Abstract
本发明公开了一种生猪无抗饲料及其制备方法,所述无抗饲料包括乳酸菌和基础饲粮,所述基础饲粮包括玉米、膨化玉米、发酵豆粕、膨化大豆、鱼粉、乳清粉、白砂糖、氯化胆碱、食盐、碳酸氢钙、石粉、L‑赖氨酸盐酸盐、DL‑蛋氨酸、L‑苏氨酸、L‑色氨酸和预混料。本发明的无抗饲料由基础饲粮中添加5×1010cfu/kg罗伊氏乳杆菌LR1得到,可显著提高断奶早期仔猪生长性能,对断奶后期猪和肥育猪生长性能无显著影响,但有一定的改善作用,由此表明,罗伊氏乳杆菌LR1可作为抗生素替代物之一。
Description
技术领域
本发明涉及饲料制备技术领域,尤其是一种生猪无抗饲料及其制备方法。
背景技术
乳杆菌是益生菌中最常见、应用最广泛的一类,作为饲料添加剂之一,在养殖业中被用来提高动物生长性能、降低腹泻率和改善养殖环境等。乳酸杆菌对氨基酸转运载体的上调作用可能有助于肠道形态的完整及营养物质的吸收,进而有利于动物生长。
养猪业中,过去常常大量应用抗生素来降低病原菌感染、提高断奶仔猪的生长性能。有研究结果表明,饲粮中添加75mg/kg金霉素和100mg/kg喹乙醇显著提高了断奶仔猪日增重和采食量,生长性能促进作用显著。然而,在生产中,经常会超量添加抗生素,基于大量使用抗生素会带来抗生素残留、细菌耐药性增加、环境污染等严重问题的考虑,欧盟早在1999年9月已经宣布禁止喹乙醇作为饲料抗菌和生长促进剂,中国也考虑到了它的负面作用,并在2017年8月起禁止喹乙醇的使用。为解决禁用抗生素仔猪断奶后的高发病率和死亡率,大量抗生素替代物应运而生,主要有益生菌、益生元(主要是低聚糖)、有机酸、其他碳水化合物或酶制剂等。然而,目前市场上仍然欠缺能显著增加断奶仔猪前期体重的无抗生素饲料。
发明内容
基于上述问题,本发明的目的在于克服上述现有技术的不足之处而提供一种可显著增加断奶仔猪前期体重的无抗饲料,其不会增加人畜对细菌的耐药性,对环境绿色无污染。
为实现上述目的,本发明采取的技术方案为:一种生猪无抗饲料,所述饲料包括乳酸菌和基础饲粮,所述基础饲粮包括玉米、膨化玉米、发酵豆粕、膨化大豆、鱼粉、乳清粉、白砂糖、氯化胆碱、食盐、碳酸氢钙、石粉、L-赖氨酸盐酸盐、DL-蛋氨酸、L-苏氨酸、L-色氨酸和预混料。其中,所述氯化胆碱是质量百分比50%的水溶液。
优选地,所述生猪的体重为7~11kg时,所述基础饲粮还包括大豆皮、大豆油和血浆蛋白粉。
优选地,所述预混料包括VA、VD3、VE、VK、VB12、VB1、VB2、烟酸、D-泛酸、叶酸、VB6、生物素、FeSO4·H2O,CuSO4·5H2O,MnSO4·H2O),ZnSO4·H2O,CaI2O6和Na2SeO3。
优选地,所述生猪的体重为11~25kg时,所述基础饲粮还包括去皮大豆粕。
优选地,所述预混料包括VA,VD3,VE,VK,VB12,VB1,VB2,,烟酸,D-泛酸,叶酸,VB6,生物素,FeSO4·H2O,CuSO4·5H2O,MnSO4·H2O,ZnSO4·H2O,CaI2O6和Na2SeO3。

作为本发明的另一个方面,本发明提供了上述无抗饲料的制备方法,包括如下步骤:所述基础饲粮分成两部分,一部分制成粉料,另一部分经85℃制成粒料,粉料中添加所述乳酸菌,然后将粉料和粒料混合即得所述无抗饲料,其中,所述粉料和粒料的质量比为1:1或1:2。由此,益生菌(罗伊氏乳杆菌LR1)加在了粉料里,不经过高温,所以能保持活菌数量,这样能更好的发挥益生菌的益生作用。
综上所述,本发明的有益效果为:
本发明的无抗饲料由基础饲粮中添加5×1010cfu/kg罗伊氏乳杆菌LR1制得,可显著提高断奶早期仔猪生长性能,对断奶后期猪和肥育猪生长性能无显著影响,但有一定的改善作用,由此表明,罗伊氏乳杆菌LR1可作为抗生素替代物之一。
具体实施方式
本试验以21日龄断奶仔猪为对象,通过研究饲粮中添加罗伊氏乳杆菌LR1和抗生素对断奶仔猪生长性能、肠道形态、免疫应答、肠道屏障功能的影响,以揭示LR1发挥保护及促生长作用的可能机制,为养猪生产中使用益生菌提供一定的科学依据。
为更好的说明本发明的目的、技术方案和优点,下面将结合具体实施例对本发明作进一步说明。如无特别说明,本发明中的实验方法均为常规方法;如无特别说明,本发明中的试剂浓度均为质量浓度。
实施例1
本发明中生猪无抗饲料的一种实施例,用于饲养体重为7~11kg的生猪,该无抗饲料由基础饲粮和罗伊氏乳杆菌LR1组成,罗伊氏乳杆菌LR1的含量为5×1010cfu/kg基础饲粮,基础饲粮中含有以下质量百分含量的成分:玉米35.31%、膨化玉米15%、发酵豆粕9%、膨化大豆10%、鱼粉4%、乳清粉11%、大豆皮5%、大豆油1.2%、血浆蛋白粉3%、白砂糖2%、氯化胆碱(50%)0.2%、食盐0.45%、碳酸氢钙0.62%、石粉0.65%、L-赖氨酸盐酸盐0.6%、DL-蛋氨酸0.22%、L-苏氨酸0.21%、L-色氨酸0.04%和预混料1.5%;

实施例3
1、试验动物与设计
试验采用单因素完全随机试验设计,选取144头21日龄杜洛克×长白×大白三元杂交断奶仔猪(初始体重为6.49±0.02kg),公母各半,随机分配至三个处理组:对照组(CON),饲喂基础饲粮;抗生素组(OA),基础饲粮+100mg/kg喹乙醇+75mg/kg金霉素;罗伊氏乳杆菌组(LR1),基础饲粮+5×1010cfu/kg罗伊氏乳杆菌LR1(按照实施例1的方法制备),每组8个重复,每个重复6头仔猪,试验期划分三个阶段,分别为第一阶段(试验第1~14天),第二阶段(试验第15~42天),第三阶段(试验第43~165天(出栏))。试验期:2016年11月3日~2017年4月17日。
2、试验饲粮
根据猪体重配制的基础饲粮满足美国NRC(2012)营养需要推荐量,饲粮组成和营养成分见表1。

(4)50~75kg预混料为每千克饲粮提供:VA 7,750IU,VD3 1,750IU,VE 19IU,VK3mg,VB12 25μg,VB1 1.9mg,VB2,6mg,烟酸25mg,D-泛酸9mg,叶酸0.6mg,VB6 5mg,生物素0.05mg,铁(FeSO4·H2O)72mg,铜(CuSO4·5H2O)10mg,锰(MnSO4·H2O)42mg,锌(ZnSO4·H2O)72mg,碘(CaI2O6)0.42mg,硒(Na2SeO3)0.29mg。
(5)75~100kg预混料为每千克饲粮提供:VA 7,750IU,VD3 1,750IU,VE 19IU,VK3mg,VB12 25μg,VB1 1.9mg,VB2,6mg,烟酸25mg,D-泛酸9mg,叶酸0.6mg,VB6 5mg,生物素0.05mg,铁(FeSO4·H2O)72mg,铜(CuSO4·5H2O)10mg,锰(MnSO4·H2O)42mg,锌(ZnSO4·H2O)72mg,碘(CaI2O6)0.42mg,硒(Na2SeO3)0.29mg。
(6)100~135kg预混料为每千克饲粮提供:VA 6,200IU,VD3 14,000IU,VE 15IU,VK2.5mg,VB12 20μg,VB1 1.5mg,VB2,5mg,烟酸20mg,D-泛酸8mg,叶酸0.5mg,VB6 4mg,生物素0.04mg,铁(FeSO4·H2O)60mg,铜(CuSO4·5H2O)8mg,锰(MnSO4·H2O)35mg,锌(ZnSO4·H2O)60mg,碘(CaI2O6)0.35mg,硒(Na2SeO3)0.24mg。
(7)粗蛋白质、钙、总磷、有效磷为实测值,其余为计算值。
3、饲养管理
试验地点位于广东省农业科学院动物科学研究所试验场,栏舍为全封闭建筑带有温控装置,仔猪圈养于离地的漏缝栏,于每天08:00、12:00、16:00、20:00饲喂4次,自由饮水。试验第43天将全部猪转入生长肥育猪栏舍中,自由采食。各组猪均按猪场的免疫程序进行疫苗接种、驱虫保健等。
4、仪器设备
机械磅秤(高标,广州白云);电子秤(苏州金钻称重设备系统开发有限公司)。
5、生长性能测定
(1)平均日增重(ADG):分别于试验第1天、第15天、第43天、第166天早上08:00,猪空腹12小时后称重,计算每个阶段的平均日增重。
(2)平均日采食量(ADFI):饲养阶段每组耗料量÷(试验天数×仔猪数)。
(3)料重比(F:G):ADFI÷ADG。
6、数据处理与统计分析
采用SPSS 18.0软件的单因素方差分析(one-way ANOVA)模块进行数据分析,以图基检验法(Tukey’s test)进行多重比较,显著水平为0.05。结果表示为平均值±标准误(mean±SEM),n=8。
7、结果与分析
7.1罗伊氏乳杆菌LR1对猪生长性能的影响
如表2所示,试验第1~14天,与对照组相比,添加罗伊氏乳杆菌LR1组仔猪平均日增重(ADG)提高22.73%(P<0.05),提高平均日采食量(ADFI)15.48%,差异不显著(P>0.05),而抗生素组与罗伊氏乳杆菌组相似,日增重提高29.63%(P<0.05)、采食量提高17.82%(P<0.05)。
试验第15~42天,与对照组相比,罗伊氏乳杆菌组仔猪日增重提高9.31%(P>0.05)、采食量提高10.00%(P>0.05)。而抗生素组仔猪日增重、采食量分别提高12.65%(P=0.085)和17.34%(P<0.05)。
试验第43~165天,各组仔猪日增重和采食量均无影响(P>0.05)。各组三个阶段料重比无显著影响(P>0.05)。

7.2总结
本申请的发明人之前从健康的断奶仔猪粪便中分离获得一株新的罗伊氏乳杆菌菌株LR1,并在体外试验证明具有耐酸耐胆汁性强、抗菌性能好、可改善肠道屏障功能等优良益生性能,本试验中观察到,罗伊氏乳杆菌LR1可改善断奶后两周仔猪平均日增重和平均采食量,与前人研究结果一致,与抗生素的效果相似。在断奶第3~6周,罗伊氏乳杆菌LR1虽然未对仔猪生长性能产生显著影响,但可以一定程度上提高仔猪的日增重和采食量。罗伊氏乳杆菌对肥育猪阶段结束体重也有一定的提高,但影响也不显著。而罗伊氏乳杆菌对断奶后更长时期生长性能的影响不大。仔猪断奶时经过环境和饲料等因素改变,肠道发生巨大改变,需要时间以适应新的环境。因此,在断奶早期,运用益生菌能更好地改善断奶应激。
而在断奶后更长时期,仔猪肠道已经逐渐适应外部环境的改变并建立起正常的肠道功能。因此,猪利用罗伊氏乳杆菌的效率与猪的断奶日龄有关。但是,罗伊氏乳杆菌LR1的益生效应是持久的,表现在进一步影响了肥育猪阶段的生长性能,并可能对猪健康、肉质产生潜在的有益影响,从投入与产出的角度讲是值得的。这些结果表明,罗伊氏乳杆菌LR1可作为抗生素替代物,以改善断奶仔猪的生长性能,并影响肥育猪生长。
综上所述,饲粮中添加5×1010cfu/kg罗伊氏乳杆菌LR1,且放在粉料中,粉料和粒料比为1:1或1:2情况下,可显著提高断奶早期仔猪生长性能,对断奶后期猪和肥育猪生长性能无显著影响,但有一定的改善作用。罗伊氏乳杆菌LR1可作为抗生素替代物之一。
最后应当说明的是,以上实施例仅用以说明本发明的技术方案而非对本发明保护范围的限制,尽管参照较佳实施例对本发明作了详细说明,本领域的普通技术人员应当理解,可以对本发明的技术方案进行修改或者等同替换,而不脱离本发明技术方案的实质和范围。

1.一种生猪无抗饲料,其特征在于,所述无抗饲料包括乳酸菌和基础饲粮,所述基础饲粮包括玉米、膨化玉米、发酵豆粕、膨化大豆、鱼粉、乳清粉、白砂糖、氯化胆碱、食盐、碳酸氢钙、石粉、L-赖氨酸盐酸盐、DL-蛋氨酸、L-苏氨酸、L-色氨酸和预混料。
2.根据权利要求1所述的无抗饲料,其特征在于,所述生猪的体重为7~11kg时,所述基础饲粮还包括大豆皮、大豆油和血浆蛋白粉。
3.根据权利要求2所述的无抗饲料,其特征在于,所述预混料包括VA、VD3、VE、VK、VB12、VB1、VB2、烟酸、D-泛酸、叶酸、VB6、生物素、FeSO4·H2O,CuSO4·5H2O,MnSO4·H2O),ZnSO4·H2O,CaI2O6和Na2SeO3。
4.根据权利要求1所述的无抗饲料,其特征在于,所述生猪的体重为11~25kg时,所述基础饲粮还包括去皮大豆粕。
5.根据权利要求4所述的无抗饲料,其特征在于,所述预混料包括VA,VD3,VE,VK,VB12,VB1,VB2,,烟酸,D-泛酸,叶酸,VB6,生物素,FeSO4·H2O,CuSO4·5H2O,MnSO4·H2O,ZnSO4·H2O,CaI2O6和Na2SeO3。
6.根据权利要求1所述的无抗饲料,其特征在于,所述乳酸菌为罗伊氏乳杆菌LR1。
7.根据权利要求6所述的无抗饲料,其特征在于,所述罗伊氏乳杆菌LR1的用量为5×1010cfu/kg饲粮。
8.根据权利要求1所述的无抗饲料,其特征在于,所述生猪的体重为7~11kg时,所述基础饲粮中含有以下质量百分含量的成分:玉米35.31%、膨化玉米15%、发酵豆粕9%、膨化大豆10%、鱼粉4%、乳清粉11%、大豆皮5%、大豆油1.2%、血浆蛋白粉3%、白砂糖2%、氯化胆碱0.2%、食盐0.45%、碳酸氢钙0.62%、石粉0.65%、L-赖氨酸盐酸盐0.6%、DL-蛋氨酸0.22%、L-苏氨酸0.21%、L-色氨酸0.04%和预混料1.5%;
在每千克基础饲粮中,所述预混料含有以下含量的成分:VA 12,400IU,VD32,800IU,VE30IU,VK 5mg,VB12 40μg,VB1 3mg,VB2 10mg,烟酸40mg,D-泛酸15mg,叶酸1mg,VB6 8mg,生物素0.08mg,FeSO4·H2O 120mg,CuSO4·5H2O 16mg,MnSO4·H2O 70mg,ZnSO4·H2O 120mg,CaI2O6 0.7mg和Na2SeO3 0.48mg。
9.根据权利要求1所述的无抗饲料,其特征在于,所述生猪的体重为11~25kg时,所述基础饲粮中含有以下质量百分含量的成分:玉米47.55%、膨化玉米13%、发酵豆粕8.5%、去皮大豆粕9%、膨化大豆6%、鱼粉4%、乳清粉6%、白砂糖2%、氯化胆碱0.18%、食盐0.45%、碳酸氢钙0.6%、石粉0.74%、L-赖氨酸盐酸盐0.54%、DL-蛋氨酸0.2%、L-苏氨酸0.21%、L-色氨酸0.03%和预混料1%;
在每千克基础饲粮中,所述预混料含有以下含量的成分:VA 12,400IU,VD32,800IU,VE30IU,VK 5mg,VB12 40μg,VB1 3mg,VB2,10mg,烟酸40mg,D-泛酸15mg,叶酸1mg,VB6 8mg,生物素0.08mg,FeSO4·H2O 90mg,CuSO4·5H2O 12mg,MnSO4·H2O 53mg,ZnSO4·H2O 90mg,CaI2O60.53mg和Na2SeO3 0.36mg。
10.权利要求1~9任一项所述的无抗饲料的制备方法,其特征在于,包括如下步骤:
所述基础饲粮分成两部分,一部分制成粉料,另一部分经85℃制成粒料,粉料中添加所述乳酸菌,然后将粉料和粒料混合即得所述无抗饲料,其中,所述粉料和粒料的质量比为1:1或1:2。
