本发明公开了一种羧酸乙酯的制备方法,将五氟苯甲酰乙酸乙酯与原甲酸三甲酯在醋酐催化作用下发生取代反应,反应完成减压回收溶剂,减压蒸干后与环丙胺继续发生亲核取代,产物加入到甲苯溶剂中,以碳酸钠作为缚酸剂,脱去8号位上的氟原子生成喹啉酮结构,最后得到羧酸乙酯;本发明收率高,污染少,具有实际使用和推广价值。

本发明属于化合物合成技术领域,具体是一种羧酸乙酯的制备方法。
背景技术
1-环丙基-5,6,7,8-四氟-1,4-二氢-4-氧代-3-喹啉羧酸乙酯是一种重要医药中间体,主要用于奥比沙星的合成,奥比沙星一种新型动物专用氟喹诺酮类抗菌药,奥比沙星合成路线已有文献报道,但是有关此中间体的合成报道较少。目前的合成方法中反应步骤较长,所用试剂价格昂贵不易得,因此无法在工业化生产上广泛应用。
发明内容
本发明的目的就是提供一种收率高,污染少的羧酸乙酯的制备方法。
本发明将五氟苯甲酰乙酸乙酯与原甲酸三甲酯在醋酐催化作用下发生取代反应,反应完成减压回收溶剂,减压蒸干后与环丙胺继续发生亲核取代,产物加入到甲苯溶剂中,以碳酸钠作为缚酸剂,脱去8号位上的氟原子生成喹啉酮结构,最后得到羧酸乙酯。
本发明收率高,污染少,具有实际使用和推广价值。

1.一种羧酸乙酯的制备方法,其特征在于:将五氟苯甲酰乙酸乙酯与原甲酸三甲酯在醋酐催化作用下发生取代反应,反应完成减压回收溶剂,减压蒸干后与环丙胺继续发生亲核取代,产物加入到甲苯溶剂中,以碳酸钠作为缚酸剂,脱去8号位上的氟原子生成喹啉酮结构,最后得到羧酸乙酯。
