Abstract
本发明公开了一种蜈蚣藻多糖‑纳米硒的制备方法及其应用,属于功能化纳米硒技术领域,该方法将蜈蚣藻多糖溶液与亚硒酸钠溶液混合,向混合液中滴加抗坏血酸溶液,超声反应即可得到纳米硒;本发明将硒制备到纳米级别,得到的纳米硒具有很高的生物活性和生物利用率。纳米硒会比传统硒产品更低毒,为开发补硒产品的上提供了新思路与新发展。将多糖与纳米硒结合,保留纳米载体的给药优点,又保留模板活性。多糖‑纳米硒成为制备食品或者医疗纳米硒的新方法。

技术领域
本发明涉及功能化纳米硒技术领域,特别是涉及一种蜈蚣藻多糖-纳米硒的制备方法及其应用。
背景技术
蜈蚣藻(Grateloupia filicina C.Ag.)属于红藻门、真红藻纲、隐丝藻目、海膜科、蜈蚣藻属藻类,藻体多为红色或紫色,片状,质柔软或稍硬,丛生,多生长在海潮线边的岩礁上或和石沼中。海藻对生长环境有特定的要求,生长在高压、高盐、少光、少氧等海洋环境。自古以来就作为一种海洋中药,具有清热解毒和驱虫之功效,并且资源丰富,在全国沿海均有着广泛的分布,并被当地人们所食用。多糖作为蜈蚣藻的主要化学成分,以半乳聚糖硫酸酯为主,国内外的研究充分表明蜈蚣藻多糖具有良好的抗凝血、抗病毒、抗肿瘤等多种生物活性,说明其有较大的开发利用价值。
硒是人和动物所必需的微量营养元素之一,合理摄入硒元素对保持健康、预防疾病具有重要作用。植物是人和动物摄入硒元素的主要食物来源。在许多生物中,包括所有动物,细胞功能都需要微量的硒盐。植物对硒的需求因物种而异,有些植物需要较多的硒,而另一些植物则不需要硒。硒是多种维生素和其他膳食补充剂中的一种成分,包括婴儿配方奶粉。许多相关的流行病学研究表明,补充硒对某些类型的啮齿类动物的某些癌症有化学预防作用。虽然硒是必需的,但大剂量的硒有毒。人对硒的摄入量十分有限,超过硒最大营养剂量时就会出现不良反应,甚至中毒。硒中毒的体征和症状包括呼吸中有大蒜气味,胃肠紊乱,脱发,剥落指甲,疲劳易怒和神经损伤。极端的硒中毒病例如肝硬化,肺水肿或者死亡。此外,硒缺乏也与一些严重或慢性疾病有关。同时,硒在癌症的治疗、手术、放化疗等用途中也发挥巨大作用。
传统方法补硒是通过膳食,例如从坚果、谷类和蘑菇中得到,但这种方法摄入的硒剂量是十分难以控制的。随着纳米技术的崛起,纳米技术应用到医药行业。通过将纳米材料与生物分子或结构相连,可以为纳米材料添加生物功能。纳米技术为定向输送药物到特定细胞提供可能性,能最大化的提高生物利用度的同时,减少药物副作用。
同时,氧化应激通常引起体内自由基含量升高,从而对蛋白质、脂质和脱氧核糖核酸产生有害的影响。另外也对神经退行性疾病、癌症、心血管疾病、炎症疾病和衰老等各种病理生理过程产生影响。抗氧化剂能降低氧化应激,有益于身体健康。因此近年来,抗氧化产品成为保健品和化妆品企业的新宠儿。
发明内容
本发明的目的是提供一种蜈蚣藻多糖-纳米硒的制备方法及其应用,以解决上述现有技术存在的问题。
为实现上述目的,本发明提供了如下方案:
本发明提供一种蜈蚣藻多糖-纳米硒的制备方法,将蜈蚣藻多糖溶液与亚硒酸钠溶液混合,向混合液中滴加抗坏血酸溶液,超声反应即可得到纳米硒。
进一步地,所述蜈蚣藻多糖溶液的质量浓度为0.5%。
进一步地,所述亚硒酸钠溶液的浓度为0.1mol/L。
进一步地,所述抗坏血酸溶液的浓度为0.1mol/L。
进一步地,所述蜈蚣藻多糖溶液、亚硒酸钠溶液和抗坏血酸溶液的质量比为1:1:3。
进一步地,所述混合和滴加过程在磁力搅拌条件下进行;所述反应在40℃恒温加热磁力搅拌条件下反应2h。
进一步地,所述蜈蚣藻多糖的制备方法,包括以下步骤:
a.将蜈蚣藻粉末置于索氏提取器中,分别用石油醚回流、乙醇回流,干燥;
b.将干燥后的蜈蚣藻粉末加入去离子水中,在超声波辅助作用下提取,离心,得蜈蚣藻多糖提取液;
提取条件:温度60-100℃,料液比1:70-110,提取时间1-5h;
c.将蜈蚣藻多糖提取液浓缩,醇沉,离心分离沉淀,风干,得到蜈蚣藻多糖。
进一步地,所述蜈蚣藻粉末是将蜈蚣藻干燥后,粉碎,过40目筛得到;
所述石油醚的沸程为60-90℃,所述石油醚回流的时间为2h;
步骤a中,所述乙醇为95%乙醇,所述乙醇回流的时间为2h;
所述干燥为60℃干燥12h;
所述超声波为发散式超声,频率40kHz,功率0-800W;
步骤b中,所述离心条件为60000rpm离心15min;
所述步骤c中浓缩为抽真空旋转蒸发浓缩至原提取液体积的1/2;醇沉为加入体积为浓缩后提取液体积2~3倍的乙醇溶液,在温度4℃~10℃静置12h~24h,乙醇溶液的浓度为体积分数90%~100%。
本发明还提供一种上述的蜈蚣藻多糖-纳米硒的制备方法提取得到的蜈蚣藻多糖-纳米硒。
本发明还提供一种上述的蜈蚣藻多糖-纳米硒在抗氧化活性及制备天然抗氧化剂中的应用。
本发明公开了以下技术效果:
本发明以均匀粉碎且稳定的蜈蚣藻粉末为原料,用去离子水为提取液,运用热水提取的方法提取蜈蚣藻多糖,配合超声波作用,超声波的空化作用可以产生极大压力造成被破碎细胞壁及整个生物体破裂,而且整个破裂过程在瞬间完成,有利于多糖的溶出;并通过提取温度、料液比和提取时间的配合,缩短了提取时间,提高了提取率。
本发明将硒制备到纳米级别,得到的纳米硒具有很高的生物活性和生物利用率。纳米硒会比传统硒产品更低毒,为开发补硒产品提供了新思路与新发展。将多糖与纳米硒结合,保留纳米载体的给药优点,又保留模板活性。多糖-纳米硒成为制备医疗纳米硒的新方法。
蜈蚣藻中富含丰富多糖,具有多种生物活性,硒也是构成抗氧化酶的一个组成部分,也具有抗氧化能力。所以,本发明用亚硒酸钠为材料、聚乙烯醇为模板制备纳米硒,且创新尝试利用蜈蚣藻多糖为模板制备纳米硒,通过对比其在体外抗氧化能力,以研究蜈蚣藻多糖和纳米硒结合是否有意义,为后续进一步研究作铺垫。

在本发明实施例中,蜈蚣藻,从广东汕尾店铺中购得。其他所用试剂均为分析纯,所用试剂来源和所用仪器来源均可由本领域技术人员根据经验或有限实验来进行选择。
实施例1蜈蚣藻多糖的合成及提取率的测定
1.葡萄糖标准曲线的制备
1.1制备不同浓度的葡萄糖标准品
准确称取0.025g葡萄糖标准品。加适量去离子水溶解后,转移至50mL容量瓶中定容至刻度线,得0.500mg/mL葡萄糖对照品溶液。分别精密吸取0.500mg/mL葡萄糖标准品溶液2.00、3.00、4.00、5.00、6.00mL于25mL容量瓶中定容,得0.04、0.06、0.08、0.10、0.12mg/mL葡萄糖溶液。
1.2苯酚-硫酸显色法
吸取上述不同浓度溶液1.0mL于试管中,在冰水浴中,缓慢地加入5%苯酚溶液1.0mL,摇匀。向试管中快速加入浓硫酸溶液5mL,振摇5min,置于沸水浴10min。加热后,取出冷却20min。在进行操作的同时以去离子水代替葡萄糖溶液按同样的步骤作为空白对照。
1.3含量测定及标准曲线的绘制
在485nm处测量吸光度值,制作标准曲线,如图1所示:横坐标为葡萄糖的浓度,纵坐标为吸光度值,并计算得到回归方程Y=9.9812X+0.0095。
2.蜈蚣藻多糖提取率的测定
取蜈蚣藻多糖提取液1.0mL替换上述1.2中的葡萄糖标准品溶液,其余操作同1.3中的苯酚-硫酸显色法。将所得吸光度值代入回归方程得到提取液中多糖化合物的质量浓度(mg/mL),然后按公式(1-1)计算多糖提取率。
式中:C为蜈蚣藻多糖提取液浓度(mg/mL);V为定容体积(mL);W为蜈蚣藻粉质量(g)。
3.蜈蚣藻多糖提取最佳工艺的确定
3.1单因素试验
在蜈蚣藻多糖的提取试验,包括以下步骤:
a.将蜈蚣藻干燥后,粉碎,过40目筛得到蜈蚣藻粉末,将蜈蚣藻粉末置于索氏提取器中,分别用沸程为60-90℃的石油醚回流2h、95%乙醇回流2h,60℃干燥12h;
b.将干燥后的蜈蚣藻粉末加入去离子水中,在发散式超声波辅助作用下提取,频率40kHz,功率400W,60000rpm离心15min,得蜈蚣藻多糖提取液;
提取条件:温度60-100℃,料液比1:70-110,提取时间1-5h;
c.将蜈蚣藻多糖提取液抽真空旋转蒸发浓缩至原提取液体积的1/2,加入浓缩后提取液体积3倍的体积分数95%的乙醇溶液,在温度8℃静置18h,6000r离心20min,分离沉淀,风干,得到蜈蚣藻多糖。
蜈蚣藻多糖的提取率可受多种因素影响,为了探究相互之间关系,设定料液比、提取时间和提取温度为三种因素。改变一种因素,其它不变。
3.1.1温度
为探究温度(℃)的变化与蜈蚣藻多糖提取率的关系,设定料液比为1:70(g/mL),加热1h。改变温度,使其为60℃、70℃、80℃、90℃、100℃。
操作如下:
(1)将反应物放恒温加热磁力搅拌器中反应,设定在上述不同的温度条件下。
(2)反应结束后将反应物在高速离心机下分离。上清液即多糖粗提取液。
(3)提取液按照3.3方法测定吸光度。并计算提取率。
结果如图2所示,由图2可知,温度从60℃升高到70℃时提取率略有下降,但相差不大。温度从70℃到100℃时,随着提取温度的升高,蜈蚣藻多糖的提取率先增大后下降。在90℃时,多糖提取率达到46.76%,比70℃时的27.65%提高了69.11%。
结果表明,提高温度可以提高多糖的提取率。但是,过高的温度可能导致多糖结构的改变,从而导致多糖生物活性降低甚至失活。因此,在固定其他因素的条件下,提取温度范围选择在90℃左右。
3.1.2料液比
为探究料液比与蜈蚣藻多糖提取率的关系,设定在90℃提取,加热时间1h。改变料液比(g/mL),使其为1:70、1:80、1:90、1:100、1:110。操作同3.1.1。
结果如图3所示,由图3可知,料液比从1:70到1:90时提取率增幅明显,从1:90到1:110趋于平缓。在1:100时,提取率达到最高,达42.01%,比1:70时的33.66%提高了24.80%。
结果表明,增加水量,能给多糖的溶解提供更多的空间,进而提高多糖的提取率。随料液比提高,多糖含量逐渐饱和,提取率增加缓慢。另外增加料液比,后续工作量和成本也会增加,考虑生产效率和工作成本。因此,在固定其他因素的情况下,料液比范围选择在1:100左右。
3.1.3提取时间
为探究时间(h)与蜈蚣藻多糖提取率的关系,设定料液比为1:70(g/mL),在90℃加热。改变加热时间,使其为1h、2h、3h、4h、5h。操作同3.1.1。
结果如图4所示,由图4可知,随着加热时间的增加,提取率缓慢的增长。当时间从1h增加到3h时,提取率从34.45%提高到39.06%。当时间为3h到5h时,提取率趋于平缓,5h时提取率达最高40.25%,比1h的提取率增加16.69%。
结果表明,提高加热时间,多糖提取率增加,但趋势缓慢。增加时间会让多糖尽可能多的渗透到提取液中。随着时间推移和浓度上升趋于饱和,趋势变缓慢。另外考虑工艺成本和时间成本,在尽可能高的提取率下,在固定其他单因素的情况下,提取时间范围选择在4h左右。
3.2正交试验
根据单因素的试验结果,选取合适的条件范围,选用L9(34)正交表进行温度、料液比、提取时间3因素3水平的正交试验,确定最佳提取条件。设计与结果如表1所示。

结合表1分析可知,不同单因素对蜈蚣藻多糖提取率的影响从大到小分别为:温度>提取时间>料液比。为得出显著性,运用SPSS软件重新分析,得方差分析表如表2所示。

由表2可知,温度因素的p值等于0.01,说明有极显著性差异;料液比和时间因素的P值小于0.05,说明有显著差异。最佳工艺参数如下:90℃、提取5h、1:100(g/mL)。在该条件下,蜈蚣藻多糖化合物的提取率为50.34%,用苯酚-硫酸法测得粗多糖含量为83.46%(0.417g/mL)。
3.3验证试验
为了检验计算所得的最优条件与真实情况是否一样,进行近似验证性实验。将蜈蚣藻多糖的最佳提取工艺条件下进行3次平行试验。提取液按照2方法测定提取率,得到蜈蚣藻多糖的平均提取率为50.07%。与理论预测值比较,相对偏差为0.27%,重复性较好,说明结果可靠。
实施例2蜈蚣藻多糖-纳米硒的制备与表征
1.溶液的配制
1.1 0.1M亚硒酸钠溶液
精密称取亚硒酸钠1.729g,以去离子水为溶剂,在烧杯中用适量去离子水溶解后转移到100mL容量瓶中,补充去离子水定容至刻度线。
1.2 0.1M抗坏血酸溶液
精密称取抗坏血酸0.440g,以去离子水为溶剂,在烧杯中用适量去离子水溶解后转移到25mL容量瓶中,补充去离子水定容至刻度线。
1.3 0.5%聚乙烯醇溶液
精密称取聚乙烯醇0.500g,以去离子水为溶剂,在烧杯中用适量去离子水溶解后转移到100mL容量瓶中,补充去离子水定容至刻度线。
1.4 0.5%蜈蚣藻多糖溶液
精密称取蜈蚣藻多糖0.500g,以去离子水为溶剂,在烧杯中用适量去离子水溶解后转移到100mL容量瓶中,补充去离子水定容至刻度线。
2.纳米硒的制备
在磁力搅拌下,将浓度为0.5wt%聚乙烯醇溶液与浓度为0.1M亚硒酸钠溶液混合,向混合液中滴加浓度为0.1M抗坏血酸溶液;聚乙烯醇溶液:亚硒酸钠溶液:抗坏血酸溶液的质量比例为1:1:3;将反应混合液置于40℃恒温加热磁力搅拌锅下,采用探头式超声强化辅助制备,探头式超声的固定频率为25KHz,功率为1000W,反应2h,即得纳米硒。
探头式超声法具有制备时间短、纳米颗粒尺寸均匀、分散性好的优点。
3.纳米硒的表征
将所得到的纳米硒产物,在MERLIN扫描电子显微镜下观察产物的粒径和形貌。反应得到的纳米硒在扫描电镜下,如图5和图6所示,制得纳米硒为均匀的球形,直径105nm左右。聚乙烯醇在水溶液中具有较好的溶解能力,由于其长链分子互相盘旋缠绕,所构成的微环境具有良好的悬浮、乳化、稳定作用。生成的单质硒被聚乙烯醇吸附包裹,从而有效阻止了初始形成的硒粒子间互相聚合、团聚,并可起到减缓、控制硒粒子生长的作用。反应得到纳米硒为红色。
4.蜈蚣藻多糖-纳米硒的制备
在磁力搅拌下,将浓度为0.5wt%蜈蚣藻多糖溶液与浓度为0.1M亚硒酸钠溶液混合,向混合液中滴加浓度为0.1M抗坏血酸溶液;蜈蚣藻多糖溶液:亚硒酸钠溶液:抗坏血酸溶液的质量比例为1:1:3;将反应混合液置于40℃恒温加热磁力搅拌锅下,采用双频复合超声强化辅助制备,双频复合超声由槽式超声和探头式超声组合而成,双频复合超声工作时,将探头式超声的探头深入到槽式超声槽中以及同时发出超声工作,其中槽式超声的频率为33KHz,功率为500W可调,探头式超声的固定频率为25KHz,功率为1000W可调,变幅杆端面的直径为10mm,反应2h,即得蜈蚣藻多糖-纳米硒。
双频复合超声工作时,槽式超声和探头式超声同时工作并发出超声波,双频复合超声法具有超声场均匀,不存在死角,蜈蚣藻多糖-纳米硒获得均匀的超声场,能在较短时间内制备,得出的纳米复合颗粒尺寸均匀、分散性好的优点。
5.蜈蚣藻多糖-纳米硒的表征
反应得到的多糖-纳米硒在扫描电镜下,如图7,得到50nm左右小球,部分小球又粘连成100nm左右的球。反应生成的单质硒被多糖原位吸附包裹,有效地阻止了初始形成的粒子间的团聚,并减缓了粒子的生长,使多糖-纳米硒形成稳定的无定型纳米结构存在于溶液中。反应得到的多糖-纳米硒为橙色。
实施例3蜈蚣藻多糖、纳米硒、蜈蚣藻多糖-纳米硒抗氧化试验
3.1DPPH自由基清除实验
3.1.1溶液的配制
(1)精密称取1,1-二苯基-2-三硝基苯肼(DPPH)0.0025g,用少量70%乙醇溶解后转移至50ml棕色容量瓶中,并用70%乙醇定容,得0.05mg/mLDPPH试液,放置冰箱中保存,现配现用;
(2)取上述蜈蚣藻粗多糖溶液、纳米硒溶液和多糖-纳米硒溶液,分别配制成浓度0.5、1.0、1.5、2.0和2.5mg/mL的溶液。
3.1.2清除率测定
分别精密吸取样品液5.00mL,加入用DPPH溶液(0.05mg/mL)5.00mL,充分振摇,于暗处静置30min。在517nm处测定试液的吸光度值Ai。将样品液5.00mL与溶剂5.00mL充分混合后,在517nm处的吸光度值Aj。将DPPH溶液5.00mL与溶剂5.00mL充分混合后,在517nm处的吸光度值Ac。
将得到的A值整理代入公式(2-2)计算DPPH自由基的清除率。
式中Ai为样品液与DPPH溶液混合液的吸光度;Aj为样品液与溶剂70%乙醇混合液的吸光度;Ac为DPPH溶液与溶剂70%乙醇混合液的吸光度。
结果如图8所示,由图8可知:(1)三者清除率都是随浓度的增加而增加。(2)多糖的清除DPPH自由基能力最弱。清除率随浓度先上升后趋于平缓。在多糖浓度为2mg/mL时,清除率达最大12.88%。(3)纳米硒的清除DPPH自由基能力在三者中间水平。用SPSS分析得,纳米硒清除DPPH的IC50为0.662mg/mL。(4)多糖硒的清除DPPH自由基能力最强,多糖硒清除DPPH的IC50为0.377mg/mL。
结果表明,清除DPPH自由基的能力:多糖硒>纳米硒>多糖。说明,多糖与纳米硒的结合后比单独多糖的清除DPPH清除自由基能力显著提高,结合有意义。
3.2羟基自由基清除实验
3.2.1溶液的配制
(1)6mmol/L的FeSO4溶液:精密称取FeSO4·7H2O粉末0.0840g,以去离子水为溶剂,加少量去离子水溶解后移至50ml棕色容量瓶中,并用去离子水定容至刻度,避光保存,现配现用;
(2)6mmol/L的水杨酸-乙醇溶液:精密称取水杨酸粉末0.0420g,以无水乙醇为溶剂,加少量无水乙醇溶解后移至50mL容量瓶中,并用无水乙醇定容至刻度,即得;
(3)6mmol/LH2O2溶液:精密吸取30%H2O2溶液4.60mL于25mL容量瓶中,用去离子水定容至刻度,即得;
(4)蜈蚣藻粗多糖溶液、纳米硒溶液和多糖-纳米硒溶液,分别配制成浓度0.00、0.25、0.5、0.75和1.00mg/mL的溶液。
3.2.2清除率测定
准确吸取6mmol/L Fe2+溶液2.00mL和6mmol/L的水杨酸-乙醇溶液2.00mL于试管中。分别在试管中加入2mL的样品溶液,充分摇匀后,静置10min。加入6mmol/L H2O2溶液2.00mL启动反应,立即置于37℃水浴加热30min。冷却。于510nm处测吸光度值Ai;用去离子水代替H2O2溶液,操作不变,测得吸光度值为Ai0。用去离子水代替样品液,操作不变,测得吸光度值为A0。
将所得数值整理代入清除率的计算公式如公式(2-3)。
式中Ai为样品溶液吸光度值;Ai0为样品溶液空白对照吸光度值;A0为空白溶液吸光度值。
结果如图9所示,由图9可知:(1)多糖的清除羟基自由基能力最弱。(2)纳米硒的清除能力最强,随浓度的增加而增强。用SPSS分析,纳米硒对羟基自由基的IC50为0.468mg/mL。(3)多糖硒的清除羟基自由基能力比单独多糖的能力显著提高。随浓度的增加,清除率增强。多糖硒对羟基自由基的IC50为0.686mg/mL。
结果表明,对羟基自由基的清除率:纳米硒>多糖硒>多糖。说明,多糖与纳米硒的结合后比单独多糖的清除羟基清除自由基能力显著提高,结合有意义。
3.3超氧阴离子自由基清除实验
3.3.1溶液的配制
(1)pH 8.2Tris-HCl缓冲溶液:精密称取2-氨基-2-(羟甲基)-1,3-丙二醇(Tris)1.2114g,用去离子水定容至100mL,得Tris溶液(0.1mol/L)。取该Tris溶液50mL于100mL容量瓶中,加入0.1mol/L盐酸溶液22.9mL,充分混匀后用去离子水定容至刻度,即得;
(2)3mmol/L的邻苯三酚溶液:精密称取0.0191g邻苯三酚,加入少量10mmol/L的HCL溶液溶解后,移至50mL棕色容量瓶中,并用10mmol/L的HCL定容至刻度,即得;
(3)蜈蚣藻粗多糖溶液、纳米硒溶液和多糖-纳米硒溶液,分别配制成浓度0.050、0.075、0.100、0.125和0.150mg/mL的溶液。
3.3.2清除率测定
取试管,加入Tris-HCL缓冲溶液(pH 8.2)4.50mL和不同浓度的样品溶液4.20mL,混匀。移置25℃水浴20min。水浴后再加入预热至25℃的3mmol/L的邻苯三酚溶液0.30mL,迅速充分摇匀。迅速倒入比色皿中,于325nm处以0.1mol/L HCL溶液为参比测其吸光度。每隔30s测一次,连续测5min。用去离子水代替样品,其余操作不变,作为对照。
3.3.3计算
以波长为纵坐标、时间为横坐标作图,计算波长与时间的斜率,实验组为Kb,对照组为K0。
将所得数值代入超氧自由基清除率的计算公式如(2-4)所示:
结果如图10所示,分析由图10可知:(1)多糖对超氧阴离子的抑制率较低,随浓度上升而上升。(2)纳米硒对超氧阴离子的抑制率随浓度上升而上升,在0.100-0.125mg/mL时上升趋势大,后趋于平缓。运用SPSS软件分析得,纳米硒对超氧阴离子的IC50为0.101mg/mL。(3)多糖硒对超氧阴离子的抑制率随浓度上升而上升,多糖硒对超氧阴离子的IC50为0.0778mg/mL。
结果表明,三者对超氧阴离子的抑制率:多糖硒>纳米硒>多糖。多糖与纳米硒结合后比单独多糖的抑制率增强,说明结合有意义。
3.4还原力实验
3.4.1溶液的配制
(1)pH 6.6磷酸缓冲溶液:精密称取3.121g的磷酸二氢钠,用去离子水溶解后,转移到100mL容量瓶中,去离子水定容;精密称取7.164g的磷酸氢二钠,用去离子水溶解后,转移到100mL容量瓶中,去离子水定容;取62.5mL的磷酸二氢钠和37.5mL的磷酸氢二钠到100mL容量瓶中,调节pH值,即得;
(2)1%铁氰化钾溶液:精密称取1.00g的铁氰化钾,用去离子水溶解后,转移到100mL容量瓶中,去离子水定容,即得;
(3)10%三氯乙酸溶液的配制:精密称取10.00g的三氯乙酸,用去离子水溶解后,转移到100mL容量瓶中,去离子水定容,即得;
(4)0.1%的三氯化铁溶液:精密称取0.10g的三氯化铁,用去离子水溶解后,转移到100mL容量瓶中,去离子水定容,即得;
(5)蜈蚣藻粗多糖溶液、纳米硒溶液和多糖-纳米硒溶液,分别配制成浓度0.050、0.075、0.100、0.125和0.150mg/mL的溶液。
3.4.2还原力测定
取2.5mL的样品溶液、2.5mL的pH6.6磷酸缓冲溶液和2.5mL的1%铁氰化钾溶液于试管中,混匀。50℃水浴20min。急冷却。再加入2.5mL的10%三氯乙酸摇匀,在3000r/min的速度下离心10min。取5mL上清液加入4mL去离子水水和1mL0.1%三氯化铁,混匀,反应10min。在700nm波长下测吸光度。
还原力与吸光度值相关。吸光度越大,还原力越强,由此比较。
结果如图11所示,抗氧化剂(还原剂)是通过自身的还原作用,给出电子而清除自由基的,还原能力越强,抗氧化性越强。由图11可知:(1)相比另外两者,多糖在此浓度范围的还原力弱,随浓度的升高先上升后下降。(2)纳米硒和多糖硒在该浓度下还原力明显,两者都随浓度上升而增加,但多糖硒还原力略强于纳米硒。能够证明二者都具有较强的还原能力。
结果表明三者还原能力:多糖硒>纳米硒>多糖。说明了多糖与纳米硒结合比单独多糖的还原力强,进一步的说明了结合有意义。
以上所述的实施例仅是对本发明的优选方式进行描述,并非对本发明的范围进行限定,在不脱离本发明设计精神的前提下,本领域普通技术人员对本发明的技术方案做出的各种变形和改进,均应落入本发明权利要求书确定的保护范围内。
Claims (3)
1.一种蜈蚣藻多糖-纳米硒在制备天然抗氧化剂中的应用,其特征在于,所述蜈蚣藻多糖-纳米硒的制备步骤为:在磁力搅拌下,将浓度为0.5wt%蜈蚣藻多糖溶液与浓度为0.1M亚硒酸钠溶液混合,向混合液中滴加浓度为0.1M抗坏血酸溶液;蜈蚣藻多糖溶液:亚硒酸钠溶液:抗坏血酸溶液的质量比例为1:1:3;将反应混合液置于40℃恒温加热磁力搅拌锅下,采用双频复合超声强化辅助制备,双频复合超声由槽式超声和探头式超声组合而成,双频复合超声工作时,将探头式超声的探头深入到槽式超声槽中以及同时发出超声工作,其中槽式超声的频率为33KHz,功率为500W可调,探头式超声的固定频率为25KHz,功率为1000W可调,变幅杆端面的直径为10mm,反应2h,即得所述蜈蚣藻多糖-纳米硒。
2.根据权利要求1所述的蜈蚣藻多糖-纳米硒在制备天然抗氧化剂中的应用,其特征在于,所述蜈蚣藻多糖的制备方法,包括以下步骤:
a.将蜈蚣藻粉末置于索氏提取器中,分别用石油醚回流、乙醇回流,干燥;
b.将干燥后的蜈蚣藻粉末加入去离子水中,在超声波辅助作用下提取,离心,得蜈蚣藻多糖提取液;提取条件:温度60-100℃,料液比1:70-110,提取时间1-5h;
c.将蜈蚣藻多糖提取液浓缩,醇沉,离心分离沉淀,风干,得到蜈蚣藻多糖。
3.根据权利要求2所述的蜈蚣藻多糖-纳米硒在制备天然抗氧化剂中的应用,其特征在于,
所述蜈蚣藻粉末是将蜈蚣藻干燥后,粉碎,过40目筛得到;
所述石油醚的沸程为60-90℃,所述石油醚回流的时间为2h;
步骤a中,所述乙醇为体积分数95%乙醇,所述乙醇回流的时间为2h;
所述干燥为60℃干燥12h;
所述超声波为发散式超声,频率40kHz,功率0-800W;
步骤b中,所述离心条件为60000rpm离心15min;
所述步骤c中浓缩为抽真空旋转蒸发浓缩至原提取液体积的1/2;醇沉为加入体积为浓缩后提取液体积2~3倍的乙醇溶液,在温度4℃~10℃静置12h~24h,乙醇溶液的浓度为体积分数90%~100%。
