本发明属于癌症治疗的领域。它属于用于癌症治疗的毒素的领域。它属于微囊藻毒素和节球藻毒素以及它们在治疗疾病例如癌症、代谢性疾病以及其它应用中的用途的领域。本发明总体上涉及分子生物学、药学和生物技术的领域,和更具体地涉及能够靶向例如肿瘤的修饰的微囊藻毒素和节球藻毒素的合成。
背景
微囊藻毒素为由蓝细菌(也称为蓝绿藻)天然产生的毒素。当过多的蓝细菌在湖泊或池塘中生长时,它们形成藻华,藻华通常表现为一层绿色浮渣。然而,并非湖上所有的绿色浮渣均为藻华,也不是所有的藻华均含有产生微囊藻毒素的蓝细菌种类。微囊藻毒素同类物很多,微囊藻毒素-LR为其中毒性更强且经过充分研究的同类物之一。微囊藻毒素为由许多蓝细菌属产生的一组环状七肽肝毒素。其中最著名和同名的为广泛的微囊藻属。在结构上,大多数微囊藻毒素由一般结构环(-D-Ala1-X2-D-MeAsp3-Y4-Adda5-D-Glu6-Mdha7-)组成。X和Y为可变L-氨基酸,D-MeAsp为D-赤型-β-甲基天冬氨酸和Mdha为N-甲基脱氢丙氨酸。然而,尽管X和Y为最可变的氨基酸,但在微囊藻毒素核心结构的所有位置均可发现变化(见图1)。Adda为蓝细菌独特的C20-β-氨基酸3-氨基-9-甲氧基-2,6,8-三甲基-10-苯基-癸-4,6-二烯酸。2和4位可变L-氨基酸的取代以及在其它组成氨基酸中较不经常发现的改变导致迄今为止100多种报道的天然微囊藻毒素。
微囊藻毒素为1型和2A型蛋白磷酸酶的有效抑制剂。例如,微囊藻毒素-LR对于1型和2A型蛋白磷酸酶的IC50分别为0.03 nM和0.04 nM。
蛋白磷酸酶1和2A为真核细胞中两种主要的磷酸酶,可将丝氨酸和苏氨酸残基去磷酸化。
蛋白磷酸酶2B被有效抑制1000倍,而6种其它测试的磷酸酶和8种测试的蛋白激酶不受影响。
节球藻毒素为与微囊藻毒素结构相关的化合物,因为它们是从微囊藻毒素起源进化而来的,并且还含有在微囊藻毒素中发现的氨基酸Adda。它们尤其是由节球藻属物种产生,并且与微囊藻毒素形成对比,它们为环状五肽,具有最常见的同类物环[-D-赤型-甲基Asp-L-Arg-Adda-D-Glu-Mdhb],其中Mdhb为N-甲基脱氢丁酸(见图1)。
微囊藻毒素和节球藻毒素可用作癌症药物。假设可分离出天然微囊藻毒素变体,它们相对于OATP1B1优先由OATP1B3型主动转运蛋白转运,以发展为具有临床上可耐受的肝毒性的抗癌剂(OATP1B3转运蛋白主要存在于癌组织,例如肝癌中)。已分离出微囊藻毒素变体,并测试了在用OATP1B1和OATP1B3转运蛋白稳定转染的癌细胞中的细胞毒性。发现了细胞毒性OATP1B1/OATP1B3 IC50比率在0.2-32之间范围(代表转运蛋白选择性的150倍范围)的微囊藻毒素变体。由于微囊藻毒素结构对转运蛋白的选择性具有显著影响,因此有可能开发出具有甚至更明显的OATP1B3选择性的类似物,从而使其能够开发成为抗癌药物。然而,更特异性的递送方法将为优选的。一种这样的方法涉及本文公开的新颖概念,即添加靶向部分。对于避免脱靶毒性的靶向和高度特异性癌症疗法而言,理想的是微囊藻毒素和节球藻毒素的结构变体将带有靶向部分(例如癌症特异性单克隆抗体),并且不是由所有OATP转运蛋白亚型转运或转运不良,或者仅由或主要由癌症特异性OATP 1B3亚型转运。
微囊藻毒素难以化学合成。一种获得微囊藻毒素的较便利的方法涉及通过蓝细菌体内产生微囊藻毒素。
学院团体的先前实验旨在通过供给在由所供给菌株合成的相应微囊藻毒素的至少一种结构变体中掺入的氨基酸来增加天然产生的非核糖体肽(此处为微囊藻毒素)的产物产量。更具体地讲,将氨基酸亮氨酸(L, Leu)或精氨酸(R, Arg)供给产生微囊藻毒素(MC)变体MC-LR和MC-RR (L代表亮氨酸,R代表精氨酸)的蓝细菌菌株取决于所供给的氨基酸而影响两种变体的产量。此外,供给在由所供给菌株合成的相应微囊藻毒素的至少一种结构变体中掺入的氨基酸也可影响生物量的产生。
另外,还已经表明,供给代表天然掺入由所供给菌株产生的相应非核糖体肽中的氨基酸的略微修饰形式的氨基酸,也可掺入相应非核糖体肽中。该方法通常称为突变合成。然而,对于蓝细菌非核糖体肽,迄今为止,该方法仅限于天然氨基酸的简单类似物,比如高酪氨酸代替酪氨酸(仅相差一个亚甲基)或卤化氨基酸(仅相差一个卤素原子)比如氯酪氨酸代替酪氨酸。迄今为止尚未报道经较广泛修饰的氨基酸或与天然掺入非核糖体肽中的氨基酸不同的氨基酸及其类似物的供给。此外,在文献中已经描述了不可能供给修饰的氨基酸以掺入微囊藻毒素。
对修饰的微囊藻毒素和节球藻毒素存在需要。对产生修饰的微囊藻毒素和节球藻毒素以及将微囊藻毒素和节球藻毒素偶联至靶向单元(例如,结合用于癌症靶向疗法的抗体-药物缀合物的构建)的方法存在需要。
发明概述
通过将一种或多种修饰的底物掺入微囊藻毒素和/或节球藻毒素中来产生修饰的微囊藻毒素和节球藻毒素,该问题得到解决。
本发明涉及一种修饰的微囊藻毒素和/或节球藻毒素化合物,其包含一种或多种修饰的底物,其中至少一种修饰的底物包含直接可及或可转化以用于点击化学的锚定基团,用于连接靶向部分和/或标记和/或用于另外的结构修饰。
定义

在本文中,节球藻毒素为由环[-D-赤型-甲基Asp (异键)-L-Arg-Adda-D-Glu (异键)-Mdhb]组成的单环五肽的化合物,其中Mdhb代表N-甲基脱氢丁酸和Adda为特定的C20-氨基酸:3-氨基-9-甲氧基-2,6,8-三甲基-10-苯基癸-4,6-二烯酸,而所有位置均可天然略微修饰,如图1所示。节球藻毒素在结构和生物学活性方面非常类似于微囊藻毒素。
微囊藻毒素和节球藻毒素的修饰不应发生于Adda和D-Glu的位置处,因为这两个位置对于针对PP1和PP2A的抑制活性至关重要。

在本文中,靶向部分为蛋白质(主要是抗体及其片段)、肽、核酸(适体)、小分子或其它(维生素或碳水化合物)以及纳米颗粒。优选的是单克隆抗体(mAb)作为护航分子,用于靶向递送经改变和修饰的微囊藻毒素或节球藻毒素。然而,小分子也可充当靶向部分,因为它们可能影响所述肽的物理化学性质。对此的一个实例为与亲水性部分比如糖类偶联,以例如增加所述肽的水中溶解度。此外,连接的小分子可具有改变肽的体内药代动力学特性的目的,例如连接易于体内代谢的官能团可增加肝清除率和降低肝毒性,或者可影响转运蛋白的选择性,从而影响细胞对修饰的非核糖体肽的(主动)摄取。
在本文中,ADC (ADC代表抗体-药物缀合物)为直接或经接头(L)连接至靶向部分(TM)的CA,而根据ADC的定义,靶向部分为抗体。
本文中的术语抗体(AB)以最广义使用,并且具体地讲涵盖单克隆抗体、多克隆抗体、二聚体、多聚体、多特异性抗体(例如双特异性抗体)和抗体片段,只要它们呈现出期望的生物学活性即可。抗体可为鼠、人、人源化、嵌合的或衍生自其它物种。抗体为由免疫系统产生的能够识别并与特定抗原结合的蛋白。靶抗原通常具有被多种抗体上的互补决定区(CDR)识别的众多结合位点,也称为表位。与不同表位特异性结合的每种抗体具有不同的结构。因此,一种抗原可具有一种以上相应的抗体。抗体包括全长免疫球蛋白分子或全长免疫球蛋白分子的免疫活性部分,即含有免疫特异性地结合目标靶标的抗原或其部分的抗原结合位点的分子,这种靶标包括(但不限于)癌细胞、微生物细胞或产生与自身免疫性疾病相关的自身免疫抗体的细胞。免疫球蛋白可属于免疫球蛋白分子的任何类型(例如IgG、IgE、IgM、IgD和IgA)、类别(例如IgG1、IgG2、IgG3、IgG4、IgA1和IgA2)或子类。免疫球蛋白可源自任何物种,包括人、鼠或兔来源。
抗体片段(AB片段)包含全长抗体的一部分,通常为其抗原结合或可变区。抗体片段的实例包括Fab、Fab'、F(ab')2和Fv片段;双功能抗体;线性抗体;由Fab表达文库产生的片段、抗独特型(anti-Id)抗体、CDR (互补决定区)和以上任一项的表位结合片段(其与癌细胞抗原、病毒抗原或微生物抗原免疫特异性结合)、单链抗体分子;由抗体片段形成的多特异性抗体。
接头通过共价键将抗体或AB片段或靶向部分或标记连接于CA。接头为双功能或多功能部分,其可用于连接一个或多个药物部分(D,而D = CA)和抗体单元(Ab)以形成抗体-药物缀合物(ADC)。接头(L)在细胞外侧(即胞外)可为稳定的,或者其可通过酶活性、水解或其它代谢条件进行切割。抗体-药物缀合物(ADC)可使用具有用于与药物部分(在此为CA)和抗体结合的反应性官能团的接头便利地制备。在本文中,ADC为连接于靶向部分的CA。接头还可包括间隔子,其可具有在接头、药物和靶向部分之间获得有利空间距离的优势。
如下所述的抗体(AB)的半胱氨酸硫醇、胺例如N-末端或氨基酸侧链(比如赖氨酸)或任何其它修饰可与接头试剂、药物部分(D)或药物-接头试剂(DL)的官能团形成键。接头优选地在细胞外为稳定的。在转运或递送至细胞中之前,抗体-药物缀合物(ADC)优选地为稳定的并保持完整,即抗体保持与药物部分连接。接头在靶细胞外为稳定的,并且可在细胞内以一定速率切割。有效的接头将:(i) 维持抗体的特异性结合特性;(ii) 使得缀合物或药物部分能够细胞内递送;(iii) 保持稳定和完整,即不被切割,直至缀合物已被递送或转运至其靶部位;和(iv) 维持CA的细胞毒性、细胞杀伤作用或细胞抑制作用。ADC的稳定性可通过标准分析技术(比如质谱、HPLC和分离/分析技术LC/MS)进行测量。抗体和CA的共价连接需要接头具有两个反应性官能团,即反应性意义上的二价。已知可用于连接两个或更多个功能或生物活性部分(比如肽、核酸、药物、毒素、抗体、半抗原和报告基团)的二价接头试剂。
在另一个实施方案中,接头可被磺酸酯取代基或其它可增加试剂水溶性和促进接头试剂与抗体或CA的偶联反应、或者促进AB-L与D或D-L与AB的偶联反应的取代基取代,这取决于用于制备ADC的合成路线。抗体上的亲核基团包括(但不限于):(i) N-末端胺基,(ii) 侧链胺基,例如赖氨酸,(iii) 侧链硫醇基,例如半胱氨酸,和(iv) 在抗体被糖基化时的糖羟基或氨基。胺、硫醇和羟基为亲核性的,并且能够与接头部分、接头试剂和CA (=D)上的亲电子基团反应形成共价键,所述亲电子基团包括:(i) 活性酯,比如NHS酯、HOBt酯、卤甲酸酯和酰卤;(ii) 烷基和苄基卤化物,比如卤乙酰胺;(iii) 醛、酮、羧基和马来酰亚胺基团。某些抗体具有可还原的链间二硫键,即半胱氨酸桥。通过用还原剂比如DTT (二硫苏糖醇)处理,可使抗体具有与接头试剂缀合的反应性。因此,每个半胱氨酸桥理论上将形成两个反应性硫醇亲核体。通过赖氨酸与2-亚氨基硫杂环戊烷(Traut试剂)的反应导致胺转化为硫醇,可将另外的亲核基团引入到抗体中。可通过引入1、2、3、4或更多个半胱氨酸残基(例如制备包含一个或多个非天然半胱氨酸氨基酸残基的突变抗体),将反应性硫醇基团引入到抗体(或其片段)中。US 2007/0092940通过引入反应性半胱氨酸氨基酸来工程化抗体。
修饰的底物意指任何氨基酸和任何带有至少一个氨基和一个羧基的相关化合物,其使得在相应的非核糖体肽中肽结合形成修饰的底物,并且其天然不掺入由特定蓝细菌菌株合成的非核糖体肽中。
修饰的氨基酸或修饰的底物可包含氨基酸接头组分(包括天然存在的那些)以及次要氨基酸和非天然存在的氨基酸类似物(比如瓜氨酸)。可设计氨基酸接头组分并对其通过特定酶例如肿瘤相关蛋白酶、组织蛋白酶B、C和D或纤溶酶蛋白酶的酶促切割的选择性进行优化。氨基酸侧链包括天然存在的那些以及次要氨基酸和非天然存在的氨基酸类似物(比如瓜氨酸)。
发明详述
发明人首次将修饰的氨基酸掺入微囊藻毒素中,所述微囊藻毒素带有所谓的可点击锚定基团,使得能够快速且易于将整个分子与例如肽接头或其它功能单元(例如抗体)结合(见图4-10、15/16),或者所供给的底物带有对可点击锚定基团可易于进行另外修饰的官能团(见图11-18)。
显示供给可点击底物与例如在微囊藻毒素和/或节球藻毒素中天然存在的氨基酸、这些氨基酸的修饰形式或任何其它底物的任何组合可能潜在地导致将所供给的底物组合掺入非核糖体肽中(见图15-18)。
发明人首次显示成功供给的底物(例如修饰的氨基酸)在结构上不一定与天然掺入微囊藻毒素或节球藻毒素中的底物(例如相应的未修饰的氨基酸)直接相关(见图7/8、13/14、19-22)。这意味着在过去成功的供给仅被视为直接衍生自天然(自然)掺入的氨基酸的结构变体(例如o-甲基-酪氨酸或氯-酪氨酸代替酪氨酸或者高精氨酸代替精氨酸)。考虑到发明人的新结果,显而易见的是,如果还可使用结构上不是直接衍生自天然掺入相应的非核糖体肽中的底物和也不是天然掺入的底物被官能团(其不直接可及或可转化以用于缀合化学,包括点击化学,用于连接靶向部分或标记)取代的底物,则通过供给产生的非核糖体肽的结构和功能多样性显著增加。

在本发明的一个实施方案中,一种或多种修饰的底物在微囊藻毒素和/或节球藻毒素的确定的位置掺入。
CA可在CA中在其中在自然中氨基酸不存在的位置带有这样的氨基酸。氨基酸可以是修饰的。
令人惊讶地,发明人可修饰除了微囊藻毒素的所谓的可变位置2和4或节球藻毒素的相应的Arg2位置之外的多个位置。
本发明进一步涉及一种化合物,其中在除了Adda5和DGlu6以外的微囊藻毒素的任何位置掺入一种或多种修饰的底物,其具有以下一般结构:

而且,优选地如果CA是微囊藻毒素,修饰的位置为X2和Z4,并且修饰的底物为修饰的氨基酸(见图15-18)。
在节球藻毒素中,优选地,修饰的位置是Arg2和修饰的底物是修饰的氨基酸。
修饰的底物(优选地为修饰的氨基酸)优选地含有直接可及或可转化以用于缀合化学(包括点击化学)的锚定基团,用于连接靶向部分或标记或用于另外的结构修饰(见图4-22)。
在本发明的方法中,可点击底物的缀合化学反应(包括点击化学反应)选自包括以下的反应:铜(I)催化的叠氮化物-炔烃环加成、应变促进的叠氮化物-炔烃环加成、炔烃-叠氮化物环加成或炔烃-四嗪逆需求Diels-Alder反应。另外的缀合化学可选自利用伯胺、硫醇、醛、羧基和肟的特定反应性的反应。因此,至少一种修饰的底物的锚定基团,其直接可及而用于缀合化学(包括点击化学),用于连接靶向部分,可选自:
• 可随后例如通过与炔烃、活化的烯烃或膦反应进行修饰的叠氮基,而细胞毒素的叠氮基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与叠氮化物反应进行修饰的炔基(例如炔丙基或二芳基应变的环辛炔),而细胞毒素的炔基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与炔烃或烯烃反应进行修饰的四嗪,而细胞毒素的四嗪基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与异硫氰酸酯、异氰酸酯、酰基叠氮化物、NHS酯、磺酰氯、醛、乙二醛、环氧化物、环氧乙烷、碳酸酯、芳基卤化物、亚氨酸酯、碳二亚胺、酸酐、膦或氟苯基酯反应进行修饰的伯胺,而细胞毒素的氨基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与马来酰亚胺、卤乙酰基、吡啶基二硫化物、硫代磺酸酯或乙烯基砜反应进行修饰的硫醇,而细胞毒素的硫醇基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与胺、氨基硫醇、埃尔曼试剂、烷氧基胺、酰肼或硫醇反应进行修饰的醛,而细胞毒素的醛基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与碳二亚胺反应进行修饰的羧基,而细胞毒素的羧基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。
• 可随后例如通过与苯乙酮(比如对乙酰基苯丙氨酸)反应进行修饰的肟,而细胞毒素的肟基与接头、抗体或其它功能性分子(比如荧光染料或聚合物基质)的相应官能团反应。

在本发明的方法中,可点击底物的缀合化学反应(包括点击化学反应)选自铜(I)催化的叠氮化物-炔烃环加成、应变促进的叠氮化物-炔烃环加成、炔烃-叠氮化物环加成或炔烃-四嗪逆需求Diels-Alder反应。另外的缀合化学可选自利用伯胺、硫醇、醛、羧基和肟的特定反应性的反应。

然而,关于通过引入修饰的底物来修饰微囊藻毒素和节球藻毒素的CA,最优选的为微囊藻属(Microcystis)、浮丝藻属(Planktothrix)、颤藻属(Oscillatoria)、念珠藻属(Nostoc)、鱼腥藻属(Anabaena)、束丝藻属(Aphanizomenon)、软管藻属(Hapalosiphon)、节球藻属(Nodularia)。

一种方法用于治疗哺乳动物的癌症,其中癌症的特征为ErbB受体的过表达。哺乳动物任选地对用未缀合的抗ErbB抗体的治疗不反应或反应不良。方法包括给予哺乳动物治疗有效量的抗体-药物缀合物化合物。通过给予患者与所述生长因子受体和化学治疗剂特异性结合的本发明的ADC,可抑制过表达生长因子受体(比如HER2受体或EGF受体)的肿瘤细胞的生长,其中所述抗体-药物缀合物和所述化学治疗剂各自以在患者中有效抑制肿瘤细胞生长的量给予(见图29)。

本发明的进一步的实施方案是微囊藻毒素和/或节球藻毒素化合物,其包含一种或多种修饰的底物,其中一种或多种修饰的底物不直接衍生自天然掺入的底物,例如优选地,在自然中不掺入所述非核糖体肽的特定位置和也不是天然掺入的底物被官能团取代的氨基酸或修饰的氨基酸,所述官能团不直接可及或可转化以用于缀合化学,包括点击化学,用于连接靶向部分或标记。
实施例
在不同的培养系统和规模下进行修饰的底物的成功供给,使得能够筛选(小至10ml规模;见图2 A-D)和生产(2-20 L规模;见图24、25)修饰的非核糖体肽。不同的筛选规模包括:
在约2.2 ml深孔微量滴定板(dw-MTP)中培养1.6 ml培养物,同时通过600 rpm的剧烈振荡和在dw-MTP上方顶部空间中的5%恒定CO2浓度确保CO2供给。一天24小时经LED面板或经荧光灯泡进行照明。根据菌株及其生长阶段将光强度调整在35-250 µmol/s*m2之间。温度在20℃-30℃之间菌株特异性变化。
因此,该方法的培养为优选的,其中振荡在400-800 rpm之间和恒定CO2浓度为在顶部空间中1-10%,优选地在顶部空间中3-8%。
在40 ml聚苯乙烯管中培养10 ml培养物,同时通过250-350 rpm的剧烈振荡和培养容器下面的5%恒定CO2浓度来确保CO2供给。特此,经培养容器底部的可渗透CO2的聚丙烯膜将CO2引入到培养物中。一天24小时经荧光灯泡进行照明,并根据菌株及其生长阶段将光强度再次调整在35-250 µmol/s*m2之间。温度再次在20℃-30℃之间菌株特异性变化。
因此,该方法的培养为优选的,其中一天24小时经荧光灯泡进行照明,并且在20-450 µmol/s*m2之间。
在玻璃烧瓶中培养50 ml培养物,同时以5%恒定CO2浓度通过鼓泡确保CO2供给。经用磁力搅拌棒以100 rpm搅拌来混合培养物。一天24小时经荧光灯泡进行照明,并根据菌株和生长阶段将强度调整在35-250 µmol/s*m2之间。
另外,还以2 L-20 L之间的生产规模进行了供给实验,同时通过以0.5-5.0%的恒定CO2浓度鼓泡来确保CO2供给和混合。经荧光灯泡进行照明,并根据菌株和生长阶段将强度调整在35-250 µmol/s*m2之间。
任选地,在如上所述的天期间,于相同的光强度下,以16小时光照/8小时的昼夜循环进行培养。
任选地,用不同的光源(例如LED灯或硫等离子灯)和使用菌株特异性的光强度、CO2浓度、振荡/搅拌强度和培养基组成的变化进行培养。
10 ml规模的示例性供给方案:
根据之前确定的菌株特异性培养条件,所有菌株均在BG11培养基(见下文)中进行培养。
将细胞在弱光条件(30 µmol/s*m2)下的锥形烧瓶中,于25℃下和70 rpm的振荡器上预培养4天。
对于10 ml规模的供给实验,将细胞以750 nm处的光密度(OD750 nm)为0.5接种于约40 ml聚苯乙烯管中。培养基通过添加TES至培养基中的浓度为10 mM进行缓冲。任选地,添加DMSO至培养基中的浓度为1%。
接种时通过在培养基中添加相应的修饰的底物至浓度为10 mM开始供给培养物。通过每天供给另外的10 µM (第1-4天),每天添加的修饰的底物在4天之内保持恒定。或者,在接种之后的第1和第3天通过每一天在培养基中供给修饰的底物至浓度为30 µM来添加修饰的底物。每天通过测量750 nm处的光密度(OD 750 nm)来监测培养物的生长。通过向培养物中添加甲醇至最终浓度为20%来完成培养。随后使用C18改性的硅胶柱经标准固相提取程序进行提取。
对于上述其它规模,方案相似并且只是略微不同。例如,在2和20 L规模下,培养基并不总是被缓冲,并且由于生长速度较慢,培养时间再延长一周。此外,在一些情况下,为了提高修饰的非核糖体肽的产率,使用了增加量的所添加修饰的底物,直至300 µM培养基浓度(如果菌株耐受这种浓度的话)。

所有修饰的微囊藻毒素均可用两种MS方法检测到。然而,由培养基中未添加1%DMSO的供给所产生的大多数样品用MALDI-ToF-MS不能检测到,但用ESI-IT-ToF-MS可以。因此,建议使用DMSO进行小规模培养物(1-10 ml之间的培养物体积)筛选中的供给实验,尤其是如果修饰的非核糖体肽的MS检测为基于MALDI-ToF-MS的话。
