背景技术
黄蜀葵花是一种多功能的中药,成分复杂,药用广泛。黄蜀葵花中含有杨梅素具有强效的杀菌消炎的作用,因此黄蜀葵花在治疗口腔溃疡和外伤感染过程中效果极佳,黄蜀葵花中还含有大量酮类,具有极强的抗氧化性,能有效清除机体内部自由基,缓解机体的衰老并对慢性肾炎有很强的治疗效果,黄蜀葵花中黄酮物质的提取以及应用已称为近年研究的热点。
黄蜀葵花在国内具有广泛分布,但其往往作为一味中药进行使用,较少应用于保健品领域。目前公开的采用黄蜀葵花成分的保健饮品,对黄蜀葵花有效成分尤其是黄酮物质利用率低。
微胶囊作为一种包埋技术已成为在食品加工领域里的热门技术之一,微胶囊技术在食品以及保健品方面的应用在很大程度上推动了食品和保健品生产、加工工业由低级向高级产业进行转变,微胶囊包埋技术使用的壁材大多数是一些无毒、稳定、通透性较强、能够成膜的物质,如海藻酸钠、阿拉伯胶、各种糊精等,通过对所需要包埋的材料进行包埋,使其稳定性能得到较大的提升,此外还能够保护所需的物质避免受到外界因素的影响,延长功效作用时间。

一种黄蜀葵花黄酮保健品,以质量份计,由以下成份组成:玉米微孔淀粉10~25份、黄蜀葵花黄酮物质提取物5~12.5份、β-环糊精5~15份、甜叶菊0.5~1.5份。
黄蜀葵花黄酮微胶囊制备步骤如下:
1)制备玉米微孔淀粉;2)提取黄蜀葵花黄酮物质提取物;3)将制得的玉米微孔淀粉与黄蜀葵花黄酮提取物按照3:1的质量比例进行混匀搅拌,搅拌均匀后加入质量分数为 40 %的β-环糊精溶液;4)继续搅拌待表面的乙醇溶液挥发后加入一定份数甜叶菊即可得到颗粒状的淡黄色黄蜀葵花黄酮微胶囊。

1)准确称取一定质量的玉米淀粉,向其中加入一定量的柠檬酸缓冲液,制成PH=5~6的淀粉混浆;2)将淀粉混浆放入水浴锅中加热,水浴温度为55~58°C,待温度到达后加热 12~17min;3)加入α淀粉酶和糖化酶在超声波清洗器中反应一定时间;反应完成后加入1mol/L的 NaOH溶液终止反应;4)使用抽滤机将浆液进行抽滤,收集淀粉,分别再用水和乙醇洗涤抽滤;5)抽滤后的产物放进烘箱中烘干,干燥的产物过80目筛,即制得玉米微孔淀粉。
其中黄蜀葵花黄酮提取物的提取过程如下:
1)称取一定质量的黄蜀葵花粉末放入锥形瓶中,加入 85% 的乙醇后进行超声提取,收集上层提取液,再向残渣中加入体积比为 85%的乙醇 进行再次超声提取,合并提取液;将提取液放入离心机内,以 4000 r/min的转速离心,时间为15~20min,收集上清液;2)将上清液减压浓缩回收乙醇,待溶液浓缩至原来体积 2~5% ;将所得膏状物冷冻干燥得到干燥固体粉末,即为黄蜀葵花黄酮物质提取物。
优选的玉米微孔淀粉的制备步骤中α淀粉酶和糖化酶与玉米淀粉的质量比为1:100,其中α淀粉酶和糖化酶用量比例为1:1;
优选的玉米微孔淀粉的制备步骤中超声波清洗器温度为55°C,超声功率为110W,超声反应时间为35min。
优选的黄蜀葵花黄酮提取物的提取步骤中黄蜀葵花粉末与85% 的乙醇料液比例为1:30,优选的黄蜀葵花黄酮提取物的提取步骤中超声提取功率为100W提取温度、55°C,提取时间30min。
优选的黄蜀葵花黄酮提取物的提取步骤中离心机离心时间20min。
发明有益效果:
1、本发明提供的一种黄蜀葵花黄酮保健品及其微胶囊制备方法,为高效利用黄蜀葵花资源提供了一种新的途径。可充分利用黄蜀葵花黄酮具有抗氧化、消炎杀菌、保护心脑血管系统,缓解疲劳等保健作用,服务于人民大众。使用微胶囊技术对黄蜀葵花黄酮进行包埋,在不添加防腐剂的情况下增加了黄酮的保存时间和稳定性。
2、本发明中对黄蜀葵花黄酮物质的提取采用优化后的超声辅助乙醇提取法,与传统的双水相提取法和微波提取法相比具有提取率高、操作简单、提取成本低等有点适用于大规模工业生产。
3、通过本发明制备方法制得的黄蜀葵花黄酮微胶囊具有包埋率高,操作简便。本发明制备的黄蜀葵花黄酮微胶囊可在人体内长时间多次发挥作用,避免了一次摄入过多黄酮物质对机体的损害,适之更适合老年人服用。

图1所示为黄蜀葵花黄酮微胶囊在不同温度下的稳定性曲线图,结果显示黄蜀葵花黄酮微胶囊在一定温度范围内可以保持稳定;
图2所示为黄蜀葵花黄酮微胶囊的缓释曲线图,结果显示黄蜀葵花微胶囊可在人体内持续发挥作用,有效避免一次摄入过多黄酮对人体的危害。
具体实施方式
下面通过具体实施方式,结合附图对本发明技术方案做进一步的详细描述。
实施例1
本发明黄蜀葵花黄酮保健品,以质量份计,包括玉米微孔淀粉10~25份、黄蜀葵花黄酮物质提取物5~12.5份、β-环糊精5~15份、甜叶菊0.5~1.5份。
实施例2
本实施例的黄蜀葵花黄酮保健品,含有玉米微孔淀粉21份,黄蜀葵花中黄酮物质提取物7份;β-环糊精7份、甜叶菊1份。
实施例3
本实施例的黄蜀葵花黄酮保健品,含有玉米微孔淀粉15份,黄蜀葵花中黄酮物质提取物5份;β-环糊精6份。
实施例4
本实施例的黄蜀葵花黄酮保健品,含有玉米微孔淀粉24份,黄蜀葵花中黄酮物质提取物7份;β-环糊精7份、甜叶菊1.5份。
实施例5
本实施例的黄蜀葵花黄酮保健品,含有玉米微孔淀粉10份,黄蜀葵花中黄酮物质提取物12.5份;β-环糊精10份。
实施例6
本实施例的黄蜀葵花黄酮保健品,含有玉米微孔淀粉20份,黄蜀葵花中黄酮物质提取物10份;β-环糊精12份、甜叶菊0.5份。
实施例7
本实施例为黄蜀葵花黄酮保健品微胶囊制备方法,包含步骤如下:
1)制备玉米微孔淀粉;
2)提取黄蜀葵花黄酮物质提取物;
3)将制得的玉米微孔淀粉与黄蜀葵花黄酮提取物按照3:1的质量比例进行混匀搅拌,搅拌均匀后加入质量分数为 40 %的β-环糊精溶液;
4)继续搅拌待表面的乙醇溶液挥发后加入一定份数甜叶菊即可得到颗粒状的淡黄色黄蜀葵花黄酮微胶囊。
其中,玉米微孔淀粉由以下步骤制得:
1)准确称取一定质量的玉米淀粉,向其中加入一定量的柠檬酸缓冲液,制成PH=5~6的淀粉混浆;

本实施例所述的黄蜀葵花黄酮保健品微胶囊制备方法,与实施例7的区别在于,进一步的:α淀粉酶和糖化酶与玉米微孔淀粉的质量比为1:100,其中α淀粉酶和糖化酶用量比例为1:1;超声波清洗器温度为54~57°C,超声功率为95~110W,超声反应时间为30~40min。
实施例9
本实施例的黄蜀葵花黄酮保健品微胶囊制备方法,与实施例7或实施例8不同的是:黄蜀葵花黄酮提取物的提取过程如下:
1)称取一定质量的黄蜀葵花粉末放入锥形瓶中,加入 85% 的乙醇后进行超声提取,收集上层提取液,再向残渣中加入体积比为 85%的乙醇进行再次超声提取,合并提取液;将提取液放入离心机内,以 4000 r/min的转速离心,时间为15~20min,收集上清液;
2)将上清液减压浓缩回收乙醇,待溶液浓缩至原来体积 2~5%;将所得膏状物冷冻干燥得到干燥固体粉末,即为黄蜀葵花黄酮物质提取物。
其中,黄蜀葵花粉末与85% 的乙醇料液比例为1:30,超声提取功率为95~105W,提取温度48~55°C,提取时间25~35min。离心机离心时间为15~20min。
实施例10
本实施例公开了一种黄蜀葵花黄酮保健品及其微胶囊的实验室制备方法,所述的黄蜀葵花黄酮保健品,其原料组分为:玉米微孔淀粉60g,黄蜀葵花中黄酮物质提取物20g、β-环糊精20g。
制备方法如下:
取上述配方量的玉米微孔淀粉、黄蜀葵花黄酮提取物和β-环糊精,将玉米微孔淀粉、黄蜀葵花黄酮提取物混匀搅拌。搅拌均匀后加入质量分数为40 %的 β-环糊精溶液 。继续搅拌待表面的乙醇溶液挥发后即可得到的颗粒状的淡黄色颗粒即为黄蜀葵花黄酮微胶囊。
通过本法制得的黄蜀葵花黄酮微胶囊具有包埋率高,稳定性高,操作简便等优点。如图1所示,黄蜀葵花黄酮微胶囊在一定温度范围内其稳定性可以保持不变。
实施例11
一种黄蜀葵花黄酮保健品原料组分为:玉米微孔淀粉40g,黄蜀葵花中黄酮物质提取物14g、β-环糊精15g、甜叶菊2g。制备方法操作步骤如实施例10所述。
通过本法制得的黄蜀葵花黄酮微胶囊滋味微甜,使用代糖甜叶菊替换蔗糖,在增加风味的同时使之更适合糖尿病、高血糖患者服用。
实施例12
一种黄蜀葵花黄酮保健品原料组分为:其原料组分为:玉米微孔淀粉30g,黄蜀葵花中黄酮物质提取物8g、β-环糊精9g、甜叶菊1.5g。操作步骤如实施例10所述。
本实施例黄蜀葵花黄酮保健品,减少了黄酮提取物的添加含量,结合微胶囊缓释性的特点使得本配方更适合对消化功能底下或老年人服用,如图2所示,黄蜀葵花微胶囊在人体内随着摄入时间的延长可持续发挥作用,可有效避免一次摄入过多黄酮造成对人体的危害。
Claims (10)
1.一种黄蜀葵花黄酮保健品,其特征在于:以质量份计,包括玉米微孔淀粉10~25份、黄蜀葵花黄酮物质提取物5~12.5份、β-环糊精5~15份、甜叶菊0.5~1.5份。
2.根据权利要求1所述的黄蜀葵花黄酮保健品,其特征在于:含有玉米微孔淀粉21份,黄蜀葵花中黄酮物质提取物7份;β-环糊精7份、甜叶菊1份。
3.根据权利要求1所述的黄蜀葵花黄酮保健品,其特征在于:含有玉米微孔淀粉15份,黄蜀葵花中黄酮物质提取物5份;β-环糊精6份。
4.根据权利要求1所述的黄蜀葵花黄酮保健品,其特征在于:含有玉米微孔淀粉24份,黄蜀葵花中黄酮物质提取物7份;β-环糊精7份、甜叶菊1.5份。
5.一种权利要求1所述黄蜀葵花黄酮保健品微胶囊制备方法,包含步骤如下:
1)制备玉米微孔淀粉;
2)提取黄蜀葵花黄酮物质提取物;
3)将制得的玉米微孔淀粉与黄蜀葵花黄酮提取物按照3:1的质量比例进行混匀搅拌,搅拌均匀后加入质量分数为 40 %的β-环糊精溶液;
继续搅拌待表面的乙醇溶液挥发后加入一定份数甜叶菊即可得到颗粒状的淡黄色黄蜀葵花黄酮微胶囊。
6.根据权利要求5所述的黄蜀葵花黄酮保健品微胶囊制备方法,其特征在于:玉米微孔淀粉由以下步骤制得:
准确称取一定质量的玉米淀粉,向其中加入一定量的柠檬酸缓冲液,制成PH=5~6的淀粉混浆;
将淀粉混浆放入水浴锅中加热,水浴温度为55~58°C,待温度到达后加热 12~17min;

7.根据权利要求6所述的黄蜀葵花黄酮保健品微胶囊制备方法,特征在于:α淀粉酶和糖化酶与玉米微孔淀粉的质量比为1:100,其中α淀粉酶和糖化酶用量比例为1:1;超声波清洗器温度为54~57°C,超声功率为95~110W,超声反应时间为30~40min。
8.根据权利要求5、6或7所述的黄蜀葵花黄酮保健品微胶囊制备方法,特征在于:黄蜀葵花黄酮提取物的提取过程如下:
