背景技术
来自基因型HLA匹配的兄弟姐妹的造血干细胞移植(HSCT)改善了患有血液癌症恶性肿瘤和骨髓衰竭综合征的患者的长期存活。每年有超过10,000名美国人患上危及生命的疾病,治愈的唯一希望就是从无关的捐献者或脐带血单位进行骨髓移植。然而,超过70%的可能从同种异体干细胞移植中受益的患者没有匹配的同胞供体。这些情况延误了治疗,因此有必要采用不太理想的部分不匹配供体,这最终会导致移植物抗宿主病(GVHD)、移植失败和复发的发生率增加,所有这些都会大大减少患者生存。

B.扩增记忆样NK细胞的方法
人NK细胞是由CD56或CD16的表达和不存在T细胞受体(CD3)定义的外周血淋巴细胞的子集。NK细胞感知并杀死缺乏主要组织相容性复合物(MHC)-I类分子的靶细胞。NK细胞活化受体尤其包括天然细胞毒性受体(NKp30、NKp44和NKp46)和凝集素样受体NKG2D和DNAM-1。它们的配体在受胁迫、转化或感染的细胞上表达,但在正常细胞上不表达,使正常细胞对NK细胞杀伤具有抗性。NK细胞活化通过抑制性受体负调节,例如杀伤性免疫球蛋白(Ig)样受体(KIR)、NKG2A/CD94和白细胞Ig样受体-1(LIR-1)。一种抑制性受体的接合可足以阻止靶裂解。因此,NK细胞有效地靶向表达许多应激诱导的配体和很少MHC I类配体的细胞。

C.组合物

Claims (21)
1.一种增加记忆样NK细胞数量的方法,包括:a)通过使至少一种NK细胞与至少一种或多种刺激性细胞因子接触来预活化NK细胞;和b)通过使所述细胞与PM21颗粒、EX21外泌体或FC21饲养细胞接触,扩增步骤a)的预活化的NK细胞。
2.根据权利要求1所述的方法,其中所述NK细胞获自未经选择的外周血单核细胞群。
3.根据权利要求1所述的方法,其中所述至少一种或多种刺激性细胞因子选自由IL-12、IL-15和IL-18组成的组。
4.根据权利要求3所述的方法,其中所述方法包括使所述NK细胞与3种刺激性细胞因子接触。
5.根据权利要求3所述的方法,其还包括使所述NK细胞与选自由以下组成的组中的细胞因子接触:4-1BBL、IL-2、IL-21、MICA/B、ULBP2、ICAM-1、2B4、BCM1/SLAMF2、CD155、CD112、Jagged1、Jagged2、δ-样1(Dll1)、δ-样3(Dll3)、δ-样4(Dll4)、Pref-1、DNER、Jedi、SOM-11、wingless、CCN3、MAGP2、MAGP1、TSP2、YB-1、EGFL7、CCR7、DAP12和DAP10。
6.根据权利要求1所述的方法,其中使所述NK细胞与IL-12、IL-15或IL-18在体外、体内或离体接触。
7.根据权利要求1所述的方法,其中使所述NK细胞与所述一种或多种刺激性细胞因子接触约6至约24小时。
8.根据权利要求1所述的方法,其中所述PM21颗粒、EX21外泌体或FC21饲养细胞包含与膜插入肽偶联的一种或多种刺激性细胞因子。
9.根据权利要求8所述的方法,其中所述膜插入细胞因子包含能够膜插入的融合肽,对脂质双层具有亲和力,并且其中所述融合肽包含IG4、CD4的片段或其组合。
10.根据权利要求8所述的方法,其中与膜插入肽偶联的所述一种或多种刺激细胞因子是由重组DNA编码的融合蛋白。
11.根据权利要求8所述的方法,其中所述膜插入肽包含人Fc、GPI、跨膜T细胞受体或pHLIP。
12.根据权利要求8所述的方法,其中所述一种或多种刺激性肽选自由4-1BBL、IL-2、IL-12、IL-18、IL-21、MICA/B、ULBP2、ICAM-1、2B4、BCM1/SLAMF2、CD155、CD112、Jagged1、Jagged2、δ-样1(Dll1)、δ-样3(Dll3)、δ-样4(Dll4)、Pref-1、DNER、Jedi、SOM-11、wingless、CCN3、MAGP2、MAGP1、TSP2、YB-1、EGFL7、CCR7、DAP12和DAP10组成的组。
13.根据权利要求1所述的方法,其中使所述NK细胞与PM21颗粒、EX21外泌体或FC21饲养细胞在体外、体内或离体接触。
14.根据权利要求1所述的方法,其中使步骤a的NK细胞与PM21颗粒、EX21外泌体或FC21饲养细胞接触7天、8天、9天、10天、11天、12天、13天、14天、15天、16天、17天、18天、19天、20天、21天、22天、23天、24天、25天、26天、27天、28天、29天、30天、45天或60天。
15.根据权利要求1所述的方法,其进一步包括在步骤b)之后静息扩增的记忆样NK细胞。
16.根据权利要求15所述的方法,其中所述细胞是静息了至少(在其之间或不超过)1天、2天、3天、4天或5天的记忆样NK细胞。
17.根据权利要求1所述的方法,其中NK细胞的来源是NK细胞前体来源,其中NK细胞前体来源选自由脐带血、诱导多能干细胞和胚胎干细胞组成的组。
18.用于增加记忆样NK细胞数量的试剂盒,其包含一种或多种细胞因子和包含NK细胞效应剂的一种或多种囊泡。
19.根据权利要求18所述的试剂盒,其中所述一种或多种细胞因子选自由IL-12、IL-15和IL-18组成的组。
20.根据权利要求18所述的试剂盒,其中包含NK细胞效应剂的所述一种或多种囊泡是PM21颗粒、EX21外泌体或FC21饲养细胞。
21.根据权利要求20所述的试剂盒,其中所述NK细胞效应剂选自由-1BBL、IL-2、IL-15、IL-21、MICA/B、ULBP2、ICAM-1、2B4、BCM1/SLAMF2、CD155、CD112、Jagged1、Jagged2、δ-样1(Dll1)、δ-样3(Dll3)、δ-样4(Dll4)、Pref-1、DNER、Jedi、SOM-11、wingless、CCN3、MAGP2、MAGP1、TSP2、YB-1、EGFL7、CCR7、DAP12和DAP10组成的组。
