Description

Traditional Chinese medicine composition containing clinacanthus nutans and preparation method thereof
Technical Field
The invention belongs to the technical field of medicines or health-care foods, and particularly relates to a traditional Chinese medicine composition containing clinacanthus nutans and a preparation method thereof.
Background
Along with the development of economy, the urbanization process of China is obviously accelerated, the life style of people is changed due to urbanization, physical activity is obviously reduced, the dietary structure is unscientific and healthy in life, the eating habit of a large amount of high carbohydrate and fat is realized, and the people are in a stress environment for a long time along with the acceleration of rhythm, so that the prevalence rates of hypertension, hyperglycemia, hyperlipidemia and tumors are higher and higher.
In general cognition, clinacanthus nutans is not a traditional Chinese medicinal material in the traditional sense, but is a wild vegetable. People in clinacanthus nutans growing areas often eat clinacanthus nutans leaves as vegetables and also drink fresh leaves by squeezing juice. It is generally found that people who like to drink Clinacanthus nutans juice exhibit stronger body resistance than people who do not like. However, the medicinal value of clinacanthus nutans has not been taken into consideration, and related compatibility and application of clinacanthus nutans are rarely studied. The existing clinacanthus nutans products mainly focus on clinacanthus nutans tea (with a food production license), fresh clinacanthus nutans leaves and clinacanthus nutans powder, more than one flavor beverage, not only the sugar content is high, but also the effective amount and the action effect of the clinacanthus nutans are all strong.
Disclosure of Invention
The invention aims to provide a Chinese medicinal composition containing clinacanthus nutans, which solves the problems of high sugar content, low effective content, poor action effect and the like of the conventional clinacanthus nutans product.
In order to solve the technical problems, the invention adopts the following technical scheme:
a traditional Chinese medicine composition containing clinacanthus nutans is characterized by comprising the following components in parts by mass: 10-15 parts of clinacanthus nutans, 2-4 parts of Chinese dates and 1-2 parts of ginger.
Further, the composition comprises the following components in percentage by mass: 12 parts of clinacanthus nutans, 3 parts of Chinese dates and 1 part of ginger.
The pharmacological efficacy of the raw materials selected by the invention is analyzed as follows:
clinacanthus nutans, school name: crocodile flower, also known as Qianli Zhui, Rou Shao Cao, Han Di Cao, Shaba Shea. Sweet, pungent, slightly bitter and neutral in nature, entering liver and kidney meridians. Contains lupeol, betulin, pentacyclic triterpene compounds, Beta sitosterol, calcium element, and flavonoid with high content. The whole plant is used as a medicine, has the effects of clearing heat and removing toxicity, eliminating stasis and swelling, diminishing inflammation and relieving alcoholism, preventing cancer and resisting cancer and the like, can improve the blood flow state of the whole body, enhance the myocardial contractility and improve the blood pressure, contains rich protein and has extremely high nutritional value. The traditional Chinese medicine composition is used for treating nephritis, renal atrophy, renal failure and renal calculus, is a salvage for patients with kidney diseases, and also can be used for treating swelling and pain of throat, hepatitis, jaundice, skin diseases, hypertension, hyperglycemia, hyperlipidemia, gastritis and rheumatic arthralgia, and has a good treatment effect on various cancers.
Fructus Jujubae is also known as fructus Jujubae, dried fructus Jujubae, and semen Ziziphi Spinosae. Is rich in protein, fat, saccharide, carotene, vitamin B, vitamin C, vitamin P, calcium, phosphorus, ferrum, cyclic adenosine monophosphate, etc., and has the important effects of reducing blood lipid and cholesterol.
The ginger is fresh rhizome of perennial herb ginger of Zingiberaceae, and is named as ginger root, Baicaoyun, hooked finger, Yidilong, Yanliangzao and fresh ginger. Ginger is pungent in flavor and slightly warm in nature. It enters lung, spleen and stomach meridians. Has effects of relieving exterior syndrome, dispelling cold, warming spleen and stomach, relieving vomit, warming lung, relieving cough, and removing toxic substance, and can be used for treating wind-cold type common cold, spleen and stomach cold syndrome, stomach cold type emesis, lung cold type cough, and fish and crab toxin.
Since clinacanthus nutans can show certain therapeutic effects on various diseases by themselves, it is generally drunk as a single tea for health care or treatment. However, it has been found that clinacan herbs are cold in nature and generally used only for the treatment of febrile patients, although effective components (including lupeol, betulin, pentacyclic triterpene compounds, flavonoids, etc.) contained therein show good therapeutic effects on hypertension, hyperglycemia, hyperlipidemia, and various cancers. If the patient is unable to take Clinacanthus nutans because the disease is already debilitated, or is prone to adverse reactions such as further debilitation and resistance decline. Therefore, through multi-party compatibility attempts, the invention finally determines that the Chinese date has the effects of tonifying middle-jiao and Qi, nourishing blood and soothing nerves when the Chinese date and the ginger are combined. The ginger has the effect of warming middle energizer and arresting vomiting, and the two medicines are compatible with the clinacanthus nutans, so that the cold property of the clinacanthus nutans which easily injure spleen and stomach can be adjusted, the cold property of the clinacanthus nutans per se can be effectively improved, the blood pressure, blood sugar and blood fat reducing effects of the clinacanthus nutans can be synergistically enhanced, and the excellent effect of the clinacanthus nutans can be more widely applied.
A preparation method of the traditional Chinese medicine composition containing clinacanthus nutans comprises the following steps:
A. cleaning the clinacanthus nutans, the Chinese dates and the gingers respectively, and removing impurities;
B. mixing the Chinese dates, the ginger and a proper amount of water, decocting, and filtering to obtain a decoction;
C. extracting the clinacanthus nutans with alcohol to obtain an extracting solution;
D. concentrating the extracting solution to obtain a concentrated solution;
E. uniformly mixing the boiled liquid and the concentrated solution to obtain a mixed solution;
F. and mixing the mixed solution with a proper amount of maltodextrin to complete granulation.
In order to solve the cold nature and enhance the efficacy of clinacan herbs, jujube and ginger are added for compatibility. For convenience of operation, the method of extracting fructus Jujubae, rhizoma Zingiberis recens and Chinese herbal medicines such as Clinacanthus nutans with alcohol or decocting is common, but neither of them is the best method. Nutraceut clinacanthus as a main medicinal material, certain active ingredients of the nutraceut clinacanthus are sensitive, and loss can be caused in the long-time boiling process. If a method of extracting alcohol by using a plurality of medicinal materials together is adopted, the use amount of the organic solvent is increased, the alcohol content of a subsequent product is higher, and adverse effects are caused to a user. Moreover, the active ingredients of different medicinal materials can form a competitive relationship in the same solvent, so that incomplete release is caused, and the drug effect is influenced. The best method is to adopt different extraction modes according to different drug properties and effects. The method adopts the modes of boiling Chinese dates and gingers and extracting clinacanthus nutans by alcohol to respectively process different medicinal materials.
Further, in the step F, the granulation is a one-step granulation, and the specific operations are as follows: and putting the maltodextrin into a one-step granulating machine set, heating, spraying the mixed solution into the one-step granulating machine set through a pump, mixing with the maltodextrin, granulating and drying.
Further, the specific gravity of the maltodextrin and the mixed solution is 1: 1.
In the existing granulating process, sugar is mostly adopted as a viscous substance, and when one-step granulating is adopted, the medicinal powder is firstly put into a one-step granulating machine set for granule blowing, and then the sugar-containing adhesive is sprayed on the medicinal powder particles to complete the adhesion of the medicinal powder particles and complete the granulating. Thus, although granulation is simple, the resulting product is not suitable for patients with hyperglycemia and is likely to aggravate symptoms of patients with hyperglycemia. Therefore, the present invention uses safe and reliable maltodextrin instead of viscous material and improves the granulation process. Because the actual compatibility of the clinacanthus nutans, the Chinese dates and the ginger after being prepared into the medicinal powder is poor (the three medicines are bonded by the adhesive and cannot interact with each other), the three medicines need to be extracted and then mixed in advance, but the conventional granulation process is difficult to complete granulation. The invention creatively puts the adhesive maltodextrin into the one-step granulating machine set and heats the adhesive maltodextrin to form fluid granules, the fluid granules have strong adsorbability to surface liquid, the mixed liquid medicine is sprayed on the maltodextrin through a pump and is quickly absorbed by the maltodextrin, and the dry medicine granules are obtained after cooling.
Further, in the step B, the boiling temperature is 100-.
Further, in the step B, the specific gravity of the water is 3-4 times of the sum of the Chinese dates and the gingers.
Ginger is a medicinal material with certain irritation, and can cause strong irritation and uncomfortable symptoms such as excessive internal heat and the like when being directly used for people with weak, deficiency and cold. The nutrient components in the Chinese dates are released by boiling and are fully mixed with the ginger juice in the co-boiling process for co-release, so that the irritation of the ginger juice is reduced, and the original warm-tonifying medicinal properties of the Chinese dates are further enhanced. Under the action of ginger, the ginger can cause reflex excitation of the blood vessel movement center of gastric mucosa and sympathetic nerves, promote blood circulation, stimulate stomach function, accelerate the sufficient absorption of nutrient components in Chinese dates, and play a role in strengthening the body resistance of weak and cold people.
Further, in the step C, the alcohol extraction specifically comprises: putting the clinacanthus nutans into an extraction tank, extracting twice, adding an ethanol solution with 7-9 times of the specific gravity of the clinacanthus nutans for the first time, immersing and continuously stirring for 2-3 hours to obtain a primary immersion liquid; adding the ethanol solution of 5-7 times of the specific weight of the clinacanthus nutans for the second time, immersing and continuously stirring for 1-2 hours to obtain a secondary immersion liquid; combining the primary immersion liquid and the secondary immersion liquid to obtain the extracting solution; the concentration of the ethanol solution is 90-95%.
Effective medicinal components of clinacanthus nutans comprise lupeol, betulin, pentacyclic triterpene compounds, flavonoids and the like, which are all organic matters, and the actual extraction efficiency and effect of the conventional method using water as an extractant are not good, because the solubility of part of the organic matters in water is low, the change loss of the effective components can not be greatly improved even if the temperature is raised. Extracting effective medicinal components of Clinacanthus nutans with ethanol as extractant for two times, and extracting to obtain high content extractive solution.
Further, in the step D, the concentration is specifically carried out by pumping the extracting solution into a concentration unit for concentration until the density is 1.1-1.15g/cm3And obtaining the concentrated solution.
The step is mainly used for removing the excessive ethanol in the extracting solution, reducing the content of the alcohol in the product and reducing the irritation of the product.
After concentration, the decoction can be mixed with the concentrated solution. Because the ethanol and the water can be mutually dissolved in any proportion, various effective components in the mixed solution can be uniformly dissolved and can play a mutual synergistic effect. Warming and invigorating components in the boiled liquid can obviously weaken the cold and cool property of clinacanthus nutans. The ginger not only accelerates the absorption of the nutrient components in the Chinese date by the patient, but also accelerates the absorption of the active ingredients of the clinacanthus nutans by the patient. The ginger can inhibit colon cancer hyperplasia and angiogenesis of colon cancer endothelial cell tubule, prevent blood from being transported to tumor via angiogenesis, and enhance the anti-tumor effect of Clinacanthus nutans synergistically. Through the compatibility of clinacanthus nutans, Chinese dates and ginger, the effects of strengthening body resistance and cultivating vigor, reducing blood pressure, blood sugar and blood fat and resisting tumors are achieved.
A Clinacanthus nutans extract product obtained by the preparation method of the Clinacanthus nutans-containing traditional Chinese medicine composition is provided.
Compared with the prior art, the implementation of the invention has the following beneficial effects:
1. clinacanthus nutans has the functions of clearing heat and removing toxicity, removing stasis and reducing swelling, diminishing inflammation and alleviating hangover, preventing cancer and resisting cancer and the like, but the medicine property is cold and cool and easily damages spleen and stomach. The Chinese date has the effects of tonifying middle-jiao and Qi, nourishing blood and soothing nerves. The ginger has the efficacy of warming middle energizer and arresting vomiting, and the two medicines are combined with clinacanthus nutans and can adjust the cold and cool characteristics of the clinacanthus nutans which easily injure spleen and stomach. The Chinese date, the ginger and the clinacanthus nutans are scientifically matched, and the effects of reducing blood sugar, blood fat, blood pressure and tumors of the clinacanthus nutans are synergistically enhanced.
2. According to different properties of the medicine, the Chinese date, the ginger and the clinacanthus nutans raw material are treated by different means, so that the effective components of the Chinese medicinal materials are fully dissolved out, and the efficacy of the pharmaceutical composition is improved.
3. Maltodextrin is used as a binding agent, and medicines are sprayed into the binding agent, so that sugar-free granulation is realized, and the problem of overhigh sugar content of the existing product is solved.

27 nude mice with age of 7-9 weeks and weight of 20-22 g are all fed with high-sugar feed. The product is divided into 9 groups, and each group comprises 3. One week later, the first blood lipid test was performed, and 3 of each group were averaged and recorded.
One group is used as a blank control group and is continuously fed with high-sugar feed; the other groups were fed with the clinacanthus nutans extract product granules (granule product 1g, dissolved in 10ml of water, twice a day) of examples 1 to 3 and comparative examples 1 to 5, respectively, while feeding with a high-sugar feed. A second blood pressure test was performed one week later, and 3 of each group were averaged and recorded. The results are reported in table 1:
TABLE 1
Figure GDA0002493277400000091
As can be seen from Table 1, examples 1-3 of the present invention have significant hypoglycemic effects, while comparative examples 1-2 also exhibit some hypoglycemic effects, but due to the defects in the extraction method, the effective content is low and the anti-cancer effect is poor. In contrast, comparative examples 3 to 4 had limited effect of lowering blood sugar only by the pharmacological effect of clinacanthus nutans itself, because of no proper compatibility.
Effect example 2
27 nude mice with age of 7-9 weeks and weight of 20-22 g are all fed with high-fat feed. The product is divided into 9 groups, and each group comprises 3. One week later, the first blood lipid test was performed, and 3 of each group were averaged and recorded.
One group is used as a blank control group, and high-fat feed feeding is continued; the other groups were fed with the clinacanthus nutans extract product granules (granule product 1g, dissolved in 10ml of water, twice a day) of examples 1 to 3 and comparative examples 1 to 5, respectively, while being fed with a high-fat diet. A second blood pressure test was performed one week later, and 3 of each group were averaged and recorded. The results are reported in table 2:
TABLE 2
Figure GDA0002493277400000101
As can be seen from Table 2, examples 1 to 3 of the present invention have significant hypolipidemic effect, and are effective in reducing the Total Cholesterol (TC) and total Triglyceride (TG) levels in blood. The comparative examples 1-2 show a certain blood fat reducing effect, but have low effective content and poor cancer inhibiting effect due to the defects of the extraction mode. In contrast, in comparative examples 3 to 4, the effect of reducing blood lipid was limited only by the pharmacological effect of clinacanthus nutans itself because of incorrect compatibility.
Effect example 3
27 spontaneous hypertensive rats with age of 7-9 weeks and weight of 20-22 g are selected as male rats and fed normally. The product is divided into 9 groups, and each group comprises 3. The first blood pressure test was performed, and 3 of each group were averaged and recorded.
One group is used as a blank control group and is continuously and normally fed; the other groups were fed separately with the clinacanthus nutans extract product granules (granule product 1g, water 3ml, twice a day) of examples 1 to 3 and comparative examples 1 to 5, while feeding normally. A second blood pressure test was performed one week later, and 3 of each group were averaged and recorded. The results are reported in table 3:
TABLE 3
Figure GDA0002493277400000102
As can be seen from Table 3, examples 1-3 of the present invention have significant blood pressure lowering effect, while comparative examples 1-2 also exhibit certain blood pressure lowering effect, but have low effective content and poor cancer suppressing effect due to the defect of the extraction method. In contrast, comparative examples 3 to 4 had a limited effect of lowering blood pressure only by the pharmacological effect of clinacanthus nutans itself, because of no proper compatibility.
Effect example 4
27 nude mice with age of 7-9 weeks and weight of 20-22 g are all male mice, inoculated with in-situ 4T1 tumor and fed normally. The product is divided into 9 groups, and each group comprises 3. After 3 days, one group is used as a blank control group and normal feeding is continued; the other groups were fed separately with the clinacanthus nutans extract product granules (granule product 1g, dissolved in 10ml of water, twice a day) of examples 1 to 3 and comparative examples 1 to 5, while feeding normally. The experiment was performed for one week, during which time only the dead mice were dissected. One week later, the non-dead mice were all sacrificed for dissection. The results of the anatomical observations are shown in table 4:
TABLE 4
Figure GDA0002493277400000111
As can be seen from Table 4, examples 1-3 of the present invention can significantly inhibit tumor growth and metastasis, while comparative examples 1-2 also exhibit a certain cancer-inhibiting effect, but due to the defects in the extraction method, the effective content is low and the cancer-inhibiting effect is poor. In contrast, in comparative examples 3 to 4, the effect of inhibiting tumor was limited only by the drug effect of Clinacanthus nutans itself, because of the incorrect compatibility.
The above disclosure is only for the purpose of illustrating the preferred embodiments of the present invention and is not to be construed as limiting the scope of the present invention, therefore, the present invention is not limited by the appended claims.
