The aster keemun root and the aster keemun extract obtained in the extraction methods of examples 1 to 15 and comparative examples 1 to 7 were weighed, and the extraction rate was calculated according to the following formula:
the extraction rate (mass of aster qishiensis extract/mass of aster qishiensis root) x 100;
the asterone content analysis was performed on the aster keemun extracts obtained by the extraction methods of examples 1 to 15 and comparative examples 1 to 7, and the analysis method was as follows:
(1) preparing a reference solution by precisely weighing a proper amount of asterone reference, and dissolving with absolute ethanol to obtain a solution with the concentration of 0.200mg/mL as the reference solution;
(2) preparing a sample solution: dissolving the Aster keemun extract obtained by the extraction methods of examples 1-15 and comparative examples 1-7 with anhydrous ethanol to obtain a solution containing 0.36mg of Aster keemun extract per 1mL as a sample solution;
(3) preparation of a standard curve: precisely sucking 0.0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0mL of the reference substance solution, respectively placing in a 25mL volumetric flask, and diluting to the scale with absolute ethyl alcohol. By ultraviolet spectrophotometry atThe absorbance of the sample is measured at the position of the maximum absorption wavelength of 390nm, and the regression equation and the related coefficient are calculated as follows: y ═ 0.0051X-5 × 10-5,r=0.9991(n=7)
(4) Determination of sample solution: the absorbance of each of the sample solutions of examples and comparative examples was measured under the above conditions.

As can be seen from the data in the above table, the extraction rate of the preparation methods of examples 1 to 15 is greatly improved compared with the preparation methods of comparative examples 1 to 7, and simultaneously, the content of asterone in the keemun aster extract is also significantly improved, which indicates that the preparation method of the present invention not only improves the extraction rate, but also significantly improves the content of asterone in the keemun aster extract. In the preparation method of example 3, the extraction rate of the aster keemun extract is the highest and can reach 3.52%, the extraction rate is improved by 15.41-32.33% compared with that of the extract obtained in the comparative examples 1-7, the content of asterone in the obtained aster keemun extract is also the highest and can reach 83.82%, and the content of asterone in the comparative examples 1-7 is improved by 38.98-97.55%.
The reason why the extraction rate and the asterone content in the aster keemun extract were low in the preparation methods of comparative examples 1 to 7 as compared with the preparation methods of examples 1 to 15 is that the preparation methods of comparative examples 1 to 7 are different from the preparation method of example 3 in that: comparative example 1 preparation method the aster keemun root was directly freeze-dried without adding the first mixed solvent for homogenization after washing in step S1; comparative example 2 preparation method in step S1 aster keemun roots were washed and then added to an aqueous t-butanol solution without adding an aqueous sucrose solution; comparative example 3 preparation method in step S1 aster keemun roots were washed and then added to the sucrose aqueous solution without adding the tert-butanol aqueous solution; preparation method of comparative example 4 aster keemun powder was added to ethanol without adding polydimethylsiloxane and ethyl acetate in step S3; in the production method of comparative example 5, ethyl acetate was not added to the second mixed solvent in step S3; in the preparation method of comparative example 6, polydimethylsiloxane was not added to the second mixed solvent in step S3; in the production method of comparative example 7, no ethanol was added to the second mixed solvent in step S3. In the preparation method, the keemun aster root is cleaned, the first mixed solvent is added for homogenate, the mixture is frozen and dried, the obtained keemun aster powder is mixed with the second mixed solvent, and then the supercritical carbon dioxide extraction is carried out, so that the extraction rate of the keemun aster extract is improved, the content of the asterone in the keemun aster extract is obviously improved, and the yield of the asterone in the keemun aster extract is higher.

s2, freeze-drying the homogenate to obtain Qiquan powder;
s3, adding the Qishiwan aster powder into a second mixed solvent, and uniformly mixing to obtain a mixed material;
the second mixed solvent comprises the following components in parts by weight:
0.2-0.5 part of ethyl acetate, 2.7-5.2 parts of polydimethylsiloxane and 5.6-8.3 parts of ethanol;
s4, continuously and circularly extracting the mixed material by using supercritical carbon dioxide fluid, wherein the extraction temperature is 38-42 ℃, the extraction pressure is 25-30 MPa, the flow rate is 22-25 kg/h, and the extraction time is 1-1.5 h;
s5, analyzing the extracted carbon dioxide fluid, wherein the flow rate of the carbon dioxide fluid is 5-13 kg/h, the analyzing pressure is 6-8 MPa, the temperature is 40-45 ℃, the analyzing time is 1.5-2.5 h, and discharging is carried out to obtain the Qizi extract.
