本发明提供一种用于尿酸定量检测的干片试剂,包括外壳,所述外壳内依次贴合有扩散层、试剂层、树脂层、基材,所述外壳上表面中心位置开有样品滴加孔,底部中心位置开有透光孔,所述扩散层、试剂层、树脂通过涂布工艺从上到下依次分层贴合在基材上,所述该扩散层主要由二氧化钛、醋酸纤维素组成,所述试剂层主要包括尿酸酶、过氧化物酶、色素、缓冲液、交联剂、凝胶、酶保护剂。通过本发明,以解决现有技术存在的湿化学检测试剂不稳定、测试结果误差较大的问题。
Description
技术领域
本发明涉及一种尿酸检测试剂,具体涉及一种用于尿酸定量检测的干片试剂。
背景技术
所谓的“干片试剂”是传统的“湿化学“相对比较而言的。它是以被检测样品中的液体作为反应媒介,待测物直接与固定于载体上的试剂反应的一种方式,它与传统湿化学的最大区别就在于参与化学反应的媒介不同。随着生物化学中的酶的分离,提纯,储存等技术的发展,传感器,光度计,和电极技术的进步,以及计算机的普及,干化学也迅速发展起来。
干式化学相对湿式化学有其独特的优越性,就试剂而言,干化学试剂无需做测前准备工作,无需定标,试剂稳定,可直接测全血;就仪器而言无需给排水系统,无需配备纯水系统,操作简单,维护容易。
发明内容
本发明提供一种用于尿酸定量检测的干片试剂,以解决现有技术存在的湿化学检测试剂不稳定、测试结果误差较大的问题。
为解决上述技术问题,本发明提供一种用于尿酸定量检测的干片试剂,包括外壳,所述外壳内依次贴合有扩散层、试剂层、树脂层、基材,所述外壳上表面中心位置开有样品滴加孔,底部中心位置开有透光孔,所述扩散层、试剂层、树脂通过涂布工艺从上到下依次分层贴合在基材上,所述该扩散层主要由二氧化钛、醋酸纤维素组成,所述试剂层主要包括尿酸酶、过氧化物酶、色素、缓冲液、交联剂、凝胶、酶保护剂。
所述树脂层是水性环氧树脂。
所述基材是pet材料,用于提供支撑试剂的骨架,所述基材的透明度大于80%。
所述外壳是塑料材质,用于保护试剂在运输、储存、测试的过程中由于各种外力导致的形变,从而避免试剂的损坏。
所述扩散层以醋酸纤维素为立体网络骨架,立体网络骨架中包裹着相应的试剂。
试剂层是以凝结干燥的明胶为载体包裹着相应的试剂。
本发明带来的有益效果:本发明提供一种建立在湿化学基础上,用于测试尿酸的干化学方法的干片试剂,该干片试剂性能优越,储存时间长,测试结果准确,可显著提高尿酸在检测和存储过程中的各性能指标,弥补湿化学的不足。

为使本发明的目的、技术方案和优点更加清楚,以下结合附图及具体实施例,对本发明作进一步地详细说明。
本发明提供一种建立在湿化学基础上,用于测试尿酸的干化学方法的干片试剂,可显著提高尿酸在检测和存储过程中的各性能指标,弥补湿化学的不足。
如图1所示,本试剂干片包括:
扩散层2,该扩散层包括二氧化钛、醋酸纤维素等必要材料,主要作用包括均匀扩散样本,过滤样本中大分子物质和干扰物质。扩散层2是以醋酸纤维素为立体网络骨架,立体网络骨架中包裹着二氧化钛等相应的试剂,结构类似海绵里包裹着二氧化钛等相应的试剂。每片干片中扩散层各成分的含量如下表1所述:

试剂层3,该试剂层主要包括尿酸酶,过氧化物酶,色素,缓冲液,交联剂,凝胶,酶保护剂等必要生化试剂。试剂层是以凝结干燥的明胶为载体包裹着相应的试剂,结构类似果冻;每片干片中试剂层各成分的含量如表2所述:

树脂层4,该树脂层可以有效的使试剂层附着在基材上,可以降低对基材的材质和性能的要求,可使用未作任何处理的基材,避免处理基材对检测试剂的干扰,树脂层是水性环氧树脂,结构类似普通的液体胶,原厂采购直接使用。
基材5,该基材主要提供支撑试剂的骨架,基材的透明度必须大于80%,基材是pet材料,结构类似硬质塑料纸片。
外壳6,该外壳层主要是保护试剂在运输,储存,测试的过程中由于各种力导致的形变,从而避免试剂的损坏,尤其是在上机测试中,无论是自动或半自动的将干片试剂送入检测机器中,干片不可避免的受到相应的力,试剂外壳可以避免试剂干片的破损。外壳6是塑料,干片试剂的整体结构如图2所示。
本发明干片试剂制备方法如下:
准备树脂材料涂敷在基材上;
在树脂上涂敷试剂,试剂层根据需要和工艺有不同的涂敷方法;
在试剂上涂敷扩散层。
综上所述,本发明是将试剂以薄膜的形式涂附在透明塑料薄膜上制作成干片,通过检测反射光得出对应目标物质浓度的干片试剂。通过该方法制作可以提供性能优越,储存时间长的干化学试剂。
该干片试剂检测的具体操作如下:如图1所示将10ul待测样品1加入扩散层表面,样品1通过扩散层会横向、纵向同时均匀的向下扩散,进入试剂层,样品中的目标物质尿酸与干片内的相应物质反应并产生颜色变化,反应方程式如表3所示,反应过程具体来说是,样本中的尿酸在尿酸酶的催化下,尿酸被氧化产生过氧化氢和尿囊素。随后过氧化物酶催化染剂前体进行氧化联结反应,生成一种染剂。该染剂的强度通过反射光测定,其强度与血尿液中葡萄糖的浓度成正比例关系。加样后用760nm波长的光打向基材中心的位置,并记录加样后干片反色光的变化,反色光密度与目标物质尿酸的浓度层反比,通过计算机算出对应的浓度即样本对应尿酸的检测浓度。

1.一种用于尿酸定量检测的干片试剂,其特征在于,包括外壳(6),所述外壳内依次贴合有扩散层(2)、试剂层(3)、树脂层(4)、基材(5),所述外壳(6)上表面中心位置开有样品滴加孔,底部中心位置开有透光孔,所述扩散层(2)、试剂层(3)、树脂(4)通过涂布工艺从上到下依次分层贴合在基材上,所述该扩散层(2)主要由二氧化钛、醋酸纤维素组成,所述试剂层(3)主要包括尿酸酶、过氧化物酶、色素、缓冲液、交联剂、凝胶、酶保护剂。
2.如权利要求1所述的用于尿酸定量检测的干片试剂,其特征在于,所述树脂层(4)是水性环氧树脂。
3.如权利要求1所述的用于尿酸定量检测的干片试剂,所述基材(5)是pet材料,用于提供支撑试剂的骨架,所述基材的透明度大于80%。
4.如权利要求1所述的用于尿酸定量检测的干片试剂,所述外壳(6)是塑料材质,用于保护试剂在运输、储存、测试的过程中由于各种外力导致的形变,从而避免试剂的损坏。
