本发明公开了一种长效型玻尿酸制备工艺,具体包括如下步骤:S1)透明酸质提纯:将透明酸质原料放入搅拌机,搅拌状态下加入2~3倍于透明酸质原料重量的、纯度为90%以上的乙醇,再次取得沉淀物,按上述步骤重复多次提纯;最后将沉淀物粉碎后依次用无水乙醇、丙酮脱水,并在80℃以下真空干燥,制得纯透明质酸,并消毒;S2)解离:得到的纯玻尿酸沉淀物中加入解离液氯化钠溶液进行解离,搅拌至溶液中无颗粒物,按上述步骤重复多次解离,经过后处理得到低蛋白玻尿酸,并进行整体消毒;S3)混料:制得的低蛋白玻尿酸中加入0.1‑0.2倍于玻尿酸的甘露醇,混合,并消毒制得成品。本发明利用独创的生产提纯工艺,使玻尿酸的作用时间,较普通玻尿酸提高300%‑600%。

一种长效型玻尿酸制备工艺
技术领域
本发明涉及一种长效型玻尿酸制备工艺,具体涉及一种利用多次提纯技术、降低蛋白质含量来延长玻尿酸使用寿命的玻尿酸制备工艺。
背景技术
玻尿酸具有很好的吸水性,同时作为人体存在的物质,和人体存在较小的排异性,所以用于微整形领域,被当做填充材料展现出非常理想的效果。然而目前的玻尿酸由于技术水平所限制,用于填充和面部微调时候,其效果往往只能保持3个月时间。本发明提供一种长效型整形用玻尿酸的生产工艺,利用独创的生产提纯工艺,以及产品配方,使玻尿酸的作用时间,较普通玻尿酸提高300%---600%,解决了玻尿酸领域的技术瓶颈,具有较高的科技含量,和市场应用潜力。
发明内容
针对现有技术的上述缺陷,本发明提供一种利用多次提纯技术、降低蛋白质含量来延长玻尿酸使用寿命的玻尿酸制备工艺;利用独创的生产提纯工艺,以及产品配方,使玻尿酸的作用时间,较普通玻尿酸提高300%---600%,解决了玻尿酸领域的技术瓶颈,具有较高的科技含量,和市场应用潜力。
为了解决上述技术问题,本发明提供了如下的技术方案:
本发明一种长效型玻尿酸制备工艺,包括如下步骤:
S1)透明酸质提纯:将透明酸质原料放入搅拌机,搅拌状态下加入2~3倍于透明酸质原料重量的、纯度为90%以上的乙醇,再次取得沉淀物,按上述步骤重复多次提纯;最后将沉淀物粉碎后依次用无水乙醇、丙酮脱水,并在80℃以下真空干燥,制得纯透明质酸,并消毒;
S2)解离:得到的纯玻尿酸沉淀物中加入解离液氯化钠溶液进行解离,搅拌至溶液中无颗粒物,按上述步骤重复多次解离,经过后处理得到低蛋白玻尿酸,并进行整体消毒;
S3)混料:制得的低蛋白玻尿酸中加入0.1-0.2倍于玻尿酸的甘露醇和0.05-0.1倍于玻尿酸的利多卡因,混合,并消毒制得成品。
作为本发明的一种优选技术方案,步骤S2)中所述解离液的加入量为玻尿酸产量的150倍~200倍体积量;
作为本发明的一种优选技术方案,步骤S2)中所述氯化钠溶液的浓度≥0.35mol/L,优选为0.35mol/L~0.6mol/ L。
作为本发明的一种优选技术方案,步骤S1)中重复提纯至少三次。
作为本发明的一种优选技术方案,步骤S2)中重复解离至少两次。
作为本发明的一种优选技术方案,在步骤S1)、S2)、S3)中消毒方式为:放入消毒柜用臭氧消毒或采用包括紫外照射、Co60照射等其他消毒方法。
本发明一种长效型玻尿酸制备工艺具有以下优点:
1、交联技术:
将单项和双向交联技术相结合的制作工艺。
制造出三次元立体构造排列的高粘度的透明质酸。
2、使用经过三次提纯的高纯度的透明质酸为原料,增加安全性。
3、使用尖端的制作工艺,最大限度的降低了肉毒素和蛋白质的的含量。
4、经过精细的提纯工艺,最大限度的降低了BDDE的残留量,从而降低过敏等负作用。
5、添加了抗氧化成分甘露醇,预防玻尿酸被氧化,从而延长玻尿酸的持久时间。
6、立体灭菌+全链条独立灭菌(指的是在生产的每个环节都做消毒,而不是最后产品出来之后整体消毒),360度无死角灭菌工艺,减少了微生物的感染等危险因素。

本发明一种长效型玻尿酸制备工艺,包括以下步骤:
S1)透明酸质提纯:将透明酸质原料放入搅拌机,搅拌状态下加入2~3倍于透明酸质原料重量的、纯度为90%以上的乙醇,再次取得沉淀物,按上述步骤重复多次提纯;最后将沉淀物粉碎后依次用无水乙醇、丙酮脱水,并在80℃以下真空干燥,制得纯透明质酸,并消毒;
S2)解离:得到的纯玻尿酸沉淀物中加入解离液氯化钠溶液进行解离,搅拌至溶液中无颗粒物,按上述步骤重复多次解离,经过后处理得到低蛋白玻尿酸,并进行整体消毒;
S3)混料:制得的低蛋白玻尿酸中加入0.1-0.2倍于玻尿酸的甘露醇和0.05-0.1倍于玻尿酸的利多卡因,混合,并消毒制得成品。
进一步的,步骤S2)中所述解离液的加入量为玻尿酸产量的150倍~200倍体积量;
进一步的,步骤S2)中所述氯化钠溶液的浓度≥0.35mol/L,优选为0.35mol/L~0.6mol/ L。
进一步的,步骤S1)中重复提纯至少三次。
进一步的,步骤S2)中重复解离至少两次。
进一步的,在步骤S1)、S2)、S3)中消毒方式为:放入消毒柜用臭氧消毒或采用包括紫外照射、Co60照射等其他消毒方法。
在本发明中,对原料进行多次提纯,形成高纯度的透明质酸,从而保证了原料的使用安全性。经过精细的提纯工艺,最大限度的降低了BDDE的残留量,从而降低过敏等负作用。
并且添加了适量的抗氧化成分甘露醇和利多卡因,预防玻尿酸被氧化,从而延长玻尿酸的持久时间,并能减少治疗时的痛苦。
同时选取合适的解离次数,使制得的玻尿酸中的蛋白质含量明显降低,达到现行最高质量标准要求的蛋白质含量控制方法,使制得的玻尿酸原料药中的蛋白质含量低于0.05wt%。最大限度的降低了蛋白质的的含量。
最后采用了立体化的消毒,在每一生产工艺中加入消毒步骤,形成独立、全链条独立灭菌,减少了微生物的干扰等危险因素。
本发明具有如下优点:
1、交联技术:

Claims (6)

1.一种长效型玻尿酸制备工艺,其特征在于,包括以下步骤:
S1)透明酸质提纯:将透明酸质原料放入搅拌机,搅拌状态下加入2~3倍于透明酸质原料重量的、纯度为90%以上的乙醇,再次取得沉淀物,按上述步骤重复多次提纯;最后将沉淀物粉碎后依次用无水乙醇、丙酮脱水,并在80℃以下真空干燥,制得纯透明质酸,并消毒;
S2)解离:得到的纯玻尿酸沉淀物中加入解离液氯化钠溶液进行解离,搅拌至溶液中无颗粒物,按上述步骤重复多次解离,经过后处理得到低蛋白玻尿酸,并进行整体消毒;
S3)混料:制得的低蛋白玻尿酸中加入0.1-0.2倍于玻尿酸的甘露醇和0.05-0.1倍于玻尿酸的利多卡因,混合,并消毒制得成品。
2.根据权利要求1所述的一种长效型玻尿酸制备工艺,其特征在于,步骤S2)中所述解离液的加入量为玻尿酸产量的150倍~200倍体积量。
3.根据权利要求1-2所述的一种长效型玻尿酸制备工艺,其特征在于,步骤S2)中所述氯化钠溶液的浓度≥0.35mol/L,优选为0.35mol/L~0.6mol/ L。
4.根据权利要求1所述的一种长效型玻尿酸制备工艺,其特征在于,步骤S1)中重复提纯至少三次。
5.根据权利要求1所述的一种长效型玻尿酸制备工艺,其特征在于,步骤S2)中重复解离至少两次。
6.根据权利要求1所述的一种长效型玻尿酸制备工艺,其特征在于,在步骤S1)、S2)、S3)中消毒方式为:放入消毒柜用臭氧消毒或采用包括紫外照射、Co60照射等其他消毒方法。
