Abstract
本发明涉及一种全紫苏籽(粕)活性肽营养品及其制备方法,是将紫苏籽(粕)蛋白质转化为人体能直接吸收的活性肽,含有2—90%的重量比的全紫苏籽(粕)活性肽,其余为黄酮类物质、亚麻酸、亚油酸、油酸、二十二碳六烯酸、二十碳五烯酸、锌、铁、钙、镁、磷、硒、钾、钠、VE、VB1和平衡量的水,选取榨过油的、无霉变的紫苏粕或紫苏籽,用4—7倍的纯净水浸泡1—4小时后采用二级研磨,将研磨细分后的浆液在温度为118—119℃,0.2—0.3Mpa的压力下脱气钝化4—5分钟,再置于100MPa的超高压均质机中进行超高压处理一遍,浆液经过管式换热器冷却至55—65℃打入专有生化系统,将生化后的浆液灭酶灭菌、调整PH、三级析出、三级浓缩和成品处理即得全紫苏籽(粕)活性肽产品。
Description
技术领域
本发明是一种运用生物工程技术和现代物理学纳米技术原理以全紫苏籽(粕)为主要原料生产的紫苏籽(粕)肽营养品及其制备方法。其活性肽一次性缩合率即活性肽与游离氨基酸的比例高达90%以上。
背景技术
紫苏全株均有很高的营养价值,它具有低糖、高纤维、高胡萝卜素、高矿物质元素等。紫苏种子中含大量油脂,出油率高达45%左右,油中含亚麻酸62.73%、亚油酸15.43%、油酸12.01%。种子中蛋白质含量占25%,内含18种氨基酸,其中赖氨酸、蛋氨酸的含量均高于一般高蛋白植物。此外还有谷维素、维生素E、维生素B1 、结合脂及游离脂肪酸。结合脂中包含卵磷脂(lecithin),溶血卵磷脂(lysolecithin),单半乳糖基甘油二酯(monogalactosyldiglyceride),脑甙脂(cere-broside),脑磷脂(cephalin)及磷酯酰丝氨酸(phosphatidylserine);甾醇中主要为β-谷甾醇...等。此外为十八碳二烯酸,十八碳一烯酸,十六碳酸及十八碳酸)等。由于紫苏籽(粕)中含有丰富的α-亚麻酸,故其具有增强学习记忆功能、降血脂、降血压、抑制血小板聚集、抗癌、防腐、抗氧化、抑制霉菌等作用。紫苏籽的营养成分十分丰富,但目前人们只局限于用紫苏籽榨油,然后将榨油后的粕作为饲料,这对于紫苏籽的利用及开发是极大的浪费,紫苏籽榨油后粕中含蛋白质达到54%以上,且18种氨基酸含量合理,其中赖氨酸、蛋氨酸的含量均高于高蛋白植物籽粒苋。
发明内容
本发明所要解决的问题是提供一种以全紫苏籽或粕为原料运用生物工程技术中的酶工程、组织蛋白工程和现代物理学中的纳米技术原理制备的紫苏籽(粕)肽营养品及其制备方法,可将紫苏籽(粕)蛋白质转化为人体能直接吸收的活性肽(低聚肽、多肽和短肽)等营养成分。所述的肽是低聚肽、短肽和多肽等化合物的混合物(含肽量为2—90%)。为实现上述目的需要解决以下科技难题:一是解决紫苏籽(粕)中脂肪降解转化各种有机酸问题;二是解决紫苏籽(粕)蛋白质不易被酶切断和蛋白链不易打开的难题;三是解决通常蛋白质降解易转化为大量氨基酸而缩合成少量肽的转化为肽的转化率低下的难题;四是解决紫苏籽(粕)肽析出的难题。
全紫苏籽(粕)活性肽营养品包括全紫苏籽(粕)活性肽口服液、全紫苏籽(粕)活性肽粉、全紫苏籽(粕)活性肽含片、全紫苏籽(粕)活性肽胶囊、全紫苏籽(粕)活性肽高能运动饮和全紫苏籽(粕)活性肽针剂。
实施步骤:
1、 选料:选取榨过油的、无霉变的紫苏粕(含油量10—15%左右)或紫苏籽;
2、浸泡软化:将紫苏籽(粕)置于带搅拌的浸泡槽中用纯净水洗干净后,用4—7+倍的纯净水浸泡1—4小时;
3、研磨细分:采用二级研磨法,第一次用细度为120—150目的胶体磨研磨一遍,然后在通过细度为350—400目的胶体磨研磨两遍;
4、钝化、脱气:将研磨细分后的浆液置入特制的五连脱气、钝化系统中,在温度为118—119℃,0.2—0.3Mpa的压力下脱气钝化4—5分钟,旨在脱去紫苏所含有的不良气味和使蛋白质钝化变性,初步克服掉紫苏蛋白质不易被蛋白酶切断的固有抑酶性及钝化掉紫苏籽(粕)中等有害物质活性;
5、高压钝化:将上述物料再置于100MPa(即压力为1000kg)的超高压均质机中进行超高压下的无限细分钝化一遍,进一步钝化紫苏蛋白不易被蛋白酶切断降解的抑酶作用;
6、换热降温:将上述处理后的浆液经过管式换热器冷却至55—650C;
7、酶解切断:将冷却降温后的浆液打入专有生化系统,首先加入0.01—0.04%的食用级稀盐酸破坏紫苏蛋白不易被蛋白酶切断的抑酶性后,再加入0.02—0.06%脂肪酶,在36转/分钟的转速下搅拌2—3分钟,静置0.5—1小时,旋即加入0.6—0.9%的酸性蛋白酶,并调温为46~49℃,对物料做2800转/分的微观分子动力学振荡处理2—3分钟,实现三个目的:(1)使紫苏蛋白的抑酶性在振荡中充分与盐酸接触从而更有效地被破坏;(2)使各种酶制剂与物料更无限地充分融合合一;(3)通过动态分子微粒的充分交融可最大限度地将脂肪转化为二十二碳六烯酸(DHA)、二十碳五烯酸(EPA)和亚麻酸、亚油酸、油酸等各种有机酸和最大限度地将蛋白一次性降解为氨基酸进而失去水分子缩合为活性肽(低聚肽、多肽和多肽),实现一次性降解蛋白质转化为活性肽的缩合率大于90%远远高于国际水平(≤50%)这一令人意想不到的神奇效果。然后恒温静置生化4—5小时;
8、灭酶灭菌:将生化后的浆液升温至 85~95℃,作灭酶灭菌处理30~40 秒;
9、调整PH:将上述物料加入适量的食用级碱性物质,调整PH至中性;
10、一级析出:将调整后的物料打入卧螺式离心分离机进行析出处理;
11、二级析出:将上述处理后的物料再打入蝶式分离机进行二级分离析出处理;
12、三级析出:将二级析出后的物料温度调到71~85℃后打入已经涂好食品级助滤剂硅藻土的板框析出系统作第三级析出,得2—4.5%的全紫苏籽(粕)肽原液,须知食品级硅藻土的预涂量=板框面积数值×(0.5—0.8%)kg;
13、三级浓缩:上述经三级析出处理的全紫苏籽(粕)活性肽原液,其含活性肽量为2—4.5%左右,然后进行三级浓缩处理,得含量达到6%以上的全紫苏籽(粕)活性肽液。
14、成型处理:将浓缩后的肽液分别打入口服液分装系统,真空喷雾干燥系统、片成型系统、胶囊成型系统、高能运动饮分装系统和针剂处理系统,依次获得全紫苏籽(粕)活性肽口服液、全紫苏籽(粕)活性肽粉、全紫苏籽(粕)活性肽含片、全紫苏籽(粕)活性肽胶囊、全紫苏籽(粕)活性肽高能运动饮和全紫苏籽(粕)活性肽针剂;
15、二次灭菌:针对不同产品特点分别作二次灭菌处理;
16、检验合格入库;
17、上市。
本发明通过上述独特而富有科学的创造性工艺技术方法步骤及关键有效参数的实施,不仅使人类第一次取得了富含为人体直接、完全、快速吸收且不会对人体肝、肾、脾和大脑造成高渗透压的特殊生理保健功能:(1)抑制细胞变性,增强人体免疫力;(2)激活细胞活性,有效清除人体有害的自由基;(3)修复人体变性细胞,改善细胞新陈代谢;(4)促进、维护细胞正常新陈代谢及所派生的增强生命活力、免疫力、清除自由基、抗氧化性、非变应性促进脂代谢、降血压、降胆固醇、降血糖、降血脂、抗衰老、抗疲劳、抗癌、消炎和美容护肤等得天独厚的保健功能2—90%重量比的活性肽(低聚肽、短肽和多肽)外,还含有黄酮类物质、亚麻酸、亚油酸、油酸、二十二碳六烯酸、二十碳五烯酸、锌、铁、钙、镁、磷、硒、钾、钠、VE、VB1和平衡量的全紫苏籽(粕)活性肽营养品;同时使蛋白一次性降解成氨基酸进而失去水分子缩合为活性肽的转化率≥90%,远远超过≥49%的国际水平这一令人意想不到的神奇效果。这充分体现了本发明工艺技术方法步骤和关键有效参数的先进性和效果的显著性,体现出极富实用性和创造性。
具体实施方式:
以下结合实施例对本发明的内容加以详细说明。
实施例:
(1)选取榨过油的、无霉变的紫苏粕300kg;
(2)将300kg紫苏粕处理干净后置于带搅拌功能的浸泡槽系统加入1200kg的纯净水浸泡1.5小时;
(3)先用胶体磨将就浸泡搅拌后上述物料磨研磨一遍至120目后,再通过调整后的胶体磨研磨2遍至细度为350目;
(4)将研磨细分后的浆液置入特制的五连脱气、钝化系统中,在温度为118℃,0.2Mpa的压力下脱气钝化4分钟;
(5)将上述物料再置于100MPa的超高压均质机中进行超高压下处理一遍;
(6)将高压钝化后的物料通过管式换热器降温至55℃;
(7)将降温后的浆液打入专有生化系统,并加入0.01%的食用级稀盐酸后,再加入0.02%脂肪酶,在36转/分钟的转速下搅拌2分钟,静置0.5小时,旋即加入0.6%的酸性蛋白酶,调温到46℃,对物料做2800转/分的微观分子动力学振荡处理2后分钟,再恒温静置生化4小时;
(8)将生化后的浆液升温至 85℃,灭酶时间 30 秒;
(9)将上述物料加入适量的食用级碱性物质,调整PH至中性;
(10)将调整后的物料打入卧螺式离心分离机进行一级析出处理;
(11)将上述处理后的物料再打入蝶式分离机进行二级分离析出处理;
(12)将二级析出后的物料温度调到71℃后打入涂好食品级助滤剂硅藻土的板框析出系统作第三级析出,得含肽3.9%的全紫苏粕活性肽原液860kg;
(13)将所得活性肽原液打入三级浓缩系统进行浓缩得含肽6%以上的全紫苏粕活性肽液570kg;
(14)将浓缩后的肽液打入口服液分装系统,可得含肽>6%,50ml/支的全紫苏粕活性口服液11400支;
(15)将上述分装后的11400支口服液通过二次灭菌系统进行二次灭菌处理;
(16)检验合格入库;
(17)上市。
Claims (2)
1.一种全紫苏籽(粕)活性肽营养品,是指全紫苏籽(粕)活性肽口服液、全紫苏籽(粕)活性肽粉、全紫苏籽(粕)活性肽含片、全紫苏籽(粕)活性肽胶囊、全紫苏籽(粕)活性肽高能运动饮和全紫苏籽(粕)活性肽针剂,其特征是它含有2—90%的重量比的全紫苏籽(粕)活性肽(低聚肽、短肽和多肽),其余为黄酮类物质、亚麻酸、亚油酸、油酸、二十二碳六烯酸、二十碳五烯酸、锌、铁、钙、镁、磷、硒、钾、钠、VE、VB1和平衡量的水。
2.权利要求 1 所述全紫苏籽(粕)活性肽营养品的制备方法,包括以下步骤:
(1) 选料:选取榨过油的、无霉变的紫苏粕(含油量10—15%左右)或紫苏籽;
(2)浸泡软化:将紫苏籽(粕)置于带搅拌的浸泡槽中处理干净后,用4—7倍的纯净水浸泡1—4小时;
(3)研磨细分:采用二级研磨法,第一次用细度为120—150目的胶体磨研磨一遍,然后在通过细度为350—400目的胶体磨研磨两遍;
(4)钝化脱气:将研磨细分后的浆液置入特制的五连脱气、钝化系统中,在温度为118—119℃,0.2—0.3Mpa的压力下脱气钝化4—5分钟;
(5)高压钝化:将上述物料再置于100MPa(即压力为1000kg)的超高压均质机中进行超高压处理一遍;
(6)换热降温:将上述处理后的浆液经过管式换热器冷却至55—650C;
(7)酶解切断:将降温后的浆液打入专有生化系统,首先加入0.01—0.04%的食用级稀盐酸破坏紫苏蛋白不易被蛋白酶切断的抑酶性后,再加入0.02—0.06%脂肪酶,在36转/分钟的转速下搅拌2—3分钟,静置0.5——1小时,旋即加入0.6—0.9%的酸性蛋白酶,并调温为46~49℃,对物料做2800转/分的微观分子动力学振荡处理2—3分钟,然后恒温静置生化4—5小时;
(8)灭酶灭菌:将生化后的浆液升温至 85~95℃,灭酶灭菌时间 30~40 秒;
(9)调整PH:将上述物料加入适量的食用级碱性物质,调整PH至中性;
(10)一级析出:将调整后的物料打入卧螺式离心分离机进行析出处理;
(11)二级析出:将一级析出后的物料打入蝶式分离机进行二级分离析出处理;
(12)三级析出:将二级析出后的物料温度调到71~85℃后打入已经涂好食品级助滤剂硅藻土的板框析出系统作第三级析出,得含肽4—4.5%的全紫苏籽(粕)肽原液,须知食品级硅藻土的预涂量=板框面积数值×(0.5—0.8%)kg;
(13)三级浓缩:上述所得的全紫苏籽(粕)活性肽原液进行三级浓缩处理,得含量达到6%以上的全紫苏籽(粕)活性肽液;
(14)成型处理:将三级浓缩后的全紫苏籽(粕)活性肽液分别依市场需要分别打入各种对应的口服液分装系统、真空喷雾干燥系统、片成型系统、胶囊成型系统、高能运动饮分装系统和针剂处理系统,即得全紫苏籽(粕)活性肽口服液、全紫苏籽(粕)活性肽粉、全紫苏籽(粕)活性肽含片、全紫苏籽(粕)活性肽胶囊、全紫苏籽(粕)活性肽高能运动饮和全紫苏籽(粕)活性肽针剂。
