Abstract
The invention discloses a gel dressing for preventing and inhibiting HPV virus infection, which consists of hydrogel and traditional Chinese medicine extracts uniformly dispersed in the hydrogel, wherein the traditional Chinese medicine extracts comprise the following components: the gel comprises a sophora flavescens extract, a scouring rush extract, a brucea javanica extract, a fructus cnidii extract, a celosin, carrageenan, a natural balance factor and borneol, wherein the hydrogel comprises sodium hydroxymethyl cellulose, ethylparaben, propylene glycol, mannitol, antibacterial peptide, glycerol, aqueous lubricating powder, diethanolamine and distilled water. By adopting the technical scheme, the anthocyanin is added, so that the effects of moistening skin, protecting heart, conditioning premenstrual syndrome and the like are achieved; the carrageenan is added, so that the immunity of a human body can be effectively improved, the genital tract is moistened, the mucous membrane is protected, and the symptoms of vaginal dryness and the like are effectively relieved.
Description
Technical Field
The invention relates to the technical field of female sanitary nursing products, in particular to a gel dressing for preventing and inhibiting HPV (human papillomavirus) virus infection and a preparation method thereof.
Background
Human Papillomavirus (HPV) infection is one of the most common gynecological infections. It has been reported that the onset of cervical malignancy may be associated with HPV infection, which induces cervical epithelial sarcoma and malignancy mainly by invasion of cervical epithelial cells. HPV viruses can be classified into low-risk types and high-risk types according to the severity of lesions caused by the HPV viruses. Wherein, the treatment with interferon is the most common, while the traditional Chinese medicine treatment can relatively reduce adverse reaction, avoid damaging normal organs and reduce the psychological burden of patients, thereby having certain advantages.
At present, the medicament for treating gynecological diseases is mainly administered through vagina, and the dosage forms of the common vagina administration preparations mainly comprise suppository, effervescent tablets, lotion, capsules and the like, but the dosage forms have some defects more or less and influence the exertion of the medicament. For example, the suppository has poor drug release, small contact surface with the vagina grinding, difficult infiltration of grinding folds, easy deformation at higher temperature and poor stability; for example, in the case of capsules, the drug is not easily released, and the coated surface is poor due to the disintegration of the capsules.
Disclosure of Invention
The invention provides a gel dressing for preventing and inhibiting HPV virus infection and a preparation method thereof.
In order to solve the technical problems, the invention adopts the technical scheme that the gel dressing for preventing and inhibiting HPV virus infection consists of hydrogel and traditional Chinese medicine extracts uniformly dispersed in the hydrogel, wherein the traditional Chinese medicine extracts comprise the following components in parts by weight: 15-25 parts of a sophora flavescens extract, 6-10 parts of a scouring rush extract, 4-7 parts of a brucea javanica extract, 12-15 parts of a fructus cnidii extract, 20-26 parts of a celosin, 6-10 parts of carrageenan, 3-5 parts of a natural balance factor and 4-6 parts of borneol, wherein the hydrogel comprises sodium hydroxymethyl cellulose, ethylparaben, propylene glycol, mannitol, antimicrobial peptide, glycerol, aqueous lubricating powder, diethanolamine and distilled water.
Furthermore, the hydrogel comprises the following components in parts by weight: 7-10 parts of sodium carboxymethylcellulose, 2-4 parts of ethylparaben, 12-16 parts of propylene glycol, 5-8 parts of mannitol, 5-8 parts of antibacterial peptide, 12-16 parts of glycerol, 9-15 parts of water-based lubricating powder, 3-6 parts of diethanolamine and 20-30 parts of distilled water.
The preparation method of the gel dressing for preventing and inhibiting HPV virus infection comprises the following steps,
(1) preparing a sophora flavescens extract: selecting radix sophorae flavescentis, cleaning with clear water, fishing out, drying, crushing, sieving with a 20-60-mesh sieve to obtain radix sophorae flavescentis powder, soaking with 2-4 times of 60-80% ethanol in volume for 20-30 h, and percolating at the flow rate of 5ml/min to obtain the radix sophorae flavescentis extract.
(2) Preparing a scouring rush extract: selecting scouring rush, removing impurities, drying in a dryer, crushing, sieving with a 120-200 mesh sieve to obtain scouring rush powder, putting the scouring rush powder in an ultrasonic extraction device, wherein the extracting solution is 60-80% ethanol, the extraction temperature is 35-45 ℃, and the material-liquid ratio is 1: 25-1: 15, so as to obtain the scouring rush extract.
(3) Preparing the brucea javanica extract: selecting brucea javanica seeds, removing impurities, drying in a dryer, crushing, sieving with a 120-200-mesh sieve to obtain brucea javanica powder, putting the brucea javanica powder in an ultrasonic extraction device to extract brucea javanica oil for 2-4 times, wherein the material-liquid ratio is 31-71, the extraction time is 15-25 hours, and the extraction temperature is 50-70 ℃ to obtain the brucea javanica extract.
(4) Preparing a common cnidium fruit extract: selecting fructus cnidii, removing impurities, drying in the sun, crushing, sieving with a sieve of 80-100 meshes to obtain fructus cnidii powder, soaking in a 200-time volume of 1% NaOH solution at 22-25 ℃ for 50-70 min, carrying out ultrasonic extraction for 8-12 min, adding 10% HCL into an extracting solution, acidifying, centrifuging, taking a precipitate, adding 5-10 times methanol into the precipitate for dissolving to obtain a dissolving solution, and filtering the dissolving solution by using a 0.45-micrometer microporous membrane to obtain the fructus cnidii extract.
(5) Preparing a hydrogel: in a preparation container, sequentially adding propylene glycol and antibacterial peptide according to parts by weight, stirring until compatibilization is transparent, adding mannitol, glycerol, sodium carboxymethylcellulose and ethylparaben, stirring uniformly, standing for 15-20 min, sequentially adding glycol amine, borneol, water-based lubricating powder and distilled water, and stirring uniformly to obtain the hydrogel.
(6) Preparing a traditional Chinese medicine extract mixture: and (3) uniformly mixing the traditional Chinese medicine extracts obtained in the steps (1) - (4) for later use.
(7) Preparing a gel dressing: adding the traditional Chinese medicine extract mixture into the hydrogel according to the parts by weight, uniformly stirring, then adding the anthocyanin, the carrageenan and the natural balance factor, mixing and stirring, and uniformly stirring to obtain the gel dressing.
Furthermore, in the step (7), after the gel dressing is uniformly stirred, the gel dressing is irradiated and disinfected by adopting an ultraviolet irradiation disinfection mode, wherein the irradiation time is 30-60 min.
By adopting the technical scheme, the anthocyanin is added, the anti-free radical oxidation capacity of the anthocyanin is 50 times that of vitamin E and 20 times that of vitamin C, and the anthocyanin can be completely and rapidly absorbed and has the effects of moistening skin, protecting heart, conditioning premenstrual syndrome and the like; the carrageenan is added, so that the immunity of a human body can be effectively improved, the genital tract can be moistened, the mucous membrane can be protected, and the symptoms of vaginal dryness, keratosis and the like can be effectively relieved; because the natural balance factors are added, the effect of activating balance is achieved; the scouring rush is added, and silicate and tannin contained in the scouring rush have an astringent effect and have the effects of diminishing inflammation and stopping bleeding on a contact part; adding brucea javanica extract: oleum fructus Bruceae has effects of clearing away heat and toxic materials.
Detailed Description
The following further describes the embodiments of the present invention. It should be noted that the description of the embodiments is provided to help understanding of the present invention, but the present invention is not limited thereto. In addition, the technical features involved in the embodiments of the present invention described below may be combined with each other as long as they do not conflict with each other.
Fructus cnidii is a plant, also called wild carrot seed, which is picked in summer and autumn, and has mild temperament and bitter taste. The fructus Cnidii extract is extracted from fruit of cnidium monnieri (L.) DC of Umbelliferae, and contains pinene, isovalerian borneol ester, parsley phenol methyl ether, dihydrocarveol, bergapten, osthole, and isoanisic apigenin. The extract of the fructus cnidii mainly contains osthole, and the external application of the traditional Chinese medicine can treat trichomonas vaginitis, but the single fructus cnidii has poor efficacy and needs to be synthesized with other components into a compound preparation for treatment. The extraction of the cnidium fruit generally adopts a water decoction method, an ethanol ultrasonic extraction method and the like, and different extraction methods have different extraction results on the osthole. The table below shows the osthole extraction content for different extraction methods.

Therefore, the alkali-dissolution acid-precipitation ultrasonic extraction method is the best extraction method of osthole.
The first embodiment is as follows:
the gel dressing for preventing and inhibiting HPV virus infection consists of hydrogel and traditional Chinese medicine extracts uniformly dispersed in the hydrogel, wherein the traditional Chinese medicine extracts comprise the following components in parts by weight: 15 parts of coconut extract, 15 parts of sophora flavescens extract, 6 parts of scouring rush extract, 4 parts of brucea javanica extract, 12 parts of fructus cnidii extract, 20 parts of anthocyanin, 6 parts of carrageenan, 3 parts of natural balance factor and 4 parts of borneol, wherein the hydrogel comprises sodium hydroxymethyl cellulose, ethylparaben, propylene glycol, mannitol, antibacterial peptide, glycerol, aqueous lubricating powder, diethanolamine and distilled water.
The hydrogel comprises the following components in parts by weight: 7 parts of sodium carboxymethylcellulose, 2 parts of ethylparaben, 12 parts of propylene glycol, 5 parts of mannitol, 5 parts of antibacterial peptide, 12 parts of glycerol, 9 parts of aqueous lubricating powder, 3 parts of diethanolamine and 20 parts of distilled water.
The preparation method of the gel dressing for preventing and inhibiting HPV virus infection comprises the following steps,
(1) preparing a sophora flavescens extract: selecting radix Sophorae Flavescentis, cleaning with clear water, taking out, oven drying, pulverizing, sieving with 20 mesh sieve to obtain radix Sophorae Flavescentis powder, soaking in 2 times volume of 60% ethanol for 20 hr, and percolating at flow rate of 5ml/min to obtain radix Sophorae Flavescentis extract.
(2) Preparing a scouring rush extract: selecting scouring rush, removing impurities, drying in a dryer, crushing, sieving with a 120-mesh sieve to obtain scouring rush powder, putting the scouring rush powder in an ultrasonic extraction device, wherein the extracting solution is 60% ethanol, the extraction temperature is 35 ℃, and the material-liquid ratio is 1:25 to obtain the scouring rush extract.
(3) Preparing the brucea javanica extract: selecting brucea javanica seeds, removing impurities, drying in a dryer, crushing, sieving with a 120-mesh sieve to obtain brucea javanica powder, putting the brucea javanica powder in an ultrasonic extraction device to extract brucea javanica oil for 2 times, wherein the material-liquid ratio is 31, the extraction time is 15 hours, and the extraction temperature is 50 ℃ to obtain the brucea javanica extract.
(4) Preparing a common cnidium fruit extract: selecting fructus Cnidii, removing impurities, sun drying, pulverizing, sieving with 80 mesh sieve to obtain fructus Cnidii powder, soaking with 200 times volume of 1% NaOH solution at 22 deg.C for 50min, ultrasonic extracting for 8min, adding 10% HCl into the extractive solution, acidifying, centrifuging, collecting precipitate, adding 5 times methanol into the precipitate for dissolving to obtain solution, and filtering with 0.45 μm microporous membrane to obtain fructus Cnidii extract.
(5) Preparing a hydrogel: in a preparation container, sequentially adding propylene glycol and antibacterial peptide according to parts by weight, stirring until compatibilization is transparent, adding mannitol, glycerol, sodium carboxymethylcellulose and ethylparaben, stirring uniformly, standing for 15min, sequentially adding glycol amine, borneol, water-based lubricating powder and distilled water, and stirring uniformly to obtain the hydrogel.
(6) Preparing a traditional Chinese medicine extract mixture: and (3) uniformly mixing the traditional Chinese medicine extracts obtained in the steps (1) - (4) for later use.
(7) Preparing a gel dressing: adding the traditional Chinese medicine extract mixture into the hydrogel according to the weight part, uniformly stirring, then adding the anthocyanin, the carrageenan and the natural balance factor, mixing and stirring, uniformly stirring, and then irradiating and disinfecting the gel dressing by adopting an ultraviolet irradiation disinfection mode for 30-60 min to obtain the gel dressing.
Example two:
the gel dressing for preventing and inhibiting HPV virus infection consists of hydrogel and traditional Chinese medicine extracts uniformly dispersed in the hydrogel, wherein the traditional Chinese medicine extracts comprise the following components in parts by weight: 16 parts of coconut extract, 20 parts of sophora flavescens extract, 8 parts of scouring rush extract, 5 parts of brucea javanica extract, 13 parts of fructus cnidii extract, 23 parts of anthocyanin, 8 parts of carrageenan, 4 parts of natural balance factor and 5 parts of borneol, wherein the hydrogel comprises sodium hydroxymethyl cellulose, ethylparaben, propylene glycol, mannitol, antibacterial peptide, glycerol, aqueous lubricating powder, diethanolamine and distilled water.
The hydrogel comprises the following components in parts by weight: 8 parts of sodium carboxymethylcellulose, 3 parts of ethylparaben, 14 parts of propylene glycol, 6 parts of mannitol, 6 parts of antibacterial peptide, 14 parts of glycerol, 12 parts of aqueous lubricating powder, 5 parts of diethanolamine and 25 parts of distilled water.
The preparation method of the gel dressing for preventing and inhibiting HPV virus infection comprises the following steps,
(1) preparing a sophora flavescens extract: selecting radix Sophorae Flavescentis, cleaning with clear water, taking out, oven drying, pulverizing, sieving with 40 mesh sieve to obtain radix Sophorae Flavescentis powder, soaking in 3 times volume of 70% ethanol for 25 hr, and percolating at flow rate of 5ml/min to obtain radix Sophorae Flavescentis extract.
(2) Preparing a scouring rush extract: selecting scouring rush, removing impurities, drying in a dryer, crushing, sieving with a 160-mesh sieve to obtain scouring rush powder, putting the scouring rush powder in an ultrasonic extraction device, wherein the extracting solution is 70% ethanol, the extraction temperature is 40 ℃, and the material-liquid ratio is 1:20 to obtain the scouring rush extract.
(3) Preparing the brucea javanica extract: selecting brucea javanica seeds, removing impurities, drying in a dryer, crushing, sieving with a 160-mesh sieve to obtain brucea javanica powder, putting the brucea javanica powder in an ultrasonic extraction device to extract brucea javanica oil for 3 times, wherein the material-liquid ratio is 51, the extraction time is 20 hours, and the extraction temperature is 60 ℃ to obtain the brucea javanica extract.
(4) Preparing a common cnidium fruit extract: selecting fructus Cnidii, removing impurities, sun drying, pulverizing, sieving with 90 mesh sieve to obtain fructus Cnidii powder, soaking with 200 times volume of 1% NaOH solution at 23 deg.C for 60min, ultrasonic extracting for 10min, adding 10% HCl into the extractive solution, acidifying, centrifuging, collecting precipitate, adding 8 times volume of methanol into the precipitate for dissolving to obtain solution, and filtering with 0.45 μm microporous membrane to obtain fructus Cnidii extract.
(5) Preparing a hydrogel: in a preparation container, sequentially adding propylene glycol and antibacterial peptide according to parts by weight, stirring until compatibilization is transparent, adding mannitol, glycerol, sodium carboxymethylcellulose and ethylparaben, stirring uniformly, standing for 18min, sequentially adding glycol amine, borneol, water-based lubricating powder and distilled water, and stirring uniformly to obtain the hydrogel.
(6) Preparing a traditional Chinese medicine extract mixture: and (3) uniformly mixing the traditional Chinese medicine extracts obtained in the steps (1) - (4) for later use.
(7) Preparing a gel dressing: adding the traditional Chinese medicine extract mixture into the hydrogel according to the weight part, uniformly stirring, adding the anthocyanin, the carrageenan and the natural balance factor, mixing and stirring, uniformly stirring, and irradiating and disinfecting the gel dressing by adopting an ultraviolet irradiation disinfection mode for 45min to obtain the gel dressing.
Example three:
the gel dressing for preventing and inhibiting HPV virus infection consists of hydrogel and traditional Chinese medicine extracts uniformly dispersed in the hydrogel, wherein the traditional Chinese medicine extracts comprise the following components in parts by weight: 18 parts of coconut extract, 25 parts of sophora flavescens extract, 10 parts of scouring rush extract, 7 parts of brucea javanica extract, 15 parts of fructus cnidii extract, 26 parts of anthocyanin, 10 parts of carrageenan, 5 parts of natural balance factor and 6 parts of borneol, wherein the hydrogel comprises sodium hydroxymethyl cellulose, ethylparaben, propylene glycol, mannitol, antibacterial peptide, glycerol, aqueous lubricating powder, diethanolamine and distilled water.
The hydrogel comprises the following components in parts by weight: 10 parts of sodium carboxymethylcellulose, 4 parts of ethylparaben, 16 parts of propylene glycol, 8 parts of mannitol, 8 parts of antibacterial peptide, 16 parts of glycerol, 15 parts of aqueous lubricating powder, 6 parts of diethanolamine and 30 parts of distilled water.
The preparation method of the gel dressing for preventing and inhibiting HPV virus infection comprises the following steps,
(1) preparing a sophora flavescens extract: selecting radix Sophorae Flavescentis, cleaning with clear water, taking out, oven drying, pulverizing, sieving with 60 mesh sieve to obtain radix Sophorae Flavescentis powder, soaking in 4 times volume of 80% ethanol for 30 hr, and percolating at flow rate of 5ml/min to obtain radix Sophorae Flavescentis extract.
(2) Preparing a scouring rush extract: selecting scouring rush, removing impurities, drying in a dryer, crushing, sieving with a 200-mesh sieve to obtain scouring rush powder, putting the scouring rush powder in an ultrasonic extraction device, wherein the extracting solution is 80% ethanol, the extraction temperature is 45 ℃, and the material-liquid ratio is 1:15 to obtain the scouring rush extract.
(3) Preparing the brucea javanica extract: selecting brucea javanica seeds, removing impurities, drying in a dryer, crushing, sieving with a 200-mesh sieve to obtain brucea javanica powder, putting the brucea javanica powder in an ultrasonic extraction device to extract brucea javanica oil for 4 times, wherein the material-liquid ratio is 71, the extraction time is 25 hours, and the extraction temperature is 70 ℃, so as to obtain the brucea javanica extract.
(4) Preparing a common cnidium fruit extract: selecting fructus Cnidii, removing impurities, sun drying, pulverizing, sieving with 100 mesh sieve to obtain fructus Cnidii powder, soaking with 200 times volume of 1% NaOH solution at 25 deg.C for 70min, ultrasonic extracting for 12min, adding 10% HCl into the extractive solution, acidifying, centrifuging, collecting precipitate, adding 10 times volume of methanol into the precipitate, dissolving to obtain solution, and filtering with 0.45 μm microporous membrane to obtain fructus Cnidii extract.
(5) Preparing a hydrogel: in a preparation container, sequentially adding propylene glycol and antibacterial peptide according to parts by weight, stirring until compatibilization is transparent, adding mannitol, glycerol, sodium carboxymethylcellulose and ethylparaben, stirring uniformly, standing for 20min, sequentially adding glycol amine, borneol, water-based lubricating powder and distilled water, and stirring uniformly to obtain the hydrogel.
(6) Preparing a traditional Chinese medicine extract mixture: and (3) uniformly mixing the traditional Chinese medicine extracts obtained in the steps (1) - (4) for later use.
(7) Preparing a gel dressing: adding the traditional Chinese medicine extract mixture into the hydrogel according to the weight part, uniformly stirring, then adding the anthocyanin, the carrageenan and the natural balance factor, mixing and stirring, uniformly stirring, and then irradiating and disinfecting the gel dressing by adopting an ultraviolet irradiation disinfection mode for 30-60 min to obtain the gel dressing.
150 subjects (all the cervical parts have erosive surfaces, namely, the possibility of HPV infection exists) are invited to use the gel dressings for preventing and inhibiting HPV virus infection prepared in the first example, the second example and the third example respectively, and the feedback results of the subjects after 1 month are obtained as follows:

Claims (4)
1. A gel dressing for preventing and inhibiting HPV viral infection, characterized in that: the hydrogel comprises hydrogel and a traditional Chinese medicine extract uniformly dispersed in the hydrogel, wherein the traditional Chinese medicine extract comprises the following components in parts by weight: 15-25 parts of a sophora flavescens extract, 6-10 parts of a scouring rush extract, 4-7 parts of a brucea javanica extract, 12-15 parts of a fructus cnidii extract, 20-26 parts of a celosin, 6-10 parts of carrageenan, 3-5 parts of a natural balance factor and 4-6 parts of borneol, wherein the hydrogel comprises sodium hydroxymethyl cellulose, ethylparaben, propylene glycol, mannitol, antimicrobial peptide, glycerol, aqueous lubricating powder, diethanolamine and distilled water.
2. The gel dressing for preventing and inhibiting HPV viral infection according to claim 1, wherein: the hydrogel comprises the following components in parts by weight: 7-10 parts of sodium carboxymethylcellulose, 2-4 parts of ethylparaben, 12-16 parts of propylene glycol, 5-8 parts of mannitol, 5-8 parts of antibacterial peptide, 12-16 parts of glycerol, 9-15 parts of water-based lubricating powder, 3-6 parts of diethanolamine and 20-30 parts of distilled water.
3. The method for preparing a gel dressing for preventing and inhibiting HPV viral infection according to claim 1, wherein the method comprises the following steps: comprises the following steps of (a) carrying out,
(1) preparing a sophora flavescens extract: selecting radix sophorae flavescentis, cleaning with clear water, fishing out, drying, crushing, sieving with a 20-60-mesh sieve to obtain radix sophorae flavescentis powder, soaking with 2-4 times of 60-80% ethanol in volume for 20-30 h, and percolating at the flow rate of 5ml/min to obtain a radix sophorae flavescentis extract;
(2) preparing a scouring rush extract: selecting scouring rush, removing impurities, drying in a dryer, crushing, sieving with a 120-200 mesh sieve to obtain scouring rush powder, putting the scouring rush powder in an ultrasonic extraction device, wherein the extracting solution is 60-80% ethanol, the extraction temperature is 35-45 ℃, and the material-liquid ratio is 1: 25-1: 15 to obtain the scouring rush extract;
(3) preparing the brucea javanica extract: selecting brucea javanica seeds, removing impurities, drying in a dryer, crushing, sieving with a 120-200-mesh sieve to obtain brucea javanica powder, putting the brucea javanica powder in an ultrasonic extraction device to extract brucea javanica oil for 2-4 times, wherein the material-liquid ratio is 31-71, the extraction time is 15-25 hours, and the extraction temperature is 50-70 ℃ to obtain a brucea javanica extract;
(4) preparing a common cnidium fruit extract: selecting fructus cnidii, removing impurities, drying in the sun, crushing, sieving with a sieve of 80-100 meshes to obtain fructus cnidii powder, soaking in a 200-time volume of 1% NaOH solution at 22-25 ℃ for 50-70 min, carrying out ultrasonic extraction for 8-12 min, adding 10% HCL into an extracting solution, acidifying, centrifuging, taking a precipitate, adding 5-10 times methanol into the precipitate for dissolving to obtain a dissolving solution, and filtering the dissolving solution by using a 0.45-micrometer microporous membrane to obtain a fructus cnidii extract;
(5) preparing a hydrogel: sequentially adding propylene glycol and antibacterial peptide in parts by weight in a preparation container, stirring until the volume is increased and transparent, adding mannitol, glycerol, sodium carboxymethylcellulose and ethylparaben, stirring uniformly, standing for 15-20 min, sequentially adding glycol amine, borneol, water-based lubricating powder and distilled water, and stirring uniformly to obtain hydrogel;
(6) preparing a traditional Chinese medicine extract mixture: uniformly mixing the traditional Chinese medicine extracts obtained in the steps (1) - (4) for later use;
(7) preparing a gel dressing: adding the traditional Chinese medicine extract mixture into the hydrogel according to the parts by weight, uniformly stirring, then adding the anthocyanin, the carrageenan and the natural balance factor, mixing and stirring, and uniformly stirring to obtain the gel dressing.
4. The method for preparing a gel dressing for preventing and inhibiting HPV viral infection according to claim 1, wherein the method comprises the following steps: and (7) after uniformly stirring, irradiating and disinfecting the gel dressing by adopting an ultraviolet irradiation disinfection mode, wherein the irradiation time is 30-60 min.
