Abstract
本发明提供一种具抗真菌功能的兰花萃取物及其组合物,属于天然提取物技术领域,包含至少一具有抗真菌活性的菲类衍生物,其制备方法为:将兰花粉加入水中,加入葡聚糖酶、甘氨酸和/或丝氨酸,微波复合酶解反应后干燥,即得混合物Ⅰ;将混合物Ⅰ加入乙醇或甲醇中,经提取获得提取液,干燥,即得提取物II;将提取物II用乙醇或甲醇溶解,经乙酸乙酯超声萃取,合并萃取液,干燥,得到兰花萃取物。本发明兰花萃取物具有数个功能特性的活性萃取物,对于枝孢属、曲霉属、青霉属等的霉菌,毕赤酵母属、酵母属等的酵母等真菌发挥优良的抗菌效果,还有具有抗氧化效果和安全性,可用于饮食品、化妆品、抗真菌感染药物的研发等领域。

技术领域
本发明属于天然提取物技术领域,具体涉及具抗真菌功能的兰花萃取物及其组合物。
背景技术
兰科植物是被子植物的最大科之一,全世界约有700属20000种,广泛分布于除两极和极端干旱沙漠地区以外的各种陆地生态系统中,热带地区分布最多。我国兰科植物为194属1388种,在全国各省市、自治区均有分布,主要集中于我国的西南地区和台湾地区,尤以喜马拉雅山脉东段、横断山脉地区、西双版纳地区,滇东-桂西山地、台湾东部山地、海南岛南部、黔桂交界山区、鄂西渝东山地、秦岭伏牛山一带最为丰富。在兰科植物中有很多珍稀的药用植物,如天麻(Gastrodia elata)、金线莲、金钗石斛、山慈菇等都是具有较高的药用价值的品种。兰科植物具有清热解毒、活血调经、润肺化痰、生津止渴、止血、敛疮的功效。兰科植物资源广泛,药用价值高,然而在2015年版药典中收录的仅有白及、天麻、金钗石斛、石斛和山慈菇5个兰科药材。在兰科植物中菲类化合物分布较广泛,此外还含有黄酮、甾醇、三萜、小分子酚酸、蒽醌、多糖类化合物。目前为止,对于兰科植物详细的化学成分研究的主要集中在白及属、杜鹃兰属、独蒜兰属、石斛属、金石斛属、毛兰属、贝母兰属、石仙桃属、石豆兰属、手参属、天麻属、开唇兰属、竹叶兰属、绶草属的一些种。菲类的生理活性,除早期发现的的抗菌活性外,近年来又发现一些菲类化合物有抗肿瘤作用、抗病毒作用、抗炎作用、抗变态反应、降血脂等多种生理活性。这与兰科植物的抗菌、抗癌、镇咳平喘、降血脂、抗氧化等作用基本吻合。可以说,菲类化合物是兰科植物的特征性成分。
申请号为201510295194.7的发明专利申请提供一种兰花萃取物及其制备方法和应用,其中该兰花为蝴蝶兰属,兰花萃取物由下列步骤所制备而得:(1)萃取:将兰花与溶剂进行混合粉碎或者破壁得到兰花浆;其中溶剂为水或醇类或其混合物中的至少一种;(2)固液分离:将步骤(1)的兰花浆进行固液分离,并将杂质去除,留下液态得兰花滤液;(3)活性划分:将步骤(2)的兰花滤液以特定分子筛薄膜处理纯化,得到兰花萃取筛分液;(4)浓缩:将步骤(3)的兰花萃取筛分液以分子筛薄膜处理浓缩,或者真空或蒸煮方式进行部分浓缩,得到活性沉淀物与具活性的液态活性萃取液。申请号为201510295194.7的活性沉淀物与具活性的液态活性萃取液具有美白功能、抗氧化力、以及抗皱(抗老化)能力。申请号为201710586576.4的发明专利申请提供一种兰花萃取物包含至少一抗真菌成分,而该萃取物萃取自一兰花部位,且该抗真菌成分具有抗真菌的活性。一种兰花萃取物添加产品为将该兰花萃取物添加于一原材料内,以制成一抗真菌组成物。
发明内容
本发明的目的在于提供一种含有其上亲水氨基酸甘氨酸和/或丝氨酸存在的菲类衍生物、多糖基物质的具抗真菌功能的兰花萃取物,对于枝孢属、曲霉属、青霉属等的霉菌,毕赤酵母属、酵母属等的酵母等真菌发挥优良的抗菌效果,可用于饮食品、化妆品、抗真菌感染药物的研发等领域。
本发明为实现上述目的所采取的技术方案为:
具抗真菌功能的兰花萃取物,包含至少一具有抗真菌活性的菲类衍生物。
本发明兰花萃取物中含有的菲类衍生物对于枝孢属、曲霉属、青霉属等的霉菌,毕赤酵母属、酵母属等的酵母等真菌发挥优良的抗菌效果,可用于饮食品、化妆品、抗真菌感染药物的研发等领域。
上述菲类衍生物为菲呋喃类衍生物。
上述菲类衍生物为结构式(I)或结构式(II)所示的化合物;
其中R是H或CH2OH。该菲类衍生物上亲水氨基酸甘氨酸和/或丝氨酸的存在,使菲类衍生物具有较好的具有良好的生物、生理活性,可在真菌表面形成穿孔,造成原生质体泄漏,使真菌裂解死亡,具有优良的抗菌效果。

本发明还公开具抗真菌功能的兰花萃取物的制备方法,包括如下步骤制备:
S1:将兰花粉加入水中,加入葡聚糖酶、甘氨酸和/或丝氨酸,微波复合酶解反应后干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ加入乙醇或甲醇中,经提取获得提取液,干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经乙酸乙酯超声萃取,合并萃取液,干燥,得到兰花萃取物。
本发明制备方法中葡聚糖酶、亲水氨基酸甘氨酸和/或丝氨酸的存在一方面在微波的作用下提高了兰花水萃物中结构式(I)或结构式(II)所示的菲类衍生物的含量,另一方面能够通过酶解反应能够将兰花多糖分解为低聚糖或葡萄糖,且在亚临界水萃取过程中,产生的低聚糖或葡萄糖可与兰花中的游离氨基酸以及酚酸类进一步反应生成多糖基物质,在多糖物质上引入大量的-NH2和-COOH,不但能通过渗透到微生物的细胞核中与DNA相结合,干扰和抑制mRNA以及蛋白质的合成,从而赋予木兰萃取杀菌作用,而且还可以作为螯合剂,选择性结合痕量金属,从而抑制真菌的生长,能够与菲类衍生物发挥增益作用,进一步提高兰花萃取物的抗真菌功能。
其中,兰花粉包括花和/或叶和/或茎和/或根;
葡聚糖酶的添加量为兰花粉重量0.20-0.30‰,甘氨酸的添加量为兰花粉重量0-1.2‰,丝氨酸的添加量为兰花粉重量的0-1.2‰;
微波复合酶解温度为50-60℃、功率为600-1000W、时间为5-15min。
本发明还公开具抗真菌功能的兰花萃取物的组合物,包含上述兰花萃取物。
上述组合物还包含脂肪酸酯类。本发明的脂肪酸酯类没有特别限定,优选为选自由蔗糖脂肪酸酯、单甘油脂肪酸酯、失水山梨醇脂肪酸酯和聚甘油脂肪酸酯组成的组中的至少一种,更优选为蔗糖脂肪酸酯或单甘油脂肪酸酯。另外,若考虑到提高对真菌的抗菌效果,则蔗糖脂肪酸酯中的脂肪酸优选为碳原子数12-16的脂肪酸,单甘油脂肪酸酯中的脂肪酸优选为碳原子数8-12的脂肪酸。另外,若考虑到将添加到饮食品中时对风味的影响抑制为低水平,则本发明的脂肪酸酯类特别优选为蔗糖肉豆蔻酸酯。
与现有技术相比,本发明的有益效果为:本发明兰花萃取物具有数个功能特性的活性萃取物,如其上亲水氨基酸甘氨酸和/或丝氨酸存在的菲类衍生物、多糖基物质等,对于枝孢属、曲霉属、青霉属等的霉菌,毕赤酵母属、酵母属等的酵母等真菌发挥优良的抗菌效果,还有具有抗氧化效果和安全性,具有实际的商品应用性,可用于饮食品、化妆品、抗真菌感染药物的研发等领域;本发明兰花萃取物的制备方法制得的兰花萃取物具有较好的具有良好的生物、生理活性,可在真菌表面形成穿孔,造成原生质体泄漏,使真菌裂解死亡,具有优良的抗菌效果。
本发明采用了上述技术方案提供具抗真菌功能的兰花萃取物及其组合物,弥补了现有技术的不足,设计合理,操作方便。

本发明的具抗真菌功能的兰花萃取物的制备方法,包括如下步骤制备:
S1:将兰花粉加入水中,加入葡聚糖酶、甘氨酸和/或丝氨酸,微波复合酶解反应后干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ加入乙醇或甲醇中,经提取获得提取液,干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经乙酸乙酯超声萃取,合并萃取液,干燥,得到兰花萃取物。
本发明制备方法中葡聚糖酶、亲水氨基酸甘氨酸和/或丝氨酸的存在一方面在微波的作用下提高了兰花水萃物中结构式(I)或结构式(II)所示的菲类衍生物的含量,另一方面能够通过酶解反应能够将兰花多糖分解为低聚糖或葡萄糖,且在亚临界水萃取过程中,产生的低聚糖或葡萄糖可与兰花中的游离氨基酸以及酚酸类进一步反应生成多糖基物质,在多糖物质上引入大量的-NH2和-COOH,不但能通过渗透到微生物的细胞核中与DNA相结合,干扰和抑制mRNA以及蛋白质的合成,从而赋予木兰萃取杀菌作用,而且还可以作为螯合剂,选择性结合痕量金属,从而抑制真菌的生长,能够与菲类衍生物发挥增益作用,进一步提高兰花萃取物的抗真菌功能。
其中,兰花粉包括花和/或叶和/或茎和/或根。
其中,兰花萃取物具体通过如下步骤制备:
S1:将兰花原料于110-120℃的温度下烘干,然后粉碎至20-100目,得兰花粉,以增加兰花粉与萃取溶剂的接触表面积,借此提升后续萃取的萃取效率,当原料粒径为40-60目,活性物质的萃取率达到最大,而后,随着粒径的减小,萃取率反而逐渐减小。这是由于原料兰花粉的细胞外层是由纤维素和半纤维素等物质构成的细胞壁,质壁坚硬,犹如细胞的外壳有保护内含物的作用。若原料颗粒的粒径过大,当萃取温度一定时,则细胞壁的阻力会使萃取剂在短时间内难以快速扩散至颗粒内部,因而影响了有效成分的溶剂化和向外扩散;萃取率就减小了;当原料通过粉碎处理,原料的粒径变小了,细胞壁被破坏的程度就增大了,整个原料的颗粒被溶剂浸润的时间就缩短了,有效成分溶剂化的速度和扩散速度也就自然加快了,同时粒径减小也增加了萃取剂与原料的接触面积和萃取通道,从而有利于提高萃取率,但并不是原料的粒径越小越好,原料过细,虽然此时对原料细胞壁的破坏更加彻底,但却增加了原料的堆积密度,物料空隙率下降,通透性变差,致使萃取剂水只能沿着阻力小的线路穿过料层,形成许多针孔,使萃取显著不均匀。同时,颗粒太细,不仅会导致流路堵塞,使萃取柱柱压过高,而且还会造成原料结块,现沟流,对萃取不利,因而就出现了原料粒径过小,萃取率不再提高,反而减小的现象,因此,提取过程中原料的粒径应该适当,优选颗粒粒径为40-60目;
S2:将兰花粉加入水中,加入兰花粉重量0.20-0.30‰的葡聚糖酶(如0.21‰、0.22‰、0.23‰、0.24‰、0.26‰、0.28‰等)、兰花粉重量0-1.2‰的甘氨酸(如0.1‰、0.2‰、0.5‰、0.7‰、0.8‰、1.0‰等)和/或兰花粉重量0-1.2‰的丝氨酸(如0.1‰、0.2‰、0.5‰、0.7‰、0.8‰、1.0‰等),在50-60℃(如51℃、52.5℃、53℃、56℃、57℃、59℃等)、600-1000W(如620W、750W、800W、830W、880W、950W等)下微波复合酶解反应5-15min(如6min、7min、8min、11min、12.5min、14min等),干燥,即得混合物Ⅰ;
S3:将混合物Ⅰ用75-85%(如76%、77.2%、78%、79%、82%、83.4%等)乙醇回流提取两次,第一次以20-30倍量(如21倍量、22.4倍量、23倍量、26倍量、27倍量、28.5倍量等)的乙醇回流提取1-2h,第二次以10-20倍量(如12倍量、13倍量、14.5倍量、17倍量、18倍量、19倍量等)的乙醇回流提取0.5-1.5h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S4:将提取物ⅠI用乙醇或甲醇溶解,经2.5-3.5倍量硅胶拌样,用35-45倍乙酸乙酯超声萃取2-3次,每次各30-40min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
本发明的具抗真菌功能的兰花萃取物的组合物,包含上述兰花萃取物。
其中,组合物还包含脂肪酸酯类。本发明的脂肪酸酯类没有特别限定,优选为选自由蔗糖脂肪酸酯、单甘油脂肪酸酯、失水山梨醇脂肪酸酯和聚甘油脂肪酸酯组成的组中的至少一种,更优选为蔗糖脂肪酸酯或单甘油脂肪酸酯。另外,若考虑到提高对真菌的抗菌效果,则蔗糖脂肪酸酯中的脂肪酸优选为碳原子数12-16的脂肪酸,单甘油脂肪酸酯中的脂肪酸优选为碳原子数8-12的脂肪酸。另外,若考虑到将添加到饮食品中时对风味的影响抑制为低水平,则本发明的脂肪酸酯类特别优选为蔗糖肉豆蔻酸酯。
关于本发明的组合物中的脂肪酸酯类的含量,以兰花萃取物总重量为基准,优选以干燥重量计为0.1-2.0wt%、更优选为0.5-1.2wt%。
若考虑到抗菌性的提高,则本发明的抗真菌剂优选还含有乙醇等水性有机溶剂,本发明的组合物中的乙醇的含量没有特别限定,优选为50-90wt%、更优选为60-70wt%。
本发明的抗真菌剂除上述外还可以含有饮食品、化妆品或医药上能够允许的添加剂等。作为该添加剂,没有特别限定,可举出例如:保存材料、抗氧化剂、增稠稳定剂、乳化剂、食物纤维、pH调节剂等。
下面,结合具体实施例对本发明实施方式作进一步说明。
实施例1:
兰花萃取物通过如下步骤制备:
S1:将20目兰花粉加入水中,加入兰花粉重量0.20‰的葡聚糖酶、兰花粉重量1.2‰的甘氨酸,在50℃、600W下微波复合酶解反应5min,干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ用75%乙醇回流提取两次,第一次以20倍量的乙醇回流提取1h,第二次以10倍量的乙醇回流提取0.5h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经2.5倍量硅胶拌样,用35倍乙酸乙酯超声萃取2次,每次各30min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
实施例2:
兰花萃取物通过如下步骤制备:
S1:将60目100g兰花粉加入水中,加入兰花粉重量25mg的葡聚糖酶、兰花粉重量60mg的甘氨酸和兰花粉重量60mg的丝氨酸,在55℃、700W下微波复合酶解反应10min,干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ用80%乙醇回流提取两次,第一次以25倍量的乙醇回流提取1.5h,第二次以15倍量的乙醇回流提取1h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经3.0倍量硅胶拌样,用40倍乙酸乙酯超声萃取2次,每次各35min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
实施例3:
兰花萃取物通过如下步骤制备:
S1:将100目兰花粉加入水中,加入兰花粉重量0.30‰的葡聚糖酶、兰花粉重量1.2‰的丝氨酸,在60℃、1000W下微波复合酶解反应15min,干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ用85%乙醇回流提取两次,第一次以30倍量的乙醇回流提取2h,第二次以20倍量的乙醇回流提取1.5h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经3.5倍量硅胶拌样,用45倍乙酸乙酯超声萃取3次,每次各40min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
对比例1:
兰花萃取物通过如下步骤制备:
S1:将60目100g兰花粉加入水中,加入兰花粉重量25mg的葡聚糖酶,在55℃、700W下微波复合酶解反应10min,干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ用80%乙醇回流提取两次,第一次以25倍量的乙醇回流提取1.5h,第二次以15倍量的乙醇回流提取1h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经3.0倍量硅胶拌样,用40倍乙酸乙酯超声萃取2次,每次各35min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
对比例2:
兰花萃取物通过如下步骤制备:
S1:将60目100g兰花粉加入水中,加入兰花粉重量60mg的甘氨酸和兰花粉重量60mg的丝氨酸,在55℃、700W下微波复合酶解反应10min,干燥,即得混合物Ⅰ;
S2:将混合物Ⅰ用80%乙醇回流提取两次,第一次以25倍量的乙醇回流提取1.5h,第二次以15倍量的乙醇回流提取1h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经3.0倍量硅胶拌样,用40倍乙酸乙酯超声萃取2次,每次各35min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
对比例3
兰花萃取物通过如下步骤制备:
S1:将60目100g兰花粉用80%乙醇回流提取两次,第一次以25倍量的乙醇回流提取1.5h,第二次以15倍量的乙醇回流提取1h,合并提取液,浓缩,真空干燥,即得提取物ⅠI;
S2:将提取物ⅠI用乙醇或甲醇溶解,经3.0倍量硅胶拌样,用40倍乙酸乙酯超声萃取2次,每次各35min,合并萃取液,浓缩,真空干燥,得到兰花萃取物。
试验例1:
兰花萃取物的抗真菌效果
滤纸片法测定抑菌圈:将配制好灭过菌的固体琼胶培养基熔化后倒入培养皿中,每皿20mL,待冷却凝固后,用无菌吸管分别加入0.1mL霉菌或酵母菌球菌的悬液(菌体含量为1×105~1×106 CFU/mL),用无菌涂布器将菌液涂布均匀,制成含菌平板。选择吸水性强的定性滤纸,用打孔器打成若干直径为6mm的圆形滤纸片,分装于洁净干燥的小培养皿内,干热灭菌后,浸在浓度为20ug/mL、50ug/mL、100ug/mL、200ug/mL兰花萃取物溶液中,用无菌镊子镊取浸有兰花萃取物溶液的滤纸片贴在含菌平板上,每个培养皿放置3个纸片。放入培养箱于37℃恒温倒置培养24h,取出后测定滤纸片抑菌圈直径大小,抑菌效果如表1所示。由表1可知,相同浓度的兰花萃取物溶液的条件下,抑菌圈的大小排序为:实施例2>对比例2>对比例1>对比例3,获得的兰花萃取物的抑菌效果好于对比例1获得的兰花萃取物。这说明本发明的兰花萃取物具有优良的抗真菌功能,且制备方法中葡聚糖酶、亲水氨基酸甘氨酸和/或丝氨酸的存在能提高兰花萃取物的抗真菌功能。

以上实施方式仅用于说明本发明,而并非对本发明的限制,本领域的普通技术人员,在不脱离本发明的精神和范围的情况下,还可以做出各种变化和变型。因此,所有等同的技术方案也属于本发明的范畴,本发明的专利保护范围应由权利要求限定。
Claims (10)
1.具抗真菌功能的兰花萃取物,其特征在于:包含至少一具有抗真菌活性的菲类衍生物。
2.根据权利要求1所述的具抗真菌功能的兰花萃取物,其特征在于:所述菲类衍生物为菲呋喃类衍生物。
3.根据权利要求1或2所述的具抗真菌功能的兰花萃取物,其特征在于:所述菲类衍生物为结构式(I)或结构式(II)所示的化合物;
其中R是H或CH2OH。
4.权利要求1或2或3所述的兰花萃取物的用途,其特征在于:在制备饮食品、化妆品或抗真菌感染制剂中的用途。
5.权利要求1或2或3所述的具抗真菌功能的兰花萃取物的制备方法,其特征在于:包括如下步骤:
S1:将兰花粉加入水中,加入葡聚糖酶、甘氨酸和/或丝氨酸,微波复合酶解反应后干燥,即得混合物Ⅰ;
S2:将所述混合物Ⅰ加入乙醇或甲醇中,经提取获得提取液,干燥,即得提取物ⅠI;
S2:将所述提取物ⅠI用乙醇或甲醇溶解,经乙酸乙酯超声萃取,合并萃取液,干燥,得到兰花萃取物。
6.根据权利要求5所述的具抗真菌功能的兰花萃取物的制备方法,其特征在于:所述兰花粉包括花和/或叶和/或茎和/或根。
7.根据权利要求5所述的具抗真菌功能的兰花萃取物的制备方法,其特征在于:所述葡聚糖酶的添加量为兰花粉重量0.20-0.30‰,甘氨酸的添加量为兰花粉重量0-1.2‰,丝氨酸的添加量为兰花粉重量的0-1.2‰。
8.根据权利要求5或7所述的具抗真菌功能的兰花萃取物的制备方法,其特征在于:所述微波复合酶解温度为50-60℃、功率为600-1000W、时间为5-15min。
9.具抗真菌功能的兰花萃取物的组合物,其特征在于:包含权利要求1-8任一项所述的兰花萃取物。
10.根据权利要求9所述的具抗真菌功能的兰花萃取物的组合物,其特征在于:所述组合物还包含脂肪酸酯类。
