Heating to boil, extracting for 1.5 hr, filtering, collecting extractive solution, extracting with water once, mixing extractive solutions, heating, and concentrating to obtain extract (YBL). The yield of the extract was 3.24%.
2. Experimental study on weight-reducing effect of jackfruit extract
An animal experiment research scheme for the weight-reducing effect of the wild pineapple extract is designed by referring to an evaluation method of the weight-reducing efficacy of the health food established by the weight-reducing function evaluation method of food and drug administration [2012] 107.
70 male mice of SPF grade 16-20g aged 7 weeks C57BL/6 were selected. After 5 days of adaptive feeding, 60 mice were fed with TP23300 type high-fat high-sugar obesity model feed to induce obesity model, and another 10 mice were fed with TP23302 type control feed as blank control group, which were all provided by Nantong Telofia feed science and technology Co.
The obesity model was fed on week 4 (day 24) with 60 mice ranked by weight gain, and 1/3 obesity resistant mice with lower weight gain were eliminated. The 40 screened obesity sensitive mice were randomly divided into 4 groups: the model group, the YBL high dose group, the YBL medium dose group and the YBL low dose group are continuously fed with the obesity model feed, and the blank control group is continuously fed with the low-fat low-sugar control feed. All mice were housed in a single cage. The obesity model was fed to animals at 5 weeks (day 30) with YBL dissolved in drinking water to a concentration of 2.4mg/mL,1.2mg/mL, 0.6mg/mL, and administered to the YBL high, medium, and low dose groups for drinking. The food intake, food consumption, food remaining, water intake, and weight change were recorded for each group of animals every week.
After feeding the obesity model with the feed for 13 weeks (day 54 of YBL intervention), all mice were fasted for 12h, then the mice were sacrificed by removing cervical vertebrae, the abdomen was opened, the abdominal fat accumulation was recorded by photographing, the perirenal fat and the peritesticular fat were taken and weighed, and the fat/body ratio was calculated. Taking a part of liver tissue, storing in 4% paraformaldehyde, preparing pathological sections, and observing pathological changes after HE staining; the other part was stored at-80 ℃ for testing.
3 results of the experiment
3.1 Effect of Jacob's apple extract on weight, feed, fat/body ratio, and weight gain in obese model mice
Mice fed the high-fat high-sugar obesity model diet for 4 weeks had a significant weight gain (p <0.05) compared to normal mice fed the control diet, after which the difference in body weight became greater and the fat/body weight ratio increased significantly. At the end of the experiment at week 13, the intervention group mice, which had daily drinking a solution of wild pineapple extract (YBL) (concentration 2.4mg/mL,1.2mg/mL), weighed less and gained weight (p < 0.05-0.01) and had a lower fat/body ratio (p <0.05) than the obesity model group mice. The amount of feed consumed daily by each group of mice during the experiment did not differ significantly. The results are shown in detail in FIGS. 1-4.
The results show that the dry prognosis of the jackfruit extract can effectively control the weight increasing trend of the obese mice and reduce the fat proportion in the body.
3.2 Effect of wild pineapple extract on the pathological morphology of the liver tissue of the obese model mouse
When the liver tissue slices of the mice in each group were observed under an HE staining microscope, as shown in FIG. 5, it was found that the fat accumulation in the liver tissue of the mice in the obesity model was significant, the cell morphology was large, and the peripheral steatosis was observed. The YBL administration group mice reduce fatty lesion of liver to different degrees.
