And (3) performance testing:
1) Subjective efficacy testing:
100 infants with mild and moderate diaper rash (flush skin on the hip skin, chestnut-like skin rash, skin damage wave and 2-3 cm around anus, and basically consistent skin rash phenomenon) in 0-3 years old are selected and randomly divided into 10 groups, the infant soothing cream of the examples 1-3 and the infant soothing cream of the comparative examples 1-7 are respectively used, the using effect is tested, and the evaluation result is shown in the following table:
TABLE 3 subjective efficacy test results for infant buttocks
Figure BDA0002459273570000061
Note:
relieving itching: subjective evaluation was performed based on infant response.
As can be seen from Table 3: after the infant soothing cream disclosed by the invention is used for infants with mild and moderate diaper rash, the flushing of the hip skin is relieved, the red rash is reduced and weakened, the pruritus is relieved, and no adverse reaction occurs.
2) Human skin closed patch test:
the test method is based on: technical Specifications for cosmetic safety (2015 edition).
Selecting 32 volunteers of 18-30 years old who meet the test requirements, selecting a proper spot test device, adding a tested substance and a blank control (no substance is placed) into the spot test device by a closed spot test method, sticking the spot test device added with the tested substance on the inner side of the forearm of a testee, lightly pressing with a palm to uniformly stick the spot test device on the skin for 24 hours, observing skin reactions for 0.5 hour (after the indentation disappears), 24 hours and 48 hours after the spot test device of the tested substance is removed, and recording the observation results, wherein the test results are shown in the following table:
TABLE 4 test results of human skin blocking patch test
Figure BDA0002459273570000071
Figure BDA0002459273570000081
Note:
the skin response score was as follows:
0: negative reaction;
1: and (3) suspicious reaction: only faint erythema;
2: weak positive reaction (erythema reaction): erythema, infiltration, edema, and possibly pimples;
3: strong positive reaction (herpes reaction): erythema, infiltration, edema, pimples, herpes: the reaction goes beyond the test area;
4: very strong positive reaction (fusogenic herpes reaction): obvious erythema, severe infiltration, edema, fusional herpes: the reaction goes beyond the test area.
As can be seen from Table 4: after the test substance spot tester is removed, the rating grades of all volunteers are 0 at 0.5h, 24h and 48h, no adverse reaction occurs, and the infant soothing cream is safe and non-irritant.
3) Experimental test for antipruritic (preparation of guinea pig model of pruritus induced by histamine phosphate):
the utilization of histamine receptors is a class of G protein-coupled receptors that leads to a mechanism of pruritus by activating transient receptor potentials, which in turn activate a series of intracellular signaling. An allergic pruritus model is prepared by adopting a histamine phosphate-induced pruritus method, 40 guinea pigs are taken, the right and back instep are shaved 1 day before the experiment, and the hair removal area is about 2cm 2 The guinea pigs were randomly divided into 4 groups and depilated at 0.1g/cm for 2 consecutive days 2 The dosage of the composition is respectively and uniformly coatedThe infant soothing cream of examples 1 to 3 and comparative example 7, wherein the model control group was applied with an equal volume of physiological saline, and on day 3 of the experiment, an appropriate amount of histamine phosphate was weighed and prepared with a gradient concentration of 0.01% (w/v), 0.02% (w/v), 0.03% (w/v), 0.04% (w/v), 0.05% (w/v), 0.06% (w/v), 0.07% (w/v), 0.08% (w/v), 0.09% (w/v) and 0.10% (w/v) with physiological saline just before use, and the right and rear dorsum of the foot of the guinea pig was bruised with coarse sandpaper to have an area of about 1cm 2 After the sample is locally reapplied for 1 time, 0.05mL of 0.01% (w/v) phosphate group is dripped on the abraded part after 10min, whether the action of back foot of the guinea pig after back head licking appears within 3min is observed, if the action does not appear, 0.02% (w/v) histamine phosphate is dripped on the abraded part continuously, whether the action of foot adding appears within 3min is observed, the concentration is increased according to the gradient, 0.05mL of histamine phosphate solution is dripped on the abraded part every 3min until the action of back foot of the guinea pig after back head licking appears, the accumulated total amount of the histamine phosphate dripped at the moment is taken as the itch threshold, the itch threshold of each group is calculated, the difference of the index among the groups is compared, and the test data of the itch relieving experiment are shown in the following table:
TABLE 5 antipruritic Experimental test data
Figure BDA0002459273570000091
From table 5 it can be seen that: the infant soothing cream of examples 1-3 had better antipruritic effect than the infant soothing cream of comparative example 7, and the infant soothing cream of example 3 had much better antipruritic effect than the infant soothing cream of comparative example 7, with significant difference (P < 0.05), with statistical significance of the difference. Therefore, compared with a control sample, the infant soothing cream has a certain instant itching relieving and soothing effect, histamine medium is released during mosquito bite, the experiment is a simulated antihistaminic experiment, and the test result shows that the infant soothing cream has a good inhibition effect on pruritus caused by histamine stimulation.
The above embodiments are preferred embodiments of the present invention, but the present invention is not limited to the above embodiments, and any other changes, modifications, substitutions, combinations, and simplifications which do not depart from the spirit and principle of the present invention should be construed as equivalents thereof, and all such changes, modifications, substitutions, combinations, and simplifications are intended to be included in the scope of the present invention.
