Abstract

本发明涉及一种增加香石竹花朵直径的基因与应用,属于植物基因工程技术领域。本发明所述的增加香石竹花朵直径的基因的RNA序列如SEQ ID NO.1所示,该基因命名为OSDLa。采用该基因增加香石竹花朵直径的方法具体为:抑制所述OSDLa基因的基因表达量,得到花朵直径增大的转基因植物。本发明首次在香石竹中发现了一个控制香石竹花朵直径的基因,该基因控制花朵直径增大的功能为植物中首次发现,为其他物种利用同源基因克隆相关基因提供了候选基因。本发明基因也可以为石竹科以及其他双子叶植物的花朵直径增大的基因研究提供依据。

一种增加香石竹花朵直径的基因与应用
技术领域
本发明属于植物基因工程技术领域,具体涉及一种增加香石竹花朵直径的基因与应用。
背景技术
花卉是具有观赏价值的植物,花卉主要供观赏的部位有花色、花形、花香、花的大小、数目及花瓣数目等。花朵大小是重要的观赏性状,培育花朵大的花卉新品种是观赏植物育种的重要目标。在拟南芥中发现了一些与花朵大小相关的候选基因,但是大多数基因是在模式植物中进行研究。目前发现拟南芥HWS和CUC1基因调节细胞数量和花瓣的大小,影响细胞增殖(Gonzalez-Carranza ZH, Zhang X, Peters JL, Boltz V, Szecsi J,Bendahmane M, Roberts JA. (2017) HAWAIIAN SKIRT controls size and floralorgan number by modulating CUC1 and CUC2 expression. 香石竹主要分布于欧洲温带以及中国大陆的福建、湖北等地,原产于地中海地区。香石竹是肯尼亚也是美洲哥伦比亚主要的出口花卉品种,在亚洲的中国、日本、韩国、马来西亚等国都有大量栽培。在欧洲,德国、匈牙利、意大利、波兰、西班牙、土耳其、英国和荷兰等国栽培的规模都很大,是世界上应用最普遍的花卉之一。香石竹既可以作为盆栽植物来观赏,也可以当做切花的材料,同时在药用方面也有比较广泛的应用。香石竹花朵大小是重要的观赏性状,有关控制香石竹花朵大小的基因还未被发现。因此,有必要提供一种调控香石竹花朵大小的基因,进而通过转基因的方法培育一些花朵大的花卉新品种。
发明内容
本发明的目的是为了解决现有技术的不足,提供一种增加香石竹花朵直径的基因与应用。
为实现上述目的,本发明采用的技术方案如下:
本发明第一方面提供一种增加香石竹花朵直径的基因,所述的增加香石竹花朵直径基因的RNA序列如SEQ ID NO.1所示。该基因命名为OSDLa
本发明第二方面提供含有所述的增加香石竹花朵直径的基因的表达载体、表达盒、转基因细胞系或重组菌。
本发明第三方面提供所述的增加香石竹花朵直径的基因在调控植物花朵大小表型中的应用。
进一步,优选的是,所述的植物为石竹属植物。
本发明第四方面提供所述的增加香石竹花朵直径的基因在植物育种中的应用。
进一步,优选的是,所述的植物为石竹属植物。
本发明第五方面提供一种增加香石竹花朵直径的方法,包括如下步骤:
抑制所述增加香石竹花朵直径的基因的基因表达量,得到花朵增大的转基因植物。
本发明所述的抑制方法包括RNAi干扰技术和基因编缉技术,但不限于以上基因敲除方法。

实施例1 香石竹OSDLa基因RNAi表达载体构建
1)人工合成OSDLa基因正向及反向互补插入片段
OSDLa基因非保守区域设计插入片段,OSDLa正向插入片断为162 bp,序列如SEQ IDNO.2所示;OSDLa反向插入片断为162 bp,序列如SEQ ID NO.3所示,DNA片断由上海捷瑞生物有限公司人工合成。
2)正向插入片段的连接
30μL 150 ng/μL PJL10的载体,载体序列为SEQ ID NO.4,1μL KpnI和1μL XhoI酶,5μL共用Buffer,加水补齐至50μL体系,放入恒温培养培养箱2-3h后,回收所需的14 kb左右的目的条带备用。
30μL 50 ng/μL待切的正向插入片段,序列如SEQ ID NO.2所示,1μL KpnI和1μLXhoI酶,5μL共用Buffer,加水补齐至50μL体系,放入恒温培养培养箱2-3h后,回收所需的150bp左右的目的条带备用;将上述回收到的片段和载体进行连接。片段和载体进行连接的反应体系如下:

补水到总体积15 μL。
22℃连接1小时。连接液直接去转化。挑取单克隆,提取质粒,质粒提好后测序鉴定,鉴定正确的载体命名为OSDLa-FOR。
3)反向互补插入片段的连接
30μL 150ng/μL待切的含正向插入片断的PJL10表达载体OSDLa-FOR,1μL ClaI和1μLXbaI酶,5μL共用Buffer,加水补齐至50μL体系,放入恒温培养培养箱2-3h后,回收所需的14kb左右的目的条带备用。
30μL 50ng/μL待切的反向插入片段,序列如SEQ ID NO.3所示,1μL ClaI和1μLXbaI酶,5μL共用Buffer,加水补齐至50μL体系,放入恒温培养培养箱2-3h后,回收所需的150bp左右的目的条带备用;将上述回收到的片段和已酶切好的含正向插入片断的载体OSDLa-FOR进行连接。片段和载体进行连接的反应体系如下:

22℃连接1小时。连接液直接去转化,挑取单克隆,提取质粒测序鉴定。验证正确的RNAi载体命名为pJL10-OSDLa-RNAI。
实施例2 香石竹OSDLa基因遗传转化
1)植物表达载体转化农杆菌
采用冻融法将构建好的表达载体转入农杆菌,具体操作如下:向冰上刚融化的40微升农杆菌感受态细胞中加入2µg pJL10-OSDLa-RNAI载体质粒DNA,混匀后冰上放置40min,液氮速冻5min,37 ℃水浴3min至全融,冰上放置2min,加入到1ml YEB液体培养基中(酵母提取物1g,胰蛋白胨5g,牛肉膏5g,七水硫酸镁0.5g,加水定容到1L,pH 7.0),28 ℃、200 rpm/min恢复培养3小时,5000rpm/min离心3分钟,收集菌体,均匀涂于事先加有50mg/LRif和50mg/L Kan的YEB固体培养基平板上(酵母提取物1g,胰蛋白胨5g,牛肉膏5g,七水硫酸镁0.5g,10g琼脂粉,加水定容到1L,pH7.0),28℃倒置避光培养2d。
从转化平板上随机挑取10个单菌落,分别接种于50ml含50mg/LRif和50mg/LKan的YEB液体培养基中,28℃,200rpm/min恒温振荡培养2d,取少量菌液进行PCR扩增鉴定,通过pjL10-pdk-939F(5’-gttagaaattccaatctgcttgtaa-3’,SEQ ID NO.5)和OSD1-REV-88R(5’-acggtacttggctaggttgc-3’,SEQ ID NO.6)引物检测。PCR扩增的体系及程序为:质粒50 ng,pjL10-pdk-939F(10µmol/L)和OSD1-REV-88R(10µmol/L)引物各1µL,10×PCR Buffer(Mg2+ plus)2µL,2.5 mM dNTP Mixture 1.6µL,5U/µL Taq 0.15µL,双蒸水补充至20 µL。PCR反应条件:95℃ 5min;95℃ 30s,58℃ 30s,72℃ 1min,35个循环(从步骤2到步骤4);72℃5min。将鉴定的含目的载体阳性农杆菌与相同体积50%(体积百分比)的甘油混合均匀,置于-80℃冰箱内保存。
2)香石竹遗传转化体系
将-80℃保存的农杆菌接种到加有50mg/L Rif和50mg/L Kan的YEB固体培养基平板上,将平板在28℃倒置培养2d,待菌落长出后,挑取单菌落接种到含50mg/L Rif和50mg/L Kan的YEB液体培养基中,在28℃,200rpm/min摇床上振荡培养过夜,得到预活化菌液;再取20µL预活化菌液接种于30mL YEB液体培养基,继续在28℃,200rpm/min摇床上振荡,当OD600为0.5时,用于农杆菌浸染。
在超净工作台上,取无菌的香石竹组培苗放入体视显微镜下,用镊子小心的剥去顶端的叶片,露出茎尖;将茎尖切去生长点,然后将茎尖横切成1毫米的薄层,薄层背面朝下接种于MS+TDZ 0.3mg/L +NAA 0.1mg/L的预培养基中,培养5d(光照强度1000-2000lux),得到外植体。再浸入之前已活化备好的农杆菌菌液中30min,此过程放在摇床上,低速(30-50rpm)不断振荡以使菌液与外植体充分接触。将外植体放在灭过菌的滤纸上吸干多余的菌液,转入共培养的培养基MS+TDZ 0.3 mg/L+NAA 0.1 mg/L+AS 30 mg/L中在24 ℃条件下暗培养4d,再转入筛选培养基MS+TDZ 0.3 mg/L+NAA 0.1 mg/L+Cef 400 mg/L+kan 80 mg/L中进行筛选培养,光照强度1000-2000lux,40天转接一次,待分化出再生芽,将再生芽转入MS+BA 0.2 mg/L+NAA 0.1 mg/L+Kan 50 mg/L增殖培养基中进行增殖筛选培养,光照强度1000-2000lux。上述所有培养基中含蔗糖30g/L,琼脂10g/L,pH为5.8。
3)香石竹阳性转基因植株的验证
pJL10-OSDLa-RNAI载体阳性转基因植株采用PCR法验证,首先提取的转基因植株DNA,通过pjL10-pdk-939F和OSD1-REV-88引物检测,引物序列如上所述。引物均扩出含目的条带的片断为RNAi载体阳性转基因再生植株。图3PCR扩增的转基因植株为22株,其中1、2、7、9、12、17、18和22扩增出目的条带的为转基因阳性植株,共8株。
实施例3 香石竹转基因阳性植株OSDLa表达和表型
采用实时定量荧光PCR,对OSDLa基因的表达进行分析,发现OSDLa基因在转基因阳性植株中的表达量下降(图1),通过构建OSDLa基因RNAi载体获得转基因植株8株并进行组培及扦插扩繁。转基因植株5-3-B1花朵平均直径7.32cm、3-1-B1平均6.89cm、15-2-A3平均7.09cm、12-2-C4平均6.65cm、12-2-C2平均7.11cm、12-2-C1平均7.33cm、1-3-A4平均7.08cm、12-2-B1平均7.23cm,而对照非转基因植株‘Noglte’花朵平均直径5.9 cm。与野生型相比花直径增加,花朵明显变大(图2)。本发明公开的内容可为研究OSDLa基因在植株中调控花瓣数量打下基础。
