The components are as follows: 18kg of astragalus membranaceus, 7kgg of epimedium herb, 4.5kg of glossy privet fruit, 0.04kgg of chromium picolinate, 0.028kg of zinc methionine, 0.03kg of lutein, 0.15kg of grape seeds, 0.15kg of morinda officinalis, 0.10kg of schisandra chinensis, 0.03kg of 1,25 dihydroxyvitamin D, 0.005kg of trace elements, 0.08kg of soybean lecithin and 0.1kg of chitosan nanoparticle suspension.
The trace elements are composed of calcium, iron, selenium and molybdenum according to a molar ratio of 2:1:1: 1.
The preparation method of the chitosan nanoparticle suspension comprises the following steps: dissolving water-soluble chitosan in 0.5% citric acid solution, adjusting pH to 6, and preparing into chitosan mixed solution with chitosan solution content of 2 mg/ml; taking a proper amount of sodium tripolyphosphate to prepare 1mg/ml sodium tripolyphosphate aqueous solution; then at 750--1Slowly dripping the sodium tripolyphosphate aqueous solution into the chitosan mixed solution at a rotating speed, carrying out ionic crosslinking and continuously stirring for 10-15min to prepare the chitosan nanoparticle suspension.
The preparation method of the premix for improving the sperm motility of the boars in summer comprises the following steps:
(1) pulverizing radix astragali, herba Epimedii, and fructus Ligustri Lucidi, decocting in 8-10 times of water, and filtering to obtain decoction A and residue A;
(2) mixing grape seed, radix Morindae officinalis, and fructus Schisandrae, pulverizing, adding into the decoction A, stirring at 50-60 deg.C for 30-40min, and filtering to obtain Chinese medicinal extractive solution A and residue B; mixing residue A and residue B, adding 0.12-0.15 wt% of compound microbial inoculum, fermenting at 25-30 deg.C for 20-24 hr, and sterilizing to obtain fermented product;
(3) adding trace elements into the Chinese medicinal extract A, adding 8-10 times of 70% methanol solution, mixing, reacting at 60-70 deg.C for 1-1.2 hr, and recovering methanol to obtain mixed extract A;
(4) adding 6-8 times of 80% ethanol into the fermented product, reflux extracting at 50-60 deg.C for 40-50min, filtering, recovering ethanol from the filtrate, and concentrating at 40-50 deg.C to obtain Chinese medicinal extract; mixing the Chinese medicinal extract and soybean phospholipid, adding 1-2 times of anhydrous ethanol, reacting at 45-50 deg.C under magnetic stirring for 1.5-2 hr, and recovering ethanol to obtain mixed extract B; wherein the magnetic stirring speed is 150-;
(5) mixing the mixed extract A and B, concentrating at 40-50 deg.C to obtain paste, spray drying to obtain powder, adding xanthophyll and chitosan nanoparticle suspension, mixing, and treating with ultrasonic at 25-30kHz for 3-5min to obtain mixture; and adding chromium picolinate and zinc methionine, and mixing uniformly to obtain the premix.
The composite microbial inoculum is a liquid microbial inoculum, which contains lactobacillus acidophilus, bacillus subtilis and trichoderma; the viable count of Lactobacillus acidophilus is 6-8 × 108cfu/ml, viable count of Bacillus subtilis is 4-6 × 106cfu/ml, viable count of Trichoderma 6-8 × 104cfu/ml。
Example 4
10kg of astragalus membranaceus, 5kg of epimedium herb, 5kg of glossy privet fruit, 0.03kg of chromium picolinate, 0.05kg of zinc methionine, 0.03kg of lutein, 0.01kg of grape seeds, 0.01kg of morinda officinalis, 0.01kg of schisandra chinensis, 0.002kg of 1,25 dihydroxyvitamin D, 0.003kg of trace elements, 0.06kg of soybean lecithin and 0.1kg of chitosan nanoparticle suspension.
The preparation method is the same as example 1.
Example 5
The components are as follows: 20kg of astragalus membranaceus, 10kg of epimedium herb, 3kg of glossy privet fruit, 0.05kg of chromium picolinate, 0.02-0.05 part of zinc methionine, 0.05kg of lutein, 0.015kg of grape seeds, 0.015kg of morinda officinalis, 0.008kg of schisandra chinensis, 0.004kg of 1,25 dihydroxyvitamin D, 0.005kg of trace elements, 0.08kg of soybean lecithin and 0.08kg of chitosan nanoparticle suspension.
The preparation method is the same as example 3.
Comparative example 1
The difference between the comparative example 1 and the example 1 is that no trace elements and soybean lecithin are added in the preparation process to react with the traditional Chinese medicine extracting solution A and the traditional Chinese medicine extract, but the traditional Chinese medicine extracting solution A and the traditional Chinese medicine extract are directly concentrated into paste, and then are spray-dried to prepare powder. The rest of the process is the same.
Comparative example 2
Comparative example 2 is different from example 1 in that the chitosan nanoparticle suspension is not added in step (5), and the rest of the process is the same.
Comparative example 3
The difference between the comparative example 3 and the example 1 is that the filter residue A and the filter residue B are directly mixed and extracted without adopting the complex microbial inoculum, and the rest processes are the same.
Experimental example 1
The research on the traditional Chinese medicine components in the premix comprises the following steps: the traditional Chinese medicine is prepared from Chinese herbal medicines with the functions of tonifying kidney and securing essence, such as astragalus, epimedium, glossy privet fruit, Chinese yam, eucommia bark, cistanche deserticola, Chinese dodder seed, rehmannia root, medlar, mulberry, Chinese yam, lotus seed, Chinese gall, malaytea scurfpea fruit and the like; mixing different Chinese medicinal formulas with the same amount of chromium picolinate, zinc methionine, lutein, grape seed, radix Morindae officinalis, fructus Schisandrae, 1,25 dihydroxyvitamin D, microelements, soybean lecithin and chitosan nanoparticle suspension, and grinding to obtain different premixes; then adding the premix into ration according to the weight ratio of 0.4% to feed the boars; feeding 2.5kg daily ration twice a day with normal drinking water. Experiment time: 1/9/2017-25/9/2017. And counting the sperm motility indexes of the boars after the experiment.
As a result, it was found that: by adopting the traditional Chinese medicine formula of astragalus, epimedium and glossy privet fruit, the average sperm motility of the boar can reach 82.15; is 0.05-18.32 percent higher than other traditional Chinese medicine compositions.
Experimental example 2
Experiment time: 1/9/2017-25/9/2017
Experimental materials: 180 duroc breeding boars were selected for the trial and divided into 11 groups of 20, of which 10 were experimental groups and the other group was control group. The experimental groups were fed with the diets of the experimental groups in table 1, wherein the premixes in the experimental groups 1-8 were the premixes prepared in examples 1-5 and comparative examples 1-3; the experimental groups 5-6 respectively feed Chinese patent with application number CN201410338462.4, which discloses a traditional Chinese medicine formula for resisting heat stress in breeding period of breeding poultry, and Chinese patent with application number CN201110042053.6, which discloses a heat stress resisting composition for pigs; the experimental groups were fed the diets of the control group in table 1. Feeding 2.5kg daily ration twice a day with normal drinking water. After statistical experiments, the energy and activity indexes of the boars in each group are detected, and the results are shown in table 2:

Note: weighing the amount of the semen by using an electronic balance; the sperm motility is measured by a sperm automatic analyzer; the sperm density is measured by a sperm automatic analyzer; the sperm motility rate is measured by a sperm automatic analyzer; effective sperm count is sperm amount x sperm density x sperm motility; total sperm count is the amount of sperm multiplied by sperm density.
As can be seen from the table, compared with the control group, the premixes used in the experimental groups 1 to 10 have the effect of the sperm motility and the survival rate of boars, wherein the effect of the experimental groups 1 to 5 is more significant, and the premixes are significantly different from the experimental groups 6 to 10.
It should be noted that the above examples and test examples are only for further illustration and understanding of the technical solutions of the present invention, and are not to be construed as further limitations of the technical solutions of the present invention, and the invention which does not highlight essential features and significant advances made by those skilled in the art still belongs to the protection scope of the present invention.

Claims (8)

1. The premix for improving the sperm motility of boars in summer is characterized by comprising the following raw materials in parts by weight: 10-20 parts of astragalus membranaceus, 5-10 parts of epimedium, 3-5 parts of glossy privet fruit, 0.03-0.05 part of chromium picolinate, 0.02-0.05 part of zinc methionine, 0.03-0.05 part of lutein, 0.01-0.015 part of grape seed, 0.01-0.015 part of morinda officinalis, 0.008-0.01 part of schisandra chinensis, 0.002-0.004 part of 1,25 dihydroxyvitamin D, 0.003-0.005 part of trace element, 0.06-0.08 part of soybean lecithin and 0.08-0.1 part of chitosan nanoparticle suspension.
2. The premix for improving boar summer sperm motility according to claim 1, wherein the trace elements are composed of calcium, iron, selenium and molybdenum according to a molar ratio of 2:1:1: 1.

The premix for improving boar summer sperm motility according to claim 1, wherein the preparation method of the chitosan nanoparticles comprises the following steps: dissolving water-soluble chitosan in 0.5% citric acid solution, adjusting pH to 6, and preparing into chitosan mixed solution with chitosan solution content of 2 mg/ml; taking a proper amount of sodium tripolyphosphate to prepare 1mg/ml sodium tripolyphosphate aqueous solution; then at 750--1Slowly dripping the sodium tripolyphosphate aqueous solution into the chitosan mixed solution at a rotating speed, carrying out ionic crosslinking and continuously stirring for 10-15min to prepare the chitosan nanoparticle suspension.
4. The method for preparing the premix for improving the sperm motility of boars in summer according to claim 1, comprising the following steps:
(1) pulverizing radix astragali, herba Epimedii, and fructus Ligustri Lucidi, decocting in 8-10 times of water, and filtering to obtain filtrate A and residue A;
(2) mixing grape seed, radix Morindae officinalis, and fructus Schisandrae, pulverizing, adding into filtrate A, stirring at 50-60 deg.C for 30-40min, and filtering to obtain Chinese medicinal extractive solution A and residue B; mixing residue A and residue B, adding complex microbial inoculum, fermenting at 25-30 deg.C for 20-24 hr, and sterilizing to obtain fermented product;
(3) adding trace elements into the Chinese medicinal extract A, adding 8-10 times of 70% methanol solution, mixing, reacting at 60-70 deg.C for 1-1.2 hr, and recovering methanol to obtain mixed extract A;
(4) adding 6-8 times of 80% ethanol into the fermented product, reflux extracting at 50-60 deg.C for 40-50min, filtering, recovering ethanol from the filtrate, and concentrating at 40-50 deg.C to obtain Chinese medicinal extract; mixing the traditional Chinese medicine extract and soybean lecithin, adding 1-2 times of anhydrous ethanol for compound reaction, and recovering ethanol to obtain a mixed extract B;
(5) mixing the mixed extract A and B, concentrating at 40-50 deg.C to obtain paste, spray drying to obtain powder, adding xanthophyll and chitosan nanoparticle suspension, and ultrasonic processing for 3-5min to obtain mixture; and adding chromium picolinate and zinc methionine, and mixing uniformly to obtain the premix.

The method for preparing the premix for improving the sperm motility of boars in summer according to claim 4, wherein in the step (4), the condition of the complex reaction is as follows: the reaction is carried out for 1.5 to 2 hours at the temperature of between 45 and 50 ℃ by magnetic stirring, and the magnetic stirring speed is 150-.
6. The method for preparing the premix for improving boar summer sperm motility according to claim 4, wherein the compound microbial inoculum is a liquid microbial inoculum which contains lactobacillus acidophilus, bacillus subtilis and trichoderma.
7. The method of claim 6, wherein the complex microbial inoculum comprises viable Lactobacillus acidophilusThe number of the particles is 6-8 multiplied by 108cfu/ml, viable count of Bacillus subtilis is 4-6 × 106cfu/ml, viable count of Trichoderma 6-8 × 104cfu/ml。
