如果需要,变体可以通过磷酸化、硫酸盐化、丙烯酰化、糖基化、甲基化、法尼基化、乙酰化、酰胺化等进行修饰。
对于本发明中的非限制性示例,生长激素变体可以是具有如下表1所示的SEQ IDNO.的蛋白质。SEQ ID NO:1到9的生长激素变体分别与下面描述的实施例1到9中包括的生长激素变体相同。同时,实施例10和实施例11分别包括SEQ ID NO:7和SEQ ID NO:9的生长激素变体。

如本文所使用的,术语“生长激素受体(GHR)”是指生长激素与之结合以将信号传输到细胞的受体。生长激素受体具有穿透细胞膜一次的结构。当受体被激活时,STAT二聚体通过JAK/STAT途径调控核内各种基因的转录。生长激素受体存在于全身组织中,包括肝脏、肌肉、脂肪、肾脏、早期胚胎、胎儿组织等。
当生长激素与受体结合时,随后的信号传导导致IGF(胰岛素样生长因子)-1的分泌增加。hGH分泌过多和导致的IGF-1升高可引起肢端肥大症,其是一种以手脚肿大为典型症状的慢性疾病。
如本文所用,术语“生长激素受体拮抗剂”是指拮抗生长激素与生长激素受体结合,从而抑制生长激素与生长激素受体过度结合所引起的副作用的药剂。
具体地说,生长激素受体拮抗剂可以是具有与生长激素受体的高结合效力并且能够竞争性地拮抗生长激素功效的生长激素变体。
此外,生长激素受体拮抗剂可包括与生长激素变体融合的长效载体。
如本文所用,术语“长效载体”是指能够增加体内半衰期的物质。当已知增加体内半衰期的多种长效载体与根据本发明的生长激素变体融合时,预期其将用作在拮抗生长激素受体时具有增加的体内半衰期的长效剂。
在本发明中,长效载体的非限制性示例可包括能够降低肾清除率的各种载体,具体地说,选自由聚乙二醇、脂肪酸、白蛋白或其片段、白蛋白结合物质、α-1抗胰蛋白酶或其变体、免疫球蛋白Fc或其片段、特定氨基酸序列重复单位的聚合物、抗体或其片段、FcRn结合物质、体内结缔组织或其衍生物、核苷酸、纤维粘连蛋白、转铁蛋白、糖和聚合物组成的组中的任何一种或多种。
更具体地说,α-1抗胰蛋白酶(A1AT)或其变体可以用作长效载体。
α-1抗胰蛋白酶或其变体在专利出版物KR10-2013-0136883A和KR10-2013-0029713A中公开。具体地说,A1AT是人体血浆中含量最丰富的蛋白质之一,其浓度为每升1.5-3.5克,主要在肝细胞中合成并分泌到血液中。α-1抗胰蛋白酶变异体是通过在A1AT中加入额外的突变来增加糖基化并消除其内在活性而设计的。α-1抗胰蛋白酶变体可与靶蛋白融合以延长靶蛋白的半衰期。
α-1抗胰蛋白酶变体技术的主要优点之一是非免疫原性。特别是,血浆来源的人类A1AT已经被用作治疗肺气肿病人的药物,其中肺气肿是由于A1AT缺乏引起的一种肺部疾病。它的周剂量高达60毫克/千克体重。严重的副作用尚未报道,表明A1AT作为一种治疗剂的安全性。
在本发明中,生长激素受体拮抗剂的非限制性示例可提供通过将α-1抗胰蛋白酶变体与生长激素变体融合而获得的长效hGHRA。α-1抗胰蛋白酶变体可以包括α-1抗胰蛋白酶序列中的第1个氨基酸和第25个氨基酸中的一个或多个氨基酸的取代,并且生长激素变体可以包括选自生长激素的氨基酸序列中的H18D、H21N、Q46K、F54P、R64K、G120K、R167N、K168A、D171S、K172R、E174S、F176Y和I179T的任何一个或多个取代。
更具体地说,α-1抗胰蛋白酶变体可以具有例如下表2的SEQ ID NO:10的氨基酸序列。α-1抗胰蛋白酶变体可以是“NexP”(KR10-2013-0136883A)。

与聚乙二醇化生长激素变体相比,通过将α-1抗胰蛋白酶变体与生长激素变体融合以增加血浆半衰期而获得的生长激素受体拮抗剂可能与生长激素受体具有高结合效力,并且可能强烈拮抗生长激素的功效。
具体地说,与已知的生长激素受体拮抗剂培维索孟相比,作为本发明中生长激素受体拮抗剂的非限制性示例的实施例4、6、7、8、9、10和11显示出与hGH受体的高结合能力和高抑制效力(参见图4和5,表4)。
在通过将长效载体融合到生长激素变体而获得的生长激素受体拮抗剂中,长效载体可融合到生长激素变体的N-端或C-端。
特别地,与通过将长效载体融合到其C端而获得的生长激素受体拮抗剂相比,通过将长效载体融合到特定生长激素变体的N-端而获得的生长激素受体拮抗剂,可能对生长激素受体具有显著的高结合效力或可强烈地拮抗生长激素的功效。对于本发明中的非限制性示例,即使hGH-A10和hGH-A11分别具有与hGH-A7和hGH-A9相同的氨基酸序列的生长激素变体,融合到变体的N-端的那些被证实强烈拮抗生长激素的功效(见图4C和4D,图5和表4)。
在本发明中,所述长效载体可以直接或通过连接器与所述长效载体融合。
只要在长效载体和生长激素变体之间的共价键中使用并且不影响活性,任何连接器都可以无限制地使用。具体地,该连接器可以是聚乙二醇、聚丙二醇、乙二醇和丙二醇的共聚物、聚氧乙基化多元醇、聚乙烯醇、多糖、葡聚糖、聚乙烯基乙醚、聚乳酸(PLA)、聚乳酸-乙醇酸(PLGA)、脂质聚合物、甲壳素、透明质酸和其组合的非肽基连接器,并且可以是其中连接两个或更多氨基酸的肽基连接器。非限制性示例可包括GGGGS和具有可变长度的连接器(2X、3X、4X等)。
为了实现上述目的,本发明的另一方面提供了一种用于预防或治疗由人类生长激素引起的疾病的药物组合物,该药物组合物包含生长激素受体拮抗剂。此处使用的术语与上述相同。
如本文所用,术语“由人类生长激素引起的疾病”是指由于脑下垂体的异常控制等引起生长激素分泌过多导致的疾病。例如,该疾病可能包括肢端肥大症、巨人症、癌症、糖尿病肾病、关节炎、肺部炎症、生长激素缺乏症(GHD)、特发性身材矮小、特纳氏综合征、普拉德-威利综合征、小于胎龄儿、慢性肾功能不全(CRI)等。
此外,由人类生长激素引起的疾病包括由生长激素过度作用引起的IGF(胰岛素样生长因子)-1分泌增加引起的疾病。
对于本发明中的非限制性示例,实施例(hGH-A1至hGH-A11)抑制生长激素与其受体的结合,从而用于预防或治疗由人类生长激素引起的疾病(参见试验性实施例和表4)。
为了实现上述目的,本发明的另一方面提供了一种制备生长激素受体拮抗剂的方法,包括培养细胞的步骤,所述细胞包括编码生长激素变体的多核苷酸,所述生长激素变体中的生长激素氨基酸序列中的一个或多个氨基酸被另一个氨基酸取代。此处使用的术语与上述相同。
这些细胞可以是用生长激素变体的表达载体转染的细胞,其非限制性示例可以包括CHO(中国仓鼠卵巢)-K1等。
为了实现上述目的,本发明的另一方面提供了一种预防或治疗由人类生长激素引起的疾病的方法,该方法包括向受试者施用包括生长激素受体拮抗剂的药物组合物的步骤。此处使用的术语与上述相同。
如本文所使用的,术语“给药”是指通过任何适当的方法将本发明的药物组合物引入患者体内,并且只要本发明的组合物能够到达所需组织,就可以通过诸如口服或肠外途径等各种途径实施本发明组合物的给药。根据本发明的试剂可以根据期望的给药方式制备成各种制剂。
可预防性或治疗性地实施给药。
本发明的药剂的给药频率没有特别限制,但可以一天给药一次或以分开剂量多次给药。
将被给药以根据本发明的药剂的受试者可指包括人类在内的所有动物。这些动物可能包括哺乳动物,如和人类一样需要治疗类似症状的牛、马、羊、猪、山羊、骆驼、羚羊、狗、猫等,但不限于此。
为了实现上述目的,本发明的另一方面提供了包含生长激素受体拮抗剂的药物组合物在预防或治疗由人类生长激素引起的疾病中的用途。此处使用的术语与上述相同。
本发明的技术效果
根据本发明的生长激素受体拮抗剂可以具有与生长激素受体的强结合效力,并且可以表现出长效拮抗作用。

本发明的最佳模式
在下文中,将参考以下实施例更详细地描述本发明。然而,以下实施例仅用于说明性目的,并且本发明的范围不意图被这些实施例限制。
制备实施例:生长激素受体拮抗剂的制备
1.克隆、转染和细胞培养
根据已知的方法制备hGH-NexP的cDNA克隆。通过对hGH-NexP基因定点诱变制备hGH受体拮抗剂(hGHRA-NexP)。随后,DNA测序证实了突变。用含有各hGHRA-NexP克隆的核苷酸序列的质粒瞬时转染CHO(中国仓鼠卵巢)-K1细胞。转染细胞在5%CO2增湿培养箱中在补充有10%FBS的IMDM培养基(Iscove’s Modified Dulbecco’s Medium)中生长7天。
2.hGHRA-NexP的纯化
hGHRA-NexP变体通过一系列柱层析法从瞬时转染的CHO-K1细胞中纯化。用等体积的缓冲液A(20mM磷酸钠,pH 8.0)稀释培养上清液,并将其应用于用缓冲液A平衡的离子交换柱上。用缓冲液A洗涤步骤后,用缓冲液A中的NaCl线性梯度洗脱蛋白质。然后将含有hGHRA-NexP的部分直接加载到用缓冲液B(20mM Tris-HCl,100mM NaCl,pH 7.5)平衡的亲和柱上,并用在缓冲液B中的MgCl2梯度洗脱该部分。在调整洗脱池的pH和电导率后,将其装载到第二个离子交换柱上,该离子交换柱用缓冲液C(20mM磷酸钠、80mM NaCl、pH 8.0)平衡。用Vivaspin 20浓缩仪(Sartorius)浓缩蛋白质组分,用磷酸盐缓冲液(PBS)透析。
3.SE-HPLC分析
为测定hGHRA-NexP的纯度,采用体积排阻高效液相色谱法(SE-HPLC)。将蛋白质溶液装入TSKgel G3000SWXL柱(Tosoh),在流动缓冲液(50mM磷酸钠、150mM氯化钠、0.05%叠氮化钠、pH6.8)中以0.5mL/min的流速获得色谱图。计算色谱主峰的面积百分率(%)。
实施例:生长激素受体拮抗剂的实施例
本发明的目的是通过对hGH序列的定点诱变和NexP技术制备生长激素受体拮抗剂,以提供长效性,并且制备了实施例1(hGH-A1:G120K)和实施例2(hGH-A2:G120R),每个实施例中hGH的120位置的氨基酸被取代。此外,对于上述目的,在hGH序列的hGH-A1的位点1引入额外的突变以制备实施例3至实施例7。实施例中取代的特定氨基酸如表3所示。在实施例9中,引入Q46K突变以通过引入赖氨酸诱导阳离子-π相互作用。

在表3中,hGH-A1至hGH-A9是通过将NexP融合到变体的C-端获得的,hGH-A10和hGH-A11是通过将NexP融合到变体的N-端获得的。
根据上述制备实施例,通过转染CHO-K1细胞和一系列柱层析法制备hGHRA-NexP蛋白。代表性地,第二离子交换柱色谱分析的色谱图如图3A所示。从色谱分析中获得纯蛋白质,且纯化的蛋白质在100kDa到70kDa之间迁移(图3B)。将7号部分至14号部分这些部分合并后,用磷酸盐缓冲液(PBS)对溶液进行透析。经确证,该蛋白质纯度高达95%,如SE-HPLC色谱主峰所示(图3C)。
试验性实施例:hGH功效和受体结合效力的分析
1.hGH功效分析方法
为了进行这一分析,hGH受体基因被引入HEK293F细胞的染色体中,HEK293F细胞含有荧光素酶基因,该基因可以被hGH受体信号传导诱导。制备的细胞系命名为hGHR/Luc/HEK293F。将hGH和hGHRA-NexP的系列稀释液分别加入到含有hGHR/Luc/HEK293F的96孔白板中。将该板在37℃的5%CO2培养箱中培养24小时。孵育后,每孔加入100μL荧光素酶检测试剂(Steady-荧光素酶检测系统,Promega),并将该板覆盖以防光线照射。在室温下5分钟后,使用多模式微孔板阅读器(SpectraMax M5,Molecular Devices)分析孔中的发光。
hGH、hGH-A1和hGH-A2的测量结果如图2所示。结果证实,hGH-A1和hGH-A2与hGH不同,没有显示任何功效。
2.hGH受体结合力分析
利用重组hGH受体Fc嵌合体,通过亲和性分析评价hGRA-NexP与hGH受体的结合效力。在25℃下将hGH受体嵌合物涂覆在微孔板上过夜,并用TPBS缓冲液(含有0.05%吐温-20的PBS缓冲液)洗涤,然后将样品(实施例1至11和培维索孟)装载在每个孔上。样品洗涤三次,并与抗hGH多克隆抗体生物素缀合。在额外的洗涤步骤后,向每个孔中添加3,3’,5,5’-四甲基联苯胺(TMB),以允许TMB反应。在450nm至650nm处记录来自反应的吸光度信号。
hGH-A7(●)、hGH-A9(○)和培维索孟(□)的结合概况的测量结果如图4A所示。此外,hGH-A10(▲)、hGH-A11(△)和培维索孟体(□)的结合概况的测量结果如图4C所示。
与培维索孟相比,hGH-A7和hGH-A9被发现具有与hGH受体的高结合效力。此外,与培维索孟相比,hGH-A10和hGH-A11被发现具有与hGH受体的显著高的结合效力。
3.hGH竞争性抑制效力分析
用hGH竞争分析法研究hGHRA-NexP对下游信号传导的抑制效力。将实施例1至11和培维索孟的连续稀释液分别添加到含有hGHR/Luc/HEK293F的96孔板的每个孔中。将该板在37℃的5%CO2培养箱中培养24小时。培养后,每孔加入100μL荧光素酶检测试剂(Steady-荧光素酶检测系统,Promega),并将该板覆盖以防光线照射。在室温下5分钟后,使用多模式微孔板阅读器(SpectraMax M5,Molecular Devices)分析孔中的发光。
hGH-A7(●)、hGH-A9(○)和培维索孟(□)的结合概况的测量结果如图4B所示。此外,hGH-A10(▲)、hGH-A11(△)和培维索孟体(□)的结合概况的测量结果如图4D所示。
与培维索孟相比,hGH-A7和hGH-A9被发现具有高的hGH竞争性抑制效力。此外,与培维索孟相比,hGH-A10和hGH-A11被发现具有显著高的hGH竞争性抑制效力。
测定hGH-A1与hGH-A11以及比较实施例培维索孟的相对结合效力和hGH竞争性抑制效力的结果如下表4所示。这些结果绘制在图5中。

如表4所示,与已知的培维索孟相比,hGH-A4、hGH-A6、hGH-A7、hGH-A8、hGH-A9、hGH-A10和hGH-A11具有较高的hGH受体结合效力和竞争性抑制效力。尤其是hGH-A10和hGH-A11,每一种都是通过将NexP融合到变体的N-端而制备,其显示出显著高的竞争性抑制效力。
基于上述描述,本领域技术人员将理解,在不改变本发明的技术精神或基本特征的情况下,本发明可以以不同的具体形式实现。因此,应当理解,上述实施例不是限制性的,而是在所有方面都是说明性的。本发明的范围由所附权利要求而不是由其前面的描述来定义,因此,在权利要求的范围内的所有变更和修改,或此类范围的等价物,都意图包含在权利要求中。
Claims (18)
1.一种生长激素受体拮抗剂,包括生长激素变体,其中生长激素的氨基酸序列中的一个或多个氨基酸用另一个氨基酸取代。
2.根据权利要求1所述的生长激素受体拮抗剂,其中所述用另一个氨基酸取代包括用赖氨酸取代自N-端起第46个氨基酸、用赖氨酸或精氨酸取代自N-端起第120个氨基酸、或其组合。
3.根据权利要求2所述的生长激素受体拮抗剂,其中所述用另一个氨基酸取代还包括在选自由自N-端起第18个氨基酸、第21个氨基酸、第54个氨基酸、第64个氨基酸、第167个氨基酸、第168个氨基酸、第171个氨基酸、第172个氨基酸、第174个氨基酸、第176个氨基酸和第179个氨基酸组成的组中的任一个或多个位置处的取代。
4.根据权利要求2所述的生长激素受体拮抗剂,其中所述用另一个氨基酸取代还包括用丝氨酸取代自N-端起第174个氨基酸、用天冬酰胺取代自N-端起第21个氨基酸、或其组合。
5.根据权利要求1所述的生长激素受体拮抗剂,其中所述用另一个氨基酸取代包括选自由以下组成的中选择的任一种或多种取代:用天冬氨酸取代自N-端起第18个氨基酸、用天冬氨酸取代自N-端起第21个氨基酸、用赖氨酸取代自N-端起第46个氨基酸、用脯氨酸取代自N-端起第54个氨基酸、用赖氨酸取代自N-端起第64个氨基酸、用赖氨酸或精氨酸取代自N-端起第120个氨基酸、用天冬酰胺取代自N-端起第167个氨基酸、用丙氨酸取代自N-端起第168个氨基酸、用丝氨酸取代自N-端起第171个氨基酸、用精氨酸取代自N-端起第172个氨基酸、用丝氨酸取代自N-端起第174个氨基酸、用酪氨酸取代自N-端起第176个氨基酸、以及用苏氨酸取代自N-端起第179个氨基酸。
6.根据权利要求1至5中任一项权利要求所述的生长激素受体拮抗剂,其中长效载体与所述生长激素变体连接。
7.根据权利要求6所述的生长激素受体拮抗剂,其中所述长效载体融合至所述生长激素变异体的N-端或C-端。
8.根据权利要求6或7所述的生长激素受体拮抗剂,其中所述长效载体选自由聚乙二醇、脂肪酸、白蛋白或其片段、白蛋白结合物质、α-1抗胰蛋白酶或其变体、免疫球蛋白Fc或其片段、特定氨基酸序列重复单位的聚合物、抗体或其片段、FcRn结合物质、体内结缔组织或其衍生物、核苷酸、纤维粘连蛋白、转铁蛋白、糖和聚合物组成的组中。
9.根据权利要求7所述的生长激素受体拮抗剂,其中所述长效载体为α-1抗胰蛋白酶或其变体。
10.根据权利要求9所述的生长激素受体拮抗剂,其中α-1抗胰蛋白酶变体包含一个或多个氨基酸用另一个氨基酸取代,且所述取代包含自N-端起第1个氨基酸和第25个氨基酸中一个或多个位置的取代。
11.根据权利要求9或10所述的生长激素受体拮抗剂,其中α-1抗胰蛋白酶变体包含选自由以下组成的组中的任一种或多种取代:用天冬酰胺取代自N-端起第9氨基酸、用丝氨酸取代自N-端起第232氨基酸、用天冬酰胺取代自N-端起第37个氨基酸、用苏氨酸取代自N端起第359个氨基酸。
12.根据权利要求6至11中任一项权利要求所述的生长激素受体拮抗剂,其中所述生长激素变体直接或通过连接器与所述长效载体融合。
13.根据权利要求12所述的生长激素受体拮抗剂,其中所述连接器为肽基连接器或非肽基连接器。
14.根据权利要求13所述的生长激素受体拮抗剂,其中所述非肽基连接器为聚乙二醇、聚丙二醇、乙二醇和丙二醇的共聚物、聚氧乙基化多元醇、聚乙烯醇、多糖、葡聚糖、聚乙烯基乙醚、聚乳酸(PLA)、聚乳酸-乙醇酸(PLGA)、脂质聚合物、甲壳素、透明质酸和其组合。
15.根据权利要求13所述的生长激素受体拮抗剂,其中所述肽基连接器包含两个或更多个氨基酸。
16.一种用于预防或治疗由人类生长激素引起的疾病的药物组合物,其包括根据权利要求1至15中任一项权利要求所述的生长激素受体拮抗剂。
17.根据权利要求16所述的药物组合物,其中所述疾病选自由以下组成的组中:肢端肥大症、巨人症、癌症、糖尿病肾病、关节炎、肺部炎症、生长激素缺乏症(GHD)、特发性身材矮小、特纳综合征、普拉德-威利综合征、小于胎龄儿和慢性肾功能不全(CRI)。
18.一种制备生长激素受体拮抗剂的方法,所述方法包括培养细胞的步骤,所述细胞包括编码所述生长激素变体的多核苷酸,其中生长激素的氨基酸序列中的一个或多个氨基酸用另一个氨基酸取代。
