Abstract

本发明涉及一种止咳贴及其制备方法，所述止咳贴包括基材和涂布于基材上的浸膏，所述浸膏中的活性成分包括以下按重量份数计的组分：伊贝母提取物800～2000份、维生素D 0.06～0.2份、维生素C 40～100份、维生素B6 3～6份、维生素B12 0.04～0.08份。本发明将伊贝母提取物，与维生素D、维生素C、维生素B6、维生素B12选择合适的量结合在一起，制备成贴剂，用于治疗咳嗽，特别是可适合于治疗儿童咳嗽，其使用方便，解决伊贝母提取物作为药物不利于儿童口服的阻碍。

Description

一种止咳贴及其制备方法
技术领域
本发明属于医疗器械技术领域，特别涉及一种含伊贝母提取物的止咳贴及其制备方法。
背景技术
伊贝母(Bulbus fritillariae Pallidiflorae)为百合科植物新疆贝母或伊犁贝母的干燥鳞茎，主要分布于新疆伊犁河流域，伊贝母为药用贝母之一，是一种与川贝、浙贝齐名的贵重中药材。其味苦、甘；性微寒，归肺经，具有清热润肺、化痰止咳功效。伊贝母常与其他中药成分一起，制备用于止咳的中药制剂。本申请的发明人长期研究伊贝母这种物质，希望能开发更多、更有利于大众咳嗽用药，特别是适用于儿童的用药，进一步推广伊贝母的使用功效。
发明内容
基于此，本发明的目的之一是提供一种止咳贴，其使用方便，无任何副作用，特别适用于儿童。
实现上述目的的技术方案如下。
一种止咳贴，包括基材和涂布于基材上的浸膏，所述浸膏中的活性成分包括以下按重量份数计的组分：伊贝母提取物800～2000份、维生素D 0.06～0.2份、维生素C 40～100份、维生素B6 3～6份、维生素B12 0.04～0.08份。
在其中一些实施例中，还包括薄荷脑2～3份、纳米氧化铁1～2份、羟甲基纤维素钠6～12份。
在其中一些实施例中，所述浸膏中的活性成分包括以下按重量份数计的组分：伊贝母提取物800-1000份、维生素D 0.08-0.12份、维生素C 50-70份、维生素B6 3.5-4.5份、维生素B12 0.05-0.07份。
在其中一些实施例中，所述浸膏中的活性成分包括以下按重量份数计的组分：伊贝母提取物800份、维生素D 0.1份、维生素C 60份、维生素B6 4份、维生素B12 0.06份。
在其中一些实施例中，伊贝母提取物的制备方法如下：得到伊贝母细粉；伊贝母细粉用85％-96％的乙醇于80℃恒温水浴锅中回流提取5-7h，提取1-3次，抽滤，合并滤液，减压回收乙醇得浸膏，备用。
本发明的另一目的是提供上述的止咳贴的制备方法。
所述止咳贴的制备方法，包括以下步骤：
在伊贝母提取物中加入所述维生素D、维生素C、维生素B6、维生素B12，搅拌混匀，得伊贝母提取物混合物；加入贴剂赋形剂，制备成型。
在其中一些实施例中，所述赋形剂为薄荷脑、纳米氧化、和羟甲基纤维素钠。
在其中一些实施例中，将所述维生素D、维生素C、维生素B6、维生素B12、薄荷脑、纳米氧化铁、羟甲基纤维素钠加入到所述伊贝母浸膏中，搅拌均匀后，浓缩得稠浸膏，室温下相对密度为1.2；高压微射处理，取浸膏涂抹于贴布上，再在浸膏上贴上离型纸，封装即可。
本发明将伊贝母提取物，与维生素D、维生素C、维生素B6、维生素B12选择合适的量结合在一起，制备成贴剂，具有良好的止咳作用，特别是可适合于治疗儿童咳嗽，其使用方便，解决伊贝母提取物作为药物不利于儿童口服的阻碍。而且，发明人发现，伊贝母提取物与上述几种维生素能具有协同促进止咳的效果，能够促进伊贝母的止咳药效，在此基础上，制成膏贴剂，拓展伊贝母提取物在止咳中的进一步应用。
具体实施方式
除非另有定义，本发明所使用的所有的技术和科学术语与属于本发明的技术领域的技术人员通常理解的含义相同。本发明的说明书中所使用的术语只是为了描述具体的实施例的目的，不用于限制本发明。本发明所使用的术语“和/或”包括一个或多个相关的所列项目的任意的和所有的组合。
本发明以下实施例中所涉及的伊贝母提取物的制备方法如下：称伊贝母药材磨碎呈细粉，并经过筛网筛得颗粒均匀的药物粉末；取伊贝母粉适量置于圆底烧瓶中，加入95％乙醇500ml，于80℃恒温水浴锅中回流提取6h，提取3次，抽滤，合并滤液，减压回收乙醇得浸膏，提取率大约为50％，即1600克的药材得到800克的提取物，备用。
实施例1
本实施例所述止咳贴，它是由以下重量份的原料制成：伊贝母提取物800g、维生素D 0.1g、维生素C 60g、维生素B6 4g、维生素B12 0.06g，薄荷脑2.5g、纳米氧化铁1.5g、羟甲基纤维素钠8g。
所述止咳贴的制备如下：将所述维生素D、维生素C、维生素B6、维生素B12、薄荷脑、纳米氧化铁、羟甲基纤维素钠加入到伊贝母浸膏中，搅拌均匀后，浓缩得稠浸膏，相对密度为1.2；高压微射处理，取浸膏涂抹于贴布上，做成10个膏贴，再在浸膏上贴上离型纸，封装即可。
实施例2
本实施例所述止咳贴，它是由以下重量份的原料制成：伊贝母提取物1000g、维生素D 0.12g、维生素C 50g、维生素B6 5g、维生素B12 0.05g，薄荷脑2.5g、纳米氧化铁1.5g、羟甲基纤维素钠8g。
制备方法如实施例1。
实施例3：氨水引咳实验
1、动物给药及引咳
取昆明种小鼠40只，随机分为空白组、伊贝母提取物组、混合维生素组和实施例1中所述伊贝母提取物混合物(伊贝母提取物+混合维生物)组，每组10只，标记。其中混合维生素包括维生素D、维生素C、维生素B6、维生素B12，比例为0.1：60：4：0.06。各组分别按下表1剂量制作贴剂。给药前一天剪去小鼠腹部毛，将贴剂贴于小鼠腹部，每天1剂，连续7天，于末次给药后4小时开始实验，将小鼠置于引咳仪中，往雾化杯中加入25％浓氨水1ml，雾化15s(雾化速率0.15ml/min)，计数3min内的咳嗽数。
2、结果
各组的咳嗽次数和镇咳率见表1，用SPSS 19.0软件进行统计分析，实验数据以x±s表示,组间比较用t检验。P＜0.05为差异有统计学意义。

从上述结果中可以看出，伊贝母提取物组，咳嗽次数相对于空白组有显著差异，说明伊贝母提取物有良好的镇咳作用，镇咳率为36.6％；混合维生素组没有镇咳作用。混合维生素与伊贝母提取物混合(实施例1)后，小鼠的咳嗽次数显著减少，镇咳率显著提高，与空白组比较，差异更显著，实施例1伊贝母提取物混合物组与伊贝母提取物组比较，也有统计学的差异，说明维生素能够促进伊贝母提取物的止咳药效，该混合物表现出了更强的药效。
以上所述实施例仅表达了本发明的几种实施方式，其描述较为具体和详细，但并不能因此而理解为对发明专利范围的限制。应当指出的是，对于本领域的普通技术人员来说，在不脱离本发明构思的前提下，还可以做出若干变形和改进，这些都属于本发明的保护范围。因此，本发明专利的保护范围应以所附权利要求为准。

Claims (7)

1.一种止咳贴，其特征是，包括基材和涂布于基材上的浸膏，所述浸膏中的活性成分由以下按重量份数计的组分制备而成：伊贝母提取物800～2000份、维生素D 0.06～0.2份、维生素C 40～100份、维生素B6 3～6份、维生素B12 0.04～0.08份。
2.根据权利要求1所述的止咳贴，其特征是，所述浸膏中还包括薄荷脑2～3份、纳米氧化铁1～2份、羟甲基纤维素钠6～12份。
3.根据权利要求1所述的止咳贴，其特征是，所述浸膏中的活性成分由以下按重量份数计的组分制备而成：伊贝母提取物800-1000份、维生素D 0.08-0.12份、维生素C 50-70份、维生素B6 3.5-4.5份、维生素B12 0.05-0.07份。
4.根据权利要求3所述的止咳贴，其特征是，所述浸膏中的活性成分由以下按重量份数计的组分制备而成：伊贝母提取物800份、维生素D 0.1份、维生素C 60份、维生素B6 4份、维生素B12 0.06份。
5.根据权利要求1-4任一项所述的止咳贴，其特征是，所述伊贝母提取物的制备方法如下：取伊贝母细粉用85％-96%的乙醇于80℃恒温水浴锅中回流提取5-7h，提取1-3次，抽滤，合并滤液，减压回收乙醇得伊贝母提取物，备用。
6.权利要求1所述的止咳贴的制备方法，其特征是，包括以下步骤：
在所述伊贝母提取物中加入所述维生素D、维生素C、维生素B6、维生素B12，搅拌混匀，得伊贝母提取物混合物；加入贴剂赋形剂，制备成型。
7.根据权利要求6所述的止咳贴的制备方法，其特征是，所述赋形剂为薄荷脑、纳米氧化铁和羟甲基纤维素钠。
