技术领域
本发明属于生物医药领域,涉及一种5-氨基酮戊酸水凝胶组合物其制备方法和在皮肤美容、荧光成像、光动力治疗等方面的应用。
背景技术
5-氨基酮戊酸是一种光动力前药,可以在亚铁血红素酶的作用下转化为原卟啉,原卟啉可以吸收光,并将光能转换为荧光或活性氧的形式释放能量。5-氨基酮戊酸具有广泛的应用前景,比如可以应用于皮肤美容、荧光成像、光动力治疗癌症中。在荧光成像技术中,生物组织中的5-氨基酮戊酸分子在亚铁血红素酶的作用下转化为原卟啉,原卟啉发射的荧光信号的强度在一定的范围内与原卟啉的量成线性关系。从而可以利用此成像技术来实时连续动态监测体内的各种生物学过程,该技术具有方便、直观、敏感性高的优点。在光动力治疗中,5-氨基酮戊酸作为一种FDA批准的药物,已经在临床治疗中得到了广泛使用。与传统的放射治疗和化学治疗相比较,光动力治疗因为其高选择性及微创性,对正常细胞的危害较小,过程简单,并且远程可控、治疗时间短,从而大大降低了副作用的产生,成为癌症治疗的一个强有力的技术。此外,光动力治疗技术只需满足三个基本要素就可以实施:一是激光光源,二是具有光转换性能的光敏分子,三是氧气。5-氨基酮戊酸在光动力治疗中即作为光敏分子发挥作用。
用于荧光成像和光动力治疗的5-氨基酮戊酸需要在病变部位有一定的保留时间。由于5-氨基酮戊酸是水溶性的分子,药物向周围皮肤和组织很快扩散,无法实现较长时间的持续给药,降低了治疗效果。5-氨基酮戊酸的水溶液稳定性较差,如中性条件下,其失活半衰期约15小时,极大地影响了5-氨基酮戊酸水溶液的保存和使用。此外,5-氨基酮戊酸分子在静脉给药和口服给药后半衰期较短,导致生物利用度低等问题。因此,发明一种新型的5-氨基酮戊酸剂型用于解决此问题显得尤为重要。基于此,我们发明了5-氨基酮戊酸水凝胶组合物,用于增强5-氨基酮戊酸的稳定性,实现持续释放给药,提高治疗效果、降低副作用。
蛋白是生物体天然存在的重要成分,机体所有重要的组成部分都需要蛋白的参与。蛋白是由氨基酸脱水缩合而成,可以形成不同的二级结构,进一步地在合适的条件下自组织和缠绕形成三维凝胶网络结构。分子间氢键、静电和疏水作用可以提高它们的稳定性。这些凝胶网络结构在药物分子的递送中已经展示了一些优势,可以用于化疗、光疗、免疫疗法中。
多糖是由多个单糖分子缩合、失水而成,凡符合高分子化合物概念的碳水化合物及其衍生物均称为多糖,广义分为均一性多糖和不均一性多糖。多糖的种类繁多,分布广泛且具有重要作用,由于它的高生物相容性、低免疫原性、和生物降解性在药物递送方面发挥重要作用,目前作为化药和光疗药物的载体得到飞速发展。
除了蛋白和多糖等生物分子,一些生物相容性好的共聚物,如PLGA(聚乳酸-羟基乙酸共聚物)、PLGA-PEG(聚乙二醇)-PLGA、PEO(聚氧化乙烯)-PPO(聚苯醚)-PEO等,也已经在临床得到了一些应用。
基于5-氨基酮戊酸分子在荧光成像和光动力治疗中的应用,将该分子装载在蛋白、多糖、共聚物形成的水凝胶中,有望通过水凝胶与5-氨基酮戊酸的相互作用,提升5-氨基酮戊酸的稳定性和生物利用度,实现其持续的释放和转化,提高在皮肤美容和诊断治疗方面的潜力。
发明内容
本发明的目的在于提供一种5-氨基酮戊酸水凝胶组合物,用于美容和生物医药方面;本发明形成的水凝胶组合物具有稳定性好、生物安全性高、生物利用度高的特性,可用于皮肤美容、荧光成像、光动力治疗等方面。
为达此目的,本发明采用以下技术方案:
第一方面,本发明提供了一种5-氨基酮戊酸水凝胶组合物,所述水凝胶组合物含有5-氨基酮戊酸与凝胶因子,所述凝胶因子选自蛋白、多糖、共聚物,所述共聚物为PLGA(聚乳酸-羟基乙酸共聚物)、PLGA-PEG(聚乙二醇)-PLGA、PEO(聚氧化乙烯)-PPO(聚苯醚)-PEO中的任意一种或至少两种的混合物;任选的,所述水凝胶组合物含有或不含有除了水之外的其它成分。
在本发明的一个优选实施方式中,所述5-氨基酮戊酸与凝胶因子的比例为1~10:1~10。
在本发明的一个优选实施方式中,所述的凝胶因子含有蛋白,所述是丝素蛋白、胶原蛋白、明胶中的任意一种或至少两种的混合物。通过采用上述蛋白作为凝胶因子,相比于其它凝胶因子而言,可以极大的增强5-氨基酮戊酸的稳定性,这样的技术效果是其它凝胶因子所不具备的。
在本发明的另一个优选实施方式中,所述多糖是海藻糖、壳聚糖、几丁糖、透明质酸中的任意一种或至少两种的混合物。
第二方面,本发明提供了如第一方面所述的5-氨基酮戊酸水凝胶组合物的制备,包括以下步骤:
i.称取所需量的5-氨基酮戊酸,溶解在水中,制备成5-氨基酮戊酸水溶液;
ii.将上述5-氨基酮戊酸水溶液与凝胶因子进行混合,得到5-氨基酮戊酸的水凝胶组合物;
所述的5-氨基酮戊酸水凝胶组合物中,5-氨基酮戊酸的质量含量为0.1-30%,凝胶因子的质量含量为0.1-30%。
第三方面,本发明还提供了本发明第一方面所述的5-氨基酮戊酸水凝胶组合物的应用;5-氨基酮戊酸在皮肤美容和疾病治疗中具有广泛的应用,本发明的5-氨基酮戊酸水凝胶组合物能够提高5-氨基酮戊酸在局部位置的保留,增强其向原卟啉的转化,从而增强其在皮肤美容和疾病治疗中的价值,因此,本发明的5-氨基酮戊酸水凝胶组合物可用于制备美容产品、医疗药物。
与现有的5-氨基酮戊酸注射剂型相比,本发明至少具有以下有益效果:
(1)本发明提供的以蛋白作为凝胶因子的水凝胶组合物能够延长5-氨基酮戊酸的稳定性。5-氨基酮戊酸在水溶液中的失活半衰期约15小时,极大地影响了5-氨基酮戊酸水溶液的保存和使用。通过水凝胶与5-氨基酮戊酸的相互作用,能够显著提升5-氨基酮戊酸的稳定性,失活半衰期可延长至100小时以上。这种通过以蛋白作为凝胶因子的水凝胶提升5-氨基酮戊酸稳定性的策略,还未见文献报道,有利于5-氨基酮戊酸的保存和使用。
(2)本发明提供的水凝胶组合物能够延长5-氨基酮戊酸分子的保留时间,在局部涂抹后,保留时间延长一倍以上;
(3)本发明提供的5-氨基酮戊酸水凝胶组合物能够增强5-氨基酮戊酸向原卟啉的转化,在局部注射后,原位转化生成的原卟啉的量相比于5-氨基酮戊酸水溶液增加一倍以上。

下面对本发明进一步详细说明。但下述的实例仅仅是本发明的简易例子,并不代表或限制本发明的权利保护范围,本发明的保护范围以权利要求书为准。
具体实施方式
以下通过实施例对本发明进行详细说明,但本发明并不仅限于下述实施例。
实施例1
取1mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与2mg胶原蛋白进行混合,制备得到水凝胶组合物。通过拍照得到实物图1,表明该设计可以成功形成5-氨基酮戊酸蛋白水凝胶复合物。
实施例2
取2mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与10mg几丁糖进行混合,制备得到水凝胶组合物。通过原子力显微镜表征得到图2,其表明复合物的形貌为纳米纤维。
实施例3
取5mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与20mg丝素蛋白进行混合,制备得到水凝胶组合物。通过圆二色谱光谱仪测得CD图3,说明复合物的二级结构为β折叠结构。
实施例4
取10mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与50mg明胶进行混合,制备得到水凝胶组合物。将该水凝胶组合物原位注射到荷瘤小鼠肿瘤部位,通过小动物荧光成像测得5-氨基酮戊酸在体内随时间形成的原卟啉荧光,结果见图4,说明水凝胶组合物能够原位形成原卟啉。
实施例5
取50mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与100mg透明质酸进行混合,制备得到水凝胶组合物。通过流变仪测得5-氨基酮戊酸水凝胶组合物的流变图5,确定复合物的凝胶特性。
实施例6
取100mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与10mg壳聚糖进行混合,制备得到水凝胶组合物。通过扫描电子显微镜表征5-氨基酮戊酸水凝胶组合物的形貌图6,确定复合物的纤维形貌。
实施例7
取150mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与20mg海藻糖进行混合,制备得到水凝胶组合物。通过透射电子显微镜表征5-氨基酮戊酸水凝胶形貌,见图7,确定复合物为相互缠绕的三维纤维网络。
实施例8
取4mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与5mg丝素蛋白进行混合,制备得到水凝胶组合物。测试该水凝胶组合物在室温下保存时5-氨基酮戊酸含量的变化,并与5-氨基酮戊酸的水溶液进行比较。结果如图8,水溶液中的5-氨基酮戊酸快速降解,而水凝胶中的5-氨基酮戊酸降解速度较慢,说明水凝胶能够增强5-氨基酮戊酸的稳定性。
实施例9
取10mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与10mg壳聚糖进行混合,制备得到水凝胶组合物。测试该水凝胶组合物中5-氨基酮戊酸的降解行为,结果如图9所示,其半衰期大于100小时。
实施例10
取15mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与15mg PLGA进行混合,制备得到水凝胶组合物。测试该水凝胶组合物中5-氨基酮戊酸的降解行为,结果如图10所示。
综合对比图8~10可知,采用蛋白作为凝胶因子的实施例8对于5-氨基酮戊酸稳定性的提高效果最佳。
实施例11
取10mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与10mg胶原蛋白进行混合,制备得到水凝胶组合物。将该水凝胶组合物原位注射到荷瘤小鼠肿瘤部位,通过小动物荧光成像测得5-氨基酮戊酸在肿瘤部位形成的原卟啉荧光强度;采用同样方法向控制组小鼠肿瘤部位注射5-氨基酮戊酸水溶液,并进行荧光测试。肿瘤部位荧光强度的变化如图11所示,结果表明,水凝胶组合物增强了5-氨基酮戊酸的保留时间,且原位转化生成的原卟啉的量相比于5-氨基酮戊酸水溶液增加一倍以上。
实施例12
取2mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与100mg PLGA-PEG-PLGA进行混合,制备得到水凝胶组合物。
实施例13
取10mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与10mg PEO-PPO-PEO进行混合,制备得到水凝胶组合物。
实施例14
取1mg 5-氨基酮戊酸分子加入1mL水中,制备成5-氨基酮戊酸水溶液,将此水溶液与5mg胶原蛋白进行混合,制备得到水凝胶组合物。通过液相色谱测试不同保存时间后水凝胶组合物中含有5-氨基酮戊酸的量,以5-氨基酮戊酸的水溶液为对照组。以5-氨基酮戊酸的水溶液失活半衰期为15小时,而水凝胶组合物中5-氨基酮戊酸的失活半衰期为180小时,说明水凝胶组合物能够显著提高5-氨基酮戊酸的稳定性。
申请人声明,本发明通过上述实施例来说明本发明的产品及详细制备方法,但本发明并不局限于上述产品和详细制备方法,即不意味着本发明必须依赖上述产品和详细制备方法才能实施。所属技术领域的技术人员应该明了,对本发明的任何改进,对本发明产品各原料的等效替换及辅助成分的添加、具体方式的选择等,均落在本发明的保护范围和公开范围之内。
以上详细描述了本发明的优选实施方式,但是,本发明并不限于上述实施方式中的具体细节,在本发明的技术构思范围内,可以对本发明的技术方案进行多种简单变型,这些简单变型均属于本发明的保护范围。
另外需要说明的是,在上述具体实施方式中所描述的各个具体技术特征,在不矛盾的情况下,可以通过任何合适的方式进行组合,为了避免不必要的重复,本发明对各种可能的组合方式不再另行说明。
此外,本发明的各种不同的实施方式之间也可以进行任意组合,只要其不违背本发明的思想,其同样应当视为本发明所公开的内容。
Claims (5)
1.一种5-氨基酮戊酸水凝胶组合物,所述水凝胶组合物含有5-氨基酮戊酸与凝胶因子,所述凝胶因子为蛋白;所述蛋白是丝素蛋白、胶原蛋白中的任意一种或两种的混合物;任选的,所述水凝胶组合物含有或不含有除了水之外的其它成分;5-氨基酮戊酸的质量含量为0.1-30%,蛋白的质量含量为0.1-30%。
2.权利要求1所述的5-氨基酮戊酸水凝胶组合物的制备方法,其特征在于,所述制备方法包括如下步骤:
(1)首先称量好所需量的5-氨基酮戊酸,溶解在水溶液中;
(2)将步骤(1)中制备的水溶液与能形成水凝胶的成分进行混合,制备成5-氨基酮戊酸水凝胶组合物。
3.权利要求1所述的5-氨基酮戊酸水凝胶组合物在制备美容产品或医疗药物中的用途。
4.权利要求1所述的5-氨基酮戊酸水凝胶组合物在制备治疗肿瘤的药物中的应用。
5.根据权利要求4所述的应用,所述的治疗肿瘤的药物是光动力治疗药物。
