Abstract
The invention discloses a common turnip composition with an anti-hypoxia function, a preparation method and application thereof. The invention has the advantages of unique and simple formula, high extraction rate of effective components, obvious function, no toxic or side effect, quick absorption, convenient taking and the like, integrates the functions of resisting anoxia and enhancing immunity, and can be safely taken for a long time.
Description
Technical Field
The invention belongs to the technical field of traditional Chinese medicine compositions, and particularly relates to a yuenkanin composition with an anti-hypoxia function, and a preparation method and application thereof.
Background
High altitude hypoxic environment, respiratory diseases (such as pulmonary heart disease, pulmonary infection) and cardiovascular diseases (heart failure, right-left shunt of congenital heart disease) can all cause hypoxia, the lower the blood oxygen concentration, the more severe the hypoxia, and severe hypoxia can affect the cardiovascular system.
Acute hypoxia is usually more acute. The mild patients may have dysphoria, euphoria, agitation, slow reaction, poor comprehension and orientation, rapid respiration and heart rate increase, elevated blood pressure, and tendon hyperreflexia. Serious patients may have varying degrees of disturbance of consciousness, generalized or localized seizures.
Chronic anoxia has symptoms of dizziness, headache, tinnitus, dim eyesight, weakness of limbs, palpitation, short breath, shortness of breath, and rapid heartbeat. Severe, can lead to respiratory failure and even life-threatening.
Either acute or chronic hypoxia can cause dysfunction or dysfunction of body organs, and serious can be life threatening. Therefore, the anti-hypoxia peptide has important significance.
At present, acute hypoxia needs to be sent to a hospital in time for treatment by high pressure oxygen and the like, and chronic hypoxia needs to be treated by equipment such as a breathing machine and the like, so that the use is inconvenient. The technical scheme adopted by the invention is as follows:
a common turnip composition with an anti-hypoxia function comprises the following raw materials in parts by weight:
5-15 parts of common turnip, 20-40 parts of potentilla anserina and 3-9 parts of sea buckthorn.
Further, the feed comprises the following raw materials in parts by weight:
10 parts of common turnip, 30 parts of potentilla anserina and 6 parts of sea buckthorn.
The Genkwa root (Brassica rapa L.) has rich nutrition, and has pure white, succulent, tender, light and slightly sweet taste, and slightly pungent taste, and contains abundant vitamin A, vitamin B, vitamin C, various saccharides and minerals, p-coumaric acid, caffeic acid, ferulic acid, gentisic acid, phenylpyruvic acid, and p-channelBenzoic acid, and the like. Every hundred grams of fresh product contains 1.5g of protein, 0.1g of fat, 6.3g of carbohydrate, 0.5g of dietary fiber and VA38mg、VC25mg of vitamin B10.07mg of vitamin B20.04mg, phosphorus 31mg, iron 0.5mg, calcium 41mg, and amino acids and other vitamins essential to human body. The lilac daphne root has the effects of clearing away heat and toxic materials, nourishing, enhancing immunity, resisting mutation, radiation, fatigue, aging, anoxia and the like. The common turnip contains more sugar, and people of Tibetan often use the common turnip to relieve a series of altitude anoxia reactions such as dizziness, dim eyesight, palpitation and the like.
Potentilla anserina is the underground root tuber of Potentilla anserine L (Potentilla anserine L.) of Potentilla of Rosaceae, also called herba Herminii, and is commonly used as the medicine in Qinghai plateau drug atlas. Researches show that the potentilla anserina can obviously improve the utilization rate of the organism to oxygen and has obvious anti-hypoxia effect. The ethanol extract of radix Potentillae Anserinae can prolong survival rate of mouse under reduced pressure and anoxia condition, prolong survival time of mouse under specific myocardial anoxia condition caused by asphyxia anoxia and isoproterenol, and inhibit oxidation of oxygen free radicals on erythrocyte. Potentilla anserine polysaccharide is an important active ingredient of potentilla anserine, which has been found to have the ability to enhance the immunity of mice.
Hippophae rhamnoides (Hippophae rhamnoides), Elaeagnaceae, shrubs or trees, also known as Acetoceros tamarinowii, Nitraria, Oxytropis procumbens, Ottypodium, Kittypu, and Kighui. Seabuckthorn contains more than one hundred compounds, which comprise amino acids, vitamins, trace elements, tocopherols, carotenoids, polyphenols, flavones and other active substances such as fatty acids beneficial to the body, which are necessary for the human body but cannot be synthesized by the human body. The sea-buckthorn has obvious functions in protecting cardiovascular and cerebrovascular systems, treating gastrointestinal diseases, protecting liver, resisting tumor, resisting oxidation and regulating immunity.
The invention takes the plant juice and the extract of traditional Tibetan medicine and food as main components, fully exerts the regulation effect of the plant raw materials on the physiological function of a human body through the reasonable proportioning of the daphne genkwa root, the potentilla anserina and the sea buckthorn, has the synergistic effect on the functions of resisting anoxia, enhancing immunity and the like of the flavone and the polyphenol of the mixed extract of various saccharides of the daphne genkwa root and the potentilla anserina and the sea buckthorn, can generate symptoms of dizziness, dim eyesight and the like due to insufficient oxygen supply of brain during anoxia, can supplement energy for the human body in time by sugar intake at the moment, can promote the brain to operate and relieve the symptoms of dizziness, palpitation and the like, can improve the immunity of the human body by enhancing the activity of immunocytes and accelerating the oxygen supply to the brain to relieve anoxia, therefore, the components of the invention are mutually cooperated to promote to obtain good effect, and the invention has no toxic side effect and can be eaten for a long time; the extract obtained by mixing and extracting the potentilla anserina and the sea buckthorn effectively improves the concentration of effective components, so that the extract obtains good anti-hypoxia effect, and the extract is further verified by an anti-hypoxia experiment, and simultaneously neutralizes the cold and heat properties of the extract, so that the applicable population is expanded, and the long-term eating is facilitated.
The preparation method of the radix genkwa composition with the anti-hypoxia function comprises the steps of uniformly mixing the radix genkwa with the mixed extract of the potentilla anserina and the sea buckthorn to obtain the radix genkwa composition;
the radix Genkwa is obtained by squeezing radix Genkwa, centrifuging and filtering;
the mixed extract of potentilla anserina and sea buckthorn is prepared by the following operations: mixing radix Potentillae Anserinae and fructus Hippophae, adding 70 vt% ethanol 4-10 times of the total weight of the mixture, soaking for 20-40min, heating under reflux, and extracting for 2 times.
Further, the specific preparation steps of the mixed extract of the potentilla anserina and the sea buckthorn are as follows:
mixing radix Potentillae Anserinae and fructus Hippophae, adding 5-10 times of 70 vt% ethanol, soaking for 30min, heating to 75-85 deg.C, reflux extracting for 1-3 hr, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 4-6 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 75-85 deg.C, reflux extracting for 0.5-1.5 hr, filtering, rotary steaming the filtrate until no ethanol smell exists, mixing filtrates, and concentrating.
The application of the yuenkanin composition with the anti-hypoxia function in preparing medicines, health-care products and foods.
Further, the method specifically comprises the following steps: the composition can be made into granule, capsule, tablet, pill, oral liquid, plant beverage, solid beverage or teabag.
In summary, due to the adoption of the technical scheme, the invention has the beneficial effects that:
1. according to the invention, the Tibetan medicinal and edible dual-purpose coriander raw juice and the potentilla anserina and sea backthern mixed extract are used as main components, and the regulation effect of plant raw materials on the physiological function of a human body is fully exerted through reasonable proportioning of the coriander, the potentilla anserina and the sea backthern and mixed extraction of the potentilla anserina and the sea backthern, so that the Tibetan medicinal and edible dual-purpose natural juice has no toxic or side effect and can be;
2. experiments prove that the mixed extract of the common turnip, the silverweed cinquefoil root and the sea backthern has a synergistic effect in the aspects of functions of resisting anoxia, enhancing immunity and the like, compared with the raw juice of the silverweed cinquefoil root and the sea backthern and the extract which is separately extracted from the silverweed cinquefoil root and the sea backthern, the extract which is obtained by mixing and extracting the silverweed cinquefoil root and the sea backthern effectively improves the concentration of effective components, so that the extract shows good effects in normal-pressure anoxia resistant time and acute cerebral ischemia anoxia experiments, and meanwhile, the extraction mode of mixing and extracting the silverweed cinquefoil root and the sea backthern neutralizes the cold and heat properties, thereby expanding applicable people, being beneficial to;
3. the composition has the advantages of simple components, unique formula, scientific dosage proportion, high extraction rate of effective components, remarkable function, no toxic or side effect, quick absorption, convenient taking and easy preparation, and can be prepared into various medicines, health-care products or foods by using a method acceptable in production.

The yuenkanin composition with the anti-hypoxia function provided by the preferred embodiment of the invention comprises the following components in parts by weight:
10 parts of common turnip, 30 parts of potentilla anserina and 6 parts of sea buckthorn;
is prepared by uniformly mixing the mixed extracts of radix genkwa, radix potentillae anserinae and sea buckthorn;
the radix Genkwa is obtained by squeezing radix Genkwa, centrifuging and filtering;
the mixed extract of potentilla anserina and sea buckthorn is prepared by the following operations: mixing radix Potentillae Anserinae and fructus Hippophae, adding 70 vt% ethanol 8 times of the total weight of the mixture, soaking for 30min, heating to 80 deg.C, reflux extracting for 2 hr, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 5 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 80 deg.C, reflux extracting for 1h, filtering, rotary steaming the filtrate until no ethanol smell exists, mixing filtrates, and concentrating.
Example 2
The yuenkanin composition with the anti-hypoxia function provided by the preferred embodiment of the invention comprises the following components in parts by weight:
5 parts of common turnip, 20 parts of potentilla anserina and 3 parts of sea buckthorn;
is prepared by uniformly mixing the mixed extracts of radix genkwa, radix potentillae anserinae and sea buckthorn;
the radix Genkwa is obtained by squeezing radix Genkwa, centrifuging and filtering;
the mixed extract of potentilla anserina and sea buckthorn is prepared by the following operations: mixing radix Potentillae Anserinae and fructus Hippophae, adding 70 vt% ethanol 5 times of the total weight of the mixture, soaking for 30min, heating to 80 deg.C, reflux extracting for 2 hr, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 4 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 80 deg.C, reflux extracting for 1h, filtering, rotary steaming the filtrate until no ethanol smell exists, mixing filtrates, and concentrating.
Example 3
The yuenkanin composition with the anti-hypoxia function provided by the preferred embodiment of the invention comprises the following components in parts by weight:
15 parts of common turnip, 40 parts of potentilla anserina and 9 parts of sea buckthorn;
is prepared by uniformly mixing the mixed extracts of radix genkwa, radix potentillae anserinae and sea buckthorn;
the radix Genkwa is obtained by squeezing radix Genkwa, centrifuging and filtering;
the mixed extract of potentilla anserina and sea buckthorn is prepared by the following operations: mixing radix Potentillae Anserinae and fructus Hippophae, adding 10 times of 70 vt% ethanol, soaking for 30min, heating to 80 deg.C, reflux extracting for 2 hr, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 6 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 80 deg.C, reflux extracting for 1h, filtering, rotary steaming the filtrate until no ethanol smell exists, mixing filtrates, and concentrating.
Example 4
The yuenkanin composition with the anti-hypoxia function provided by the preferred embodiment of the invention comprises the following components in parts by weight:
7 parts of common turnip, 25 parts of potentilla anserina and 5 parts of sea buckthorn;
is prepared by uniformly mixing the mixed extracts of radix genkwa, radix potentillae anserinae and sea buckthorn;
the radix Genkwa is obtained by squeezing radix Genkwa, centrifuging and filtering;
the mixed extract of potentilla anserina and sea buckthorn is prepared by the following operations: mixing radix Potentillae Anserinae and fructus Hippophae, adding 7 times of 70 vt% ethanol, soaking for 30min, heating to 80 deg.C, reflux extracting for 1h, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 5 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 80 deg.C, reflux extracting for 0.5h, filtering, rotary steaming the filtrate until no alcohol smell exists, mixing filtrates, and concentrating.
Example 5
The yuenkanin composition with the anti-hypoxia function provided by the preferred embodiment of the invention comprises the following components in parts by weight:
12 parts of common turnip, 35 parts of potentilla anserina and 8 parts of sea buckthorn;
is prepared by uniformly mixing the mixed extracts of radix genkwa, radix potentillae anserinae and sea buckthorn;
the radix Genkwa is obtained by squeezing radix Genkwa, centrifuging and filtering;
the mixed extract of potentilla anserina and sea buckthorn is prepared by the following operations: mixing radix Potentillae Anserinae and fructus Hippophae, adding 9 times of 70 vt% ethanol, soaking for 30min, heating to 80 deg.C, reflux extracting for 2 hr, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 6 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 80 deg.C, reflux extracting for 1.5 hr, filtering, rotary steaming the filtrate until no alcohol smell exists, mixing filtrates, and concentrating.
Examples of the experiments
Experimental groups: compositions prepared in example 1 were used in the low (0.05mL/10g), medium (0.2mL/10g), and high (0.5mL/10g) dose groups, respectively;
blank group: pure water;
control group 1: rhodiola sachets available on the market;
control group 2: the preparation method is the same as that of the embodiment 1, only the potentilla anserina and the sea buckthorn are respectively extracted and then mixed;
control group 3: the mixture ratio is the same as that of example 1, and the mixture of yuenkanin, potentilla anserine and sea-buckthorn raw juice is adopted.
100 Kunming white mice with half male and female bodies, 18-22g of body weight and clean grade are bred for 5 days conventionally, after the mice are adapted to the environment, except a blank group, the mice are irrigated with corresponding medicaments by intragastric administration every day, the dosage is 0.2mL/10g, and the mice are continuously bred for 7 days.
(1) Experiment of oxygen deficiency resistance under normal pressure
According to the functional test method for improving anoxia endurance in the functional evaluation test method of health food test and evaluation technical Specification (2003 edition), after the last 1 gastric lavage for administration for 60min, each group of mice is placed in 250mL ground bottles sealed by Vaseline (10 g of soda lime is placed in the bottles in advance to absorb carbon dioxide and water), 1 mouse is placed in each bottle, the time is measured immediately after the bottle is covered, and the survival time of the mice is observed and recorded by taking the last breathing time of the mice as an index.
(2) Acute cerebral ischemia and hypoxia experiment
After the administration for 60min by gavage, each group of mice broke their heads from the neck, immediately timing, and recording the time from the head breaking to the mouth opening wheezing stopping, namely the time of acute cerebral ischemic anoxia.
Statistical processing was performed using SPSS19.0 statistical software, the results of the analysis are expressed in x + -s, the comparison between groups was performed using t-test, and differences were considered statistically significant when P < 0.05.
The results of the experiments are shown in table 1 below. Compared with the blank group, the acute cerebral ischemia and hypoxia time of the control group 1 is obviously increased, has extremely obvious difference (P <0.01), and the normal pressure hypoxia tolerance time is only slightly increased; the normal pressure hypoxia tolerance time of the middle and high dose groups in the experimental group has significant difference (P <0.05), and the acute cerebral ischemia hypoxia time has significant difference (P < 0.01); the time of the normal pressure hypoxia tolerance of the control group 2 is between that of the control group 1 and that of the experimental group, while the time of the acute cerebral ischemia hypoxia is between that of the control group 1 and that of the medium-high dose group of the experimental group, and the time is significantly different (P < 0.05); the time of normal pressure hypoxia tolerance and the time of acute cerebral ischemia hypoxia of the control group 3 are slightly increased.
The result shows that the invention can obviously increase the mouse normal pressure hypoxia tolerance time and the acute cerebral ischemia hypoxia tolerance time, and compared with the commercially available medicine, the invention has the advantages that the normal pressure hypoxia tolerance time is increased, and the normal pressure hypoxia tolerance time and the acute cerebral ischemia hypoxia tolerance time of the original juice are greatly improved, which shows that the mixed extraction of the potentilla anserina and the sea buckthorn has the synergistic enhancement effect, so that the composition of the invention has the good anti-hypoxia function.

4. The method for preparing the yuenkanin composition with the anti-hypoxia function according to claim 3, wherein the mixed extract of the potentilla anserina and the sea buckthorn is prepared by the following steps:
mixing radix Potentillae Anserinae and fructus Hippophae, adding 5-10 times of 70 vt% ethanol, soaking for 30min, heating to 75-85 deg.C, reflux extracting for 1-3 hr, filtering, and rotary steaming the filtrate until no alcohol smell exists; adding 70 vt% ethanol 4-6 times of the total weight of radix Potentillae Anserinae and fructus Hippophae into the residue, heating to 75-85 deg.C, reflux extracting for 0.5-1.5 hr, filtering, rotary steaming the filtrate until no ethanol smell exists, mixing filtrates, and concentrating.
5. Use of the yuenkanin composition with anti-hypoxia function according to claim 1 or 2 in preparation of medicines, health products and foods.
6. The use according to claim 5, characterized in that it is in particular: the composition can be made into granule, capsule, tablet, pill, oral liquid, plant beverage, solid beverage or teabag.
