fructus Schisandrae chinensis, fructus Lycii, and semen Allii Tuberosi for 12h, adding cold water to submerge the materials, heating for 4h, filtering, and keeping the filtrate for use; adding cold water into the filter residue to immerse the filter residue, heating for 2h, and collecting the filtrate; mixing the two decoctions, standing for 20min, and removing bottom precipitate to obtain medicinal liquid; heating and concentrating the medicinal liquid in a concentrating pot, transferring the concentrated medicinal liquid to a stirrer, sequentially adding the Notoginseng radix extract obtained in example 2, the rhizoma Polygonati extract obtained in example 3, and a starch drying agent, and stirring to obtain a viscous substance; placing into a constant-temperature reduced-pressure drying oven, vacuumizing, drying under reduced pressure, pulverizing, and sieving with 80 mesh sieve to obtain medicinal powder; adding Mel and glyceryl monostearate into the medicinal powder, mixing, and placing in a mold to obtain the final product.
Example 6
The health care efficacy of the traditional Chinese medicine composite health care product is researched: setting the sheet-shaped traditional Chinese medicine composite health-care product prepared in the example 5 as an experimental group; the decoction of pseudo-ginseng, rhizoma polygonati, angelica, radix adenophorae, schisandra, medlar and semen allii tuberosi is used as a first control group; the commercially available functionally similar drug is control group two.
Respectively arranging 10 mice in an experimental group, a first control group and a second control group, carrying out normal feeding in the same environment, respectively feeding three medicaments for 20 days continuously, and carrying out the following index determination:

Stress response tests of three groups of mice were carried out on days 1, 10, 15 and 20 of feeding, respectively, and each group was averaged, and the time from stimulation to response was recorded as the stress time, and the average stress response time of three groups of mice was compared with that shown in FIG. 2 (unit: s).

Claims (10)

1. The preparation process of the traditional Chinese medicine composite health product is characterized by comprising the following steps of:
firstly, picking and cleaning dry roots of pseudo-ginseng, drying and then placing the pseudo-ginseng in a grinder to be ground into particles with the particle size of 2-3 mm; adding cold water and citric acid into the pseudo-ginseng particles, heating to 60-70 ℃, and leaching for 3-5 h;
step two, after filtering, respectively collecting filter residues and filtrate, adding water and citric acid into the filter residues again, heating and leaching, filtering to obtain filtrate, and combining the secondary filtrate to obtain pseudo-ginseng extract;
step three, adsorbing the pseudo-ginseng extract obtained in the step two by using resin, then performing dynamic cycle elution by using ethanol, heating and concentrating at the temperature of 60-70 ℃, and recovering the ethanol to obtain a concentrated solution;
step four, adding ethyl acetate with the volume 3-5 times of that of the concentrated solution into the concentrated solution obtained in the step three, extracting, and collecting an organic layer and a water layer respectively; adding ethyl acetate into the water layer again for extraction, and collecting an organic layer; then combining the organic layers collected twice to obtain extract liquor;
step five, concentrating and drying the extract liquid obtained in the step four to obtain a pseudo-ginseng crude extract; adding water into the pseudo-ginseng crude extract, heating for dissolving, cooling for 36-48 h at 4-6 ℃, and crystallizing to obtain crystals; drying and crushing the crystals under the protection of gas, and sieving the crystals with a 50-80-mesh sieve to obtain fine powder, namely the pseudo-ginseng extract;
step six, rhizoma polygonati pretreatment: cleaning rhizoma polygonati, removing dust and granular impurities on the surface, cutting into thin slices with the thickness of 0.2-1 mm, and drying at a low temperature of 45-60 ℃;
step seven, crushing pretreated rhizoma polygonati slices into particles with the particle size of 2-3mm, adding 75-85% ethanol solution which is 15-30 times of the weight of the rhizoma polygonati particles into the rhizoma polygonati particles, carrying out ultrasonic extraction at 50-65 ℃ for 3-5 times for 0.5-2.5 h each time, combining ethanol extracting solutions, and filtering to obtain rhizoma polygonati filtrate I and rhizoma polygonati filter residues;
step eight, adding cold water into the sealwort filter residue, heating for 2 hours, and filtering to obtain a filtrate II; adding cold water into the filter residue, heating for 2h, filtering to obtain filtrate III, mixing filtrate I, filtrate II and filtrate III, concentrating, and spin-drying with rotary evaporator to obtain crude rhizoma Polygonati extract;
step nine, diluting the crude polygonatum sibiricum extract obtained in the step eight with a proper amount of distilled water, then adding the diluted solution into prepared adsorption resin for adsorption, eluting with distilled water, then eluting with 70-85% ethanol until no extract flows out of the eluate, namely complete elution is obtained, collecting ethanol eluate, spin-drying with a rotary evaporator, and drying with a reduced pressure dryer until the weight is constant to obtain a purified polygonatum sibiricum extract;
step ten, soaking the angelica sinensis, the adenophora stricta, the schisandra chinensis, the medlar and the semen allii tuberosi for 12-15 hours, adding cold water to submerge the medicinal materials, heating for 4-5 hours, filtering to obtain filtrate and filter residues, and keeping the filtrate for later use;
step eleven, adding cold water into the filter residue obtained in the step eleven to immerse the filter residue, heating for 2-5 hours, and taking a filtrate; mixing the two decocted filtrates, standing for 20-30 min, and removing bottom precipitates to obtain a liquid medicine;
step twelve, putting the liquid medicine into a concentration pot for heating and concentrating, transferring the concentrated liquid medicine into a stirrer, sequentially adding the pseudo-ginseng extract, the rhizoma polygonati extract and the starch drying agent, and uniformly stirring to obtain a viscous substance;
thirteen, putting the viscous substance obtained in the twelfth step into a constant-temperature reduced-pressure drying box, vacuumizing and drying under reduced pressure, crushing, and sieving with a 80-100-mesh sieve to obtain medicinal powder;
step fourteen, adding the wetting agent and the anti-sticking agent into the medicinal powder obtained in the step thirteen, mixing and stirring uniformly, placing the mixture into a mould, and pressing the mixture into tablets to obtain the traditional Chinese medicine composite health-care product.

The preparation process of the traditional Chinese medicine composite health product according to claim 1, wherein in the first step, the amount of the added cold water is 2-2.5 times of the weight of the pseudo-ginseng particles, and the amount of the citric acid is 2-3% of the mass of the added water.
3. The process for preparing a composite health product of traditional Chinese medicine according to claim 1, wherein in the third step, the resin is AB-8, LX-8 or HPD826 resin.

The preparation process of the traditional Chinese medicine composite health product according to claim 1, wherein in the third step, the amount of ethanol added in the dynamic circulation elution and concentration is 3-4 times of the mass of the pseudo-ginseng particles, the concentration of the ethanol is 65%, and the dynamic circulation extraction time is 20-24 hours.
